2 ABSTRACT In the Alternative DSM-5 Model for Personality Disorders (AMPD), the diagnosis of specific personality disorders is obtained through an evaluation of the level of impairment in personality functioning and an assessment of dimensional personality traits associated in 25 "lower order" facets and 5 "higher order" domains. The Personality Inventory for DSM-5 (PID-5) was developed for examination of personality traits within this system. This Ph.D. thesis covers five relevant publications regarding AMPD, particularly PID-5. First, the authors introduce the theoretical background of the PID-5. Attention is paid on its ease of use, data interpretation and use of these data for treatment planning in different clinical settings. Two empirical studies test PID-5 psychometric properties (internal consistency, validity, discrimination capacity and unidimensional structure) in the sample of community volunteers (n=351) and a clinical sample of psychiatric patients (n=143). PID-5 was administered individually and in a group setting using pen-paper method and online data collection. 33 respondents completed the inventory twice to check test-retest reliability. Influenza viruses cause respiratory illnesses which can vary in severity depending on the strain of the virus, as well as the age and health condition of the host. Influenza remains a major threat to public health due to its nature prone to suffer mutations. As a result, vaccines have to be reformulated annually and new strains may cause sporadic global pandemics. Furthermore, the recent emergence of resistant strains of the virus against the current standard of care (oseltamivir and zanamivir) underlines the need of novel anti-influenza therapeutics. The aim of this dissertation work is to contribute to the discovery of new anti-influenza inhibitors either by rational drug-design and optimization of oseltamivir structure, or by developing screening assays suitable for the discovery of novel inhibitors of the enzymes neuraminidase or RNA-polymerase. Nela Dvořáková 19.7.2019 1 Abstract We study a representative of a diverse group of stars exhibiting the B[e] phe- nomenon, designated MWC 939. This object might be an important link be- tween the stage of the asymptotic giant branch and planetary nebula stage as it is one of a few such objects observed. Our focus is aimed at spectral analysis of the circumstellar matter around the central star. We assemble a line list for MWC 939 and observe variations of its spectrum. Electron density and temper- ature are estimated using nebular diagnostics of ratios of [S II] λλ 6716, 6731 ˚A and [O I] λλ 6300, 6364 ˚A and 5577 ˚A. These estimates are then used for detailed calculations of the structure of circumstellar envelope. 1

Title: Level and degree of association of selected motor abilities, motor skills in football players category U6. Objectives: The aim of the research is to identify the level of the selected motor abilities, motor skills and motor docility and to determine the degree of association between these performance components in football players category U6. Methods: The research group (n = 52; age = 6.1 ± 0.3 years) consisted of football players of SK Slavia Praha Youth Academy. The participants completed one-time test battery which consisted of motor docility tests (IOWA- BRACE test), five tests of the motor abilities (5m and 10m linear running acceleration, 5m flying maximum speed, standing long jump, 4x10m shuttle run) and two tests of the specific football skills (4x10m shuttle run with a ball, slalom skills test). The degree of association was determined via Pearson's correlation coefficient. Results: The part with the results provides information on the level of the players' motor abilities, motor skills based on the values obtained through performed tests. Haemoproteus u pěvců: prevalence a dynamika infekce / Haemoproteus in passerines: Prevalence and infekction dynamics

The genera Haemoproteus and Plasmodium are widespread genera of blood parasites from phylum Apicomplexa. Both genera have dixenous life cycle and their definitive hosts belong to the order Diptera. Both parasites can be found in birds where asexual division takes place. The infection with genera Haemoproteus and Plasmodium is usually asymptomatic; during long lasting chronic phase relapses of the infection can occur. We gained 1 092 blood samples from 29 trapped species of passerines in Milovice forest during seasons 2017-2019. Haemosporidians were detected in 48 % of samples. The prevalence of genus Plasmodium was 16 %, the prevalence of genus Haemoproteus was 22 % and 10 % we could not specify the parasite. We have found six new lineages of genus Haemoproteus, provisionally named Haemoproteus lineages coccoc_1, coccoc_2, coccoc_3, coccoc_4, embcit, fricoe which total prevalence was 14 %. Lineage Haemoproteus sp. coccoc_1 was the most noticed one and its prevalence was 12 %. Plasmodium relictum clone Peng14-121Br2AF and isolate Cc_P1 was the second most prevalent (13 %). Thesis Assessment and Development of Visual Perception in Preschool Children responds to the need of broader selection of standardized tools designed for the assessment of actual development of visual perception in preschool children, especially in relation to the assessment of school readiness. The main aim of the thesis is standardization of a new assessment tool. Complementary aim is also to design and evaluate a new intervention program supporting development of selected areas of visual perception for children before and at the beginning of their school attendance. In its theoretical part thesis scrutinizes current findings in the area of visual perception and its relation to basic academic skills, especially reading. The explorative part of thesis presents standardization study based on classical model of Test theory consisting of convergent validity and reliability evaluation of proposed test and setting national norms for four age groups (from 5 to 7 years). Normative group covered 1234 participants.

About 4 % women in general population and up to 19 % women in at-risk population suffer from PTSD related to childbirth. The main risk factors for postpartum PTSD are negative birth experience and obstetric emergencies. However, the findings about risk factors and prevalence rates vary across studies, which is mainly due to the inconsistency in the diagnostics of postpartum PTSD. Researchers use various PTSD measures that differ in the number and type of symptoms monitored and many of these measures do not cover all DSM criteria for PTSD. Furthermore, the PTSD diagnostic criteria have been significantly modified in the new DSM-5 and the existing instruments need to be revised. The first part of the thesis presents the concept of PTSD related to childbirth. Special attention is paid to the risk factors identified in previous research and to the analysis of PTSD diagnostic and screening measures used in the postpartum context. The aim of the second part of the thesis is to evaluate the psychometric properties of the unpublished Childbirth-Related PTSD Questionnaire on a sample of 620 Czech women. Master’s thesis DNA markers for bacterial infection diagnosis engages in matters of pathogenic bacteria that cause urinary tract infections and in matters of diagnostic methods for this bacteria with focus on the usage of DNA probes. The aim of this thesis was to find, design and test suitable DNA probes by using quantitative Real-Time PCR. Probes were designed based on specific bacterial sequences of 16S rRNA gene fragment. Afterwards, testing and optimisation of Real-Time PCR was conducted. Successful optimisation was managed for two probes out of twelve – one for S. aureus, second for P. aeruginosa. These results represent the basis for the follow-up experimental work to ensure improvement and reproducibility of the experiments.

Studium transferinu jako markeru dědičných poruch glykosylace / Study of transferrin as a marker of congenital disorders of glycosylation

Congenital disorders of glycosylation (CDG) represent a heterogeneous group of mul- tisystemic metabolic disorders which are caused by defects in biosynthetic pathways of glycoproteins. The screening test for N-glycosylation disorders is the analyses of sialylated isoforms of serum transferrin (Tf) by means of isoelectric focusing (IEF). Two distinct pathological IEF patterns of Tf are observed. A type I pattern is cha- racterized by a decrease of tetra- and an increase of di- and asialotransferrin, whereas a type II pattern shows in addition an increase of tri- and monosialotransferrin. The aims of diploma thesis were: 1) to evaluate reference range for spectrum of sialylated forms of Tf separated by IEF and 2) to perform biochemical and molecular analyses in three patients (P1-P3) with clinical suspicion for CDG. Serum and genomic DNA from three patients with clinical suspicion for CDG and family members of P1 were analysed. Sera from 99 healthy volunteers within the age range of 2-42 years served as a control group. Tf was analysed by IEF with direct immunofixation, SDS-PAGE and Western blot using specific antibody against human Tf (Dako). Profiles of Tf were quantified by AlphaEaseFC software (Alpha Innotech). The diploma thesis focuses on the elaboration of proposals for materials for clients of the Aphasia Club, which were used in group therapy. The diploma thesis is divided into theoretical and practical part. The theoretical part contains three main chapters, in which the concept of aphasia is defined, the symptomatology and etiology of this imaired communication ability are described. Then are listed possible classifications of aphasia, available tests for diagnosis and directions in the therapy of aphasia. The practical parts sets the main goal of the thesis, together with three research questions. As research methods were chosen case studies, which were prepared on the basis of medical reports of individual clients of the Aphasia Club. With information of medical reports were created materials for group therapies of Aphasia Club. The materials were used in group therapies. Using method of observation, research questions focused on the use of materials, their modification and practical signifikance in the development of impaired communication functions were subsequently answered.
