Neste trabalho foram avaliados o comportamento do ácido gálico e de adjuvantes tecnológicos frequentemente empregados em formas farmacêuticas sólida, e das suas misturas físicas, através de métodos termoanalíticos e por espectroscopia de infravermelho. Foi investigada também a influência da compactação sobre as misturas físicas equiponderais. Os adjuvantes avaliados foram amidoglicolato de sódio, celulose microcristalina, croscarmelose sódica, crospovidona, dióxido de silício coloidal, estearato de magnésio e polimetacrilato. O ácido gálico apresentou um comportamento térmico diferenciado nas misturas, assumindo, provavelmente, uma forma instável com menor ponto de fusão. Os resultados obtidos por DSC demonstraram interação de natureza física com mudança de entalpia para misturas do ácido gálico com celulose microcristalina, crospovidona, estearato de magnésio e polimetacrilato. A interação não pode ser confirmada por espectroscopia de infravermelho para a crospovidona e polimetacrilato, devido à sobreposição das bandas com o ácido gálico. Os demais adjuvantes também apresentaram interação física, porém, sem alteração da entalpia, confirmada por espectroscopia de infravermelho, relacionada ao estabelecimento de ligações de hidrogênio entre os componentes da mistura. A compactação demonstrou particular influência sobre a interação com celulose microcristalina, croscarmelose sódica e crospovidona. / In this work were evaluated the behavior of the gallic acid and technological excipients used in sold dosage forms and their physical powder mixtures, by Differential Scanning Calorimetry (DSC) and Thermogravimety (TGA) and infrared spectroscopy (IR). The influence of the compression force on the 1:1 (w/w) physical mixtures was also investigated. The excipients evaluated were sodium starch glycolate, microcrystalline cellulose, croscarmellose sodium, crospovidone, colloidal silicon dioxide, magnesium stearate and polymethacrylate. Gallic acid presented a different thermal behavior in the mixtures, assuming, probably, an unstable form with a lower melting point. The results obtained by (DSC) demonstrated the occurrence of physical interactions with enthalpy changes for the mixtures of gallic acid with microcrystalline cellulose, crospovidone, magnesium stearate and polymethacrylate. Os métodos termo-analíticos são ferramentas úteis na avaliação da compatibilidade entre fármacos e adjuvantes, com destaque à calorimetria exploratória diferencial. Neste trabalho foram avaliados a compatibilidade e o comportamento térmico entre a isoniazida e adjuvantes tecnológicos primários usualmente empregados em formas farmacêuticas sólidas. A compatibilidade foi examinada por meio da preparação de misturas físicas binárias do tipo fármaco/adjuvante. Foi investigada também a influência da granulação por via úmida e do processo de compactação para as misturas de isoniazida e adjuvantes com função de material de enchimento e carga e deslizante. A isoniazida apresentou um comportamento térmico não encontrado na literatura. Os adjuvantes avaliados foram: ácido esteárico, amido, celulose microcristalina, crospovidona, croscarmelose sódica, dióxido de silício coloidal estearato de magnésio, glicolato de amido sódico, hipromelose, lactose, manitol, polidona e talco. Para as misturas físicas, a maioria dos adjuvantes mostrou-se compatível com o fármaco em questão. Foram verificadas interações com o ácido esteárico, o glicolato de amido sódico, a lactose, o manitol e a povidona. A isoniazida mostrou a formação de uma mistura eutética com o manitol e de interação química com a lactose. A agregação por via úmida e o processo de compactação não mostraram influências adicionais na compatibilidade das misturas avaliadas. Os resultados observados foram confirmados por métodos não-térmicos como difratometria de raios X, espectroscopia de infravermelho e ressonância nuclear magnética. / Thermo-analytical methods, and specially Differential Scanning Calorimetry, are useful support for the evaluation of compatibility between drug substances and pharmaceutical excipients. In this work were studied the compatibility and the thermal behavior of isoniazid and pharmaceutical excipients, commonly used for the formulation of solid dosage forms. Colloidal silicon dioxide, corn starch, crospovidone, hypromellose, lactose, magnesium stearate, mannitol, microcrystalline cellulose, povidone, sodium croscarmellose, sodium starch glycolate, stearic acid and talc were the excipients employed in these experiments. The compatibility was analyzed testing binary physical drug/excipient admixtures. The effect of wet granulation and compression was also investigated, in this case especially between isoniazid, fillers and lubricant. For almost all excipients no incompatibilities with isoniazid were observed. Interactions were detected when the drug substance was added to stearic acid, sodium starch glycolate, lactose, mannitol and povidone. Isoniazid formed a euthetic mixture with mannitol, whereas a possible chemical reaction occurred between isoniazid and lactose. Wet granulation and compaction of the tested admixtures did not affect the results observed above. These observations were confirmed by non-thermal techniques, such as X-Ray diffractometry, infrared spectroscopy and nuclear magnetic resonance.

Method Development for Thermal Stability Analysis by Circular Dichroism : Application to the Abp1p SH3 domain from yeast

Thermal stability is an important and interesting physical property of proteins. A common method to study it by is circular dichroism (CD) spectroscopy. The aim of this study was to test methods to improve thermal stability analysis by CD spectroscopy. Experiments were performed using the Abp1p SH3 domain from yeast as a model protein. Thermal denaturation was monitored at multiple wavelengths. It was concluded that for data sets of reasonable quality the choice of wavelength does not affect the results. An approach to estimate stability of thermophilic proteins was tested where thermal stability was measured at different concentrations of the denaturant GuHCl. The thermochemical data was used to estimate the stability in absence of GuHCl by extrapolation. The results were compared to those obtained from CD spectroscopy and differential scanning calorimetry. It was found that a stabilizing effect from low concentrations of GuHCl complicated the extrapolation. It is likely that this method is more successful if there is no stabilizing effect. The effect of ΔCp in stability parameter calculations was investigated with an experimentally and theoretically determined ΔCp. This was further investigated with synthetic data sets. The ΔCp used in calculations had no notable effect, as long as there was no cold denaturation. Although ΔCp is not necessary in calculations, it is an interesting parameter itself. ΔCp can be calculated from the thermochemical data used for extrapolation. The results in this study demonstrate robustness in thermal stability analysis by CD spectroscopy and a potential for development.

Isto tem sido feito através da mudança das rotas produtivas, onde o bagaço será utilizado para a produção de etanol de segunda geração e a palha como substituta do bagaço nas caldeiras de geração de vapor. Atualmente a palha não está consolidada como combustível e, portanto, seu comportamento nos geradores de vapor permanece desconhecido. Sabendo que as incrustações devido à fusão das cinzas durante a combustão de combustíveis sólidos representa um dos maiores problemas na operação dos equipamentos de geração de vapor, neste trabalho, foi caracterizada a palha de cana como combustível a fim de prever seu possível comportamento em caldeiras industriais. Amostras de palha de cana-de-açúcar foram recolhidas em duas condições: diretamente da planta e recolhida no solo após a colheita mecanizada. Foram estudados e analisados os comportamentos das cinzas da palha integral e seus componentes (pontas, folhas verdes e folhas secas), por meio das técnicas de análise térmica simultânea (STA) e da espectrometria de energia dispersiva (EDS). Desta forma, foram identificadas temperaturas de evaporação e a composição dos principais constituintes voláteis das cinzas, representados por uma reação endotérmica com perda de massa. Igualmente, a temperatura de fusão foi detectada como reação endotérmica sem envolver perdas de massa. As análises apresentaram composição e comportamentos diferentes para cada um dos componentes da palha baseados principalmente no teor de metais álcali, cloro e das temperaturas de preparação das cinzas prévias às análises. Neste sentido, as porções correspondentes às pontas e folha verdes da cana-de-açúcar, apresentaram impactos relacionados com incrustações (fouling/slagging) superiores às de folhas secas e palha integral. Baseados nos resultados obtidos a palha de cana-de-açúcar perfila-se como uma possível biomassa substituta ao bagaço nas caldeiras de geração industriais / Abstract: The sugarcane supply chain has lately placed great interest on the comprehensive use of its main by-products: straw and bagasse (the straw constituted by leaves and tops). By changing the production routes, sugarcane bagasse will be used as an energy source for the production of second generation ethanol and straw will substitute bagasse in steam generators. However, sugarcane straw is not currently used as fuel; therefore, its behavior in industrial boilers remains unknown. Slagging and fouling depositions due to fly ash melting within the boiler during combustion of solid fuel represents one of the most significant issues in the operation and maintenance of steam generation equipment. The main objective of this work was to characterize sugarcane straw as a fuel in order to predict its performance in industrial boilers. Samples of straw were collected in two different conditions: directly from the plant and off the field after mechanized harvest. The behavior of the ashes produced at different temperatures (575°C, 785°C and 985/950/850°C) from the integral sugarcane straw and its individual components (tops, dry leaves and green leaves) were analyzed by simultaneous thermal analysis (STA) and energy dispersive spectroscopy (EDS). Evaporation temperature and composition of the main volatile constituents of ashes formed, represented by an endothermic reaction involving changes in mass were identified. Also, fusion temperature was determined to be an endothermic reaction without mass loss. Results showed different elemental compositions and behaviors of ashes for each constituent of sugarcane straw, mainly based on alkali and chlorine percentage, and ashing temperature. In this regard, the fractions corresponding to sugarcane tops and green leaves showed major impacts on slagging and fouling depositions compared to dry leaves and raw straw fraction possibly due to its high levels of potassium and chlorine. [pt] Neste trabalho, foram estudadas filmes poliméricos densos formadas por acetato de celulose (AcC) e polihidroxialkanoato de cadeia média PHAmcl, com diferentes concentrações relativas de cada polímero. Os filmes de PHAmcl/DAC foram preparadas a partir de soluções poliméricas, diluídas em dimetil cetona, na qual se realiza uma mistura com agitação contínua. Foram preparadas as amostras: AcC puro, PHAmcl puro e seis misturas com concentrações variando de 2 porcento a 12 porcento (m/v) de PHAmcl em AcC. Medidas de calorimetria diferencial exploratória e análise dinâmica mecânica foram realizadas a fim de se conhecer a miscibilidade das blendas formadas A composição química das superfícies desses filmes foi caracterizada utilizando a técnica de espectroscopia de fotoelétrons induzida por raios-X (XPS). A morfologia e as propriedades mecânicas das amostras foram estudadas com o auxílio da microscopia de força atômica (AFM). Observou-se nas misturas um enriquecimento na superfície de PHAmcl e uma alteração da morfologia para toda a superfície com a incorporação de PHAmcl. Foi observada uma diminuição na dureza com o aumento a concentração de PHAmcl no filmes. Os filmes apresentaram miscibilidade até 10 porcento PHAmcl/90porcentoAcC em volume. / [en] In this work, the study of thick films formed by cellulose acetate and medium chain length polyhydroxyalkanotes with different molar concentration of PHB is presented. The films were prepared by polymeric solutions diluted in dimethil ketone with continued agitation. The solvent evaporation occurs on a glass plate in controlled atmosphere of 300 C. Eight samples were prepared: pure AcC and PHAmcl and mixtures with PHAmcl bulk concentrations in the range of 2 to 12 percent (w/v). Differential scanning calorimetry and dynamic mechanical analysis measurements were performed in order to know the samples miscibility. The surface chemical composition of the samples was characterized by X- ray photoelectron spectroscopy (XPS). The morphology and mechanical properties were studied by atomic force microscopy (AFM). PHAmcl surface enrichment and morphology modification of the films were observed on the whole surface with the increasing of PHAmcl. A decrease in the films hardness was observed for increasing PHAmcl bulk concentration. The films were found to be miscible until 10percentPHAmcl/90percentAcC bulk concentration.

Vodní-kationické můstky v půdní organické hmotě / Water-cation bridges in soil organic matter

Degradace půdy zasolováním, zvyšující se teplota a následný úbytek mnonžství srážek v posledních letech vedou k rozšiřování aridních a semi-aridních oblastí (v současnosti pokrývají třetinu pevniny Země). Navíc představují globální environmentální problémy, které, pokud se nebudou správně řešit, můžou vést k vážné celosvětové krizi. V této diplomové práci jsme propojili tato témata a zaměřili se na lepší porozumění role sodných kationtů na vazbu vody v půdní organické hmotě v aridních a semi-aridních podmínkách. Náš cíl byl prozkoumat proces vzniku a stabilitu vodních molekulových můstků a vodních-kationických můstků v půdní organické hmotě, sílu vazby a množství vázáné vody v půdě za využití diferenční kompenzační kalorimetrie. Nasycení sodnými kationty bylo studováno ve dvou koncentračních oblastech. Na simulaci přirozených podmínek v půdách bylo použito nasycení vazebných míst sodnými kationty do 100% a nasycení nad 100% reprezentovalo vysoce zasolené půdy. Výsledky potvrdily význam schopnosti půdy získávat vodu ze vzdušné vlhkosti, podíl vody v půdě vzrůstal s relativní vlhkostí. Vyšší podíl vody v půdě pak způsoboval snížení síly vazebných interakcí, což bylo interpretováno jako fakt, že rovnováha mezi půdní a vzdušnou vlhkostí vede k optimální vlhkost půdy. Přítomnost sodných kationtů ve vzorcích do nasycení 100% měla na množství vody zanedbatelný vliv, ale v přesycených vzorcích umožňovala navázání většího množství vody. V přesycených vzorcích bylo také pozorováno zvýšení síly vazebných interakcí mezi půdou a vodou, což vede k její větší zádrži. Molekulové můstky byli stabilnější ve vzorcích, které obsahovaly Na+. Nejvyšší stabilita byla pozorována u 10% nasycených vzorcích, další zvyšování Na-saturace pak již vedlo k snižování stability molekulových můstků. Hlavním výstupem práce je, že tvorba vodních-kationických můstků stabilizuje půdní strukturu a že má vlyv na sílu a množství vody vázané v půdě, což představuje klíčové aspekty kvality půdy. Je potřebný rozsáhlejší výskum tohoto tématu, aby získané výsledky pomohli optimalizovat půdní manažment v aridních oblastech a také v ostatních oblastech, kde dochází k degradaci půdy.

Peatlands are considered to be valuable ecosystems on the Planet and play an important role in many processes globally likewise and in the landscape. They are important for maintaining a global biodiversity, play an important role in landscape hydrology and can be a source of drinking water. They can help minimize floods and contribute to climate change mitigation. The peat bogs are one of the largest terrestrial carbon reservoirs. They have been storing carbon for centuries. Many of peat functions are affected due to human activities. It can result into release of greenhouse gases into atmosphere and the excretion of organic carbon into watercourses. The subject of the thesis is understanding of the mechanisms of water binding in peat and exploring possible dependencies within the depth profile of the peatlands using thermal analytical methods, more preciously using thermogravimetry (TG) and differential scanning calorimetry (DSC). The stability of water molecule bridge (WaMB) was investigated. It was found out that the minimum temperature required for WaMB abatement decrease and the whole structure becomes less stable with increasing peat layer depth. Water release processes were evaluated with the meaning of the TGA measurements. A decreasing trend in the degradation of thermolabile substances and an increasing trend in thermostable substances within the peat profile were observed.

Platinum nanoparticles pose a risk to environmental compartments. The aim of this diploma thesis is to conduct research on the influence of platinum nanoparticles in a wide range of concentrations on soil properties at different humidities. Specifically, it is about influencing the stability of water molecular bridges, the content of aliphatic crystallites, the retention capacity of water in the soil and the strength of water binding. Furthermore, the amount of nanoparticles sorbed on the soil was also measured. These indicators give us more information about the influence of platinum nanoparticles on evapotranspiration and soil deterioration. Methods of thermal analysis (differential scanning calorimetry) and optical methods (atomic absorption spectrometry) were used for these researches. The theoretical part of the work is devoted to a comprehensive introduction to platinum and its nanoparticles, as well as aqueous molecular bridges. It has been found that 200 nm platinum nanoparticles affect the strength of aqueous molecular bridges, have no effect on aliphatic crystallite content, have minimal effect on soil water retention, and that the water bond strength decreases due to platinum nanoparticles at high relative humidity. With 200 nm platinum nanoparticles, complete adsorption to soil occurs over the entire concentration range. With 3 nm nanoparticles, the soil is supersaturated at higher concentrations and larger amounts are not adsorbed. The results show that the larger the size of the platinum nanoparticles, the lower the negative effect on evapotranspiration and soil function itself.
