Chez l’ensemble des organismes, la capacité des cellules à se polariser est une propriété nécessaire à la croissance, à la division, à la mobilité, à la différenciation ou encore au trafic intracellulaire. Les GTPases de la famille Rho jouent des rôles prépondérants dans la régulation de ces mécanismes. Chez la levure Saccharomyces cerevisiae, nous étudions la manière dont le régulateur RhoGAP Rgd1p des protéines Rho3 et Rho4 est lui-même régulé au cours de la croissance polarisée. Nous avons montré que la protéine Rgd1 est localisée au fond du bourgeon lors de la croissance isotropique du bourgeon puis sous forme d’un anneau au cou du bourgeon lors de la cytocinèse. Nous avons également montré que le trafic intracellulaire, notamment l’exocytose et les phosphoinosotides tels que le PdtIns(4)P et le PdtIns(4,5)P2 ont un rôle majeur dans la régulation spatio-temporelle de la protéine Rgd1. Ces mécanismes permettraient d’acheminer la protéine RhoGAP au niveau des sites de croissances afin d’agir sur les protéines Rho3 et Rho4. / In all organisms, the ability of cells to polarize is a property necessary for growth, cell division, mobility, cell differentiation or intracellular trafficking. GTPases of the Rho family play central roles in the regulation of these mechanisms. In the yeast Saccharomyces cerevisiae, we study how the regulator RhoGAP Rgd1p of Rho3 and Rho4 proteins may be itself regulated during polarized growth. We showed that the protein Rgd1 is located at the bud tip during isotropic growth of the bud and form a ring at the bud neck during cytokinesis. We also showed that the intracellular traffic, especially the exocytosis and phosphoinosotides as the PdtIns(4)P and PdtIns(4,5)P2 have a major role in the spatio-temporal regulation of the Rgd1 protein. These mechanisms would deliver the RhoGAP protein at growth sites to act on Rho3 and Rho4 proteins.

The main aim of this thesis was to create a business plan to open an Italian bakery and coffee bar and discover the viability of the plan. Thesis is divided into theoretical and practical part. The practical part is formed on basis of theoretical information that characterizes business, environment of business in the Czech and Italy and main points of structure of the business plan. The practical part is formed by own business plan. This part describes goals, marketing plan, financial analysis, SWOT analysis, risk analysis and supplement. In part of conclusion readers learns if the business plan will be suitable for implementation.

This diploma thesis studies the marketing strategy of a social business Mléčný bar Bílá vrána. The theoretical part of this work presents the process of creation, implementation and control of a marketing strategy and features definitions of social business and its difference in comparison to classical businesses. The practical part of this work analyses the real strategy of Mléčný bar Bílá vrána. The strategy is researched and evaluated here. This work brings suggestions for improvements of this strategy.

La población de Lima metropolitana se caracteriza por tener un ritmo de vida ajetreado que lleva a elecciones de comida rápida y muchas veces poco saludables, que tiene como consecuencia el aumento de índices de sobrepeso u obesidad. En la actualidad se ha observado un aumento de la preocupación de las personas sobre su salud y alimentación y se ha evidenciado por el aumento de locales que ofrecen servicios y alimentos saludables en la ciudad. Sin embargo, según nuestro estudio de mercado no hemos encontrado que esta tendencia haya llegado a los locales de entretenimiento nocturno como son los bares o lounge-bar. En el presente trabajo planteamos un plan de negocio que incluye alimentación saludable, entrenamiento y que busca rescatar las interacciones interpersonales perdidas por el aumento del uso de la tecnología mediante la diversión basada en juegos lúdicos. “Cheers Lounge-bar” es un espacio innovador que busca la comodidad del cliente y promueve un estilo de vida saludable. / Lima city's population its characterized by a busy lifestyle that leads to unhealthy fast food choices, with increased obesity and overweight index. Nowadays, there has been an increased concern about health and nutrition and there has been an increased amount of locations in the city that offer healthy food and services. However, according to our market research, we haven't found that this trend has reached nightlife venues such as bars or lounge-bars. In this work we propose a business plan that includes healthy eating, training; that seeks to rescue the interpersonal interactions lost by the increase in the use of technology through fun and playful games. "Cheers Lounge-bar" is an innovative space that seeks the clients comfort and promotes a healthy lifestyle. / Trabajo de investigación

The diploma thesis follows the urban planning analysis of the BVV area, which was elaborated in the subject TG02 –Specializovanýateliér. The Diploma Project processes an architectural study of the hotel with a congress center. The request for the proposal was the classification of the building into First Class (**** hotel) according to the Official Uniform Classification of Accommodation Facilities of the Czech Republic, as well as the number of rooms 250. The congressional part should serve more than 2,000 persons. The design responds to the historical urbanism of the BVV area, where it retains and optically complements the view axis to the Z-pavilion. The building also reacts to the height next to the standing mansion. In its close proximity is the lower part of the building with the congress center, the height part with rooms is located on the opposite side, at the sports hall. The object is optically divided into two parts - a lower base and a "growing" height part with room. The congress and the hotel part are optically separated by planing offsets and façade solutions. Lower, congress, part orbit horizontal aluminum lamellas, instead, the hotel creates vertical lamellas. These lamellas serve not only as optical separation but also as shading.

The aim of the thesis is implementation of the barcode system for tracking goods to a specific business organization. Based on ascertained knowledge, the specific request for installation of the barcode is presented to firms dealing with this subject. Submitted proposals are subsequently evaluated. Then the proposal is selected which meets the company´s demands in the best way. Further, the procedure is suggested for marking goods with the barcode, possible manipulation with the marked goods and interconnection of used data with the company´s information system.

The theme of diploma thesis is the newly built sports center in Uherský Brod. The building is partially cellar with two above ground floors. The building is designed from Porotherm system and from prestressed concrete floor slab Spiroll. The roof is warm flat roof. There are 4 squash court, lasergame hall, fitness room, gym, bar and restaurant with bowling.

This Diploma Thesis solves design of the pension with wine bar. The building is located on the edge of the of village Šardice. It is a single storey basement building with attic that is roofed by a gable roof with dormers. Floor plan of the building is rectangular with the entrance facing northeast. There is a restaurant with facilities on the first floor and guest rooms where two of them are designed as wheel chair accessible. On the second floor there are only guest rooms. The construction of the building is structural masonry system. Bearing vertical structures are made of permanent formwork in the ground floor, for external walls are used aerated concrete blocks and for the inner bearing masonry are used lime sand-lime blocks. Horizontal bearing structures consists of prestressed hollow core slab, bearing structure of the roof consists of purlin roof.

Diploma thesis focuses to design anti roll-bar for Formula Student car Dragon II build in FME. This work follows the proposal anti roll-bar car Dragon I of Jan Krasula. Task of this work is analyses last design, introduce more effective method treatment, create own design with parameters which will be better then last design. An integral part design is prepare manufacture documentation which will be base for manufacture of this komponent
