Caracterização de possíveis homólogos aos fatores de iniciação da tradução eIF4G e eIF4A de Leishmania major

Neste processo, atuam proteínas designadas eIFs (fatores de iniciação da tradução). Destas destaca-se o complexo eIF4F eIF4E, eIF4A e eIF4G que permite associar o mRNA ao ribossomo. Identificamos no genoma de Leishmania major seqüências que apresentam homologia aos componentes do eIF4F. Este trabalho contempla a caracterização de um homólogo do eIF4A (LmeIF4A2) e três homólogos eIF4G (LmeIF4G1-3). O gene LmeIF4A2 foi clonado, expresso e utilizado na produção de anticorpos. Ensaios de Western-blot sugeriram não haver expressão do LmeIF4A2 na fase promastigota de Leishmania major. Em seguida realizamos construções com os LmeIF4G1-3, fusionando-os a GST, permitindo a realização de ensaios de pull down visando investigar sua associação com as proteínas LmeIF4A1-2. O LmeIF4A2 não interage com nenhum dos LmeIF4G1-3 e o LmeIF4A1 interage especificamente com o LmeIF4G3. Em outra etapa produzimos anticorpos contra as proteínas LmeIF4G1-3 para avaliar sua expressão na forma promastigota do parasita. A expressão do LmeIF4G3 foi confirmada, todavia não detectamos a expressão dos LmeIF4G1-2. A interação eIF4G/eIF4E foi investigada em ensaios onde homólogos LmeIF4G1-3 foram incubados a homólogos LmeIF4E1-3, para tentar reconstituir parcialmente o complexo eIF4F in vitro, e testados quanto a sua afinidade pelo cap sintético. Estes resultados se mostraram inconclusivos. A utilização de novas abordagens e a caracterização dos demais fatores será importante na elucidação da tradução nestes protozoários

Development of new approaches for the synthesis of flavaglines / Développement de nouvelles approches pour la synthèse des flavaglines

Nous avons développé deux méthodes originales de synthèse de cyclopenténones fonctionnalisées et découvert de nouveaux réarrangements d’alcools 1-styryl propargyliques catalysés par des acides.Ce travail de thèse a aussi permis de mettre en évidence les limitations d’une méthode de synthèse des flavaglines développée par des chercheurs de la compagnie Bayer. De plus, la resynthèse de certaines flavaglines et la synthèse d’une sonde de fluorescence originale ont permis de mieux caractériser le mode d’action et le potentiel thérapeutique des flavaglines pour leurs effets anticancéreux (notamment dans la résistance aux inhibiteurs de B-RAF), la régulation du canal TRPM6 et l’infection par le virus du Chikungunya. / We have developed two novel methods to prepare functionalized cyclopentenones and also discovered new acid-catalyzed rearrangements of 1-styryl propargylic alcohols. This work also explored the limitations of a method for the synthesis of flavaglines reported by Bayer researchers. Moreover, the re-synthesis of some flavaglines and the synthesis of an original fluorescence probe allowed some advances in the characterization of the mechanism of action and therapeutic potential of flavaglines for their anti-cancer effects (especially in the resistance to B6RAF inhibitors), the regulation of TRPM6 channel and the infection by Chikungunya virus.

Nous avons développé trois accès synthétiques performants à des cyclopentènones fonctionnalisées en exploitant des réactivités inattendues que nous avons découvertes. Nous avons aussi effectué la première synthèse d’isostères des flavaglines substitués par un groupement formylamino ou mésylamino en position 1b et ainsi démontré l’importance de l’hydroxyl en cette position pour la cytotoxicité de ces composés. De plus, nous avons aussi contribué à l’exploration du potentiel thérapeutique des flavaglines et d’un autre ligand des prohibitines, la fluorizoline, dans le traitement des cancers et de l’inflammation chronique des intestins, ainsi que dans la prévention des effets adverses des chimiothérapies au niveau cardiaque. / We have developed three novel synthetizes of functionalized cyclopentenones based on unexpectedreactivities that we discovered.We also developed the first synthesis of flavaglines isostere substituted by a formylamino or mesylaminogroup on the position of 1b, and demonstrated the importance of a hydroxyl group on this position forcytotoxicity.Moreover, we contributed to the exploration of the therapeutic potential of flavaglines and another ligand ofprohibitins, fluorizoline, in the treatment of cancers and intestinal chronic inflammation, and also in theprevention of the cardiac adverse effects in anticancer treatments.
