Endothelial Progenitor Cells (EPCs) are thought to participate in the healing of injured vascular endothelium by incorporating into the defect sites to mediate endothelial recovery. Recently, osteopontin (OPN) was shown to be fundamental in accelerating estrogen-dependent healing of injured blood vessels. Here, we are investigating the effect OPN has on EPC behavior. Late outgrowth human EPCs (LEPCs) were derived from circulating monocytes isolated by leukophoresis, and grown in culture until passage six. L-EPCs were then assayed for adhesion, spreading, chemotaxis, and haptotaxis, as well as resistance to detachment by flow electric cellsubstrate
impedance sensing (ECIS). The results of standard and ECIS methods showed both dose and time dependent responses in cell adhesion and spreading. In addition, OPN promoted haptotactic migration of EPCs in Boyden chamber assays. LEPCs seeded onto 10μM OPN substrates and exposed to laminar flow had grater survival and higher resistance to detachment than OPN/static and flow only conditions. CD44 and !1 integrins were only responsible for approximately 50% of LEPCs
adhesion to OPN compared to the unblocked condition. Western blots showed that Rho GTPases were activated in L-EPCs seeded on OPN. However, this activation could not be completely blocked by either CD44 or !1 integrin antagonists. These data confirm the direct effects of OPN on EPCs adhesion, and suggest that OPN works by mediating cell adhesion during vascular injury.

Användarupplevelser av affärssystem: Inlärningsmetodens påverkan på de åsikter som slutanvändare bildarEmnegard, Sara, Emtö, Malin January 2023 (has links)
Införskaffning av affärssystem är en komplex process där nära hälften av samtliga implementeringsprojekt misslyckas. Forskning har visat att en av huvudorsakerna till att postimplementationsfasen av ett affärssystemimplementeringsprojekt misslyckas beror på att systemet inte accepteras av slutanvändarna och den främsta orsaken till detta menar vissa är ett underutnyttjande av systemet till följd av otillräcklig användarutbildning. Forskare menar att individer som har lite eller ingen tidigare kunskap om ett visst ämne har det svårt att lära sig ämnet via enbart text. Därav finner det sig vara intressant att undersöka huruvida en översättning av textuella användarmanualer till hybridvarianter av text blandat med visuella representationer skulle kunna användas i utbildningssammanhang. Det har därför skapats en användarmanual enligt EPC-notation för att testa om användares åsikter kring ett visst system differentierar mellan användare som använder textuella användarmanualer jämfört med de som använder den skapade modellmanualen. Deltagarna i studien fick först besvara en kort enkät i syfte att få en bild av deras demografiska profil, baserat på detta har de delats upp i två grupper och utfört samma uppgifter i affärssystemet Microsoft Dynamics 365 Business Central men med olika användarmanualer. Efter utförande av uppgifterna har deltagarna blivit intervjuade om deras respektive upplevelser. Intervjuerna spelades in och transkriberades för att sedan analyseras genom en tematisk analys. Under den tematiska analysen kodades gruppernas svar initialt var för sig för att erhålla en bild av respektive grupp, för att sedan identifiera gruppöverskridande kategorier och teman. Resultatet av studien visar att deltagarna har ungefär samma generella systemkrav, men upplevelserna av det använda systemet differentierar grupperna emellan. Både positiva och negativa åsikter uttrycktes från båda grupperna, men deltagarna som använt den textuella manualen har generellt en sämre uppfattning om systemet än deltagarna som använt modellmanualen. Även de uttryckta känslorna kring systemet differentierade mellan grupperna på samma sätt. Detta indikerar att typ av användarmanual de facto kan påverka användarupplevelsen av ett system, och stärker tidigare forsknings tes kring visuell kontra textuell inlärning. / Procurement of business systems is a complex process where nearly half of all implementation projects fail. Research has shown that one of the main reasons why implementation projects fail is that the system is not accepted by the end users and the main reason for this is believed by some to be underutilization of the system as a result of insufficient user training. Researchers believe that individuals who have little, or no previous knowledge of a certain subject find it difficult to learn the subject via text alone. Hence, it is interesting to investigate whether a translation of textual user manuals into hybrid variants of text mixed with visual representations could be used in an educational context. A user manual has therefore been created according to EPC notation to test whether users' opinions about a certain system differentiate between users who use textual user manuals compared to those who use the model manual created. The participants in the study were first asked to answer a short survey in order to receive information about their demographic profile. Based on this information they have been divided into two groups who, with different user manuals at hand, performed the same tasks in the business system Microsoft Dynamics 365 Business Central . After completing the tasks, the participants were interviewed about their respective experiences. The interviews were recorded and transcribed, and the data has then been analysed using thematic analysis. During the thematic analysis, the groups’ responses were initially coded separately to obtain an understanding of each group. Based on this, cross-group categories and themes were identified. The findings of this study indicate that the participants share similar overall system requirements, but that their experience with the system differentiate the groups from each other. Both positive and negative opinions were expressed from both groups, but the participants who used the textual manual generally have a more negative perception of the system than the participants who used the model manual. The expressed feelings about the system also differentiated between the groups in the same way. This suggests that the type of user manual can indeed influence the user experience of a system, thereby reinforcing previous research findings on the effectiveness of visual versus textual learning.

90Y (T1/2 = 2,67 dias; Eβmáx = 2,28 MeV) é um radionuclídeo com eficácia estabelecida para diversas terapias de câncer, marcando biomoléculas e no tratamento da radiosinovectomia. Devido às suas propriedades nucleares, é obtido através do decaimento do 90Sr (T1/2 = 28 anos), na forma de um gerador. Vários tipos de geradores de 90Sr/90Y foram desenvolvidos, e os mais empregados são os que usam resinas de troca catiônica, onde Sr e Y são adsorvidos e 90Y é seletivamente eluído com acetato ou EDTA. A desvantagem deste tipo de gerador é a radiólise, que degrada o seu uso. O gerador eletroquímico é uma solução proposta devido ao fato de não haver efeitos significativos da radiação sobre o próprio gerador. Neste conceito, a diferença entre os potenciais eletroquímicos dos elementos Y e Sr é utilizada para se obter uma rápida separação do 90Y do 90Sr. A produção de 90Y via formação de colóides é o método mais simples para a separação, baseando-se na formação de colóides de Y em pH altamente alcalino, podendo ser filtrado e separado do Sr, sendo posteriormente dissolvido em HCl. O objetivo deste trabalho consistiu no desenvolvimento de tecnologias para o preparo de geradores de 90Sr/90Y, onde foram desenvolvidos três tecnologias, a saber: geradores do tipo coluna utilizando resinas catiônicas, geradores via formação de colóides e geradores eletroquímicos. Foram também avaliadas metodologias para o controle de qualidade radionuclídico do 90Y dos geradores desenvolvidos: cintilação líquida, identidade radionuclídica, cromatografia por extração em papel (EPC) utilizando complexantes para 90Y e técnica por Espectrometria de Emissão Ótica com Plasma Indutivamente Acoplado (ICPOES). Os resultados mostraram que os geradores utilizando resinas catiônicas obtiveram os melhores resultados em relação ao rendimento e eficiência (~ 83%) de eluição, reprodutibilidade e a pureza radionuclídica. O gerador eletroquímico mostrou um potencial para o desenvolvimento, tendo a vantagem de não sofrer os efeitos da radiólise do par 90Sr/90Y, como acontece com a resina. Previous issue date: 2013-01-18 / O estudo tem por objetivo descrever, analisar e propor um conjunto de tecnologias aplicáveis ao auxílio da gestão da aquisição, do diligenciamento, da inspeção e da contratação de itens de suprimento que compõem um Empreendimento Complexo para, assim, permitir que as empresas EPCistas no Rio de Janeiro tenham ganhos de produtividade, sem perda da qualidade inerente ao processo. Os conceitos-chave que permeiam o trabalho giram em torno da tecnologia envolvida para otimizar a gestão de suprimentos de empresas que compõem a cadeia produtiva EPC. De modo a permitir uma melhor leitura do comportamento dos EPCistas no Rio de Janeiro e, consequentemente identificar as melhores práticas e ferramentas interativas mais aplicáveis a esse nicho, foi realizado um estudo aprofundado dos principais stakeholders que influenciam ou são influenciados por essa cadeia, tais como: visão do contratante, do fornecedor, do parceiro e de instituições de classe que atuam promovendo boas práticas de gestão do setor. O estudo, realizado através de entrevistas direcionadas aos principais stakeholders dos EPCistas, trás elementos que fundamentam a necessidade de implementação de soluções tecnológicas mais adequadas à evolução pela qual essa cadeia passará, influenciada especialmente pelas descobertas do Pré-Sal. / The present study aims to describe, analyze and propose a set of technologies applicable to aid the management of the acquisition, the expediting, inspection and contracting supply items that compose a complex undertaking to thereby allow companies in EPCistas Rio de Janeiro have productivity gains without loss of quality inherent in the process. The key concepts that permeate the work revolve around the technology involved to optimize the supply management of companies that make up the supply chain EPC. To enable a better reading of the behavior of EPCistas in Rio de Janeiro and thus identify best practices and interactive tools most applicable to that niche, we performed a detailed study of the major stakeholders that influence or are influenced by the chain, such as: Vision of the contractor, supplier, partner and working-class institutions by promoting good management practices in the industry. The study, conducted through interviews of key stakeholders aimed to EPCistas, behind elements that underlie the need to implement technological solutions to the most appropriate evolution through which the chain will, especially influenced by the pre-salt discoveries.
