Análise dos níveis relativos de transcrição de genes antioxidantes e da cadeia de transporte de elétrons de Aspergillus fumigatus / Transcription relative levels analysis of Aspergillus fumigatus antioxidant and electrons transport chain genes

Aspergillus fumigatus é um fungo oportunista, sendo uma importante causa de morbidade e mortalidade entre os pacientes imunossuprimidos, acometidos com aspergilose invasiva. O sistema de defesa antioxidante do fungo atua como fator primordial de sua patogenicidade, mitigando os efeitos deletérios de espécies reativas produzidas pelo hospedeiro. Este trabalho teve o objetivo de avaliar o nível de transcrição relativo dos genes associados às proteínas mitocondriais e antioxidantes na presença de diferentes agentes pró-oxidantes para hifas e germinantes de cepas controle (CEA17) e nocaute para oxidase alternativa (?aoxA) de A. fumigatus. Utilizando qPCR, foram analisados 23 genes para as cepas CEA17 e mutante, dentre os quais, 5 associados à cadeia de transporte de elétrons (CTE), 4 relacionados aos componentes alternativos mitocondriais e 14 associados às proteínas antioxidantes. Os resultados foram obtidos através da normalização com o gene constitutivo gapdh utilizando a cepa CEA17 sem tratamento de cada fase de desenvolvimento celular como calibrador. As diferenças de transcrição da maioria dos genes estudados variaram com o pró-oxidante utilizado e com a fase de desenvolvimento celular (germinantes e hifas). A CTE em germinantes apresentou maiores níveis de transcritos dos complexos II e III em CEA17 do que em ?aoxA tratadas com menadiona. Com os demais complexos e atpase não foram observadas diferenças significativas quando comparados os resultados de ambas as cepas. Entretanto, os níveis de transcritos de atpase não foram superiores ao controle em nenhuma das cepas, sugerindo sua regulação na presença de ucp. ucp-like teve seus níveis de transcritos aumentados no mutante, sugerindo o desacoplamento da CTE em relação ao controle e à CEA17, que apresentou alto nível de transcritos de nde. Nesta cepa, os níveis de aoxA e ucp não foram significativamente diferentes. Esse fato, associado a níveis menores de ndiA e transcritos dos complexos I e II, sugerem manutenção do fluxo de elétrons na mitocôndria. Em relação às enzimas antioxidantes, houve aumento significativo nos níveis de cat2, catA, prx1, trx1 e grx1, indicando provável resposta à ausência de AOX. Os transcritos relacionados à CTE em hifas não apresentou diferença significativa nos níveis de complexo III e atpase em CEA17 e ?aoxA. No entanto, complexo IV se mostrou superior no mutante. Nesta fase de desenvolvimento, o aumento dos níveis de ucp-like está relacionado ao tratamento com menadiona e não com a presença de aoxA, explicando os mesmos níveis de transcritos em CEA17 e ?aoxA. Os níveis de nde sugerem atividade coordenada de sua proteína com AOX, por seus níveis estarem modulados pela presença de aoxA. A participação da Nde e da AOX na detoxificação das espécies reativas de oxigênio é sugerida ao se obter altos níveis de seus transcritos em hifas tratadas com menadiona. Diferentemente de germinantes, transcritos de sod3 foram predominantes em hifas de CEA17, enquanto transcritos de sod2 foram maiores em ?aoxA. Entre as catalases, os níveis de catA foram maiores no mutante, bem como a atividade das catalases como um todo. prx1 e trx1 foram maiores em CEA17 e ii grx1 em ?aoxA. Porém, trx1 atingiu os mais altos níveis de transcritos em germinantes, indicando possível redução de proteínas tiólicas e ribonucleotídeos por diferentes vias, de acordo com a fase de desenvolvimento do fungo. As enzimas antioxidantes clássicas, como superóxido dismutase, catalase e enzimas tiólicas, apresentaram aumento de transcrição na maioria das condições estudadas. No entanto, menadiona e paraquat promoveram maiores variações nos transcritos do que o observado com peróxido de hidrogênio gerado pelo sistema glicose/glicose oxidase. / Aspergillus fumigatus is an opportunistic fungus which has been referred to as a major cause of morbidity and mortality in imunosupressed patients affected with invasive aspergillosis. The antioxidant defense system of the fungus plays an important role in its pathogenicity by overcoming the negative effects of the oxygen reactive species produced by host. This work aims evaluating the transcription of genes associated to mitochondrial and antioxidant proteins in the presence of different prooxidants for hyphae and germinants of control (CEA17) and alternative oxidase knockout (?aoxA) strains. Using qPCR, 23 genes have been analyzed, 5 of which are electron transport chain (ETC), 4 are related to alternative components of the mitochondria, and 14 are associated to antioxidant proteins. The results were obtained via normalization with the constitutive gene gapdh using untreated CEA17 strain in each developmental phase as calibrator. The differences in the transcription of most analyzed genes varied with the prooxidant and the developmental phase (germinants and hyphae). The ETC in germinants presented higher amounts of transcripts of the complex II and III in CEA17 than in ?aoxA treated with menadione. With other complexes and the atpase no significant difference has been noted between both strains. However, the transcript levels of ATP synthase were not higher than control in any strain, which is related to its regulation in the presence of UCP. UCP-like had increased transcription in mutant, suggesting uncoupling from the ETC, relative to control and CEA17, which showed high levels of nde. In this strain, aoxA and ucp-like transcripts were not significantly different. This fact associated the lower levels of ndiA and transcripts of the complexes I and II suggest the maintenance of the electron flow in the mitochondria. Concerning the antioxidant enzymes, there was significant increase in the transcript production of cat2, catA, prx1, Ttrx1 and Ggrx1, suggesting a response to the absence of AOX. The ETC transcripts in hyphae did not present significant difference in the levels of complex III and atpase in CEA17 and ?aoxA. Yet the complex IV has shown higher in mutant than in CEA17. In this developmental phase, the increase of ucp-like is related to the treatment with menadione and not with the presence of aoxA, what explains the equivalent amount of transcripts in CEA17 and ?aoxA. The levels of nde suggest coordinated action of the protein with AOX, because it is modulated by the presence of aoxA. The role of Nde and AOX in the detoxification of reactive oxygen species is suggested by their high levels of transcripts in hyphae treated with menadione. Different from germinants, transcripts of sod3 was predominant in CEA17 hyphae, whereas transcripts of sod2 were higher for ?aoxA. Among catalases, levels of catA were higher in mutant, as was the activity of catalase as a whole. prx1 and trx1 transcripts were higher in CEA17 and grx1 in ?aoxA. Ru(II) under illumination: a study of charge and energy transfer elementary processes / Complexes de ruthénium sous illumination

Une compréhension sans cesse plus pointue des processus élémentaires de transferts de charges et d’énergie, qui sont à la base même de nombreux processus biologiques, permet non seulement l’élaboration mais aussi l’amélioration de la mise au point de molécules photoactives utiles dans différentes applications. C'est le cas (i) de systèmes moléculaires et supramoléculaires destinés à mimer efficacement la photosynthèse, ou encore (ii) de molécules photoactives capables d’interagir avec des macromolécules biologiques et d’induire une transformation de ces biomolécules. C’est dans ce cadre général que s’inscrit l’élaboration de nouveaux complexes polyazaaromatiques de Ru(II) capables d’interagir avec la double hélice d’ADN et de photoréagir avec sa base la plus réductrice, la guanine, par transfert d’électron photoinduit. C’est sur la base de ces processus que des nouveaux agents antitumoraux photoactivables ont pu être développés. L’utilisation de complexes de Ru(II) dans le design d’entités supramoléculaires polymétalliques destinées à jouer le rôle de collecteurs de lumière et permettant ainsi de mimer les systèmes d’antennes naturels s’intègre également dans cette démarche. <p><p>L’ensemble de notre travail s’est concentré sur ces deux domaines d’applications. Par l’étude de différents processus de transfert de charges/d’énergie au sein des complexes seuls (processus intramoléculaires) ou en interaction avec un environnement spécifique (processus intermoléculaires), nous avons souhaité mettre en évidence l’intérêt de l’utilisation d’un nouveau ligand plan étendu, le tpac, au sein de complexes du Ru(II). Un tel ligand permet en effet de conférer d’une part une affinité élevée des complexes résultants pour l’ADN, et d’autre part, de par sa nature pontante, de connecter des unités métalliques entre elles au sein d’entités supramoléculaires de taille importante. <p><p>Les propriétés photophysiques de quatre complexes basés sur le ligand plan étendu tpac, le [Ru(phen)2tpac]2+ (P) et son homologue dinucléaire le [(phen)2Ru tpac Ru(phen)2]4+ (PP) (à base de ligands ancillaires phen), ainsi que le [Ru(tap)2tpac]2+ (T) et son homologue dinucléaire le [(tap)2Ru tpac Ru(tap)2]4+ (TT) (à base de ligands ancillaires tap), ont été étudiées et comparées entre elles.<p><p>L’examen de ces propriétés, d’abord pour les complexes seuls en solution, en parallèle avec celles de complexes dinucléaires contenant un ligand pontant PHEHAT, a permis de mettre en évidence l’importance de la nature du ligand pontant utilisé. Ces résultats ont ainsi révélé qu’un choix judicieux du ligand pontant permet de construire des entités de grande taille capables de transférer l’énergie lumineuse vers un centre (cas du ligand PHEHAT), ou, au contraire, de relier entre elles des entités ne s’influençant pas l’une l’autre d’un point de vue photophysique (cas du ligand tpac). <p><p>Les propriétés des complexes du tpac, étudiés cette fois en présence de matériel génétique (mononucléotide GMP, ADN ou polynucléotides synthétiques), se sont révélées très différentes selon que le complexe portait des ligands ancillaires phen (P, PP) ou tap (T, TT). Seuls les complexes à base de tap sont en effet photoréactifs envers les résidus guanine. Nous avons dès lors focalisé cette partie de notre travail sur les deux complexes T et TT. Cette photoréaction, ainsi que le transfert d’électron photoinduit entre ces complexes excités et la guanine, ont pu être mis en évidence par différentes techniques de spectroscopie d’émission tant stationnaire que résolue dans le temps, ainsi que par des mesures d’absorption transitoire dans des échelles de temps de la nano à la femto/picoseconde. L’étude du comportement photophysique des complexes en fonction du pH a en outre révélé de manière très intéressante que, pour des études en présence d’ADN, la protonation des états excités des complexes devait être considérée. Les résultats de cette étude nous ont fourni des pistes quant à l’attribution des processus observés en absorption transitoire.<p><p>Le transfert d’électron a également fait l’objet d’une étude par des méthodes théoriques. Ces calculs ab initio ont permis de mettre en évidence une faible influence de l’énergie de réorganisation sur la vitesse de transfert d’électron, qui semble dépendre plus sensiblement de la non-adiabaticité du processus, mais surtout de l’énergie libre de la réaction et d’un éventuel couplage à un transfert de proton. <p><p>L’ensemble des résultats obtenus avec les complexes T et TT en présence de matériel génétique, qui, de manière assez inattendue, sont très semblables, indiquent que ces complexes présentent tous deux un grand intérêt pour le développement de nouvelles drogues antitumorales photoactivables.<p> / Doctorat en Sciences / info:eu-repo/semantics/nonPublished

Étude expérimentale du guidage du faisceau d’électrons dans le cadre de l’allumage rapide de cibles de fusion

Les travaux de cette thèse s’inscrivent dans le cadre de l’allumage rapide pour la fusion par confinement inertiel (FCI), pour la production d’énergie. Dans ce schéma les phases de compression et d’allumage sont découplées. Au cours de la seconde phase, le faisceau d’électrons doit parcourir une distance de 300 µm dans le combustible dense avantde déposer son énergie au coeur de la cible et d’initier les réactions de fusion. Le principal défaut de ce schéma réside dans la divergence du faisceau d’électrons au cours de son transport dans la matière dense. Parmi plusieurs schémas proposés pour réduire cette divergence, nous considérons ici, les schémas sans cône basés sur la collimation des électrons dans un champ magnétique. En particulier, A.P.L. Robinson et ses collaborateurs [Phys. Rev. Lett. 100, 025002, 2008] ont proposé une méthode simple à mettre en place pour contrôler la divergence du faisceau d’électrons :utiliser une séquence de deux impulsions laser. La première impulsion permet de créer un environnement magnétique favorable au confinement du faisceau d’électrons engendré par la seconde interaction. La validation de cette proposition est le sujet de cette thèse. Nous présenterons les résultats expérimentaux et les modélisations théoriques motivées par cette proposition. L’expérience du guidage d’un faisceau d’électrons avec deux impulsions laser a été réalisée sur l’installation laser petawatt Vulcan au Rutherford Appleton Laboratory (RAL) à Didcot en Angleterre. Elle est basée sur la proposition d’un groupe international dans le cadre du projet FCI HiPER. Cette expérience nous a permis d’obtenir les conditions de guidage en fonction du rapport des intensités et du délai entre les deux impulsions. Les résultats de l’expérience ont été modélisés par le code hydrodynamique CHIC couplé au module de transport de particules chargées M1. L’interprétation des résultats expérimentaux nous a permis d’expliquer la base de la physique du guidage du faisceau d'électrons et d'en définir les conditions magnétiques favorables. / The work presented in this thesis is realised in the framework of the fast ignition of inertial confinement fusion for energy production. In this scheme the compression and the ignition phases are decoupled. During the second phase, the electron beam must cross over 300 µm in the dense fuel to deposit its energy in the dense core and ignite the fusion reactions.The major problem of the scheme is related to the divergence of the electron beam while it crosses the dense matter. Among the different propositions to inhibit the electron divergence we consider here the schemes without cone that are based on the effect of magnetic collimation. In particular, A.P.L. Robinson and his co-authors [Phys. Rev. Lett. 100, 025002, 2008] suggested a simple way to control the electron beam divergence by using a sequence of two laser pulses. The first one creates a magnetic background favourable for the confinement of the second electron beam resulting from the second interaction. The validation of this scheme is the major goal of this thesis.We present the results of experimental sudies and numerical modeling of the electron beam guiding with help of two consequent laser pulses. The experiment was performed on the Vulcan facility at the Rutherford Appleton Laboratory at Didcot in UK, based on the proposal submitted by an international group of scientists in the framework of the European project for inertial fusion energy HiPER. This experiment allowed us to define a combination of laser and target parameters where the electron beam guiding takes place. The analysis of experimental data and numerical modelling is realised with the hydrodynamic code CHIC coupled to the charged particules transport module M1. The interpretation of the experimental results allowed us to explain the experimental data and the physical basis of guiding and to define the magnetic conditionflavourable to the electron beam guidance.
