For this, there were five tests in which the control group animals received no treatment, while animals in the treated group received the formulation within the association in a single dose of 600 mcg of abamectin and 3 mg of fluazuron per Kg of body weight by route ?pour-on?. As to acaricide efficacy, two tests were conducted, one using artificially infested cattle and other naturally infested by Rhipicephalus microplus. In the same manner, a test was conducted to evaluate the efficacy of the product in test front of Dermatobia hominis naturally infested cattle. And two tests anthelmintic efficacy in infected cattle naturally and artificially by gastrointestinal nematodes. The mean of the carrapaticide efficacy for cattle artificially infested with R. microplus over the 23 days after treatment was 87,40%. The preventive effectiveness results of 98,12%; 98,68%; 98,36%; 97,88%; 96,76%; 96,45%; 96,84%; 97,03%; 95,36% and 87,07% respectively for the experimental days +7, +14, +21, +28, +35, +42, +49, +56, +63, +70. The acaricide effectiveness for naturally infested cattle by R. microplus was 96,30%; 96,90%; 95,98%; 96,83%; 98,32%; 96,12%; 96,15%; 98,8%; 97,18%, 99,12%; 97,18%; 98,04% and 77,50%, respectively for the experimental days +7, +14, +21, +28, +35, +42, +49, +56, +63, +70, +77, +84 and +91 for the treated group. The efficacy of the test product to D. hominis was 90,70% and 96,84% for days +7 and +14. The values of anthelmintic efficacy in naturally infected cattle with gastrointestinal nematodes was 92,17%; 89,32%; 87,38% respectively for the experimental days +7, +14, +21 for the treated group. For cattle artificially infected by gastrointestinal nematodes the efficacy was 92,59%; 97,62%; 87,82%; 86,96%; 96,00% respectively for Haemonchus placei, Trichostrongylus axei, Cooperia punctata, Cooperia pectinata and Oesophagostomum radiatum. The association abamectin 0.6% with fluazuron 3.0% was effective in artificially infested cattle and naturally infested by R. microplus, as well as demonstrates efficacy for D. hominis. The test product showed efficacy also in the reduction the eggs counts per gram of feces (EPG) in cattle naturally infected with gastrointestinal nematodes, as well as shown to be effective front of artificial infections with H. placei, T. axei and O. radiatum and has presented moderately effective against C. pectinata and C. punctata / O objetivo da pesquisa foi avaliar a efic?cia da associa??o de abamectina 0.6% com fluazuron 3.0% controle de Rhipicephalus microplus, Dermatobia hominis e nemat?ides gastrointestinais em bovinos. Para isso, foram realizados cinco ensaios, nos quais os animais do grupo controle n?o receberam tratamento, enquanto que os animais do grupo tratado receberam a formula??o da associa??o na dose ?nica de 600 mcg de abamectina com 3 mg de fluazuron por Kg de peso corporal, por meio de via ?pour-on?. Quanto ? atividade carrapaticida, foram conduzidos dois ensaios, um utilizando bovinos artificialmente infestados e outro naturalmente infestados por Rhipicephalus microplus. Da mesma forma, foi realizado um ensaio para avaliar a efic?cia do produto em teste frente a Dermatobia hominis em bovinos infestados naturalmente e dois ensaios de efic?cia anti-helm?ntica em bovinos naturalmente e artificialmente infectados por nemat?ides gastrointestinais. A m?dia da efic?cia carrapaticida para bovinos artificialmente infestados por R. microplus ao longo dos 23 dias ap?s o tratamento foi de 87,40%. O resultado da efic?cia preventiva foi de 98,12%; 98,68%; 98,36%; 97,88%; 96,76%; 96,45%; 96,84%; 97,03%; 95,36%; e 87,07% respectivamente para os dias experimentais +7, +14, +21, +28, +35, +42, +49, +56, +63, +70. A efic?cia carrapaticida para bovinos naturalmente infestados por R. microplus foi de 96,30%; 96,90%; 95,98%; 96,83%; 98,32%; 96,12%; 96,15%; 98,8%; 97,18%, 99,12%; 97,18%; 98,04% e 77,50%, respectivamente para os dias experimentais +7, +14, +21, +28, +35, +42, +49, +56, +63, +70, +77, +84 e +91 para o grupo tratado. A efic?cia do produto em teste para D. hominis foi de 90,70% e 96,84%, para os dias +7 e +14. Os valores de efic?cia anti-helm?ntica em bovinos naturalmente infectados por nemat?ides gastrointestinais foi de 92,17%; 89,32%; 87,38% respectivamente para os dias experimentais +7, +14, +21 para o grupo tratado. Para bovinos artificialmente infectados por nemat?ides gastrointestinais, a efic?cia foi de 92,59%; 97,62%; 87,82%; 86,96%; 96,00% respectivamente para Haemonchus placei, Trichostrongylus axei, Cooperia punctata, Cooperia pectinata e Oesophagostomum radiatum. A associa??o de abamectina 0.6% com fluazuron 3.0% demonstrou ser eficaz em bovinos artificialmente e naturalmente infestados por R. microplus, assim como apresentou efic?cia para D. hominis. O produto em teste apresentou efic?cia tamb?m na redu??o de ovos por grama de fezes (OPG) em bovinos naturalmente infectados por nemat?ides gastrointestinais, assim como, demonstrou ser efetivo frente a infec??es artificiais por H. placei, T. axei e O. radiatum, e apresentou-se moderadamente efetivo frente a C. pectinata e C. punctata.

Parâmetros farmecocinéticos e atividade endectocida de uma nova formulação contendo avermectinas, via tópica (pour-on), em bovinos

Quanto à atividade ectoparasiticida, foram conduzidos nove ensaios, sendo quatro contra Rhipicephalus (Boophilus) microplus (três com infestação natural e um com infestação artificial). Os outros cinco tiveram como alvo Haematobia irritans (dois), dois em Cochliomyia hominivorax (miíases escrotais naturais) e o último em larvas de Dermatobia hominis (berne, infestação natural). Uma avaliação anti-helmíntica (24 necropsias), completou os 12 experimentos realizados. A associação (ivermectina + abamectina) apresentou eficácia anti-ixodídica superior a 95% até o 28o dia pós-tratamento (DPT) nos três experimentos a campo. No “stall test” alcançou 100% de eficácia em oito datas observacionais e eficácia superior a 95% do 8o a 43o DPT. Contra H. irritans os valores de eficácia foram superiores a 90% do 3o ao 14o DPT, em ambos experimentos conduzidos. Eficácia bernicida acima de 90% foi observada do 7o ao 42o DPT. A nova formulação apresentou eficácia anti-helmíntica máxima (100%) contra sete das 11 espécies de helmintos diagnosticadas. Contra Haemonchus placei e Cooperia punctata os percentuais de eficácia foram de 92,22% e 95,77%, respectivamente. Os estudos farmacocinéticos revelaram valores da área sob a curva (AUC) de 1089,34 ± 237,17 ng dia/mL e de 634,34 ± 121,49 ng dia/mL, para ivermectina 1,5% e abamectina 0,5%, respectivamente. O pico de concentração máxima (Cmax) da ivermectina 1,5% (70,62 ng/ml) foi superior ao da abamectina... / A new association containing of ivermectin 1,5% + abamectin 0,5%, topical route of administration (pour-on), was evaluated in 12 experiment, incluse the pharmacokinetic parameters and residues determination this compounds in cattle medicados. As ectoparasiticide activity, was nine study, four against Rhipicephalus (Boophilus) microplus (tree with natural infestation and one with artificial infestation). The other five had like target Haematobia irritans (two), two in Cochliomyia hominivorax and last in the larvaes of Dermatobia hominis (natural infestation). An anthelmintic avaliation (24 necropsias), completed the 12 experiments. The association (ivermectin + abamectin) presented efficacy antiixodidic higher than 95% up to the 28th day post-treatment (DPT) in the tree field experiments. In the “stall test” showed 100% of efficacy in eight experimentally time and efficacy higher than 95% on the 8th to 43th DPT. Against H. irritans the efficacy values were higher than 90% on the 3th to 14th DPT, in the both experiments conducted. Efficacy anti- D. hominis higher than 90% on the 7th to 42th DPT. The new formulation presented maximal efficacy (100%) against seven of the eleven helminth especies diagnostic. Against Haemonchus placei and Cooperia punctata the percentiles efficacy were the 92,22% and 95,77%, respectivelly. The studies pharmacokinetics showed iverrmectin, present in the association, showed values area under the plasma concentration-time curve AUC of 1089,34 ± 237,17 ng day/mL and 634,34 ± 121,49 ng day/mL, for the ivermectin and abamectin, respectivelly. Atividade endectocida, segurança clínica e farmacocinética de resíduos de uma nova alternativa terapêutica (Fluazuron + Abamectina) em bovinos

Quanto à atividade carrapaticida, foram conduzidos dois ensaios, um utilizando bovinos experimentalmente infestados e outro bovinos naturalmente parasitados por Rhipicephalus (Boophilus) microplus. Em dois outros estudos, avaliou-se a eficácia contra Haematobia irritans e contra larvas de Dermatobia hominis em bovinos naturalmente parasitados. A atividade anti-helmíntica da associação foi avaliada em 12 animais necropsiados, os quais eram portadores de nematodioses gastrintestinais (infecção natural). Para determinação de remanescentes residuais em tecidos dos bovinos medicados foram necropsiados 26 animais. Nenhuma alteração significativa (P≥0,05) foi verificada nos parâmetros hematológicos (hemogramas) e bioquímicos mensurados nos bovinos medicados com nova associação, até o 10º dia pós-tratamento. A eficácia anti-ixodídica da associação fluazuron 3,0% + abamectina 0,5%* (stall test) atingiu índices de eficácia superiores a 80,00% em 78 datas experimentais. O fluazuron 2,5%** atingiu eficácia acima de 80,00% em 77 datas. Eficácia máxima (100,00%) foi atingida em 40 datas experimentais pela associação fluazuron 3,0% + abamectina 0,5%, enquanto que o fluazuron 2,5%** atingiu este índice em 51 datas. Estatisticamente as formulações fluazuron 3,0% + abamectina 0,5%* e fluazuron 2,5%** reduziram significativamente (P<0,05) as contagens de fêmeas de R. The clinical safety and security of the above formulation were assessed by clinical examination, blood counts and biochemical measurements. As for anti-tick activity, two tests were conducted, using an experimentally infected cattle and other cattle naturally infested by Rhipicephalus (Boophilus) microplus. In two other studies evaluated the efficacy against Haematobia irritans, against larvae of Dermatobia hominis in cattle naturally infected. The anthelmintic activity of the association was evaluated in 12 animals necropsied, who suffered from gastrointestinal nematodiosis (natural infection). To determine the remaining waste in tissues of cattle treated 26 animals were autopsied. No significant change (P ≥ 0.05) was observed in blood parameters (blood counts) and biochemical measured in cattle treated with the new association, until the 10th day post-treatment. The effectiveness of combination anti-ticks fluazuron 3.0% + abamectin 0.5%* (Stall test) reached peak levels of efficiency exceeding 80.00% in 78 experimental days. The fluazuron 2.5%** reached efficiency above 80.00% in 77 dates. Maximum efficiency (100.00%) was achieved in 40 experimental days by the pool fluazuron 3.0% + abamectin 0.5%*, while the fluazuron 2.5%** this index reached in 51 dates. Statistically formulations fluazuron 3.0% + abamectin 0.5%* and fluazuron 2.5%** significantly reduced (P <0.05) scores of female R. (B.) microplus (detached) in the control group, the 5th to 100th DPT and 14th to 100th DPT, respectively.
