Factors Released From Embryonic Stem Cells Inhibit Apoptosis in h9c2 Cells Through PI3K/Akt but Not ERK Pathway

We recently reported that embryonic stem cells-conditioned medium (ES-CM) contains antiapoptotic factors that inhibit apoptosis in the cardiac myoblast H9c2 cells. However, the mechanisms of inhibited apoptosis remain elusive. In this report, we provide evidence for the novel mechanisms involved in the inhibition of apoptosis provided by ES-CM. ES-CM from mouse ES cells was generated. Apoptosis was induced after exposure with H2O2 (400 μm) in H9c2 cells followed by the replacement with ES-CM or culture medium. H9c2 cells treated with H2O2 were exposed to ES-CM, and ES-CM plus cell survival protein phosphatidylinositol 3-kinase/Akt inhibitor, LY-294002, or extracellular signal-regulated kinase (ERK1/2) inhibitor, PD-98050. After 24 h, H9c2 cells treated with ES-CM demonstrated a significant increase in cell survival. ES-CM significantly inhibited (P < 0.05) apoptosis determined by terminal deoxynucleotidyl transferase dUTP-mediated nickend labeling staining, apoptotic ELISA, and caspase-3 activity. Importantly, enhanced cell survival and inhibited apoptosis with ES-CM was abolished with LY-294002. In contrast, PD-98050 shows no effect on ES-CM-increased cell survival. Furthermore, H2O2-induced apoptosis is associated with decreased levels of phosphorylated (p)Akt activity. Following treatment with ES-CM, we observed a decrease in apoptosis with an increase in pAkt, and the increased activity was attenuated with the Akt inhibitor, suggesting that the Akt pathway is involved in the decreased apoptosis and cell survival provided by ES-CM. In contrast, we observed no change in ES-CM-decreased apoptosis or pERK with PD-98050. In conclusion, we suggest that ES-CM inhibited apoptosis and is mediated by Akt but not the ERK pathway.

Les adaptations cellulaires et moléculaires induites par les drogues jouent un rôle central dans les altérations comportementales à long terme observées dans l’addiction. Cette étude s’inscrit dans une démarche de compréhension des processus cellulaires rapidement mis en jeu par la cocaïne et susceptibles d’impacter durablement le fonctionnement neuronal et les comportements. La protéine Arc joue un rôle clé dans l’établissement de la plasticité synaptique à long-terme et la consolidation de la mémoire. Cette étude visait à caractériser l’induction de Arc dans le striatum en réponse à la cocaïne et d’analyser son rôle dans les réponses moléculaires et comportementales qu’elle induit. Notre étude a montré que l’expression de Arc est augmentée rapidement et transitoirement dans le striatum après une injection de cocaïne sous la dépendance de l’activation de la voie ERK. Nous montrons que la cocaïne induit une forte accumulation de la protéine Arc dans le noyau des neurones striataux où Arc se localise dans des zones actives de transcription, à proximité des histones H3 phosphorylées. In vitro, la surexpression de Arc diminue la phosphorylation des histones H3 induite par le glutamate indiquant qu’elle altère le remodelage de la chromatine. L’invalidation génétique de la protéine in vivo dans un modèle de souris transgénique conduit à une décompaction de la chromatine associée à une augmentation de l’activité de la RNA Polymerase II démontrant que Arc exerce un effet répresseur sur les mécanismes transcriptionnels. La perte totale d’expression de Arc favorise le développement d’altérations comportementales à long terme chez des animaux exposés à de faibles doses de cocaïne. / Molecular and cellular adaptations induced by drugs of abuse in the reward system play a key role in long-term behavioral alterations encountered in addiction. This work falls within an approach of understanding the cellular processes rapidly engaged by cocaine that could underlie the persistent alteration of neuronal physiology and behaviors. Arc protein is a major player in neuronal plasticity. Arc is induced in many behavioral paradigms and is essential for long-term synaptic plasticity and memory consolidation. The aim of this study was to characterize the profile and modality of Arc induction within the mouse striatum in response to cocaine administration. Our study shows that Arc expression is rapidly and transiently increased in the striatum after acute cocaine in an ERK-dependent fashion. This work revealed that cocaine-induced Arc protein rapidly and transiently accumulates in the nucleus of striatal neurons. In the nucleus, Arc is preferentially expressed in active transcription regions and localizes at the vicinity of phosphorylated histones H3. In vitro Arc overexpression decreased glutamate-induced Histones H3 phosphorylation showing that Arc interferes with activity-dependent chromatin remodeling. In vivo genetic invalidation of Arc expression in a transgenic mouse model was associated with a decreased chromatin compaction and increased RNA Polymerase II activity suggesting a repressive role of Arc on transcriptional mechanisms. Total Arc loss of expression leads to increased sensitivity to cocaine and promotes long-term behavioral alterations induced by low doses of cocaine.

Role of differential heparan sulphate sulphation in Fgf/Erk signalling during mouse telencephalic development

Heparan sulphate proteoglycans (HSPGs) are cell surface/secreted molecules expressed by all cells. HSPGs consist of carbohydrate side-chains attached to a core protein and are involved in regulating key signalling pathways in the developing mammalian brain via sugar-protein interactions. It has been hypothesized, in the ‘heparan sulphate (HS) code hypothesis’, that the specificity for the interaction between the HSPGs and particular signalling pathways is encoded by its HS side-chain. HS has an enormous variety of structures due to postsynthetic modification. Hs2st and Hs6st1 are enzymes involved in generating different HS structures by sulphating the 2-carbon or 6-carbon molecule of the sugar backbone respectively. Fibroblast growth factors (Fgfs) are a family of signalling molecules crucial for forebrain development. Some of its members such as Fgf8 are morphogens which pattern the forebrain via regulated gradient formation while others such as Fgf2 drive neurogenesis and cell proliferation. One of the main molecular consequences of Fgf signalling is activation of extracellular signal-regulated kinase (Erk) where the activation of Erk then drives developmental events such as neurogenesis or cell migration. Based on previous studies on the HS code hypothesis, we hypothesized that differential sulphation regulates Fgf signalling in a specific manner depending on the HS sulphation pattern. We performed binding assays on Hs2st-/- mice to ascertain the molecular mechanism behind the role of differential sulphation in Erk signalling through Fgf2 in the forebrain. We found that differential sulphation also has an important role to play in regionally targeting Fgf2/Erk signalling through regulating the formation of active signalling complexes. Studying the Fgf8/Erk signalling axis at E14.5 developing mouse corticoseptal boundary (CSB) revealed increased Fgf8 levels and Erk hyperactivation in both Hs2st and Hs6st1 null mutants. The dysregulation of Fgf8/Erk signalling at the CSB also highly correlates with the high expression of Hs2st and Hs6st1 at the CSB. A closer look into the molecular phenotypes of Hs2st-/- and Hs6st1-/- CSB revealed differences between them in which Hs6st1-/- CSB has higher Fgf8 levels compared to Hs2st-/- CSB. To elucidate the mechanisms underlying Hs2st and Hs6st1 role at the CSB, we investigated the formation and interpretation of Fgf8/Erk signalling gradient using Fgf8 bead assays in mice with Hs2st and Hs6st1 loss of function throughout development. We found that differential sulphation has a complex effect on Fgf8 gradient formation and interpretation in the forebrain in which Hs2st acts to stabilise the Fgf8 distribution through regulating Fgf8 levels through time while Hs6st1 acts to stabilise the Fgf8 distribution by maintaining the shape of the Fgf8 gradient through restricting Fgf8 levels during the formation of the Fgf8 distribution. In addition, we found Hs2st and Hs6st1 both function to increase the sensitivity of the CSB to Fgf8 for an Erk response although through different modes of action. Therefore, we conclude that differential HS sulphation plays a specific role in Fgf/Erk signalling depending on the HS sulphation pattern.

The Epidermal Growth Factor Receptor (EGFR) Is Proteolytically Modified by the Matriptase-Prostasin Serine Protease Cascade in Cultured Epithelial Cells

Prostasin is expressed at the apical surface of normal epithelial cells and suppresses in vitro invasion of cancer cells. Prostasin re-expression in the PC-3 prostate carcinoma cells down-regulated the epidermal growth factor receptor (EGFR) protein expression and EGF-induced phosphorylation of the extracellular signal-regulated kinases (Erk1/2). We report here that prostasin and its activating enzyme matriptase are capable of inducing proteolytic cleavages in the EGFR extracellular domain (ECD) when co-expressed in the FT-293 cells, generating two amino-terminally truncated fragments EGFR135 and EGFR110, at 135 and 110 kDa. Prostasin's role in EGFR cleavage is dependent on the serine active-site but not the GPI-anchor. The modifications of EGFR were confirmed to be on the primary structure by deglycosylation. EGFR135 and EGFR110 are not responsive to EGF stimulation, indicating loss of the ligand-binding domains. EGFR110 is constitutively phosphorylated and in its presence Erk1/2 phosphorylation is increased in the absence of EGF. The protease-induced EGFR cleavages are not dependent on EGFR phosphorylation. The EGFR ECD proteolytic modification by matriptase-prostasin is also observed in the BEAS-2B normal lung epithelial cells, the BPH-1 benign prostate hyperplasia and the MDA-MB-231 breast cancer cell lines; and represents a novel mechanism for epithelial cells to modulate EGF-EGFR signaling.

Intracellular Signaling Contributions to Behaviors Relevant to Nicotine Addiction

Nicotine is the primary addictive substance in tobacco, and most smokers who quit will relapse within a year. Evidence shows that cigarette craving increases over time, termed “incubation.” The purpose of these studies was to see if protracted abstinence from chronic nicotine increases rat self-administration, an animal model with good face validity for human tobacco use, and if nicotine self-administration during daily exposure/after 8+ days of abstinence is regulated by extracellular signal-regulated kinase (ERK) signaling in the nucleus accumbens (NAc) shell or anterior cingulate cortex (PFC). ERK kinase inhibitor U0126 was infused in the NAc shell or PFC of Long Evans rats immediately prior to daily self-administration sessions and following 8+ days of abstinence. U0126 in the PFC decreased responding for nicotine during daily sessions. Following 8+ days of abstinence, animals showed a robust increase in responding for nicotine, blocked by U0126 in the NAc shell, but not the PFC. Western blots revealed that nicotine treatment decreased levels of a substrate of ERK, ribosomal s6 kinase (RSK), in the NAc shell and increased it in the PFC, which occurred independent of abstinence period. In contrast, levels of RSK were increased in the NAc shell following a nicotine challenge during the abstinence period. In summary, our data show that the ERK signaling pathway plays a vital role in nicotine addiction during daily nicotine exposure and following periods of abstinence.

Role of Extracellular-signal Regulated Kinase (ERK) and cAMP Response Element Binding Protein (CREB) in the Incubation of Nicotine Craving

Nicotine Addiction is a chronic relapsing disorder. Relapse risk persists despite years of abstinence. Drug-associated cues have been demonstrated to induce craving and provoke relapse. Surprisingly, in human smokers, craving for nicotine increases or “incubates” with longer abstinence durations, a phenomenon that may explain persistent relapse liability. This incubation phenomenon also presents in animals trained to intravenously self-administer nicotine though the underlying mechanisms are unclear. Two proteins, ERK (Extra-cellular signal Regulated Kinase) and CREB (cAMP Response Element Binding protein) play important roles in learning, memory, and numerous aspects of drug addiction. We therefore examined whether changes in these proteins are associated with incubation of craving for nicotine in rats. We found increased nicotine-seeking behaviour after 14 days of abstinence (compared to 1 day) along with elevated ERK and CREB activity in the Accumbens brain region suggesting that these proteins may be involved in the incubation phenomenon.
