It also seeks to quantify the expression of messenger RNA for BMP-6 and BMP-7 in goat preantral follicles and to evaluate the effects of FSH, BMP-6 and BMP-7 on growth and gene expression in secondary follicles goats after 6 days of culture. To evaluate the stability of reference genes, secondary follicles (150-200 μm) were mechanically isolated from goat ovaries. After extraction of total RNA and synthesis of complementary DNA, the quantification of mRNA was carried out by real-time PCR using specific primers for genes of reference (GAPDH, β-tubulin, β-actin, PGK, UBQ, RPL - 19, rRNA18S), growth factors (GDF-9, BMP-15, BMP-6, FGF-2, VEGF, KL and IGF-1) and hormone receptor (FSH-R, LH-R and GH-R). To evaluate expression of BMP-6 and BMP-7, primordial follicles, primary and secondary as well as small and large antral follicles were obtained and the mRNA levels of BMP-6 and 7 were measured. For in vitro studies, the effects of BMP-6 (50 ng / mL) and BMP-7 (50 ng / mL) in the presence or absence of FSH (50 ng / mL) on the development of secondary follicles and on the expression of mRNA for BMP-6 and 7 and FSH-R was evaluated after 6 days of culture. Results showed that mRNA for growth factors (EGF, GDF-9, BMP-15, VEGF, FGF-2, BMP-6, IGF-1 and KL) and the receptors for FSH, LH and GH are expressed in at different levels in preantral follicles of goats. Moreover, among the growth factors studied, IGF-1 and EGF had higher and lower levels or RNA, respectively. UBQ and β-actin genes were the mostr stable reference in fresh and cultured preantral follicles. The level of mRNA for BMP-6 in primary and secondary follicles was significantly higher than those in primordial follicles, while the levels of mRNA for BMP-7 was higher in granulosa cells / theca in large antral follicles than in small antral follicles. After culture of secondary follicles for 6 days, FSH increased follicular diameter and FSH and BMP-7 significantly increased the levels of mRNA for BMP- 7 and FSH-R. In addition, BMP-6 in the presence or absence of FSH increased the diameter of secondary follicles after 6 days of culture. Real-time PCR shoed that FSH increased the levels of mRNA for BMP-6, while both BMP-6 and FSH and increased levels of mRNA for FSH-R, after a period of 6 days of culture. In conclusion, UBQ and β-actin are the two most stable genes for secondary follicles goats and FSH and BMPs of types 6 and 7 stimulate the growth of preantral follicles after 6 days of in vitro culture. / Este trabalho tem como objetivo avaliar a estabilidade de genes de referência e a expressão de fatores de crescimento e de receptores de hormônios em folículos secundários de caprinos. Além disso, visa quantificar a expressão dos RNA mensageiros para BMP-6 e BMP-7 em folículos pré-antrais e antrais caprinos e avaliar os efeitos de FSH, BMP-6 e BMP-7 sobre o crescimento e expressão gênica em folículos secundários caprinos durante 6 dias de cultivo. Para avaliar a estabilidade dos genes de referência, folículos secundários (150-200 µm) foram isolados mecanicamente de ovários caprinos. Após a extração do RNA total e síntese de DNA complementar, realizou a quantificação dos mRNA, por PCR em tempo real, utilizando-se primers específicos para genes de referência (GAPDH, β-tubulina, β-actina, PGK, UBQ, RPL-19, rRNA18S), fatores de crescimento (GDF-9, BMP-15, BMP-6, FGF-2, VEGF, KL e IGF-1) e receptores de hormônio (FSH-R, LH-R e GH-R). Para avaliar a expressão de BMP-6 e BMP-7, folículos primordiais, primários e secundários, bem como pequenos e grandes folículos antrais foram obtidos e os níveis de mRNA de BMP-6 e 7 foram quantificados. Nos estudos in vitro, os efeitos da BMP-6 (50 ng/mL) e BMP-7 (50 ng/mL) na presença ou ausência de FSH (50 ng/mL) sobre o desenvolvimento in vitro de folículos secundários e sobre a expressão de mRNA para BMP-6 e 7 e FSH-R foi avaliada após 6 dias de cultivo. Os resultados mostraram que UBQ e β-actina são os genes de referência mais estáveis em folículos pré-antrais caprinos fresco cultivados por 12 dias. Os RNAs mensageiros para os fatores de crescimento (EGF, GDF-9, BMP-15, VEGF, FGF-2, BMP-6, IGF-1 e KL) e os receptores de FSH, LH e GH são expressos em diferentes níveis em folículos pré-antrais de caprinos, sendo que o IGF-1 e o EGF apresentaram, respectivamente, o maior e o menor nível de mRNA. O nível de mRNA para BMP-6 em folículos primários e secundários foi significativamente maior do que aqueles em primordial, enquanto que os níveis de mRNA para BMP-7 foi maior nas células da granulosa/teca de grandes do que nos pequenos folículos antrais. Após o cultivo de folículos secundários durante 6 dias, FSH aumentou o diâmetro folicular e FSH e BMP-7 aumentou significativamente os níveis de mRNA para BMP-7 e FSH-R. Já a BMP-6 na presença ou ausência de FSH aumentou o diâmetro dos folículos secundários. Além disso, FSH aumentou os níveis de mRNA para BMP-6, enquanto ambos BMP-6 e FSH e aumentaram os níveis de mRNA para FSH-R, após o período de cultivo. Em conclusão, UBQ e β-actina são os dois genes mais estáveis para folículos secundários caprinos e o FSH e as BMPs dos tipos 6 e 7 estimulam o crescimento de folículos pré-antrais in vitro durante 6 dias de cultivo.

The dissertation was divided into two experimental phases. At first, it was performed solid surface vitrification (SSV) using different concentrations of cryoprotectant agents (CPAs) in which the effects of the 3 and 6 M concentrations of dimethylsufoxide (DMSO) and ethylene glycol (EG) were verified, as well as the association of both CPAs in the high concentration (6 M) under morphology, viability and apoptosis cell on the in situ PFs. A total of 865 PFs was analyzed before and after vitrification, it was observed an average of 80.7 ± 5.21% of morphologically normal follicles in the control group and after SSV, indifferent the CPA used, it was possible to preserve until 76.7% ± 5.4 OF PFs. At viability analysis, DMSO 3 M, DMSO 6 M, EG 3, EG 6 M (70.0%, 81.11%, 76.6% and 71.11%, respectively) presented similar values to the control group (79.0%). No apoptotic cells (TUNEL positive) were found before and after vitrification. At second, vitrification was performed using the association of CPAs, followed by xenografting of ovarian tissue in C57Bl/6 SCID Black mice. Through vaginal washing monitoring, was observed that 80% mice of the xeno-fresh group and 42% of the xeno-vitrified group returned to ovarian activity, confirmed by hormonal measures. Microscopically, primordial, primary and transitional follicles were observed in the grafts, and all had normal morphology for the species studied. However, major primordial and primary follicles were observed in transplants. The NORs revealed that after transplantation a significant reduction (1.66 ± 0.25) occurred when compared to the control groups (group fresh control: 7.19 ± 1.23 and xeno-fresh group: 9.10 ± 0.64). Apoptotic cells (TUNEL positive) were found only after transplantation of samples vitrified and the healthy follicles (TUNEL negative) observed in other groups (TUNEL negative). Thus, as the general conclusion, the use of MOIFOPA in agouti allowed the knowledge of aspects related to its reproductive morphology and physiology, enabling the germplasm conservation, with the possibility of germplasm bank formation, as the elucidation of mechanisms related to the PF survive and in vivo development / O objetivo do presente estudo foi utilizar a manipulação de oócitos inclusos em folículos ovarianos pré antrais (MOIFOPA) como ferramenta para o resgate e conservação do uso de gametas femininos de cutias (Dasyprocta leporina). A dissertação foi dividida em duas fases experimentais. Na primeira, foi realizada a vitrificação em superfície sólida (SSV) utilizando diferentes concentrações de agentes crioprotetores (ACPs), na qual foram verificados os efeitos das concentrações de 3 e 6 M de dimetilsufoxido (DMSO) e etilenoglicol (EG), bem como a associação de ambos os ACPs na concentração maior (6 M) sob a morfologia, viabilidade e apoptose celular de folículos ovarianos pré-antrais in situ (FOPAs). Um total de 865 FOPAs foi analisado antes e após a vitrificação. No grupo controle, foi observado 80,7 ± 5,21% de FOPA morfologicamente normais. Após SSV, independentemente do ACP utilizado, foram obtidos até 76,7% ± 5,4 de FOPAS. Na análise de viabilidade, DMSO 3 M, DMSO 6 M, EG 3, EG 6 M (70,0%; 81,11%; 76,6% e 71,11%; respectivamente) apresentaram valores semelhantes de FOPAS viáveis ao grupo controle (79,0%). Na segunda fase, foi realizada a SSV utilizando a associação dos ACPs (DMSO e EG), seguido do xenotransplante de tecido ovariano de cutias em camundongas C57Bl/6 SCID. Através do monitoramento do lavado vaginal, observou-se que 80% das camundongas do grupo controle e 42% do grupo vitrificado retornaram à atividade ovariana, confirmada pela dosagem hormonal. Microscopicamente, folículos primordiais, primários, transição e secundários foram observados nos enxertos, e todos tinham morfologia normal para as espécies estudadas. No entanto, os folículos primordiais e primários foram observados em maior quantidade após transplante. As Regiões organizadoras de nucléolos (NORs) revelaram que após o transplante ocorreu uma redução significativa de NORs no grupo vitrificado-xenotranplantado (1,66 ± 0,25) quando comparados aos grupos controles (grupo controle fresco: 7,19 ± 1,23; grupo controle xenotransplantado: 9,10 ± 0,64). As células apoptóticas (TUNEL positivo) foram encontradas somente após o transplante das amostras vitrificadas e folículos saudáveis foram encontrados nos outros grupos tratado (TUNEL negativo). Assim, como conclusão geral, o uso da MOIFOPA em cutias permitiu o conhecimento de aspectos relacionados a sua morfofisiologia reprodutiva, possibilitando tanto a conservação do material genético, com a possibilidade de formação de bancos de germoplasma; e ainda a elucidação dos mecanismos relacionados à sobrevivência e ao desenvolvimento dos FOPA in vivo / 2017-05-12

Folículos ovarianos pré-antrais bovinos: cultivo in vitro e xenotransplante

Para tanto, um total de 41 ovários de fetos bovinos foram obtidos em matadouro, transportados e utilizados para o cultivo in vitro (n=20) e para o xenotransplante (n=21). Após processados no laboratório em fragmentos entre 0,5 e 1 mm3 foram encaminhados para os cultivos O cultivo in vitro baseou-se em protocolo bem sucedido de cultivo de FOPA em caprinos e testou diferentes fontes de macromoléculas e a utilização do azul de tripan na viabilidade dos tecidos cultivados a uma atmosfera controlada de 5% CO2 em ar, a 38,5°C e nutridos com dMEM (300 mOsm/L, pH 7,2) suplementado com antibióticos, ITS, piruvato de sódio, glutamina, hipoxantina, dAMPc, bFSH e IGF-I. A depender do tratamento, foi adicionado BSA (0,1%) SFB (10%) ou PVA (1%). O cultivo in vivo de FOPA foi executado por xenotransplante sob a cápsula renal num total de 65 camundongas imunodeficientes. Desenvolveu-se uma técnica de biopsia e verificou-se o efeito do tempo de transplante (30, 60 e 30 e 60 dias após o transplante) sobre a percentagem e a viabilidade de FOPA bem como a possível presença de folículos antrais. Num segundo momento, 32 receptoras receberam estímulo hormonal de 10 UI de eCG (n=18) e 10 UI r-hFSH (n=14). Os resultados mostraram que o cultivo com PVA apresentou FOPA normais em percentagem semelhante aos cultivos com BSA e PVA. Quanto ao cultivo por xenotransplante, observou-se o crescimento sucessivo de FOPA até estádios antrais ao longo do tempo de transplante (> 30 dias). O resultado das estimulações exógenas apresentou folículos antrais com oócitos que apresentaram o cumulus expandido em 2/5 (40%) dos oócitos selecionados para MIV de cada um dos tratamentos propostos. Concluindo, FOPA oriundos de fetos bovinos podem ser cultivados por pelo menos 8 dias em PVA... / This study aimed to evaluate different protocols of in vitro and in vivo preantral follicles (PFs) culture from bovine fetus. Thus, a total of 41 fetal bovine ovaries, from a slaughterhouse, were collected and transported at laboratory for in vitro culture (n=20) and for xenotransplantation (n=21), after processing into small cortical pieces measuring between 0,5 and 1mm3 the slices were cultured. The in vitro culture was based on well successful protocol of preantral follicles in caprine and tested different sources of macromolecules and trypan blue viability of cultured tissues cultured at controlled atmosphere (5%CO2 in air, 39°C). The culture medium used was dMEM (300 mOsm/L, pH 7, 2) supplemented with antibiotics, ITS, sodium pyruvate, glutamine, hypoxanthine, dAMPc, bFSH, and IGF-I. Depending of treatment, was added BSA (0,1%), FCS (10%) or PVA (1%). In vivo culture of preantral follicles was carried out by xenotransplantation under renal capsule of immunodeficient females mice (total of 65) that were submitted to a biopsy technique previously developed for tissue collection and to verify the effectiveness of time of transplantation (30, 60 and 30 and 60 days post surgery) under percentage and viability as well as putative growth of PFs to antral follicles. At second stage, 32 recipient mice were submitted to hormonal stimuli with 10 IU of eCG (n=18) and 10 IU of r-hFSH (n=14). The results showed that PVA culture presented normally PF in similar distribution when compared with BSA and PVA culture. Regarding xenotransplantation, successive growth of PF until antral stages was observed belong time of transplantation. Exogenous stimulation presented oocytes with expanded cumulus in 2/5 (40%) of selected oocytes for IVM from each treatment. Conclusively PFs from fetal bovine ovaries can be cultured at PVA at least 8 days and grows until antral stages after xenotransplantation procedures... Efeito da tiroxina e do hormônio folículo estimulante no desenvolvimento in vitro de folículos pré-antrais ovinos / Effects of thyroxin and follicle stimulating hormone on the in vitro development of ovine preantral follicles

Fragments of ovarian tissue were grown for 1 or 7 days in a situation with a minimum essential (α-MEM +) alone or containing thyroxine (10, 20, 50 µg/mL) in the presence or absence of rFSH (50 ng/mL). The ovarian tissue non-grown (fresh control) and grown in different treatments were processed for analysis using the classical histology technique. When the days of culture were compared to the non-cultivated control, on the first day was found that the treatment with α-MEM+, FSH, T20 and T50 showed follicular survival percentages similar to control. When the treatments have been compared between each other within the growing season was identified that only on the 7th day, the treatment containing T50 presents a percentage of normal follicles significantly superior to the treatment with T10F. Related to the follicular diameter, the follicle in culture has not grown, since all treatments have had a similar behavior when controlling non-grown and α- MEM+, except the T10 treatment. When comparing the treatments within each growing season, it has been observed that on the first day, treatments T50, T10F and T50F have showed a follicular diameter greater when compared to T10 treatment. Similar to what happenedto follicular growth, oocitário growth has not grown either. For oocitário diameter, on the first day, there was a reduction for T10 and T20F treatments when compared to non-grown. On the 7th day of grown, FSH, T10F and T50F treatments have reduced oocitário diameter, with a progression of culture from the first day to the 7th one and even when compared to control non-grown and α-MEM+. It was concluded that high T4 concentrations (50 g / ml) have provided promising about the survival and follicular development in vitro. The association of T4 with FSH, have not presented positive results in the survival and follicular development in vitro. Although it is known the thyroxine action on ovarian physiology, there is a need to verify its importance for the in vitro development of preantral sheep follicles. / O presente experimento foi desenvolvido a fim de se verificar o efeito da tiroxina na presença ou ausência do hormônio folículo estimulante (FSH) sobre a sobrevivência e o crescimento de folículos pré-antrais de ovinos. Fragmentos de tecido ovariano foram cultivados por 1 e 7 dias em meio essencial mínimo (α-MEM+) suplementado ou não com diferentes concentrações de tiroxina (10, 20, 50 µg/mL) na presença ou ausência de rFSH (50 ng/mL). O tecido ovariano não-cultivado (controle fresco) e os fragmentos cultivados no dia 1 e 7 nos diferentes tratamentos foram processados e analisados utilizando a técnica de histologia clássica. Quando os dias de cultivo foram comparados ao controle não-cultivado, somente no dia 1 foi verificado que os tratamentos contendo α- MEM+, FSH, T20 e T50 mostraram percentuais de sobrevivência folicular semelhantes ao controle. Quando os tratamentos foram comparados entre si dentro de cada período de cultivo, foi observado que apenas no dia 7, o tratamento contendo T50 apresentou um percentual de folículos normais superior ao tratamento T10F. Com relação ao diâmetro folicular, não houve crescimento dos folículos em cultivo, uma vez que todos os tratamentos comportaram-se de maneira semelhante ao controle não-cultivado e ao α- MEM+, exceto o tratamento T10. Ao se comparar os tratamentos, dentro de cada período de cultivo, foi observado que no dia 1 os tratamentos T50, T10F e T50F apresentaram diâmetro folicular superior quando comparados ao tratamento T10. Semelhante ao crescimento folicular também não houve crescimento oocitário. Para o diâmetro oocitário, no dia 1, foi verificada uma redução para os tratamentos T10 e T20F quando comparados ao não cultivado. Já no dia 7 de cultivo os tratamentos FSH, T10F e T50F reduziram o diâmetro oocitário, com a progressão do cultivo do dia 1 para o dia 7 e ainda quando comparados ao controle não-cultivado e ao α-MEM+. Concluiu-se que altas concentrações de T4 (50 μg/ml) proporcionaram resultados promissores no que diz respeito à sobrevivência e desenvolvimento folicular in vitro. Quanto a associação da T4 com o FSH não demonstrou resultados positivos na sobrevivência e no desenvolvimento folicular in vitro. Embora seja conhecida a atuação da tiroxina na fisiologia ovariana são necessárias mais pesquisas que verifiquem sua importância para o desenvolvimento in vitro de folículos pré-antrais ovinos.

Two experiments were conducted. In experiment I, the morphology of FOPAs after cooling followed by vitrification was studied. Ovaries were collected (n=10) from five crossbred heifers aged of 14 to 16 months. At the laboratory 22 ovarian fragments were taken from the cortical region and distributed as followed: two fragments for fresh control (zero hour) submitted to histological analysis and 20 fragments cooled at 4 ºC for four and 24 hours in TCM-199+HEPES+Antibiotics medium. From the 20 fragments cooled, four were fixed as control 4 and 24 hours (two piece each) and the remaining 16 fragments were distributed in four vitrification treatments for each cooling time, respectively: Treatments V4a and V24a: TCM-199 + dimethyl sulfoxide (DMSO) 1.5M + Ethylene glycol (EG) 1.5M; Treatments V4b and V24b: TCM-199 + DMSO 1.5M + EG 1.5M + sucrose (SUC) 0.5M; Treatments V4c and V24c: TCM-199 + DMSO 1.5M + EG 1.5M + Ascorbic Acid (AA) 0.1 mM/L and treatments V4d and V24d: TCM-199 + DMSO 1.5M + EG 1.5M + SUC 0.5M + AA 0.1mM/L. After being vitrified, fragments were stored in liquid nitrogen for three days. Fragments were then heated in solution of decreasing SUC and fixed for histology. In experiment II, the viability of FOPAs after cooling followed by vitrification by Trypan Blue staining (AT) was studied. Bovine ovaries (n = 10) were collected, fragmented and distributed as described in the first experiment, subsequently submitted to mechanical follicular isolation for analysis of viability. The morphology variable was evaluated by SNK test and viability was analyzed by Chi-square Test at 5% probability or the Fisher Exact Test when the number of repetitions was less than 30 follicles. There was no difference in percentage of morphologically normal follicles between fresh control group and control group cooled for 4 h (99.3% and 96.0% respectively; P>0.05), nevertheless, there was reduction of FOPAs morphological integrity of control cooled for 24 h (86%; P<0.05). After vitrification, there was reduction (P<0.05) of normal morphology in all treatments when compared with cooled control groups. On the other hand, the viability analysis showed that V4c treatment were not significantly different when compared with control 24h treatment and, together V24c treatment, showed higher capacity of morphological preservation of FOPAs than other treatments (P<0.05), indicating that AA can reduce the toxic and osmotic damages caused by cryopreservation procedure. In conclusion, FOPAs included in the ovarian bovine tissue conserve the morphology efficiently when cooled to 4 °C for 4 hours in TCM-199 + HEPES + Antibiotics and, the association of permeable cryoprotectants DMSO and EG with the antioxidant AA improves survival rates and maintain the morphologic integrity during the vitrification. / O presente trabalho teve por objetivo avaliar o efeito do resfriamento e da associação de crioprotetores penetrantes e não penetrante acrescido de ácido ascórbico na qualidade de folículos pré-antrais (FOPAs) bovinos vitrificados. Para isso, foram realizados dois experimentos. No experimento I, foi estudada a morfologia dos FOPAs após resfriamento seguido de vitrificação. Ovários (n=10) foram coletados de cinco novilhas mestiças de 14 a 16 meses de idade. No laboratório, 22 fragmentos ovarianos foram retirados da região cortical, os quais foram distribuídos: dois para controle fresco (zero hora), sendo imediatamente fixados para análise histológica, e 20 fragmentos resfriados à 4 ºC por quatro e 24 horas em meio TCM-199+HEPES e antibióticos. Dos 20 fragmentos resfriados, quatro foram fixados como controle 4 e 24 horas (dois fragmentos cada) e os 16 restantes foram distribuídos em quatro tratamentos de vitrificação para cada tempo de resfriamento, respectivamente: Tratamentos V4a e V24a: TCM-199 + Etilenoglicol (EG) 1,5M + Dimetilsulfoxido (DMSO) 1,5M; Tratamentos V4b e V24b: TCM-199 + DMSO 1,5M + EG 1,5M + sacarose (SAC) 0,5M; Tratamentos V4c e V24c: TCM-199 + DMSO 1,5M + EG 1,5M + Ácido Ascórbico (AA) 0,1 mM/L e tratamentos V4d e V24d: TCM-199 + DMSO 1,5M + EG 1,5M + SAC 0,5M + AA 0,1mM/L. Após a vitrificação, os fragmentos permaneceram armazenados em Nitrogênio Líquido por três dias, e posteriormente aquecidos em soluções decrescentes de SAC e fixados para histologia clássica. No experimento II, foi estudada a viabilidade dos FOPAs após resfriamento seguido de vitrificação pela coloração Azul de Trypan (AT). Ovários bovinos (n=10) foram coletados, fragmentados e distribuídos nos quatros tratamentos conforme descrito no experimento I, os quais foram posteriormente submetidos ao isolamento folicular mecânico para análise da viabilidade. A variável morfologia foi avaliada pelo Teste SNK e a viabilidade foi analisada pelo teste Qui-quadrado com nível de significância de 5%, ou pelo Teste Exato de Fisher quando o número de repetições foi menor que 30 folículos. Não houve diferença na percentagem de folículos morfologicamente normais entre o grupo controle fresco e o grupo controle 4 h (99,3% e 96,0% respectivamente; P>0,05). Entretanto, houve redução da integridade morfológica dos FOPAs após 24 h de resfriamento (controle 24 h: 86%; P<0,05). Após a vitrificação, houve uma redução da integridade morfológica e viabilidade folicular em todos os tratamentos em relação aos controles resfriados (P<0,05). No entanto, o tratamento V4c manteve a viabilidade folicular semelhante ao controle 24 h (P > 0.05). Este mesmo tratamento (V4c) mostrou maior capacidade de preservação em relação aos demais tratamentos de vitrificação (P<0,05). Em conclusão, FOPAs inclusos no tecido ovariano bovino conservam a morfologia eficientemente quando são resfriados a 4 ºC por até quatro horas em meio TCM-199+HEPES+Antibióticos. Além disso, a associação dos agentes crioprotetores penetrantes DMSO / EG, com o agente antioxidante AA melhora as taxas de sobrevivência e mantêm a integridade morfológica durante a vitrificação.

The mare’s ovarian function (e.g., folliculogenesis) has similarities with woman’s cycle and based on these aspects, this species can be considered an animal model for comparative studies and techniques with preantral follicles to preserve female fertility. The purpose of the first project was to determine methodologies that might speed up the process of histological evaluation of preantral follicle population. On the second project, we evaluated the follicular population features considering the follicular density factor by means of a mathematical model, to determine the appropriate experimental design to estimate the follicular density in ovarian fragments. The third project, analyzed the effects of reproductive phases, ovary, and ovarian structures on the quality, classification, follicular and cellular densities in ovarian fragments. The results allowed an establishment of the best thickness of histological sections and the ideal number of fragments and sections to estimate the equine preantral follicle density. Furthermore, it was observed a low follicular density associated with a high heterogeneity within and among animals. Moreover, we demonstrated the best scenario (diestrous phase with the presence of a functional corpus luteum) to collect ovarian fragments with higher follicular and cellular densities. In summary, this study demonstrated: (i) advance on the knowledge of equine follicular dynamic on the preantral phase; (ii) the concept of the mare as an animal model to study the reproductive challenges on woman; and (iii) provided basic information for future biotechnology research with the aim to preserve or improve the reproductive capability as cryopreservation, in vitro culture, and transplants of ovarian tissue. / Estudos com folículos pré-antrais equinos são escassos e as características da dinâmica folicular pré-antral permanecem pouco elucidadas. A função ovariana (ex. foliculogênese) da égua possui diversas similaridades com a mulher e, baseado nestes aspectos, podemos considerá-la como modelo animal para estudos comparativos e técnicas que utilizam folículos pré-antrais na preservação da fertilidade feminina. O primeiro projeto teve a proposta de definir metodologias para agilizar o processo de avaliação histológica da população folicular pré-antral. No segundo projeto, avaliou-se as características da população folicular considerando o fator densidade folicular, determinando por meio de modelo matemático, o apropriado desenho experimental para estimar a densidade em fragmentos ovarianos. No terceiro projeto, analisou-se os efeitos da fase reprodutiva, ovário e estruturas ovarianas sobre a qualidade, classificação, densidade folicular e celular em fragmentos ovarianos. Os resultados permitiram estabelecer a melhor espessura de secção histológica e o número de fragmentos e de secções suficientes para estimar a densidade de folículos pré-antrais equinos. Adicionalmente, observou-se uma baixa densidade folicular associada a alta heterogeneidade dentre e entre os animais. Além disso, determinou-se afase de diestro com a presença de corpo lúteo ativo como o melhor cenáriopara a coleta de fragmentos ovarianos com maior densidade folicular e celular. Finalmente, dos resultados originados deste trabalho, pode-se concluir: (i) obteve-se avanço no conhecimento da dinâmica folicular pré-antral equina; (ii) o conceito da égua como modelo animal para estudos de desafios reprodutivos na mulher foi reforçado; e (iii) ocorreu a geração de informações básicas para futuras pesquisas com biotecnologias que visam preservar ou implementar a capacidade reprodutiva como a criopreservação, cultivo in vitro e transplantes de tecido ovariano.

Pair of ovaries from 6 females were collected. A half of one ovary was designated to the estimative and characterization of the follicle population, and it was immediately submitted to histology and transmission electron microscopy (TEM). The other ovary was divided into 9 fragments and one of it was immediately submitted to histology procedures, the other fragments were stored either in phosphate-buffered saline (PBS; control group) or ACP medium in isothermal Styrofoam boxes containing biological ice packs. The boxes were sealed and opened only after 12, 24, or 36 h. After opening each box, the ovarian fragments were submitted to histological analysis and TEM. The temperature into the boxes and pH of the medium were checked. A total of 33273.45 ± 3019.30 preantral follicles (PF) were estimated for each ovary. Most of them was morphologically normal (94.4%) and just a few was atretic (5.6%). At histological analysis, a lot of lipids droplets was observed at oocyte cytoplasm, which was confirmed by TEM. About short term storage, it was observed that the follicular integrity was kept until 4 h, using ACP medium. The medium pH was kept constant. This work shows, for the first time, the estimative of the PF population of the ovary, totally and in each follicle category, and it´s characterization in collared peccaries (Tayassu tajacu). It could be concluded that the ACP medium may be used as an alternative medium for the short term preservation of PF in that specie. / Foram realizados a estimativa da população folicular ovariana de catetos, e ainda a refrigeração dos mesmos utilizando PBS ou ACP por curtos períodos. Os pares de ovários de 6 catetos foram coletados. Metade de um dos ovários foi destinada a estimativa e caracterização da população folicular, sendo imediatamente submetida ao processamento para histologia clássica e microscopia eletrônica de transmissão (MET). O outro ovário foi dividido em 9 fragmentos e um deles foi imediatamente destinado à histologia, os demais foram colocados em tubos com solução salina fosfatada tamponada (PBS) ou em solução à base de água de coco em pó (ACP) e acondicionados em caixas isotérmicas de 5L. As caixas foram lacradas e abertas 4h,12h, 24h e 36h após, e os fragmentos ovarianos foram submetidos à histologia e processamento para MET. A temperatura das caixas e o pH do meio foram aferidos. Um total de 33273.45 ± 3019.30 folículos pré antrais (FOPA) foram estimados para a população em cada ovário. A maioria dos FOPA apresentava-se como morfologicamente normal (94.4%) e apenas poucos deles eram atrésicos (5.6%). Na análise histológica, uma grande quantidade de gotas de lipídios foi observada no citoplasma oocitário, os quais foram confirmados através da análise ultraestrutural. Quanto à refrigeração, verificou-se que a integridade folicular foi mantida até as 4 h utilizando-se ACP. O pH dos meios de conservação permaneceu constante. Este trabalho mostra pela primeira vez a estimativa da população de FOPA do ovário, total e por categoria, e também a caracterização destes folículos em catetos (Tayassu tajacu). E pôde-se concluir que a ACP® pode ser utilizada como um meio alternativo para conservação de folículos pré-antrais.
