Spider Aggregate Glue Sequence Characterization and ExpressionBerg, Kyle 01 May 2016 (has links)
Spider aggregate glue is secreted on to the webs of many spiders in the superfamily Araneoidea. Aggregate glue is sticky when wet and has a unique stress strain curve that varies depending on the relative humidity and speed of probe retraction. These features make it an attractive target for applications such as underwater adhesives or surgical glues. However, little is known about the genetic sequence of the glue protein itself. In this work, a method is identified to isolate the remainder of the aggregate gene sequence, and genetic constructs glue are created using known aggregate gene sequence. One construct is expressed in E. coli BL21 cells and the protein is tested for its adhesive properties.

It investigates using arches that increase and decrease in height and span width, while varying in their lean. When combined with a fabric enclosure, they create a roof that constantly changes. The hydrological modeling appears as a strong support engineering, however, is subject to a distinct set of constraints rarely explicitly considered by analysis of uncertainty to the results. The present study aims to highlight the role of hydrological modeling through a model transformation process for rainfall-runoff watershed stream Botafogo (Goiania - GO), called UFGModel1.0 and ascertain the uncertainty analysis applied in simulation flows. To achieve the goal, after exposure of the problem, we propose a methodology for uncertainty analysis using the Monte Carlo method, based on the method of estimation uncertainty GLUE. We stress the importance given in consideration of the precipitation fields, and the effect of simplifying the response of a model of a precipitation-runoff. The results demonstrate that multiple sets of parameters were considered excellent high efficiency enabling a model for events intense precipitation. Parameters studied concluded that the sealed area holds great sensitivity, significantly altering the design flow. / Os inúmeros problemas advindos do aumento da quantidade das áreas impermeabilizadas e do sistema de drenagem das águas pluviais defasado consistem em um árduo desafio para as gestões públicas. A modelagem hidrológica aparece como um forte suporte da engenharia, porém, está sujeita a um conjunto de restrições distintas, raramente consideradas explicitamente através da análise de incerteza associada aos resultados obtidos. O presente trabalho propõe-se a ressaltar o papel desempenhado pela modelagem hidrológica através de um modelo de transformação do processo precipitação-vazão para bacia hidrográfica do córrego Botafogo (Goiânia – GO), denominado de UFGModel1.0 e averiguar a análise de incerteza aplicada na simulação de vazões. Para o cumprimento do objetivo, após a exposição do problema, propõe-se uma metodologia de análise da incerteza através do método de Monte Carlo, baseada no método de estimação de incerteza GLUE. Ressalta-se a importância dada em consideração aos campos de precipitação, e ao efeito de sua simplificação na resposta de um modelo do tipo precipitação-vazão. Os resultados obtidos demonstram que vários conjuntos de parâmetros foram considerados ótimos possibilitando uma alta eficiência do modelo para eventos intensos de precipitação. O modelo apresentou maior sensibilidade ao parâmetro de área impermeabilizada e Manning do plano.

Soil formation and soil moisture dynamics in agriculture fields in the Mekong Delta, Vietnam conceptual and numerical modelsvan Quang, Pham January 2009 (has links)
<p>Previous studies of agricultural conditions in the Mekong Delta (MD) have identified soil compaction as an obstacle to sustainable production. A conceptual model for soil formation was presented to demonstrate the link between soil hydrology and plant response. Detailed studies of soil moisture dynamics in agricultural fields were conducted using a dynamic process-orientated model. Pressure head and water flow were simulated for three selected sites during a year for which empirical data were available. Daily meteorological data were used as dynamic input and measured pressure head was used to estimate parameter values that satisfied various acceptance criteria. The Generalised Likelihood Uncertainty Estimation (GLUE) approach was applied for calibration procedures with 10,000 runs, each run using random values within the chosen range of parameter values. To evaluate model performance and uncertainty estimation, re-sampling was carried out using coefficient of determination (R2) and mean error (ME) as the criteria. Correlations between parameters and R2 (and ME) and among parameters were also considered to analyse the relationship of the selected parameter set in response to increases/decreases in the acceptable simulations. The method was successful for two of the three sites, with many accepted simulations. For these sites, the uncertainty was reduced and it was possible to quantify the importance of the different parameters.</p><p> </p>

Comparação entre uso de cola biológica de fibrina e drenagem de aspiração pós-operatória na prevenção de hematoma e seroma em ritidoplastia : um estudo controlado, randomizado e duplo cegoRezende, Antonio Roberto da Rosa January 2018 (has links)
Este trabalho tem por objetivo comparar a eficácia da cola biológica de fibrina e da drenagem de aspiração pós-operatória na prevenção de complicações cirúrgicas como hematoma e seroma após a realização de ritidoplastia pela equipe da Clínica Rezende no Hospital Moinhos de Vento, na cidade de Porto Alegre/RS. Realizou-se estudo prospectivo, controlado, randomizado e duplo cego. Foram analisadas 72 pacientes, divididas em dois grupos de 36, sendo que no grupo dreno utilizou-se drenagem de aspiração e no grupo cola utilizou-se cola de fibrina. Quarenta e oito horas após a realização dos procedimentos, todas as pacientes foram submetidas a aferição ecográfica da lâmina de exsudato presente sob os retalhos cutâneos da face. O volume total médio foi de 3,21 mL no grupo dreno e 1,02 mL no grupo cola, com magnitude de efeito de 68,1% e intervalo de confiança de 55,3 a 77,2 e p <0,001. Com esses resultados, comprovou-se que a cola apresenta eficácia significativa, demonstrando que sua utilização é 68,1 % mais efetiva que a drenagem de aspiração na prevenção de hematomas ou seromas em ritidoplastia. / This study aimed to compare the efficacy of fibrin glue and suction drainage in preventing postoperative complications such as hematoma and seroma following rhytidoplasties conducted by the staff of Clínica Rezende at Hospital Moinhos de Vento in Porto Alegre, Brazil. A prospective, controlled, randomized, double-blind trial was conducted. The 72 patients assessed in the study were divided into two groups of 36 each, one treated with suction drainage and other with fibrin glue. Forty-eight hours after the procedures, all patients underwent ultrasound evaluation of the volume of exudate under facial skin flaps. The average volume of exudate was 3.21 mL in the drainage group and 1.02 mL in the fibrin glue group, with a size effect of 68.1%, 95% confidence interval of 55.3 to 77.2, and p <0.001. The results of this investigation significant favor the use of fibrin glue, showing that was 68.1 % more effective than suction drainage in preventing hematoma or seroma following rhytidoplasty.

The effect of storage time on the platelet concentration of Choukroun's platelet rich fibrin (PRF)Peck, M. Thabit January 2011 (has links)
Magister Chirurgiae Dentium (MChD) / Wound healing is a complex process characterised by the repair and reconstitution of lost or damaged tissue. By the mid 1990s, several methods were proposed to enhance wound healing of surgical sites by introducing high concentrations of human platelets to these areas. In the early 21st century, Choukroun et al (2006b) introduced a new type of platelet concentrate that was devoid of any additives, and required no specialised equipment for its production. This concentrate was termed Platelet-rich fibrin (PRF) and although various aspects of this biomaterial had been studied, very little is currently known about its storage properties. Aim: To determine whether storage time had a significant effect on the platelet concentration of Choukroun’s PRF Method: A total of 30 patients were enrolled into the study. Three blood samples of 10ml each were drawn from each patient. Two of the blood samples (Group A and Group B) were centrifuged to form PRF. The third sample was used to measure the baseline blood platelet concentration and was therefore not centrifuged. After PRF had formed in both test groups, it was removed from the test tubes at 2 different times i.e. immediately after centrifuge (Group A) or after 60 min of storage in the blood collecting tube (Group B). The remaining blood was then tested for platelet concentration and compared to each other and the baseline reading. Results: 14 males and 16 females participated in the study (average age 41.7 years). A mean blood platelet concentration of 282.8 ± 58.3 × 109/L was recorded for the baseline reading. Group A had a mean blood platelet concentration 7.9 ± 3.03 × 109/L. Group B had a mean blood platelet concentration of 4.0 ± 1.93 × 109/L. A statistically significant difference was seen between Groups A and B (p < 0.0001). Conclusions: Storage time has a significant effect of the platelet concentration of PRF. If stored over a period of 60 min, the platelet concentration of PRF increases. Further research is required to determine whether this finding is clinically significant.
