We present a multi-objective two-echelon vehicle routing problem with vehicle synchronization and "grey zone" customers arising in the context of urban freight deliveries. Inner-city center deliveries are performed by small vehicles due to access restrictions, while deliveries outside this area are carried out by conventional vehicles for economic reasons. Goods are transferred from the first to the second echelon by synchronized meetings between vehicles of the respective echelons. We investigate the assignment of customers to vehicles, i.e., to the first or second echelon, within a so-called "grey Zone" on the border of the inner city and the area around it. While doing this, the economic objective as well as negative external effects of transport, such as emissions and disturbance (negative impact on citizens due to noise and congestion), are taken into account to include objectives of companies as well as of citizens and municipal authorities. Our metaheuristic - a large neighborhood search embedded in a heuristic rectangle/cuboid splitting - addresses this problem efficiently. We investigate the impact of the free assignment of part of the customers ("grey Zone") to echelons and of three different city layouts on the solution. Computational results show that the impact of a "grey Zone" and thus the assignment of these customers to echelons depend significantly on the layout of a city. Potentially pareto-optimal solutions for two and three objectives are illustrated to efficiently support decision makers in sustainable city logistics planning processes.

Landscape genetic approaches allow to study effects of landscape to population microevolution. Landscape can influence gene flow even in large carnivores with good dispersal ability. Understanding the influence of landscape to the gene flow between populations is crucial for species conservation, especially in the species with low population densities. Aim of the study was to describe genetic structure of the grey wolf (Canis lupus) and snow leopard (Panthera uncia) in selected areas and to determine the influence of the landscape features on observed structure. Non-invasive genetic samples of snow leopard from Nepal were analysed, as well as invasive and non-invasive samples of grey wolf from Central Europe. Population structure was determined a posteriori using Bayesian clustering approaches that integrate genetic and geographical data, and compared to landscape connectivity models. Population structure of snow leopards is mostly influenced by human presence and presence of frequented roads, which represent a substantial dispersal barrier. Habitat suitable for this species is greatly restricted by altitude, however, during dispersal they are able to overcome areas with higher elevation than what is optimal for them. A doença de Parkinson (DP) é uma desordem neurodegenerativa caracterizada clinicamente por tremor, rigidez, acinesia (ou bradicinesia) e instabilidade postural. Patofisiologicamente, a DP é classificada como uma sinucleinopatia associada à perda de neurônios dopaminérgicos na substância negra (SN), mas outros neurônios do tronco encefálico podem estar degenerados na DP, contribuindo não só para as alterações motoras, mas também não motoras observadas. Dentre as alterações não motoras, as alterações respiratórias estão presentes tais como obstrução das vias aéreas superiores, pneumonia e ainda a apnéia obstrutiva do sono uma das principais causas de morte na DP. Os mecanismos que levam à degeneração de neurônios envolvidos no controle respiratório ainda não estão bem esclarecidos, mas dados recentes do nosso laboratório mostraram que no modelo de DP induzido pela injeção no caudado-putâmen (CPu) de 6-hidroxidopamina (6-OHDA), um agente neurotóxico seletivo para células catecolaminérgicas, observou-se intensa redução na frequência respiratória e ventilação basais e induzidas pela ativação do quimiorreflexo central por hipercapnia. Além disso, observou-se também intensa redução do número de neurônios bulbares envolvidos no controle neural da respiração, como os neurônios Phox2b+ da região do núcleo retrotrapezóide (RTN), que estão envolvidos com a inspiração e o quimiorreflexo central. Dessa forma, o objetivo do presente trabalho foi investigar se a existência de uma via entre os neurônios da SN e do RTN poderia ser responsável por essa neurodegeneração. Realizamos injeções de traçadores anterógrados e retrógrados na SN e no RTN de ratos para verificar a existência de projeções diretas entre essas regiões, entretanto observamos que não há projeções diretas entre a SN e o RTN, mas há projeções indiretas entre essas duas regiões, utilizando a Substância Cinzenta Periaquedutal (PAG) como região intermediária. Além disso, observamos que no modelo de DP induzido pela injeção bilateral de 6-OHDA no CPu ocorre uma redução no número de varicosidades catecolaminérgicas na PAG e de neurônios que são ativados pelo quimiorreflexo central que se projetam da PAG para o RTN. Nossos experimentos eletrofisiológicos mostraram que a inibição bilateral da PAG pela injeção de muscimol não gera alterações respiratórias basais como ocorre no modelo da DP; entretanto, nesses animais, pudemos também observar, apesar de ser menor do que ocorre com animais submetidos ao modelo da DP, inibição de alterações respiratórias induzidas por hipercapnia. Nossos dados anatômicos mostraram que a comunicação entre os neurônios da SN e do RTN envolve neurônios da PAG e que essa via pode estar reduzida no modelo da DP, o que pode contribuir para a redução de neurônios do RTN; e que a redução neuronal desta via pode alterar as respostas respiratórias frente à ativação do quimiorreflexo central. / Parkinson\'s disease (PD) is a neurodegenerative disorder characterized clinically by tremor, rigidity, akinesia (or bradykinesia) and postural instability. Pathophysiologically, PD is classified as a synucleinopathy predominantly associated with loss of dopaminergic neurons in the substantia nigra (SN), but other brainstem neurons may also be degenerate in PD, contributing not only to the motor but also non-motor alterations observed in this pathology. Among the non-motor changes observed respiratory changes are present and can be characterized as upper airway obstruction, pneumonia and obstructive sleep apnea are one of the main causes of death in PD. The mechanisms that lead to the degeneration of neurons involved in respiratory control are still not well understood but data in the literature have demonstrated the loss of receptors in a region considered to be the respiratory rate generator in postmortem brains of humans. In the model animal DP of 6-hydroxydopamine (6-OHDA), a selective neurotoxic agent for catecholaminergic cells, there was an intense reduction in basal respiratory rate and ventilation, in addition to a intense reduction of neurons involved in neural control of breathing: Phox2b+ neurons in the retrotrapezoid nucleus (RTN) region. Thus, the aim of the present study was to investigate whether the existence of a pathway between SN and RTN neurons could be responsible for this bulbar neurodegeneration. We performed experiments using the injection of anterograde and retrograde tracers in the SN and the RTN to verify the existence of direct projections between these regions in rats. However, our results showed that there are no direct projections between the SN and the RTN, but there are indirect projections between these two regions, using Periaquedutal Gray Substance (PAG) as the intermediate region. In addition, we observed that in the PD model induced by the bilateral injection of 6-OHDA in CPu, a reduction of the projections PAG neurons for RTN and that are activated by the central chemoreflex. Our electrophysiological experiments have shown that in the 6-OHDA PD model there is a reduction of the cardiorespiratory responses induced by the activation of the central chemoreflex, since the bilateral inhibition of the PG of control animals does not alter these cardiorespiratory responses. Therefore, our anatomical results showed that the communication between SN and RTN neurons involves PAG neurons and that this pathway may be reduced in the PD model, which may contribute to the reduction of RTN neurons; and that the neuronal reduction of this pathway may alter respiratory responses to activation of the central chemoreflex.

Is financial health a determinant of sport success?

The purpose of this study is to find the relationship between financial health in an ice hockey club and its sport success. The study answers the research question: How can financial health of Swedish ice hockey clubs be able to explain the sport success in the Swedish Hockey League? Based on the research question, the study uses the theory Benchmarking and a more specific benchmarking terminology called Financial benchmarking. The study selects eight financial variables in order to benchmark the icehockey clubs in the Swedish Hockey League (SHL). A particular methodology within financial benchmarking, called Grey Relational Analysis (GRA), is used in order to determine the financial health of the clubs in relation to each other and therefore be able to rank the clubs based on each individual variable. The same financial variables, with the addition of four non-financial variables and exclusion of two financial variables, are used in a selected Logistic Regression model to explain how the variables contribute to the sport success of the clubs. The main conclusions which can be drawn from the study are as follows: The variables Net sales and Net profit are the two only variables which are statistically significant and are able to contribute to sport success. Secondly, the club HV71 is overall the club with the most optimal financial health in SHL, among the 12 clubs investigated. Lastly, accounting trends within this industry affects the financial outcome and further how it explained sport success. Trends such as a minimal or no amount of long-term liabilities is common among the clubs, where instead the total amount of liabilities mainly consists of current liabilities. It can be further concluded that profitability, revenue and equity are financial corner stones in a hockey club which participates in SHL.

O medo e a ansiedade são emoções que apresentam claro valor adaptativo, e que tem suas origens nas reações de defesa que os animais exibem em resposta a situações de ameaça que podem comprometer sua integridade física ou sobrevivência. Recentes estudos têm indicado que a matéria cinzenta periaquedutal dorsal (MCPD) está envolvida na organização e expressão de comportamentos intempestivos do tipo fuga e luta, os quais são relacionados ao estado de medo, e também participa, juntamente com estruturas prosencefálicas (ex. córtex pré-frontal, sistema septo-hipocampal e amígdala), do controle de comportamentos defensivos mais elaborados e orientados relacionados à ansiedade. O presente estudo investigou a participação da MCPD na modulação de diferentes comportamentos defensivos (p. ex. fuga, esquiva e avaliação de risco) induzidos por métodos artificial (estimulação química) e naturalístico (exposição ao predador) em camundongos. Na primeira etapa, investigamos o padrão de resposta comportamental induzida pela infusão do ácido D,L-homocistéico (DLH, estímulo aversivo químico) na MCPD em diferentes situações ou ambientes, com e sem grande disponibilidade de espaço - o Mouse Defense Test Battery (MDTB) e a Arena (Experimento 1), respectivamente. Além disso, o presente estudo avaliou a habilidade dos animais de reagirem a estímulos aversivos (predador) durante o período inicial (nos 60 s iniciais) do efeito do ácido DLH (fuga explosiva) (Experimento 3), e imediatamente após esse período, no qual o animal apresente comportamento de congelamento ou imobilidade (Experimento 2). Nossos resultados indicaram que a fuga desencadeada pela estimulação química é a resposta predominante de camundongos e que sua exibição depende da disponibilidade de espaço, uma vez que a maioria dos saltos observados na arena está intimamente relacionada ao contato tátil do animal com as paredes do aparato. Esse perfil de respostas de fuga explosiva e saltos parece não representar o padrão comportamental defensivo natural, tal como acontece diante de uma ameaça proximal (ex. um predador), uma vez que durante a estimulação química os camundongos apresentaram um déficit na estratégia antipredador. A segunda etapa do estudo avaliou os efeitos da injeção intra-MCPD do agonista glutamatérgico ácido N-metil-D-aspártico (NMDA), do inibidor da enzima de síntese do óxido nítrico neuronial (NOSn), N?-propil-L-arginina (NPLA) (Experimento 4), e do agonista não seletivo de receptores do fator de liberação de corticotrofina (CRF), CRF ovino (oCRF) (Experimento 5), no comportamento defensivo de camundongos submetidos ao MDTB e ao teste de exposição ao rato (Rat Exposure Test; RET). Os resultados da segunda etapa demonstraram que a ativação de receptores NMDA na MCPD de camundongos intensifica comportamentos relacionados à esquiva do predador. De maneira interessante, essas alterações produzidas pelo NMDA foram consistentemente revertidas pelo inibidor da NOSn, previamente microinjetado no mesmo sítio. Além disso, efeitos intrínsecos do NPLA atenuaram as respostas de esquiva e de avaliação de risco em camundongos submetidos ao RET. Por fim, os resultados da segunda etapa também apontaram para um efeito proaversivo (nas respostas de salto e de esquiva) do agonista de receptores CRF, indicando uma participação dos sistemas glutamatérgico, nitrérgico e CRFérgico, localizados na MCPD, na modulação de diferentes estratégias defensivas (ex. esquiva, avaliação de risco e saltos) de camundongos submetidos ao confronto com o predador. Em conjunto, nossos resultados corroboram a hipótese de que a MPCD está envolvida tanto na organização e expressão de comportamentos intempestivos do tipo fuga e luta como também no controle de comportamentos defensivos mais elaborados e orientados, tais como a avaliação de risco e a esquiva. / The midbrain dorsal periaqueductal grey (DPAG) is part of the brain defensive system involved in active defense reactions to threatening stimuli. Many lines of evidence suggest that besides fundamentally controlling fear-like responses (fight and flight) the DPAG also controls responses related to anxiety, such as avoidance and risk assessment. This study investigated the role of DPAG on different defensive strategies (i.e. flight, avoidance and risk assessment) elicited by artificial (chemical stimulation, Experiments 1-3) and naturalistic (exposure to predator, Experiments 4 and 5) paradigms in mice. Firstly, D,L-Homocysteic acid (DLH) was infused into the DPAG and behavioral responses of mice were evaluated in two different situations, a rectangular novel chamber and a large oval runway, the Mouse Defense Test Battery (MDTB) apparatus (Exp. 1). We also investigated the ability of mice to react to a threatening stimulus (ex. a predator) during (Exp. 3) and immediately after (Exp. 2) the hyperactive responses (ex. jumping and running) induced by DLH injection. Our results indicated that running as opposed to jumping is the primary response in mice injected with DLH into the DPAG when the environment enables flight. However, mice did not react the predator during the flight reaction induced by chemical stimulation, suggesting the behavioral profile induced by DLH infusion into the DPAG is not related to a normal antipredator flight. In the Experiments 4 and 5, we evaluated the effects of three different compounds, N-methyl-D-Aspartate (NMDA), a NMDA receptor agonist, N?-propyl-L-arginine (NPLA), an neuronal nitric oxide synthase (nNOS) inhibitor as well as ovine CRF (oCRF), a nonspecific corticotropin-releasing factor (CRF) receptor agonist, injected into the DPAG of mice, in two predator-stress situations, the Mouse Defense Test Battery (MDTB), and the Rat Exposure Test (RET). Firstly, our results demonstrated that NMDA receptor activation into the mouse DPAG enhances antipredator reactivity (avoidance), an effect that was attenuated by prior infusion of NPLA into the same site. Moreover, the results from the Experiment 4 indicated that the NPLA treatment per se induces consistent anti-aversive effects on defensive behaviors (avoidance and risk assessment) of mice confronted by predator. Finally, our results pointed out a proaversive effect (e.g. increased jump escapes and avoidance behaviors) following intra-DPAG infusion of oCRF, suggesting an important role of glutamatergic, nitrergic and CRFergic systems into the DPAG on the defensive behaviors (risk assessment, avoidance and jumps) elicited by the confront to the predator. Taken together, present results are compatible with previous studies which have emphasized the role of the periaqueductal gray in the modulation of behavioral responses related to anxiety such as risk assessment and avoidance besides fundamentally controlling fear-like responses.

O algoritmo Otimizador por Lobo Cinzento (GWO) é aplicado para minimizar a função objetivo custo total do trocador de calor proposto. As variáveis de otimização adotadas são: número de passes nos tubos, diâmetro externo dos tubos, diâmetro interno do casco, espaçamento dos defletores e corte dos defletores. O método Bell-Delaware e o método de Kern são utilizados para calcular o coeficiente de transferência de calor e a perda de pressão para o lado do casco. Os resultados da otimização são comparados com o projeto original do trocador de calor e também com os resultados alcançados por outros algoritmos de otimização da literatura. Além disso, o algoritmo GWO é comparado com outras meta-heurísticas de otimização em três funções de teste distintas. Os resultados da comparação do algoritmo GWO nas funções de teste mostram um desempenho competitivo, comparado com o Algoritmo Genético, Otimização por Enxame de Partículas, algoritmo Evolução Diferencial e Algoritmo do Vaga-Lume. Os resultados da otimização do trocador de calor utilizando o método de Kern mostra um bom desempenho, com a redução do investimento de capital em 11,95%, 7,93%, 6,24%, 2,37% e 0,19% comparado ao projeto original, GA, ICA, PSO e GSA respectivamente. Além disso, o custo operacional total descontado foi menor que o projeto original e o restante das metaheurísticas exceto para o algoritmo GSA onde o algoritmo GWO obteve resultado 22,33 %, superior. No geral, a redução combinada do investimento de capital e do custo operacional total descontado obtido pela aplicação do algoritmo GWO levaram a uma redução de custo total de 20,80%, 7,28%, 6,07% e 4,06% comparado ao projeto original, GA, ICA e PSO respectivamente. Por fim, os resultados da otimização do trocador de calor utilizando o método Bell-Delaware comparado ao projeto original mostram desempenho satisfatório com redução do investimento de capital em 13,32%. O custo operacional total foi menor em 32,56%. Neste caso, a redução combinada do investimento de capital e do custo operacional total descontado com o GWO levaram a uma redução de custo total de 17,19%. / In this work, a new approach to optimizing the design of a shell and tube heat exchanger is developed. The Grey Wolf Optimizer algorithm (GWO) is applied to minimize the objective total cost function of the proposed heat exchanger. The optimization variables adopted are: number of tubes passes, tube outside diameter, shell inside diameter, baffles spacing and baffle cut. The Bell-Delaware method and the Kern method are used to calculate the heat transfer coefficient and the pressure drop from the shell side. The results of the optimization are compared with the original design and with other optimization algorithms in the literature. In addition, the GWO algorithm is compared with other optimization meta-heuristics in three different test functions. The results of the comparison of the GWO algorithm in the test functions show a competitive performance compared to the Genetic Algorithm, Particle Swarm Optimization, Differential Evolution algorithm and Firefly Algorithm. Already, the results of the optimization of the heat exchanger using the Kern method shows a good performance, with the reduction of capital investment by 11.95%, 7.93%, 6.24%, 2.37% and 0.19% compared to the original project, GA, ICA, PSO and GSA, respectively. In addition, the total discounted operating cost was lower than the original project and the rest of the metaheuristics except for the GSA algorithm where the GWO algorithm obtained 22.33% higher result. Overall, the combined reduction in capital investment and total discounted operating cost obtained by applying the GWO algorithm led to a total cost reduction of 20.80%, 7.28%, 6.07% and 4.06% compared to the original project, GA, ICA and PSO, respectively. Finally, the results of optimization of the heat exchanger using the Bell-Delaware method compared to the original design show satisfactory performance with reduction of capital investment by 13.32 %. The total discounted operating cost was lower by 32.56%. In this case, the combined reduction in capital investment and discounted total operating cost obtained by applying the GWO algorithm led to a total cost reduction of 17.19%.

Fungicide sensitivity and spatial and temporal dynamics of Botrytis cinerea and Colletotrichum spp. in conventional and organic strawberry fields / Sensibilidade a fungicidas e dinâmicas espacial e temporal de Botrytis cinerea e Colletotrichum spp.em campos de morangueiro convencionais e orgânicos

Botrytis and Anthracnose fruit rots, caused by Botrytis cinerea and Colletotrichum spp., respectively, are major strawberry diseases. This study aimed to compare organic and conventional strawberry production systems by evaluating the sensitivity of B. cinerea and Colletotrichum spp. isolates to the main single-site fungicides used in Brazil and by characterizing the spatial and temporal dynamics of the diseases caused by these pathogens using epidemiological tools. Isolates were collected from conventional and organic fields in four different Brazilian states from 2013 to 2015. Resistance to azoxystrobin, iprodione and thiophanate-methyl was found in B. cinerea isolates with EC50 values higher than 71.9, 688 and 1.2 μg/ml, respectively. Resistance to the aforementioned fungicides was observed in 87.5, 76.6 and 92.2 % of isolates from conventional fields, and 31.4, 22.9 and 51.4 % of isolates from organic fields, respectively. Moreover, frequencies of populations with multiple fungicide resistance to the three active ingredients were 75 and 8.6 %, with no resistance to any of the fungicides were 6.25 and 34.3 % for isolates collected from conventional and organic areas, respectively. Molecular analyses of the cytochrome b, beta-tubulin and bos1 genes revealed, respectively, the presence of G143A, E198A and I365N/S, Q369P or N373S mutations in resistant populations of B. cinerea. Fungicides sprayed preventively on strawberry fruit inoculated with B. cinerea failed to control resistant isolates. Isolates of C. acutatum resistant to azoxystrobin and difenoconazole were not observed. Mean EC50 values for isolates collected from organic fields were 0.44 and 0.95 μg/ml, and from conventional areas were 0.629 and 0.107 μg/ml for azoxystrobin and difenoconazole, respectively. Populations of C. acutatum showed insensitivity to thiophanate-methyl rather than resistance and EC50 values could not be determined. None of the isolates contained the most common mutations associated with fungicide resistance. The temporal and spatial dynamics of Botrytis and Anthracnose fruit rots were characterized in the 2015 strawberry season in conventional and organic farms. Temporal progress models were fitted to the cumulative incidence of diseased strawberry fruit or plants over time with non-linear regressions. The spatial pattern of the diseases was analyzed according to the index of dispersion (D), the index of aggregation (V/M), and the Taylor\'s power law. The incidence-diseased fruit density relationship was analyzed by fitting a linear model. Fruit with symptoms of Colletotrichum spp. were not observed in the organic area and were found in few assessment dates in the conventional area. Incidence of Botrytis fruit rot in symptomatic fruit and plants were generally well described by the logistic model. The organic area presented disease incidence and daily progress rate higher than the conventional area. The aggregation of diseased fruit on strawberry plants were observed for both areas. Botrytis fruit rot presented similar epidemiological patterns for both conventional and organic areas, however, disease intensity was greater for the organic area; this was likely due to differences in disease control management. This study reinforces the importance for the implementation of integrated management programs in strawberry nurseries and production fields. / Mofo cinzento e antracnose do fruto, causados por Botrytis cinerea e Colletotrichum spp., respectivamente, são as mais importantes doenças em morangueiro. Esse estudo objetivou comparar sistemas de produção orgânico e convencional em morangueiro através da avaliação da sensibilidade de isolados de B. cinerea e Colletotrichum spp. aos principais fungicidas sítio-específicos utilizados no Brasil, e da caracterização das dinâmicas espacial e temporal das doenças com o uso de ferramentas epidemiológicas. Isolados foram coletados em campos convencionais e orgânicos em quatro diferentes Estados brasileiros nos anos de 2013 a 2015. Resistência à azoxistrobina, iprodiona e tiofanato-metílico foi encontrada em isolados de B. cinerea com valores de CE50 maiores que 71,9, 688 and 1,2 μg/ml, respectivamente. Resistência aos fungicidas acima mencionados foi observada em 87,5, 76,6 and 92,2 % dos isolados provenientes de campos convencionais e em 31,4, 22,9 and 51,4 % dos isolados oriundos de campos orgânicos, respectivamente. Além do mais, frequências de populações com resistência múltipla aos três fungicidas foram 75 e 8,6 %; e com nenhuma resistência a qualquer dos produtos, 6,25 e 34,3 % para isolados coletados em campos convencionais e orgânicos, respectivamente. Análise molecular dos genes do citocromo b, beta-tubulina e bos1 revelaram, respectivamente, a presença das mutações G143A, E198A e I365N/S, Q369P ou N373S em populações resistentes. Fungicidas aplicados preventivamente em morangos inoculados com B. cinerea falharam em controlar isolados resistentes. Isolados de C. acutatum resistentes à azoxistrobina e ao difenoconazole não foram observados. Valores médios de CE50 para isolados coletados em campos orgânicos foram 0,44 e 0,95 μg/ml; e para isolados de áreas convencionais, 0,629 e 0,107 μg/ml para azoxistrobina e difenoconazole, respectivamente. Populações de C. acutatum demonstraram insensibilidade ao tiofanato-metílico e valores de CE50 não puderam ser determinados. Nenhum dos isolados analisados continha as mutações comumente associadas à resistência a fungicidas. As dinâmicas temporal e espacial das doenclas causadas por esses patógenos foram caracterizadas na safra de 2015 em campos de morangueiro convencional e orgânico. Modelos de progresso temporal foram ajustados à incidência cumulativa de frutos ou plantas doentes no tempo com regressões não-lineares. O padrão espacial das doenças foi analisado de acordo com o índice de dispersão (D), índice de agregação (V/M) e lei de Taylor. A relação incidência-densidade de frutos doentes foi analisada com o ajuste de um modelo linear. Frutos com sintomas de Colletotrichum spp. não foram observados na área orgânica e foram encontrados em poucos dias de avaliação na área convencional. Incidência de mofo cinzento em frutos e plantas foi bem descrita pelo modelo logístico. A área orgânica apresentou incidência de doença e taxa de progresso diária maiores que da área convencional. A agregação de frutos doentes em plantas foi observada para ambos os locais. O mofo cinzento apresentou padrões epidemiológicos semelhantes para as áreas convencional e orgância, porém com maior intensidade de doença no campo orgânico, provavelmente em decorrência do manejo adotado para controle de doenças. Esse estudo reforça a importância da implementação de programas de manejo integrado de doenças em viveiros de mudas e campos de produção de morangueiro.
