Evaluation de l'effet thérapeutique des cellules souches mésenchymateuses dans la sclérodermie systémique / Mesenchymal stem cell based therapy in systemic sclerosis

La sclérodermie systémique (ScS) est une maladie rare et incurable, caractérisée par une fibrose cutanée et la production d’auto-anticorps (maladie auto-immune). Le pronostic vital est engagé dans les formes diffuses et rapidement progressives de la maladie, responsables de fibrose pulmonaire. Par leurs propriétés immunomodulatrices et anti-fibrotiques, les cellules souches mésenchymateuses (CSM) constituent une approche prometteuse pour traiter la ScS. Ce travail a pour objectif d’évaluer le potentiel thérapeutique des CSM dans un modèle préclinique de forme diffuse de la maladie (d-ScS).Patients et Méthodes : Nous avons évalué l’effet de différentes modalités d’injections des CSM par voie intraveineuse dans le modèle murin de ScS-HOCl. Ce modèle, basé sur l’injection d’acide hypochloreux (HOCl), induit un phénotype proche des formes d-ScS. Nous avons notamment comparé un traitement préventif et curatif, des approches syngénique, allogénique et xénogénique, et l’utilisation de CSM d’origine adipocytaire (ASC) à celle de CSM de moelle osseuse (CSM-MO).Résultats : Le modèle ScS-HOCl est caractérisé par l’installation d’un phénotype de d-ScS, avec fibrose cutanée, pulmonaire et production d’anticorps anti-topoisomérase 1. Nous montrons l’effet bénéfique d’une injection préventive ou curative de CSM syngéniques, réduisant la fibrose cutanée et pulmonaire. Cet effet thérapeutique passe par une diminution de la réponse immune réduisant l’inflammation tissulaire et la production d’auto-anticorps, ainsi que par l’induction du remodelage tissulaire via l’activation de métalloprotéases, et l’augmentation des défenses anti-oxydantes. Un bénéfice similaire est obtenu lors d’approches allogénique et xénogénique. Les ASC présentent des capacités immunosuppressives et de remodelage supérieures aux CSM-MO.Discussion et conclusion : Nos travaux démontrent l’effet anti-fibrotique des CSM dans un modèle préclinique pertinent de ScS, mimant les formes diffuses et rapidement progressives de la maladie, pour lesquelles les besoins thérapeutiques sont importants. Le mode d’action pléiotropique des CSM, combinant propriétés anti-inflammatoires, pro-remodelage et anti-oxydantes, est prometteur en vue des applications cliniques à venir dans cette maladie.Mots-clés : sclérodermie systémique, cellules souches mésenchymateuses, HOCl / Systemic sclerosis (SSc) is a rare intractable disease characterized by skin fibrosis and autoimmunity. In this study, we aim at evaluating the therapeutic potential of MSC in a preclinical model of d-ScS.Patients and Methods: We evaluated the effects of MSC infusion in the murine model of ScS-HOCl, based on repeated injections of hypochlorite (HOCl). We compared several approaches using MSC in a preventive and curative approach, in syngeneic, allogeneic and xenogeneic settings, and using MSC isolated from adipose tissue (ASC) or bone marrow (BM-MSC).Results: ScS-HOCl is close to d-ScS phenotype, leading to skin and lung fibrosis, together with anti-topoisomerase 1 antibody production. We show beneficial effects of a preventive or curative injection of syngeneic MSCs, reducing tissue fibrosis. Fibrosis reduction following MSC treatment involves immunosuppressive effects, tissue remodeling via metalloprotease activation, and anti-oxydant activity. Similar benefits are observed in allogeneic and xenogeneic settings. ASC display greater immunosuppressive and remodeling capacities than BM-MSCs.Discussion and conclusion: Our study demonstrates the anti-fibrotic effects of MSCs in a relevant preclinical model of ScS, mimicking diffuse and rapidly progressive ScS. Impact of simultaneous stimulation of 5-lipoxygenase and myeloperoxidase in human neutrophils

Human neutrophil 5-lipoxygenase (5-LOX) oxidizes arachidonic acid (AA) to 5S-hydro(pero)xy-6E,8Z,11Z,14Z-eicosatetraenoic acid (5-H(p)ETE) and leukotriene (LT)A4, which is further converted to the chemoattractant LTB4. These cells contain also the heme enzyme myeloperoxidase (MPO) producing several potent oxidants such as hypochlorous acid (HOCl). Previously, it was shown that MPO-metabolites influence 5-LOX product formation. Here, we addressed the question, whether a simultaneous activation of MPO and 5-LOX in neutrophils results in comparable changes of 5-LOX activity. Human neutrophils were stimulated with H2O2 or phorbol 12-myristate 13-acetate (PMA) for MPO activation and subsequently treated with calcium ionophore A23187 inducing 5-LOX product formation on endogenous AA. Special attention was drawn to neutrophil vitality, formation of MPO-derived metabolites and redox status. The pre-stimulation with H2O2 resulted in a concentration-dependent increase in the ratio of 5-HETE to the sum of LTB4 + 6-trans-LTB4 in consequence of MPO activation. Thereby no impairment of cell vitality and only a slightly reduction of total glutathione level was observed. An influence of MPO on 5-LOX product formation could be suggested using an MPO inhibitor. In contrast, the pre-stimulation with PMA resulted in different changes of 5-LOX product formation leading to a reduced amount of 5-HETE unaffected by MPO inhibition. Furthermore, impaired cell vitality and diminished redox status was detected after PMA stimulation. Nevertheless, a MPO-induced diminution of LTB4 was obvious. Further work is necessary to define the type of 5-LOX modification and investigate the effect of physiological MPO activators.

Composição química e capacidade sequestrante de espécies reativas de oxigênio e nitrogênio de mel orgânico brasileiro / Chemical composition and Nitrogen and Oxygen Reactive Species scavenging activity of Brazilian organic honey

O Brasil apresenta grande potencial para a exploração da apicultura, dado ao seu vasto território e flora diversificada, o que permite diferentes variedades de méis com propriedades únicas. O estado do Paraná é um dos maiores produtores de méis do país e o investimento em produção que atenda aos mercados mais exigentes estimulou a produção do mel orgânico. O conhecimento desde a antiguidade sobre os efeitos benéficos à saúde pelo mel vem estimulando a pesquisa deste alimento nobre. Assim, este trabalho teve por objetivo estudar méis orgânicos brasileiros certificados (MO) para a caracterização do perfil fenólico, volátil, além da avaliação da capacidade de sequestro das espécies reativas de oxigênio e nitrogênio. Os méis foram coletados nos apiários de apicultores com certificação orgânica de dois municípios do sul do Paraná, General Carneiro e Turvo-PR. Nos ensaios foram utilizados extratos fenólicos dos méis, obtidos por meio da utilização da resina Amberlite® XAD®2, bem como méis brutos in natura. Os extratos apresentaram conteúdo de compostos fenólicos significativo, sendo o melato (MO5), de General Carneiro, o de maior teor (117,68± 4,40 mg EAG/g). Para as análises de sequestro das espécies reativas de oxigênio e nitrogênio, os extratos fenólicos foram sempre superiores aos méis brutos in natura. Os extratos fenólicos, de maneira geral, apresentaram alta capacidade de sequestro para o radical peroxila (ROOo), ácido hipocloroso (HOCl) e óxido nítrico (NOo). Em relação aos melatos, o extrato MO7 apresentou alta capacidade para o sequestro do HOCl (EC50= 4,83 ± 0,13 ?g/mL), enquanto que o MO5 foi melhor para o sequestro do NOo (EC50=2,16 ± 0,18 ?g/mL). Pelo método HPLC-ABTS on-line foi possível identificar e quantificar a contribuição para a atividade antioxidante do ácido ferúlico no extrato (MO1) e do flavonoide kanferol na amostra (MO4). O ácido ascórbico foi identificado e quantificado por HPLC somente nos melatos (MO3, MO5 e MO7). Pela técnica de LC-MS/MS foram identificados a presença dos seguintes compostos fenólicos: ácido caféico, rutina e hesperidina em todos os extratos. A análise de compostos voláteis por SPME-CG/EM mostrou a presença de dois compostos, encontrados apenas nos melatos, que foram o terpineno-4-ol, que possui ação antifúngica, antiparasitológica e anti-inflamatória; e o 3,4-dimetil-1-deceno, podendo assim serem utilizados como marcadores químicos destes méis. Thus, this paper aimed to study certified Brazilian organic honeys (MO) in order to determine the phenolic and volatile profiles, and also the evaluation of radical scavenging capacity against Nitrogen and Oxygen Reactive Species (RNS and ROS, respectively). The honeys were collected from apiaries from beekeepers with the organic certification from two municipalities of southern Parana, General Carneiro and Turvo, PR. In the essays, phenolic extracts were obtained from honeys by using Amberlite® XAD®2 resin, as well as crude in natura honeys. The extracts showed a significant content in phenolic compounds, with honeydew (MO5), from General Carneiro, showing the highest content (117,68 ± 4,40 mg AGE/g). For the analyzes to determine the radical scavenging capacity against RNS and ROS, the phenolic extracts always showed up superior results in comparison to crude in natura honeys. Phenolic extracts showed, in general, great capacity to scavenge peroxyl radical (ROOo), hypochlorous acid (HOCl) and nitric oxide (NOo). In relation to honeydews MO7 extract showed the highest capacity to scavenge HOCl (IC50= 4,83 ± 0,13 ?g/mL) while MO5 was the sample with better capacity to scavenge NOo (IC50=2,16 ± 0,18 ?g/mL). By using HPLC-ABTS on-line method it was possible to identify and to quantify the ferulic acid in MO1 extract, a compound with an important contribution to the antioxidant activity of this sample, as well as the flavonoid kaempferol in MO4 sample. Ascorbic acid was identified and quantified by HPLC only in the honeydew samples (MO3, MO5 and MO7). The analyzes developed by LC-MS/MS techniques indicated the presence of the phenolic compounds caffeic acid, rutin and hesperidin in all the extracts. The analysis of volatile substances developed by SPME-GC/MS promoted the identification of two compounds found only in the honeydew samples. The compounds were the terpinen-4-ol, which has antifungal, antiparasitological and anti-inflammatory activities; and 3,4-dimethyl-1-decene. Both compounds can be used as chemical markers of these honeys. The knowledge of the chemical composition of the studied honeys, as well as their bioactive phenolic composition, contributes to supply natural antioxidants to human diet, thus attenuating the negative effects of free radicals

Impact of Myeloperoxidase-derived oxidants on the product profile of human 5-Lipoxygenase

Human 5-lipoxygenase (5-LOX) oxidizes arachidonic acid to 5S-hydroperoxy-6E,8Z,11Z,14Z-eicosatetraenoic acid (5-HpETE) and leukotriene (LT) A4. In neutrophils, LTA4 is further converted to the potent chemoattractant LTB4. These cells also contain the heme enzyme myeloperoxidase (MPO), which produces several potent oxidants such as hypochlorous acid (HOCl), which are involved in pathogen defense and immune regulation. Here, we addressed the question whether MPO-derived oxidants are able to affect the activity of 5-LOX and the product profile of this enzyme. Human 5-LOX was incubated with increasing amounts of HOCl or HOBr. Afterward, arachidonic acid metabolites of 5-LOX were analyzed by reverse-phase high-performance liquid chromatography as well as by liquid chromatography-electrospray ionization-tandem mass spectrometry. The incubation of 5-LOX with the MPO-derived oxidants significantly changed the product profile of 5-LOX. Thereby, HOCl and HOBr increased the ratio of 5-H(p)ETE to 6-trans-LTB4 in a concentration-dependent manner. At low oxidant concentrations, there was a strong decrease in the yield of 6-trans-LTB4, whereas 5-HpETE did not change or increased. Additionally, the formation of 8-HpETE and 12-HpETE by 5-LOX rose slightly with increasing HOCl and HOBr. Comparable results were obtained with the MPO-H2O2-Cl– system when glucose oxidase and glucose were applied as a source of H2O2. This was necessary because of a strong impairment of 5-LOX activity by H2O2. In summary, MPO-derived oxidants showed a considerable impact on 5-LOX, impairing the epoxidation of 5-HpETE, whereas the hydroperoxidation of arachidonic acid was unaffected. Apparently, this was caused by an oxidative modification of critical amino acid residues of 5-LOX. Further work is necessary to assess the specific type and position of oxidation in the substrate-binding cavity of 5-LOX and to specify whether this interaction between 5-LOX and MPO-derived oxidants also takes place in stimulated neutrophils.
