Consórcio de milho com adubos verdes e manejo da adubação nitrogenada no cultivo de feijão em sucessão no sistema integração lavoura-pecuária no cerrado / Nitrogen fertilization management in bean cultivated after maize-legumes intercropping in the Crop-Livestock Integration system in Brazilian Savannah

Foram realizados dez experimentos envolvendo as culturas de feijão e milho consorciado com leguminosas e/ou Brachiaria brizantha, nas safras de verão de 2008/2009, inverno de 2009 e verão de 2009/2010, em dois municípios de estado de Goiás (GO). Em Ipameri-GO, a área foi cultivada com soja por vários anos e milho consorciado com B. brizantha na safra que precedeu a implantação dos experimentos. Em Santo Antônio de Goiás-GO, a área é mantida com a rotação de milho e soja com a forrageira B. brizantha, em esquema trienal. Os principais objetivos desses estudos foram avaliar: (i) a viabilidade do consórcio de milho com leguminosas, (ii) a resposta do feijoeiro cultivado em sucessão e (iii) o manejo do N nessas culturas. Foi constatado que em solos com média a alta fertilidade química, como os utilizados na experimentação, os consórcios de milho com B. brizantha ou com guandu-anão (Canajus cajan) ou crotalária (Crotalaria spectabilis) não interferiram na produtividade de grãos de milho na presença de N em cobertura, exceto no experimento conduzido em Santo Antônio de Goiás-GO, no qual a crotalária reduziu significativamente a produtividade do milho. Também, em ambos os locais, no consórcio simultâneo ou defasado de milho com crotalária ou guandu-anão não ocorreu liberação de nitrogênio (N) das leguminosas para o milho, medido pela produtividade de grãos, durante o ciclo das espécies. Já nos consórcios em que a B. brizantha foi mantida sem dessecação nas entrelinhas do milho, denominado de dessecação parcial, ocorreu decréscimo significativo na produtividade do milho, em relação ao cultivo em área totalmente dessecada. A análise de crescimento revelou maiores índice de área foliar (IAF) e acúmulo de massa de matéria seca da parte aérea (MSPA) para o guandu em monocultivo, sendo que para a crotalária cultivada na linha do milho, tanto o IAF quanto a MSPA aproximaram-se dos valores referentes ao monocultivo. As curvas de crescimento do milho revelaram a superioridade do cultivo com 90.kg.ha-1 de N tanto no IAF quanto na MSPA, quando comparado ao cultivo consorciado com leguminosas. No caso de Santo Antônio de Goiás-GO, na rotação trienal envolvendo espécies graníferas e forrageiras, registrou-se um acentuado aporte de N no solo, sendo obtidas altas produtividades de feijão cultivado em sucessão à pastagem, sem a aplicação de N mineral, ocorrendo, porém, aumento significativo na produtividade do feijoeiro nos tratamentos de palhadas que continham leguminosas. A aplicação de N mineral imediatamente antes da semeadura ou no estádio de plântulas do feijão e do milho teve efeito semelhante nas suas produtividades em relação à aplicação tradicional, ou seja, algumas semanas após a emergência dessas espécies. A aplicação de N mineral foi mais eficiente no aumento da produtividade de feijão irrigado quando feita próximo às fileiras de plantas ou quando o N foi incorporado mecanicamente no solo, comparados à aplicação a lanço. / Ten experiments were performed involving common beans and maize intercropped with legumes and/or Brachiaria brizantha, in the summer crops of 2008/2009, 2009 winter and summer of 2009/2010 in two municipalities in the Goiás (GO) state, Brazil. In Ipameri- GO, the area was planted with soybeans for several years and corn associated with B. brizantha in crop preceding the experiments. In Santo Antônio de Goiás-GO, the area is maintained with corn or soybean rotation with B. brizantha, in a three-year scheme. The main objectives of these studies were to evaluate the feasibility of intercropping maize with legumes and the response of common bean after it, in no-tillage system, on the mulching straws provided by the cultivation of maize intercropped with forage legumes and grasses and to study the management of nitrogen (N) in these two crops. It was found that in soils with medium to high chemical fertility, such as those used in the experiments, the intercropping of corn and B. brizantha or pigeonpea (Canajus cajan) and sunn hemp (Crotalaria spectabilis) had any influence on grain yield of maize in the presence of mineral N, except in the experiment conducted in Santo Antonio de Goiás-GO, where the sunn hemp significantly reduced corn productivity. Also, in both locations, simultaneous or lagged consortium of pigeonpea or sunn hemp with maize, there was no release of N from legumes to maize, as measured by grain productivity, during the cycle of the species. However, in the consortia in which B. brizantha is maintained without drying between the maize lines, called partial desiccation, there was a significant decrease in the yield of corn in relation to the totally desiccated area. The growth analysis showed higher leaf area index (LAI) and total dry matter accumulation (TDM) for pigeonpea grown in single system, but for sunn hemp cultivated in the corn rows, both LAI and TDM approached to its single cropping, confirming its competitiveness with corn in the intercropping system. Growth curves also showed the superiority of the corn crop with 90 kg N ha-1 in both LAI and TDM, compared to intercropping with legumes. In the case of Santo Antônio de Goiás-GO the three-year rotation involving grain species and grass forages caused an intake of N in the soil, and high yields of beans grown in rotation with pasture were obtained, without the application of mineral N, occurring, though, significant increase in grain yield in the treatments of straws containing legumes. The application of mineral N immediately before sowing or at the seedling stage of beans and corn had a similar effect on their productivity compared to traditional application, a few weeks after the emergence of these species. The application of mineral N was more efficient in increasing the productivity of beans when applied on the soil surface near the plants rows and in cases where the fertilizer was mechanically incorporated into the soil, comparing to surface-broadcasting.

Ces travaux de thèse constituent une contribution au développement de composites chanvre/époxy 100% bio-sourcés. Les enjeux environnementaux actuels favorisent l'émergence de matériaux issus de ressources renouvelables telles que les fibres végétales mais conduisant aussi à une large gamme de synthons bio-sourcés, notamment à l'origine de prépolymères époxydiques. Une étude approfondie des deux constituants (fibres de chanvre et matrices polyépoxydiques) est réalisée avant l'étape d'élaboration des composites. Un traitement au CO2 supercritique est appliqué sur les fibres de chanvre utilisées comme renfort dans les matériaux composites. Le résultat de ce traitement mène à une meilleure individualisation ainsi qu'à une baisse du pouvoir hygroscopique des fibres. Ces aspects, décisifs pour garantir de bonnes propriétés pour le composite final, sont néanmoins nuancés par une baisse des propriétés ultimes en traction à l'échelle des fibres mais également à l'échelle du composite. De la même façon, la diminution du pouvoir hygroscopique des fibres après traitement se répercute à l'échelle du composite, permettant ainsi d'améliorer la durabilité du composite. La synthèse des résines époxydiques utilisées dans cette étude est réalisée à partir de ressources renouvelables et abondantes telles que la lignine. Les polyépoxydes thermodurcissables ainsi préparés présentent de bonnes performances, compatibles avec le cahier des charges pour des applications composites à renfort végétal. Au regard des résultats obtenus, les composites 100% bio-sourcés sont des matériaux d'avenir. Leur développement nécessite néanmoins une étude approfondie de leur durabilité. / This thesis contributes to the development of 100% bio-based hemp/epoxy composites. Current environmental issues favor the emergence of materials derived from renewable resources such as plant fibres and a wide range of bio-based building-blocks, source of epoxy prepolymers in particular. An intensive investigation of the two constituents (hemp fibres and polyepoxidic matrix) is carried out before the composites manufacturing. Supercritical CO2 treatment is applied to hemp fibres used as reinforcement in composite materials. This treatment, not optimized, leads to better individualization as well as a decrease in the hygroscopic power of the fibres. These aspects, which are essential in order to guarantee good properties for the final composite, are nevertheless qualified by a loss of mechanical properties at the fibre scale but also at the composite scale. In the same way, the decrease in the hygroscopic power of the hemp fibres after treatment is reflected at the composite scale, thus improving the durability of the crosslinked material. The synthesis of the epoxy resins used in this study is made from abundant resources such as lignin. The thermosetting polyepoxides prepared in this study have good performance, compatible with the specification for composite applications with plant fibres reinforcement. In view of the results obtained, 100% bio-based composites are materials with a high future potential. Nevertheless, their development requires a comprehensive sustainability study.

<p>Истраживање у оквиру докторске дисертације обухвата анализу могућности производње и примене биокомпозитних термоизолационих плоча заснованих на нуспроизводима пољопривредне производње и мицелијума гљиве буковаче као везивног средства. Циљеви истраживања су развој методе за производњу биокомпозитних термоизолационих материјала и утврђивање морфолошких, физичко-хемијских, механичких и хигротермичких својстава биокомпозита. У складу са резултатима истраживања и закључцима да је могуће произвести и користити биокомпозитни термоизолациони материјал заснован на биомаси и мицелијуму гљива изведена је анализа енергетске ефикасности и животног циклуса материјала. Резултати истраживања представљају значајну полазну основу за производњу и даље унапређење биокомпозитних термоизолационих материјала у грађевинарству, са позитивном оценом мицелијума гљива као везивног средства.</p> / <p>Istraživanje u okviru doktorske disertacije obuhvata analizu mogućnosti proizvodnje i primene biokompozitnih termoizolacionih ploča zasnovanih na nusproizvodima poljoprivredne proizvodnje i micelijuma gljive bukovače kao vezivnog sredstva. Ciljevi istraživanja su razvoj metode za proizvodnju biokompozitnih termoizolacionih materijala i utvrđivanje morfoloških, fizičko-hemijskih, mehaničkih i higrotermičkih svojstava biokompozita. U skladu sa rezultatima istraživanja i zaključcima da je moguće proizvesti i koristiti biokompozitni termoizolacioni materijal zasnovan na biomasi i micelijumu gljiva izvedena je analiza energetske efikasnosti i životnog ciklusa materijala. Rezultati istraživanja predstavljaju značajnu polaznu osnovu za proizvodnju i dalje unapređenje biokompozitnih termoizolacionih materijala u građevinarstvu, sa pozitivnom ocenom micelijuma gljiva kao vezivnog sredstva.</p> / <p>Research in the thesis is focused on development and utilisation of<br />biocomposite thermal insulation panels based on agricultural biomass and<br />oyster mushroom mycelium as a binding agent. Etude des transferts couplés de chaleur et de masse dans les matériaux bio-sourcés : approches numérique et expérimentale / Study of heat and mas transfer within bio-based building materials : numerical and experimental approaches

Le travail développé dans cette thèse a pour but d’étudier le comportement hygrothermique de matériaux isolants bio-sourcés, et plus particulièrement les fibres de bois, le béton de chanvre, la laine de lin, la laine de mouton, le métisse® et les anas de lin. Ces matériaux, par essence naturels, présentent des spécificités liées à leur origine (animale ou végétale) et à leur structure (fibres, paille, matrice solide…). Leur porosité, très élevée, les rend réactifs aux variations d’humidité relative ambiante, ce qui peut impacter leurs performances thermiques et leur durabilité (comme pour tous les matériaux), mais également leur conférer des capacités de régulation. Dans un souci d’améliorer la connaissance de ces matériaux particuliers, nous proposons tout d’abord d’étudier l’impact causé par l’humidité sur leurs caractéristiques thermiques, principalement la conductivité thermique et la chaleur spécifique. Ensuite les caractéristiques hygrothermiques sont étudiées, ce qui permet de mieux comprendre les phénomènes dépendant des capacités d’adsorption, de désorption, de perméabilité ou de résistance à la vapeur d’eau. On se rend compte également de l’importance du gradient de température sur l’évolution des transferts hygriques au sein des matériaux. En plaçant les isolants bio-sourcés sous sollicitations aléatoires ou en conditions réelles d’utilisation, nous pouvons suivre leur comportement d’un point de vue expérimental. Le couplage à une approche numérique permet d’identifier les paramètres d’influence prépondérants, dans l’optique de la prédiction des transferts couplés chaleur/masse par une simulation dans des conditions particulières d’utilisation, comme la rénovation d’un habitat existant. On constate à partir de mesures in situ que ces matériaux ont une grande capacité d’adaptation à des environnements dont l’humidité relative est évolutive. / The work developed in this thesis aims to study the hygrothermal behavior of bio-sourced insulating materials, and more particularly wood fibers, hemp concrete, linen wool, sheep wool, material made of textile recycling (metisse®) and flax shives. These materials, which are essentially natural, have specific characteristics linked to their origin (animal or vegetable) and their structure (fibers, straw, solid matrix, etc.). Their very high porosity makes them reactive to the relative humidity variations, which can affect their thermal performances and their durability (as for all materials), but also give them a regulation capacities. In order to improve the knowledge of these particular materials, first, we propose to study the impact caused by moisture on their thermal characteristics, mainly thermal conductivity and specific heat. Then the hygrothermal characteristics are studied, which makes it possible to better understand the phenomena depending on the capacities of adsorption, desorption, permeability or water vapor resistance. Also, we realize the importance of the temperature gradient impact on the evolution of the hygroscopic transfers within the materials. By placing the studied bio-sourced insulation materials under random loading or under real conditions, it will be possible to follow their hygrothermal behavior from an experimental point of view. The numerical approach makes it possible to identify the preponderant influence parameters, in the context of the prediction of coupled heat and mass transfers by simulation under particular conditions of use, such as the renovation of an existing habitat. On the basis of in situ measurements, it can be seen that these materials have a high adaptability to environments whose relative humidity is evolutionary.

La fibra di canapa europea è l'unica fibra naturale con una certificazione di sostenibilità. La qualità della fibra è influenzata principalmente da genotipo, tecniche agronomiche, epoca di raccolta e metodo di macerazione. L'obiettivo principale della tesi è delineare le pratiche agronomiche e di post-raccolta per migliorare l'estrazione e la qualità della fibra, con particolare attenzione alla produzione di canapa multiuso ritardando la raccolta fino alla maturazione dei semi. Le prove di densità, fertilizzazione azotata e varietali sono state condotte in differenti ambienti Europei. La procedura di decorticazione standardizzata è stata confrontata con la linea longitudinale di estrazione della fibra per applicazioni di alto valore aggiunto. I test su bio-compositi di canapa sono stati effettuati con fibra pettinata per confrontarne le proprietà tra genotipi, epoca di raccolta e metodi di macerazione. I risultati di questa indagine suggeriscono che i) la densità di semina e la concimazione azotata ottimale sono tra 90-150 piante m-2 e 30-60 kg N ha-1; ii) le nuove varietà dallo stelo giallo presentano un'alta efficienza di decorticazione e una ridotta contaminazione da canapulo nella fibra, iii) la fibra di canapa, con proprietà comparabili a quelle del lino, si è dimostrata adatta per applicazioni in compositi di alto valore. / European hemp fibres are the only natural fibre with an established sustainability certification. Hemp fibre quality is affected by genotype, agronomic techniques, harvest time and retting method. The main objective of this thesis is to outline the agronomic and post-harvest practices for improved fibre extraction and fibre quality, with special attention to multipurpose hemp production delaying the harvest from the flowering stage until seed ripening is complete. Planting density and nitrogen fertilization trials and variety trials were conducted in contrasting environments in Europe. Stems were decorticated following a standardised procedure and longitudinal hemp line for textile and high-added values application was compared with lab-scaled decortication. Impregnated fibre bundle tests were carried out with hemp hackled fibre bundles to compare composites and back-calculated fibre properties between genotypes, harvest times and retting methods. Results of this investigation suggest that i) optimum plant density and nitrogen fertilization are between 90 and 150 plants m-2 and 30 and 60 Kg N ha-1 respectively; ii) new yellow stemmed varieties are characterized by high decortication efficiency and relative high cleanness of the extracted fibre and iii) long hemp fibre, having properties comparable to those of flax, proved to be suitable for high-tech composites applications.

The fabric of life : linen and life cycle in England, 1678-1810

'The Fabric of Life: Linen and Life Cycle in England, 1678-1810' is structured around the human life cycle to draw out the social and cultural importance of linen for all ranks of society. Human and object life cycles are juxtaposed in the thesis to analyse co-dependent activities and processes rather than focusing on one facet of daily life. For thousands of years flax was a staple fibre, used for textile production in many parts of the globe. Cotton only overtook linen as the most popular textile in England at home and on the body during the nineteenth century. This thesis examines the preceding century to reveal why linen remained a daily necessity in England between 1678 and 1810, a period which encompassed a series of significant changes in the production, trade and use of linen. Linen was ubiquitous as underwear, sheets, table linens and for logistical purposes therefore it provides a unique insight into the early-modern world; a means of understanding the multifaceted experiences of daily life, of integrating understandings of the body, domestic, social, cultural and commercial activities. This thesis is social history through the lens of linen, reading a society through its interactions with a textile.

Construction is a very dynamically developing industry that is placing ever greater demands on the properties of used building materials and structures built on them. In this context, there is a long-term trend to use construction materials made from natural quickly renewable raw materials. These tendencies are caused by natural reaction to the overly technological modern society, as well as efforts towards a greener approach in construction. In this sense, the natural materials from renewable sources means technologically processed organic material of plant origin. Dissertation focuses on the research and subsequent development of filling materials for vertical and horizontal structures based on quickly renewable materials. Pivotal quickly renewable raw material used in the production of filling material is technical hemp. Technical hemp is quickly renewable raw material with very good properties very similar to wood. Furthermore, the research focused on secondary raw materials, which partly replace the binder of the filling material. The resulting filling material has good physical-mechanical and thermo-technical characteristics.

Development of biotechnological tools for the genetic improvement of Cannabis sativa L. / Desarrollo de herramientas biotecnológicas para la mejora genética de Cannabis sativa L.

Tesis por compendio / [EN] Cannabis sativa L. (Cannabaceae) is an angiosperm, allogamous and dicotyledonous species that includes short and neutral-day varieties with dioecious specimens (males and females), and monoecious plants. Among its many applications, its industrial and medicinal uses stand out. Despite the fact that cannabis has been used by humans since ancient times and the growing interest that the C. sativa therapeutic properties have aroused in researchers around the world, the psychoactivity of some of its varieties, derived from its ¿9-tetrahydrocannabinol (THC) content, has motivated the prohibition of its cultivation for almost sixty years. The strict control to which cannabis has been subjected has prevented professionals from all over the world from carrying out genetic breeding programs for this species, which has resulted in the absence of uniform varieties. In this Doctoral Thesis, different biotechnological tools for cannabis genetic improvement have been developed. In the first place, given the lack of reproducibility of some cannabis plant in vitro regeneration protocols and the great influence that the genotype exerts on their effectiveness, plant in vitro regeneration competence of different explants was evaluated. As a result, an hormone-free protocol from C. sativa hypocotyls that presents high regeneration rates (ranging from 32.26% to 71.15%) in all the genotypes evaluated, also presenting a 17.94% of spontaneous rooting rate of regenerants has been developed. At the same time, the polysomatic pattern of different cannabis explants has been studied, and it has been possible to regenerate, from them, a significant percentage of mixoploid specimens (17.65% from cotyledons and 13.33% from hypocotyls) that, as described in the existing literature, could show a greater capacity for cannabinoid synthesis. On the other hand, given the absence of scientific publications in this regard, and the potential that this technique presents to alleviate the intrinsic variability of this species, the most in-depth study to date on the male floral biology of C. sativa has been developed. Up to 476,903 microspores and pollen grains per male flower, with in vivo microspore viability rates from 53.71 to 70.88% have been found. Furthermore, all stages of development of the microgametophyte have been correlated with an easily measurable floral morphological marker such as the bud length, identifying bud length intervals containing mostly vacuolate microspores and young bi-cellular pollen grains in all the phenotypes evaluated. In this way, and although the starch presence in C. sativa microspores and pollen grains follows a similar pattern to that observed in species recalcitrant to androgenesis, it has been possible to address the induction of microspore embryogenesis in this species, obtaining for the first time microspore-derived multicellular structures after one week long cold-shock bud pretreatment. Finally, as a prerequisite for the genetic editing of C. sativa by using the Clustered Regularly Interspaced Short Palindromic Repeats (CRISPR)/Cas systems, and taking advantage of the in vitro plant regeneration protocol which resulted from this Doctoral Thesis, it has been possible to develop for the first time a protocol for the production of stably transformed cannabis plants, which represents a historical milestone in the genetic improvement of the species. After co-culture with A. tumefaciens and subsequent culture in antibiotic-containing selective regeneration medium, hypocotyls achieved 23.1% and 5.0% of regeneration and transformation rates respectively. As a whole, the present Doctoral Thesis provides a range of biotechnological tools that will allow the development of a new generation of high-yield cannabis varieties with uniform traits, resistant to multiple biotic and abiotic stresses, and therefore being suitable for both industrial and medicinal use. / [ES] Cannabis sativa L. (Cannabaceae) es una especie angiosperma, alógama y dicotiledónea compuesta por variedades de día corto y día neutro que presentan ejemplares dioicos (machos y hembras), y plantas monoicas. Entre sus múltiples aplicaciones destacan tanto su uso industrial como su uso medicinal. A pesar de que el cannabis ha sido empleado por el ser humano desde tiempos ancestrales, la psicoactividad que presentan algunas de sus variedades, derivada de su contenido en ¿ 9 -tetrahidrocannabinol (THC), ha motivado la prohibición de su cultivo durante casi sesenta años. La estricta fiscalización a la que ha sido sometido el cannabis, ha impedido llevar a cabo programas de mejora genética de esta especie, lo que se ha traducido en la ausencia de variedades uniformes. En esta Tesis Doctoral se han desarrollado diferentes herramientas biotecnológicas para la mejora genética del cannabis. En primer lugar, dada la falta de reproducibilidad de algunos protocolos de cultivo in vitro de cannabis y la gran influencia que el genotipo ejerce en la efectividad de los mismos, se evaluó la capacidad de regeneración in vitro de diferentes explantes. Como resultado, se ha desarrollado un protocolo libre de hormonas a partir de hipocótilos de C. sativa que presenta altas tasas de regeneración (las cuales oscilan del 32,26% al 71,15%) en todos los genotipos evaluados, presentando además un 17,94% de tasa de enraizado espontáneo de los regenerantes. A su vez, se ha estudiado el patrón polisomático de diferentes explantes de cannabis y se ha conseguido regenerar, a partir de los mismos, un porcentaje significativo de ejemplares mixoploides (17,65% procedentes de cotiledones y 13,33% de hipocotilos) que, tal y como describe la bibliografía existente, podrían mostrar una mayor capacidad de síntesis de cannabinoides. Por otro lado, dada la ausencia de publicaciones científicas al respecto y el potencial que esta técnica presenta para paliar la variabilidad intrínseca de esta especie, se ha desarrollado el estudio más profundo hasta la fecha relativo a la biología floral masculina de C. sativa. Se han descrito hasta 476.903 microsporas y granos de polen por flor masculina, con tasas de viabilidad in vivo de las microsporas del 53,71 al 70,88%. Además, se han correlacionado todas las etapas de desarrollo del microgametofito con la longitud de la yema, identificando intervalos de longitud de yema que contienen mayoritariamente microsporas vacuoladas y granos de polen joven bicelular en todos los fenotipos evaluados. De este modo, y aunque la presencia de almidón en las microsporas y granos de polen de C. sativa sigue un patrón similar al observado en especies recalcitrantes a la androgénesis, ha sido posible abordar la inducción de la embriogénesis de microsporas en esta especie, consiguiendo producir por primera vez estructuras multicelulares derivadas de las microsporas tras aplicar sobre las yemas un pretratamiento de frío de una semana de duración. Finalmente, como requisito previo para la edición genética de C. sativa mediante los sistemas CRISPR/Cas, y haciendo uso del protocolo de regeneración in vitro de plantas surgido de la presente Tesis Doctoral, se ha conseguido desarrollar por primera vez un protocolo para producir plantas de cannabis transformadas genéticamente de forma estable, lo que supone un hito histórico en la mejora genética de la especie. Después del cocultivo con A. tumefaciens y el posterior cultivo en medio de regeneración selectiva con antibióticos, los hipocótilos lograron respectivamente un 23,1% y un 5,0% de tasas de regeneración y transformación. En su conjunto, la presente Tesis Doctoral proporciona un abanico de herramientas biotecnológicas que permitirán el desarrollo de una nueva generación de variedades de cannabis de alto rendimiento, que presenten caracteres homogéneos, resistentes a múltiples estreses tanto bióticos como abióticos, y siendo así aptas tanto para un uso industrial como medicinal. / [CAT] Cannabis sativa L. (Cannabaceae) és una espècie angiosperma, alógama i dicotiledònia composta per varietats de dia curt i dia neutre que presenten exemplars dioics (mascles i femelles), i plantes monoiques. Entre les seues múltiples aplicacions destaquen tant el seu ús industrial com el seu ús medicinal. Tot i que el cànnabis ha sigut emprat per l'ésser humà des de temps ancestrals, la psicoactivitat que presenten algunes de les seues varietats, derivada del seu contingut en ¿9-tetrahidrocannabinol (THC), ha motivat la prohibició del seu cultiu durant gairebé seixanta anys. L'estricta fiscalització a la qual ha sigut sotmés el cànnabis, ha impedit que professionals de tot el món puguen dur a terme programes de millora genètica d'aquesta espècie, la qual cosa s'ha traduït en l'absència de varietats uniformes. En aquesta Tesi Doctoral s'han desenvolupat diferents eines biotecnològiques per a la millora genètica del cànnabis. En primer lloc, donada la falta de reproducibilitat d'alguns protocols de cultiu in vitro de cànnabis i la gran influència que el genotip exerceix en l'efectivitat d'aquests, es va avaluar la capacitat de regeneració in vitro de diferents explants. Com a resultat, s'ha desenvolupat un protocol lliure d'hormones a partir de hipocòtils de C. sativa que presenta altes taxes de regeneració (les quals oscil·len del 32,26% al 71,15%) en tots els genotips avaluats, presentant a més un 17,94% de taxa d'arrelat espontani dels regenerants. Al mateix temps, s'ha estudiat el patró polisomàtic de diferents explants de cànnabis i s'ha aconseguit regenerar, a partir d'aquests, un percentatge significatiu d'exemplars mixoploids (17,65% procedents de cotilèdons i 13,33% de hipocòtils) que, tal com descriu la bibliografia existent, podrien mostrar una major capacitat de síntesi de cannabinoids. D'altra banda, donada l'absència de publicacions científiques sobre aquest tema i el potencial que aquesta tècnica presenta per a pal·liar la variabilitat intrínseca d'aquesta espècie, s'ha desenvolupat l'estudi més profund fins hui relatiu a la biologia floral masculina de C. sativa. S'han descrit fins a 476.903 microspores i grans de pol·len per flor masculina, amb taxes de viabilitat in vivo de les microspores del 53,71 al 70,88%. A més, s'han correlacionat totes les etapes de desenvolupament del microgametòfit amb la longitud de la gemma, identificant intervals de longitud de gemma que contenen majoritàriament microspores vacuolades i grans de pol·len jove bi-cel·lular en tots els fenotips avaluats. D'aquesta manera, i encara que la presència de midó en les microspores i grans de pol·len de C. sativa segueix un patró similar a l'observat en espècies recalcitrants a la androgènesi, ha sigut possible abordar la inducció de la embriogènesi de microspores en aquesta espècie, aconseguint produir per primera vegada estructures multicel·lulars derivades de les microspores després d'aplicar sobre les gemmes un pretractament de fred d'una setmana de duració. Finalment, com a requisit previ per a l'edició genètica de C. sativa mitjançant els sistemes CRISPR/Cas, i fent ús del protocol de regeneració in vitro de plantes sorgit de la present Tesi Doctoral, s'ha aconseguit desenvolupar per primera vegada un protocol per a produir plantes de cànnabis transformades genèticament de manera estable, la qual cosa suposa una fita històrica en la millora genètica de l'espècie. Després del cocultiu amb A. tumefaciens i el posterior cultiu en medi de regeneració selectiva amb antibiòtics, els hipocòtils van aconseguir respectivament un 23,1% i un 5,0% de taxes de regeneració i transformació. En el seu conjunt, la present Tesi Doctoral proporciona un ventall d'eines biotecnològiques que permetran el desenvolupament d'una nova generació de varietats de cànnabis d'alt rendiment, que presenten caràcters homogenis, resistents a múltiples estressos tant biòtics com abiòtics, i sent així aptes tant per a un ús industrial com medicinal. / Galán Ávila, A. This diploma thesis is focused on the study olephylic formulations – ointments and cream type of V/O. This thesis gives out information about production, use and properties 3 cosmetic ointments and about properties baby´s skin cream and the study of stability of these products in the presence of antioxidants - tocopherol acetate and rosemary extract. Ointments are made from hemp oil, olive oil and shea butter and chamomile extract. Each ointment is produced in two versions. Once the use of vitamin E as an antioxidant, a second is used as an antioxidant extract from rosemary. Among other important substances with positive effects on human skin include, for example, saturated fatty acids such as palmitic acid and stearic acid, unsaturated fatty acids such as oleic, linoleic, and more. Ointments are suitable for greasing dry to very dry skin hands, face and other body parts. The thesis also discusses the current legislation, which producer of cosmetics must comply, the details which must be placed on the label of a cosmetic product and it contains proposals of cosmetic labels Further thesis includes studying methods for determination of lipid characterizations were used for study of behaviour of ointments during time and during storage at different temperatures. They include iodine, acid, peroxide and saponification values. The ester value and the content of glycerol were counted.
