Efeitos da Paullinia cupana e de seus principais compostos ativos na modulação da resposta imune / Effects of Paullinia cupana and its main active principles on the immune response modulation

A busca por substâncias imunomoduladoras oriundas de plantas medicinais é crescente devido à grande variedade de doenças relacionadas às respostas imunológicas desordenadas, como as autoimunidades e o câncer. Neste sentido, sabe-se que a Paullinia cupana, popularmente conhecida como guaraná, é uma planta medicinal que apresenta entre outros efeitos, propriedades quimiopreventiva e antitumoral em camundongos. Entretanto, até o presente estudo, não havia relatos na literatura a respeito dos efeitos desta planta sobre o sistema imune, conhecido por desempenhar papel fundamental não só no controle de infecções, mas também no combate ao câncer. Assim, o presente estudo avaliou os efeitos do pó das sementes de guaraná e de seus principais compostos ativos, cafeína e catequina, sobre o sistema imune de camundongos C57BL/6 através de protocolos preconizados pela Organization for Economic Cooperation and Development (OECD) com modificações. Os resultados obtidos mostraram que o guaraná, devido à ação majoritária da catequina, mas com a participação da cafeína também, diminui as respostas imunes, celular e humoral, verificada através da diminuição da expansão de linfócitos T frente a um antígeno específico, redução da resposta de hipersensibilidade do tipo tardia e título de anticorpos, diminuição dos pesos relativos do baço e timo e celularidade deste último, além de aumento de células imaturas no timo. Desta forma, os resultados aqui expostos demonstram, pela primeira vez, que a ingestão de guaraná reduz a imunidade adaptativa através da diminuição das respostas celular e humoral. Assim, futuros estudos deverão ser realizados, no sentido de se verificar a possibilidade do uso desta planta ou de seus princípios ativos no controle de doenças inflamatórias e autoimunes, caracterizadas por uma resposta exacerbada do sistema imune. / Seeking for immunomodulatory compounds derived from medicinal plants is increasing due to the huge variety of diseases related to disordered immune responses, such as autoimmunity and cancer. Therefore, it is known that the Paullinia cupana, popularly known as guarana, is a medicinal plant that has among other effects, chemopreventive and antitumor properties in mice. However, until the present moment, there were no reports in the literature about the effects of this plant on the immune system, which is known to play key role not only in the infectious control, but also in combat cancer. Thus, this study evaluated the effects of guarana seed powder and its major active principles, caffeine and catechin, on the immune system of C57BL/6 mice through protocols recommended by the Organization for Economic Cooperation and Development (OECD) with modifications. The results showed that guarana, due to the majority action of catechin, but also with the participation of caffeine, decreases the cellular and humoral immune responses, as indicated by the decreased expansion of antigen-specific T lymphocyte, reduced response delayed-type hypersensitivity and titer of antibodies, reduction in relative weights of thymus and spleen cellularity, besides increase of immature cells in the thymus. Thus, the results showed here demonstrate, for the first time, that the intake of guarana decreases the adaptive immunity by reducing cellular and humoral responses. Thus, futures studies should be conducted in order to better evaluate the use of this plant and/or its main active principle in the treatment of inflammatory and autoimmune diseases, which are characterized by exarcebated immune response.

Žolinių augalų panaudojimas kietajam biokurui / Herbal plants use for solid biofuel

Šio magistro baigiamojo darbo tikslas buvo ištirti trijų energetinių žolinių augalų biometrines bei energetines savybes ir jas palyginti tarpusavyje. Augalai buvo auginami lauko sąlygomis, kiekvieną augalą tręšiant skirtingomis mineralinio azoto normomis (N0 – kontrolė, N60 – 200 kg ha-1 ir N120 – 400 kg ha-1). Buvo tiriami pagrindiniai biometriniai rodikliai: augalų aukštis, augalų stiebų skaičius, sausoji biomasė, chlorofilo indeksas. Taip pat ištirtas pasirinktų augalų šilumingumas, peleningumas, pagrindiniai elementai (C, H, N, S, O) bei šalutiniai elementai (K, Ca, Mg, P, Al, Si, S), taip pat sunkieji metalai. Visų augalų energetinės savybės palygintos tarpusavyje. Vertinant gautus duomenis, tinkamiausi žoliniai augalai biokurui yra sida ir drambliažolė, kadangi jų peleningumas yra mažiausias. Tačiau nendrinio dryžučio, nors ir išsiskyrė didesniu peleningumu, šilumingumas buvo didžiausias. Cheminės sudėties analizės metu buvo nustatyta, jog papildomas tręšimas azotinėmis trąšomis įvairių elementų kiekiui augaluose didelės įtakos neturėjo, tačiau vertinant šalutinių elementų kiekį augaluose pastebėta tai, jog kai kurių cheminių elementų padidėjimą galėjo lemti ir augalų augimo vieta (atvira pieva, šalia žvyrkelio, šalia kelio, šalia dirbamo lauko ir pan.). / This master's thesis was to analyze three energy plant grass biometric and energy properties and to compare them with each other. Plants were grown in field, every plant fertilized with different rates of nitrogen (N0 - control, N60 - 200 kg ha-1 and N120 - 400 kg ha-1). It was investigated the main biometric parameters: plant height, plant stems, dry biomass and chlorophyll index. Also plants calorific value, ash content, the main elements (C, H, N, S, O) and minor elements (K, Ca, Mg, P, Al, Si, S) and heavy metals. Plant‘s energy properties were compared with each other. Evaluating the data, the optimum herbaceous plant for biofuels are sida and Miscanthus as their ash content is lowest. Reed canary grass calorific value was highest, but it stood higher ash content too. Chemical composition analysis showed that the additional nitrogen fertilization had no major impact of the various elements in plants, but by assessing the amount of elements in plants is noticeable that plant growth and position may impact some chemical elements increase (open meadow, near the gravel, next to the road, arable land, etc.).

Determining the content of toxic elements (Pb, Cd, and As) in herbal plants collected from different sites in northern Vietnam

Plants might contain heavy metals from the surrounding polluted environment. Medicinal herb and plants, commonly used in Vietnam, may pose a risk to public health when polluted with heavy metals such as Pb, Cd and As. This study aims to investigate the content of Pb, Cd, and As in five selected herb plants (Phyllanthus urinaria L., Plantago asiatica L., Eleusine indica L., Wedelia chinensis (Osbeck) Merr, and Artemisia vulgaris L.). The samples were collected from natural sites in some provinces in northern Vietnam. The concentrations of Pb, Cd, and As were determined by the Inductively coupled plasma mass spectrometry (ICP-MS) method. The content of Pb, Cd and As was 0.247 ÷ 32.080 mg kg-1, 0.000 ÷ 1.099 mg kg-1, 0.000 ÷ 2.261 mg kg-1, respectively. A total of 50 out of the 58 investigated samples had concentrations of Pb, Cd, As lower than the permissible values of the World Health Organization (WHO). The remaining samples had contents of Pb or Cd or As significantly higher than the permissible limit defined by WHO. / Thực vật có thể bị ô nhiễm do hấp thụ kim loại nặng từ môi trường ô nhiễm xung quanh. Các loại cây thảo dược được sử dụng phổ biến làm thuốc chữa bệnh ở Việt Nam. Chúng có thể gây nguy hiểm cho sức khỏe cộng đồng khi bị ô nhiễm kim loại nặng (Pb, Cd…vv.). Nghiên cứu này nhằm điều tra hàm lượng Pb, Cd và As trong năm loại cây thảo dược (cây Diệp Hạ Châu, cây Mã Đề, cây Cỏ Mần Trầu, cây Sài Đất và cây Ngải Cứu). Các mẫu cây này được lấy từ các địa điểm mọc tự nhiên ở một số tỉnh khác nhau thuộc khu vực miền bắc Việt Nam. Hàm lượng của Pb, Cd, và As được xác định bằng phương pháp ICP-MS. Hàm lượng Pb, Cd và As trong các mẫu phân tích lần lượt nằm trong khoảng là 0.247 ÷ 32.080 mg. Kg-1, 0.000 ÷ 1.099 mg. Kg-1, 0.000 ÷ 2.261 mg. Kg-1. 50 trên 58 mẫu phân tích có hàm lượng Pb, Cd, As thấp hơn giới hạn cho phép do tổ chức y tế thế giới (WHO) quy định. Có 8 trên 58 mẫu cây phân tích có hàm lượng Pb hoặc, Cd hoặc As cao hơn tiêu chuẩn giới hạn của WHO.
