Reaching College Students Where They Live:Grayson, Andrea 17 June 2008 (has links)
The purpose of this research study was to ascertain the relative level of adoption of the use of web-based media by undergraduates, the results of which will be considered in the creation of health promotion messages and campaigns that are distributed through electronic means to a campus audience. The primary research conducted was a web-based survey of UVM undergraduates, inquiring about the extent to which they consume web delivered media programming, of both news and entertainment, and how they communicate with their peers. Through the use of descriptive statistics, it was learned that more than half of UVM undergraduates (58.7%) watch between one to five minutes of web-based video on a weekly basis, suggesting that the creation of video-based health promotion programming might indeed prove to be an effective approach to raising awareness and promoting behavior change for this population. Additional data reveals how much time respondents do a variety of media activities, as well as their most used means of communication with peers when not with them in person. The study concludes that there is a high enough adoption of the use of web-based media by undergraduates to warrant creating health promotion messages and campaigns that are distributed through electronic means to a campus audience.

Probing early stage aggregates of amyloidogenic proteins using mass spectrometry based methodsPhillips, Ashley January 2017 (has links)
Mass Spectrometry (MS) and Ion Mobility - Mass Spectrometry (IM-MS) can be used to investigate protein structure and dynamics and are ideally positioned to study intrinsically disordered and amyloidogenic proteins, whose diverse conformational space and/or oligomeric state is hard to track accurately. This thesis uses hybrid MS approaches including IM-MS, Cross-linking IM-MS and ECD-FT-ICR MS to probe the structure of alpha-Synuclein and Amyloid-beta (Abeta). For alpha-Synuclein, the effect of solution pH and ionisation polarity on the species observed by MS and IM-MS is investigated. Conformational families observed by Cross-linking IM-MS provides a link between the solution and gas phase structures of alpha-Synuclein observed here and our data correlates with that reported by other groups. MS, IM-MS and HDX-MS are used to probe alpha-Synuclein during the early stages of aggregation. A specific aggregation competent conformer is not observed suggesting that the solution constituents remain conformationally dynamic. We observe shifts in the species observed by MS and IM-MS between samples and our data contributes to an array of conflicting structural studies indicating that alpha-Synuclein adopts a diverse range of species with significant variation. For Abeta(1-42) and Abeta(1-40) Collision Induced Unfolding and ETD/ETcaD demonstrate that Abeta(1-42) adopts a compact conformation bound by intramolecular interactions. Changes to the Abeta(1-42) and Abeta(1-40) ATDs following SID are correlated to known structure influencing intermolecular interactions and demonstrate the large structural difference between Abeta(1-42) and Abeta(1-40) despite differing by only two C-terminal amino acids. IM-MS is used to classify the mode of action of anti-aggregation drug candidates on Abeta(1-42). The anti-aggregation capacity of the retro-inverso peptide, RI-OR2 is shown to result from inducing the compaction or extension of Abeta(1-42), preventing the adoption of an aggregation competent structure. In contrast, the flavonoid Rutin is shown to act solely through inducing Abeta(1-42) compaction. This thesis demonstrates the power of MS based methods to investigate the diverse range of structures of intrinsically disordered aggregating proteins implicated in disease.

<p>Opera är främst en webbläsare, men innehåller även klienter för bl.a. mail och chat. Innan den här rapporten skrevs saknades dock en klient för instant messaging-protokollet Jabber. Denna rapport beskriver arbetsgången och resultatet av utvecklandet av en Jabberklient till Opera.</p><p>Eftersom det inte fanns nog med tid att utveckla en fullfjädrad Jabberklient beslutades det att målet skulle vara ett s.k. ”proof of concept”. Kraven sattes till grundläggande instant messagingfunktionalitet, som närvarostatushantering och möjlighet att skicka och ta emot meddelanden.</p><p>Målet uppfylldes och resultatet blev en Jabberklient med grundläggande funktionalitet. I enlighet med direktiven lades relativt lite fokus på gränssnittet. Klienten har kända brister och användarvänligheten går att öka.</p><p>Resultatet skulle kunna utgöra grunden för en komplett Jabberklient. Kvar att göra är att förbättra gränssnittet för att matcha resten av Opera i användbarhet. Alla krav i Jabberspecifikationen bör också uppfyllas, speciellt eftersom företaget är stolt över att Opera följer standarder väl1. För att inte vara sämre rustad än andra Jabberklienter bör även extra funktionalitet läggas till, såsom samtalshistorik, filöverföring och möjligheten att använda andra instant messaging-protokoll via Jabberkontot.</p> / <p>Opera is first and foremost a web browser, but it also has e-mail and chat clients. Before this thesis was written however, it did not have a client for the instant messaging protocol Jabber. This thesis describes the workflow and the results of the development of a Jabber client for Opera.</p><p>Since there wasn’t enough time to develop a full-featured Jabber client, it was decided that the goal would be a proof of concept. The requirements were basic instant messaging functionality, which means the proper handling of presence information and the ability to send and receive instant messages.</p><p>The goal was achieved and the result was a Jabber client with basic functionality. In accordance with the directives, the user interface was not a primary focus. The client has known flaws and the usability is not as good as it could be.</p><p>The result could be used as the foundation of a releasable Jabber client. One of the things left to do is to improve the user interface so it matches the usability of the rest of Opera. All the requirements of the Jabber specification should also be fulfilled, especially since the company takes pride in the standards compliance of Opera2. In order to match the features of other Jabber clients, extra functionality should be added as well. Some examples would be message history, file transfer and the ability to use other instant messaging protocols through the Jabber account.</p>

Development of a MALDI-Ion Mobility-Surface-Induced Dissociation-Time-of-flight-mass spectrometer for the analysis of peptides and proteinsStone, Earle Gregory 30 September 2004 (has links)
Peptide sequencing by surface-induced dissociation (SID) on a MALDI-Ion Mobility-orthogonal-TOF mass spectrometer is demonstrated. The early version of the instrument used for proof-of-concept experiments achieves a mobility resolution of approximately 20 and TOF mass resolution better than 200. Peptide sequences of four peptides from a tryptic digest of cytochrome c (ca. 1 pmol deposited) were obtained. The advantage of IM-SID-o-TOFMS is that a single experiment can be used to simultaneously measure the molecular weights of the tryptic peptide fragments (peptide mass mapping) and partial sequence analysis, (real time tandem mass spectrometry.) Optimization of the MALDI-IM-SID-o-TOF mass spectrometer for peptide sequencing is discussed. SID spectra obtained by using stainless steel, Au grids, and fluorinated self-assembled monolayers (F-SAM) on Au are compared. Optimum collision energies differ for the various surfaces. The fragmentation patterns observed for a series of peptides and protein digests using the Nd:YAG laser (355 nm) for MALDI ion formation and an FSAM surface for ion activation is compared to the fragmentation patterns observed for CID and photodissociation. The fragmentation patterns observed in all cases are strikingly similar. Photodissociation produced a greater abundance of ions resulting from side-chain cleavages. As a general rule optimized SID spectra contain fewer immonium ions than either photodissociation or CID. Evaluation of an instrument incorporating a new hybrid drift cell is discussed. Spectra for a digest of hemoglobin is compared to that acquired with an ABI 4700 TOF-TOF. The performance of the instrument is also evaluated using a micro-crystal Nd:YAG laser (355 nm) for MALDI operated at 400 Hz. Experiments were performed to determine the sensitivity and overall performance of the instrument. The reproducibility of the MS/MS spectra for gramicidin S is shown to be 94% run-to-run. The best mobility resolution obtained for a neat deposition of the dye Crystal Violet was 60 t/∆t. Sensitivity was tested with the peptide fibrinopeptide A (m/z 1537, AA sequence ADSGEGDFLAEGGGVR). Data acquired for sixty seconds with approximately sixty femtomoles deposited. Abundant [M+H]+ ions where observed as well as [M+H]+-NH3 ions. The S/N for this short run was insufficient to identify any SID fragments

This paper was applied on the methods of ¡§Interactive Management¡]IM¡^ and Nominal Group Technique¡]NGT¡^¡¨. According to the NGT twice meetings, there are 10 strategies that can improve the development of Kaohsiung City.
The APROC plan was authorized by the Executive Yuan on June, 1995. Kaohsiung city government was assigned to renew urban land at Kaohsiung Multi-functional Commerce & Trade Park first on June, 1996.
From the study, the researcher realizes the advantages of importance, background and exploitation problem. Landlords, investors and Kaohsiung city government accorded in this exploitation plan or not that are based on understanding, negotiation and communication. Meanwhile, it was hard to break out in Three Links which have made investors lack of confidence. It makes many obstacles in improving the development of Kaohsiung city after Cross-strait in accession to the WTO. Therefore, it is necessary to explore moreover.
This paper has utilized literary reviews about Kaohsiung Multi-functional Commerce & Trade Park and interviews to collect data.¡@According to the report about ¡§Cross-strait in WTO¡¨, it is also helpful to understand the pro. Eventually, these suggestions from our study, sincerely hope, could be useful in formulating the strategies and regulations of Kaohsiung city government.

An Analysis of and Anticipation for the Influences of Penghu's Gambling Industry on Its Economy and Society AbstractLi, Yu-chu 13 July 2009 (has links)
For years Penghu has been actively developing its tourism gaming (gambling) industry since its location, climate, and soil quality have hindered the development of its agriculture, fishery, and local industries and resulted in slow economy and reduced population. Its tourism industry had not developed well despite the unique natural sceneries and local history; therefore, the local government placed hope on the gambling industry in the wish of attracting foreign investors. For the past two decades, the local effort of legalizing off-shore gambling -- the only and the most important force behind this policy, has never ceased.
The scenario adopted in this study is that the draft of the "Offshore Islands Development Act" that serves as the legal basis of Penghu's development of the gambling industry has been approved by the Legislative Yuan and is supported by the majority in a public vote. Being the only area where legalized gambling is allowed, the local government of Penghu and the central government immediately start to work on the applicable legal basis and regulations. Born and raised in Penghu, the researcher of this study also cares very much about this topic and thus wishes to come up with the recommendations for optimal local development.
In this study the "Nominal Group Technique" (NGT) of "Interactive Management" (IM) was utilized for the purpose of getting everyone's opinions across in a democratic manner and reducing egocentric and arbitrary perspectives. Through NGT, an interactive seminar is held where "feasible and specific recommendations for the influences of Penghu's gambling industry on its economy and society" are provided. The 2-stage Seminar is held based on the unique procedures of NGT: Identify problems, clarify problems, vote and select problems, and come up with 12 specific strategies:
1. Invest a certain portion of the gambling revenue in the local environment in order to maintain the local landscapes and culture.
2. Establish a Penghu Gambling Committee or a dedicated authority that carries out its duties objectively and wins public trust and support.
3. Give a certain portion of the jobs in the gambling industry to the local residents whose households have been registered in Penghu for at least 5 years.
4. Make timely plans that are dedicated to Penghu's environment and surrounding. Gambling authorities should hold studies on the size of the gambling market.
5. Establish a security system between a service provider and a security authority and invest more in software/hardware for crime prevention.
6. Reinforce personnel training and industrial-academic collaborations in order to provide more jobs.
7. Establish a Supervisory Committee that is in charge of analyzing/evaluating relevant policies and formulating tasks.
8. Formulate strategies that protect the development of local industries and create a win-win situation for them and foreign inventors.
9. Penghu County Government should formulate a public-supervision system for the gambling industry in order to prevent corruptions and political manipulation.
10. Complete the infrastructure and public transportation as quickly as possible.
11. Actively help Penghu's traditional industries and develop tourism with the gambling industry and create a win-win situation.
12. Evaluate the influences of the population-carrying capacity on infrastructure and ecosystems and determine the responses.
After comparing all the strategies, the inter-strategic relationships are reinforced using "Interpretive Structural Modeling" (ISM), and the ISM software was used to draw the diagram of the structural enhancement. After the second-stage discussions and revisions, the final, revised Diagram of Enhanced Structure was completed that clearly shows the causal relationships between each strategy and how they are ordered and ranked. The main contribution of this study is the information found provides the government with a clear, prioritized set of instructions that help the authority understand how the 12 strategies work and what they actually achieve.
Through the 2-stage interactive seminar, questionnaire surveys were conducted twice on the participants in order for us to understand their expectations for the actual procedures, participation, understanding, and interactions in the seminar, and such information helps us understand how satisfied they are towards the interactive learning and whether they have actually achieved the expected results through independent thinking, diverse perspectives, debates, listening, group discussions, brain storming, and communication. In other words, we wish to find out how effective participation, problem-defining and clarifications, and consensus-reaching benefit the entire effort.

Cross-border PAM Analysis ¡V A Study on the Integration of Public Demand with Government Policy under Regional Cooperation between Penghu and China's Economic Zone on the West Coast the Taiwan StraitWu, Cheng-yu 30 August 2009 (has links)
This study adopts a variety of research methods widely used in professional
academic fields covering regional science, management science and policy science.
The essence of each method is selected for cross-field analysis that is beyond the
framework of different fields, departments and layers. Through the public affairs
management (PAM) method, the reference architecture is integrated for analysis, and
the output of regional analysis (including comparative analysis, economic foundation
analysis, variance appropriation analysis, time series analysis and cross-sectional
analysis, gravity model) is used as input data for the project analysis (the nominal
group technique). The outputs of regional analysis and project analysis are used as
input for the interactive management (IM) method in a policy analysis; the outputs of
the first three analyses are further used as the input to the social judgments theory
(SJT) of the judgment analysis. A set of proposals is then created using VCS, and the
survey questionnaire is created using SJT and multi-attribute utility theory (MAUT)
by IM participants in generating strategies. These are sent to 53 public administrators
in the Penghu County Government for them to fill out. The results are analyzed using
the serial intention structure of the SJT method to identify the cooperation elements
among all units. The historical data in Penghu is then further analyzed using
appropriate research methods to calculate a set of objective and mutually supporting
data. The participants¡¦ psychological perception are further quantified with all kinds
of resource links swept across each node, enabling different types of energy to diffuse
to their maximum for creation of the greatest possible public values.
This study was conducted by a team of three students taking an interactive
approach to exploring issues regarding the impact of strategy in cooperation between
Penghu and China's Economic Zone on the Western Side of the Taiwan Strait. The
team worked together to conduct literature research, examining issues related to
public living within the aspects of economy, society and government. The study uses
the interactive management (IM) method widely used in policy analysis, together with
process analysis methods such as the nominal group technique (NGT) method and the
interpretive structural modeling (ISM) method, with 26 participants from various
sectors. In the NGT process, starting from idea generation, problem clarification, and
voting on problems, the issues were compared pair-wise. ISM software was used for
generating an enforcement structural diagram, which was reviewed by participants;
over half approved on the twelve strategies in the governmental aspect of cooperation
between Penghu and China¡¦s Economic Zone on the Western Side of the Taiwan Strait
study, in addition to the revised and approved enforcement structural diagram.
This paper focuses on using the IM method for analysis. During the planning
stage, information generated by varieties of research methods was used for deliberate
review by the parties of interest. During the interactive test stage, the purpose was to
make sure that the 26 participants are representative of the public. Therefore, 15-20
observers came from the same population for simultaneous voting and pair-wise
comparison of rules. The results produced were almost identical, indicating that the
participants invited in the IM method were representative enough. In the follow-up
stage, the Penghu Cable Company was supportive to air the promotional interactive
DVD, with contents connoting public welfare and social education, for free for one
month from April 1, 2009 to April 30, 2009. Afterwards, the twelve major strategies
as voted were sent to 10 administrative units, of which five replied and agreed to
adopt; More voices of the majority of people will then be heard, effective opinions
adopted and, together with cross-border governmental policies and people¡¦s needs, the
public opinions will be blended into public policies. The method will serve as a bridge
between the Government and the public, enabling people to do a final evaluation on
the government's performance or even facilitating the Central Government to listen to
people¡¦s responses when allocating subsidy funds to cities and counties. tThis study
will propose a concrete strategy on ¡§competitiveness in cooperation between Penghu
and China's Economic Zone on the Western Side of the Taiwan Strait¡¨, by surveying
county government public administrators with SJT and MAUT questionnaires. A total
of 53 county government public administrators generally agreed that the Government
should proactively expand services to improve the employment rate.
In summary, if the governmental sector is able to take the public interests and
county public services as the top priority issues, as well as expressing a willingness to
listen to people¡¦s opinion and incorporate people¡¦s needs into the policies, a
flourishing and peaceful realm of the people, by the people, for the people and based
on the people is achievable in the near future.
