Na aqüicultura, o tratamento com inseticidas pode ser realizado com a aplicação direta na água, ou administração na ração dos animais. Apesar de muito utilizados, poucos são os trabalhos sobre ecotoxicologia, comportamento e bioacumulação do diflubenzuron (DFB) e teflubenzuron (TFB) na água e seus efeitos em organismos aquáticos de clima tropical. Os objetivos deste trabalho foram: estimar a toxicidade (CL(50-96h)) do DFB e TFB para o pacu (Piaractus mesopotamicus), calcular o risco alimentar para o consumo humano, determinar resíduos de DFB e TFB na água, sedimento e músculo de pacus, após a exposição aos inseticidas na água ou na ração dos peixes e analisar a atividade hepática das enzimas glutationa-S-transefrase (GST) e catalase (CAT) em pacus alimentados com ração acrescida de DFB ou TFB. A CL50;96h do DFB para o pacu foi superior a 1200 mg.L-1 e a do TFB, superior a 1000 mg.L-1. Os resíduos de DFB foram detectáveis até 7 dias após a aplicação (com 18,3% de resíduo) e para o TFB, foram detectáveis ate o terceiro dia (8% de resíduo). No sedimento, as maiores concentrações de DFB e TFB foram observadas nas primeiras 24 h e em todos os períodos amostrados foram detectados resíduos dos inseticidas. Nos filés dos pacus foram detectados resíduos dos inseticidas após a alimentação com a ração tratada e após aplicação de DFB e TFB na água. A ingestão diária estimada (IDE) de DFB e TFB encontra-se acima dos valores de ingestão diária aceitável (IDA), mesmo para as amostras com menores valores de resíduos detectados nos filés do pacu. In aquaculture these pesticides can be injected directly in the water or in the fish food. Besides their large utilization, there are few works about the effects of these insecticides on the fish metabolism and biomagfication, especially in tropical aquatic organisms. This study aims to: evaluate the acute toxicities (LC(50-96h)) in pacu (Piaractus mesopotamicus), measure the risk food for human consumption, determinate residues of DFB and TFB in water, sediment and muscle of pacus after the management of the insect-powders on water or in the fish food and to analyze the hepatical activity of the enzymes catalase (CAT) and glutathione-S-transferase (GST) in fishes fed with ration plus DFB or TFB. The LC((50- 96h)) of DFB and TFB for the pacu was higher than 1.200 mg.L-1 and 1.000 mg.L-1, respectively. The residues TFB were detected until the thirth day (8% of residue) and DFB until seventh day after application in the water (18,3% of residue). In the sediment, the highest concentrations of DFB and TFB had been observed in the first 24 h and in all the samplings there were . In muscle of pacus, residues of the insecticides had been detected after the feeding with the ration treated and after application to DFB and TFB in the water. The daily ingestion estimate (DIE) of DFB and TFB meets above of the values of acceptable daily ingestion (DAI), exactly for the samples with lesser values of residues detected in filés of pacu. For the enzymes the CAT activity was not affected by both pesticides, but the GST activity was reduced five days after the feeding. Resumo: Uma preocupação atual consiste no uso indiscriminado de muitas substâncias sintéticas no controle de pragas na agricultura. Estudos atuais indicam que algumas dessas substâncias podem estar envolvidas em casos de intoxicação de insetos, como as abelhas, que são fundamentais em muitas culturas, devido à atividade de polinização realizada por elas. No Brasil, um dos inseticidas mais usados nas culturas de café, citrus, algodão e cana-de-açúcar é o imidaclopride, que apresenta ação neurotóxica para os insetos. Seus metabólitos circulam por todo o organismo e entram em contato com diversos órgãos. A exposição a doses subletais deste inseticida pode provocar mudanças na fisiologia e no comportamento das abelhas, sendo que tais alterações podem ser mais intensas ou não, dependendo do período de exposição. Dessa maneira, o presente estudo teve como objetivo avaliar os efeitos da exposição crônica a doses subletais (DL50/100, DL50/50 e DL50/10) do inseticida imidaclopride, no cérebro, ventrículo e túbulo de Malpighi de Apis mellifera africanizada. Para isso os órgãos estudados das abelhas expostas e dos controles foram submetidos a análises morfológica, histoquímicas e imunocitoquímica, após 1, 3, 5, 7 e 10 dias de exposição. No cérebro das abelhas expostas foi possível observar alterações morfológicas, histoquímicas e imunocitoquímicas nos lobos ópticos de todos os grupos expostos, enquanto que nos corpos pedunculados observou-se tais alterações apenas no grupo exposto à DL50/10. Já nos lobos antenais as alterações não foram observadas. No ventrículo das abelhas de todos os grupos expostos ao inseticida, a análise morfológica permitiu observar de uma maneira geral um aumento na quantidade de células eliminadas no lúmen, na quantidade de secreção apócrina eliminada no lúmen e na quantidade de halos pericromatínicos nas células ... Recent studies indicate that some of these substances are involved in cases of poisoning bees, which are fundamental in many cultures due to the activity of pollination. In Brazil, an insecticide widely used on crops of coffee, citrus, cotton and sugar cane, is imidacloprid, which is neurotoxic to insects. However, its metabolites circulate throughout the body and come into contacting with another organs. Exposure to sublethal doses of this insecticides causes changes in physiology and behavior of bees, and these changes can be more intense depending on exposure period. Therefore, this study aimed to evaluate the effects of chronic exposure to sublethal doses (DL50/100, DL50/50 and DL50/10) of imidacloprid in the brain, midgut and the Malpighian tubules of africanized bees (Apis mellifera). For this, the organs of treated and untreated bees were subjected to morphological, histochemical and immunocytochemical techniques, dissecting the bees after 1, 3, 5, 7 and 10 days. In the brains of exposed bees it was possible to observe morphological, histochemical and immunocytochemical alterations on optic lobes of all bees, whereas in the mushroom bodies, alterations was observed only in the group exposed to DL50/10. The antennal lobe presented no alterations after the treatment in all doses. In the midgut of all exposed groups of bees, morphological analysis showed in general an increase in the number of digestive cells and apocrine secretion eliminated in the lumen and in the number of peripherical chromatin halos were also increased. The histochemical techiniques confirmed the presence of pyknotic nuclei, showing the presence of nuclear fragments and degenerated areas in cytoplasm of digestive cells. Mecanismos de toxicidade do inseticida imidacloprido no fígado de rato

Orientador: Fábio Erminio Mingatto / Resumo: O imidacloprido (IMD) é um inseticida neonicotinóide largamente utilizado em diversas culturas agrícolas e em animais para o controle de pragas. O IMD é rapidamente absorvido pelo trato gastrointestinal e por contato, sendo rápida e uniformemente distribuído nos órgãos e tecidos. Dados da literatura mostram que as concentrações mais elevadas foram observadas nos órgãos de eliminação: fígado e rins. O fígado é o principal órgão envolvido na biotransformação de substâncias exógenas (xenobióticos), convertendo compostos hidrofóbicos em hidrossolúveis, mais facilmente eliminados pelo organismo. Vários estudos vêm sendo conduzidos sobre os efeitos tóxicos do IMD em animais, causando danos ao fígado. Nesse sentido, o objetivo desse estudo foi avaliar os mecanismos envolvidos na toxicidade do IMD sobre a bioenergética de mitocôndrias e hepatócitos isolados de rato e ações do IMD sobre o metabolismo de carboidratos e proteínas em fígado de rato em perfusão. Em mitocôndrias isoladas, o IMD promoveu uma diminuição dose-dependente no estado 3 da respiração e na produção de ATP, sem afetar o potencial de membrana mitocondrial. Experimentos subsequentes medindo o consumo de oxigênio mostraram que o IMD não afeta a cadeia respiratória e que seu efeito é semelhante ao da oligomicina (inibidora da FoF1-ATP sintase) e/ou ao do atractilosídeo (inibidor do translocador de nucleotídeos de adenina, ANT). FATE, TRANSPORT, AND MIXTURE TOXICITY OF TRANSGENIC BACILLUS THURINGIENSIS CORN (ZEA MAYS) AND ASSOCIATED INSECTICIDES

Corn is a dominating feature in the landscape of the Midwestern United States. Associated with this crop are a plethora of products from fertilizers to pesticides that help farmers maximize grain yield while minimizing costs. A widely accepted form of protection from major pests in the United States is genetically modified corn that has been altered so that it contains genetic material from another species, a soil bacterium, Bacillus thuringiensis Berliner (Bt corn), in order to produce proteins that are toxic to some insect pests. Additional insecticides that are commonly used to protect corn include a neonicotinoid seed coating, clothianidin, and a pyrethroid insecticide applied in-furrow, tefluthrin. The goal of my dissertation was to measure the environmental fate of the Cry1Ab Bt corn protein, clothianidin, and tefluthrin and determine if measured concentrations could be causing effects to non-target species. The Cry1Ab proteins persisted only during the corn growing season in soil, runoff water and sediment with the highest concentrations measured during pollination. Clothianidin was detected in all matrices and remained persistent throughout the year in soil pore water. Tefluthrin was consistently detected in soil, runoff water, and runoff sediment during the corn growing season, but was not found in groundwater or soil pore water. No acute toxicity was observed to any species when exposed to the Cry1Ab protein in corn plant leaf tissue, therefore little risk was anticipated. Non-target species exposed to clothianidin resulted in limited ecological risk from field exposures. Tefluthrin results indicated elevated risk for non-target species exposed to tefluthrin at concentrations that were measured in the field study. There was no increase in toxicity to tefluthrin when non-target species were exposed to a combination of all three insecticides. In summary, the genetically modified corn insecticidal proteins and clothianidin were not found at environmental concentrations exceeding benchmark values for ecological effects, but tefluthrin was consistently detected in the environment at levels that could be causing toxicity to non-target species, especially if it is able to travel off-site.
