Acute Brachial Artery Responses to Endurance and High-Intensity Interval Exercise in Young Healthy MalesMcGill, Greg M. 04 1900 (has links)
<p><strong>Purpose: </strong>Habitual aerobic exercise improves vascular function; however, the acute vascular response to exercise is poorly understood. The present investigation compared the time course of acute changes in vascular function following two different types of aerobic exercise. <strong>Methods: </strong>Ten untrained males (23 ± 2 yrs) completed one bout of sustained moderate-intensity cycling (END) (30 mins at 55% peak power) or high-intensity interval (HIT) cycling (10 one-minute intervals at 80% peak power) on different days. Endothelium-dependent dilation was assessed by brachial artery flow-mediated dilation (baFMD) at baseline, immediately post-exercise, 1 hour post-exercise and 24 hours post-exercise. Endothelium-independent dilation was assessed via nitroglycerin (NTG) at all time points, except 1 hour post-exercise. <strong>Results:</strong> baFMD values were not significantly different between END and HIT at any time point. Immediately post-exercise baFMD values were unchanged from baseline. 1 Hour post-exercise, relative (p £ 0.001) and absolute (p £ 0.05) baFMD values were attenuated compared to all other time points for both HIT (%FMD baseline: 5.9 ± 2.3%; 1 hour post-exercise: 2.5 ± 1.5%) and END (%FMD baseline: 6.8 ± 2.4%; 1 hour post-exercise: 2.6 ± 1.9%). Relative (p £ 0.05) and absolute (p £ 0.05) NTG responses were attenuated immediately post-exercise compared to baseline for both HIT (%NTG baseline: 18.8 ± 4.4%; immediately post-exercise: 12.3 ± 3.1%) and END (%NTG baseline: 18.3 ± 3.1%; immediately post-exercise: 10.9 ± 4.9%). <strong>Conclusions: </strong>Immediately post-exercise, endothelium-dependent dilation is maintained; but reduced 1 hour following exercise cessation. This thesis is missing page 63 from all copies. -Digitization Centre / Manipulating carbohydrate (CHO) availability has been shown to alter acute exercise-induced changes in metabolic gene transcription and training-induced changes in oxidative capacity. The present study examined the effect of CHO availability on signalling pathways linked to mitochondrial biogenesis in response to high-intensity interval exercise (HIE). We hypothesized that reduced CHO availability would augment phosphorylation of AMP-activated protein kinase (AMPK), calcium/calmodulin-dependent kinase II (CaMKII), and p38 mitogen-activated protein kinase (p38) in response to HIE. Ten active men performed two experimental trials in random order, separated by 2:1 wk. During each trial, subjects performed two HIE sessions separated by 3 h (AM and PM sessions). Exercise sessions consisted of 5 x 4 min cycling bouts at a workload that elicited approximately 90% V02peak, with 2 min rest periods. Between sessions, subjects ingested -1.2 g CHO/kg b.w./h (HI-HI) or a taste-matched, non-energetic placebo (HI-LO). Muscle biopsies and blood samples were obtained before (Pre) and after (Post) the AM and PM HIE sessions. AMPK, CaMKII, and p38 MAPK phosphorylation increased from AM Pre to AM Post (p<0.01). During the PM exercise session, p38 phosphorylation increased in the HI-LO condition (-4.5-fold, p<0.001), whereas the HI-HI condition remained unchanged. PM HIE significantly increased CaMKII phosphorylation independent of condition, while no exercise or condition-mediated AMPK effects were observed. In summary, restricting CHO availability following an acute session of HIE augmented the exercise-induced increase in p38 phosphorylation during a subsequent HIE session. It remains to be determined whether chronic changes in p38 MAPK signalling are mechanistically linked to altered skeletal muscle remodelling observed after CHO-restricted exercise training. / Thesis / Master of Science (MS)

Post-Exercise Hypotension in Brief ExerciseBush, Jeremiah G. 01 May 2011 (has links)
The purpose of this investigation was to examine whether a single 10 minute bout of exercise, performed at multiple intervals throughout the day to equal 30 minutes, can effectively elicit post-exercise hypotension (PEH). Secondly, it is important to explore whether a light (40% VO2R) or moderate (70% VO2R) intensity is required to elicit PEH within 10 minutes. Subjects (N=11) completed a VO2max test utilizing the Bruce Treadmill protocol. Each subject returned within 3 – 5 days to complete two separate exercising trials. A counter balanced system was employed so that each subject did not perform the same intensity rotation (Counter Balance 1 = 40% VO2R and 70% VO2R for session 1 and session 2, respectively; Counter Balance 2 = 70% VO2R and 40% VO2R for session 1 and session 2). The first session consisted of 3 sessions (morning, noon, evening) separated by an average of 3.5 hours at one of two intensities (40% VO2R or 70% VO2R). The second group of sessions were performed identical to the first, however, the intensity was altered depending upon counter balance. Baseline BP was measured prior to exercising. After each session, BP was measured at 2 intervals for the morning and noon sessions (immediately following and 20 minutes post-exercise); and at 3 intervals for the evening sessions (60 minutes post-exercise added) for both intensities. At 40% VO2R, BP decreased significantly at the morning (p = 0.007), noon (p = 0.018) and evening (p = 0.010) sessions at the 20 minute post-exercise interval. Although not significantly different, BP was observed to be lower at 60 minutes post-exercise interval. During the 70% VO2R session, BP was significantly lower at the morning 20 minute (p = .029) and evening 60 minute post-exercise measurements (p = .006) when compared to baseline. There was no significant difference noted between 40% and 70% VO2R intensities at eliciting a drop in BP at any interval at any time point. Although not statistically significant, 70% VO2R appeared to produce a further decrease at the 60 minute post-exercise measurement (102 mmHg) than did the 40% session (106 mmHg). The results of this study indicate that PEH may be elicited after a single 10 minute exercise session. Furthermore, multiple bouts of 10 minutes produce an accumulated decrease in BP that can be observed at the completion of the day.

O exercício físico aeróbio é considerado uma importante estratégia por promover melhora na função cardiovascular, respiratória e metabólica. Nenhum estudo avaliou a influência do exercício físico intervalado sobre os níveis de NO, receptor de leptina e pressões respiratórias em mulheres na menopausa. Assim, o objetivo deste estudo foi avaliar os efeitos do treinamento físico aeróbio intervalado nos níveis plasmáticos do receptor solúvel de leptina (Rb-e) e nitrito/nitrato sua correlação com a pressão arterial em mulheres no climatério. Participaram deste estudo, 23 mulheres (55±5 anos) treinadas por 12 semanas, frequência de 3 vezes por semana, duração de 30minutos, com intensidade entre 60% a 80% da frequência cardíaca de reserva. Medidas antropométricas (IMC e CA), cardio-respiratórias (teste de capacidade aeróbia e aferições da pressão arterial e respiratória) e bioquímicas dos níveis plasmáticos do receptor de leptina e nitrato/nitrito foram realizadas. Nossos resultados mostram que as mulheres no climatério apresentaram: reduções significativas da circunferência abdominal (3,8%) e redução de 6,7% na pressão arterial diastólica. Observamos também aumento no VO2max.,(25,1+0,8 para 26,5+0,9ml/kg/min) e nas pressões expiratória e inspiratória (11 e 20%, respectivamente). De maneira similar, tanto os níveis plasmáticos do receptor de leptina (basal: 47,2+3,5 TR: 54,1+ 3,6ng/ml) quando de nitrito/nitrato (55%) foram aumentados pelo treinamento físico aeróbio intervalado por 3 meses. Although its is well known that physical exercise plays an important role in preventing and/or reducing the deleterious effect of cardio-metabolic diseases, no studies have been investigated the effect of intermittent aerobic physical exercise on the plasma soluble leptin receptor, nitrite/nitrato levels, lung function in postmenopausal women. Therefore, the aim of this work was to evaluate the effect of intermittent aerobic physical training for three months on the plasma soluble leptin receptor and nitrite/nitrato levels and its relationship with pulmonary function and blood pressure in postmenopausal women. Twenty-three volunteers were enrolled in the training program, 3 days/week, 30 minutes, during 3 months. The range of the training intensity was from 60 to 80 % of reserve heart rate. Anthropometric, cardio-respiratory and biochemical parameters were evaluated at the basal and after 3 months of exercise training. Our findings show that intermittent exercise training promotes a significant decrease in the waist circumference (3.8%) as well as diastolic blood pressure (6.7%). An increase in both VO2 max (basal: 25.1+0.8 Trained: 26.5+0.9 ml/kg/min) and respiratory function (Insp: 11 and exp: 20%). Similarly, both plasma soluble leptin receptor (basal: 47.2+3.5 TR: 54.1+ 3.6 ng/ml) as well as nitrite/nitrato levels were increased after exercise training (55%) in postmenopausal women. Therefore, our findings show that intermittent aerobic training during 3 months promoted beneficial effects on the cardiovascular function that was positively associated with increase in soluble leptin receptor and nitrie/nitrato levels in postmenopausal women. Furthermore, our data suggest that plasma soluble leptin receptor might be a marker for the beneficial effect of interval exercise training

Previous issue date: 2015-03-06 / O objetivo deste estudo foi investigar os efeitos da administração crônica de digoxina e do verapamil durante treinamento físico intervalado de alta intensidade (TFI) sobre o desempenho físico, capacidade funcional e morfologia cardíaca de ratos. Para tanto, 48 ratos Wistar, com 60 dias de idade, foram aleatoriamente distribuídos em 6 grupos(N=8/grupo): controle, não treinado (C), treinado, sem administração de droga (T), digoxina sem treinamento (DIGO), verapamil sem treinamento (VERA), treinado, com administração de digoxina (TDIGO) e treinado, com administração de verapamil (TVERA). A digoxina e o verapamil foram administrados via gavagem, na dose de 30μg/kg/dia e 5,0 mg/kg/dia respectivamente, durante todo o período experimental. Os grupos T, TDIGO e TVERA foram submetidos a um programa de TFI em esteira rolante durante 60 dias. Foi aplicado o teste de esforço progressivo máximo (TEM) e determinada a concentração sérica de lactato (LAC) sanguíneo. O TFI consistiu de sessões de corrida em esteira rolante 1 h/dia, 5 dias/semana por 60 dias. A intensidade de treino foi 80% da velocidade máxima (Vmáx) atingida no teste de esforço antes do TFI por 8 min e 20% da Vmáx por 2 min. A função cardíaca foi avaliada por ecocardiograma. Foi coletado o músculo esquelético, o músculo cardíaco e a gordura corporal total (GOR) para os dados anatômicos, o ventrículo esquerdo (VE) para análise histológica e o sangue para a análise bioquímica. A comparação entre os grupos foi realizada por meio da análise de variância (ANOVA) e Kruskal Wallis para o esquema de dois fatores independentes, complementada com o teste de Bonferroni, Tukey ou Dunn. O índice de significância considerado foi de 5%. A relação VE/peso corporal final (PCF), o diâmetro diastólico do VE (DDVE) e diâmetro sistólico do VE (DSVE) foram maiores no grupo TDIGO do que o grupo T e DIGO. Os parâmetros do TEM foram maiores e a concentração de LAC foi menor em ratos treinados em relação aos não treinados. A relação GOR/PCF foi menor no TDIGO e TVERA em relação ao DIGO e VERA, respectivamente. A relação VE/PCF foi maior no TVERA em relação ao VERA. O diâmetro interno do VE (DIVE) do grupo T, TDIGO e TVERA foram maiores em relação ao C, o TDIGO teve aumento em relação ao DIGO. O colesterol total e o LDL foram maiores no TDIGO comparado ao DIGO. A área do cardiomiócito foi maior nos grupos VERA e T comparados ao grupo C. Conclusão: O Treinamento intervalado promoveu hipertrofia cardíaca do tipo excêntrica. Entretanto, a administração concomitante de digoxina ou de verapamil não afetaram a morfologia cardíaca, a função cardíaca e o desempenho físico em ratos submetidos ao treinamento. / The aim of this study was to investigate the effects of chronic administration of cardiotonic (digoxin) and the calcium channel blocker (verapamil) during high-intensity interval exercise training (IET) on physical performance, functional capacity and cardiac morphology of rats. For this study, 48 Wistar rats, 60 days old, were randomly distributed into 6 groups (N = 8 / group): control, untrained (C), trained without drug administration (T), digoxin untrained (DIGO), verapamil without training (VERA), trained with digoxin administration (TDIGO) and trained with verapamil administration (TVERA). Digoxin and verapamil were administered by gavage at a dose of 30μg/kg/day and 5.0 mg.kg-1, respectively, throughout the experimental period. The groups T, TDIGO and TVERA underwent a IET program on a treadmill for 60 days. The progressive maximal exercise test was applied (TPM) and determined the serum concentration of lactate (LAC) blood. The IET consisted of sessions running on a treadmill 1 h/day, 5 days/week for 60 days. The training intensity was 80% of the maximum velocity (Vmax) achieved in the stress test before the TAI for 8 min and 20% of Vmax for 2 min. Cardiac function was assessed by echocardiography. Was collected skeletal muscle, cardiac muscle and total body fat (TBF) for anatomical data, the left ventricle (LV) for histological analysis and blood for biochemical analysis. The comparison between groups was performed using analysis of variance (ANOVA) or Kruskal Wallis for the two independent factors, complemented by the Bonferroni test, Tukey or Dunn. The significance level considered was 5%. The ratio VE final body weight (FBW), LV diastolic diameter (LVDD) and LV systolic diameter (LVSD) were higher in TDIGO group than the group T and DIGO. The parameters MET were higher and the concentration of LAC was lower in rats training in relation to the untrained. The relationship GOR/FBW was lower in TDIGO and TVERA compared to DIGO and VERA respectively. The ratio VE/FBW was higher in TVERA compared to VERA. The of the LV inside diameter (LVID) T group, TDIGO and TVERA were higher compared to C, TDIGO had increased compared to DIGO. Total cholesterol and LDL were higher in TDIGO compared to DIGO. The area of cardiomyocytes was higher in VERA and T compared to group C. Conclusion: TAI induced cardiac hypertrophy of the eccentric type. However, concomitant administration of digoxin or verapamil did not affect the cardiac morphology, cardiac function and physical performance in rats submitted to training.

Blood pressure control occurs through the interaction between the autonomic nervous system and substances secreted by different cell types, including endothelial cells, causing changes in any of these mechanisms, leading to the development of arterial hypertension. The practice of physical exercises is shown as a non-pharmacological treatment capable of significantly reducing both systolic and diastolic blood pressure, attenuating blood pressure reactivity and increasing the bioavailability of nitric oxide. The objective of the present study was to observe blood pressure, heart rate and vasoactive (nitric oxide) response after an interval aerobic exercise session in hypertensive women. Two protocols were used: one exercise interval (sequence alternating 30 sec/ 90% and 90 sec/ 60% maximal aerobic velocity) and one control session (without exercise). As a result, it was observed that the pressure and heart rate behavior in the post-exercise were similar to the control session, and no attenuation of pressure reactivity after stress test (CPT) was observed when systolic blood pressure was observed in relation to rest: CPT: 160.9 ± 32.27 mmHg x Rest: 122.4 ± 18.58 mmHg and after 60 minutes recovery: CPT: 160.9 ± 32.27 mmHg x 60 min: 121.4 ± 13.87 mmHg. As well as were not observed in relation to the diastolic blood pressure (CPT 99.3 ± 16.19 mmHg x Rest 75.4 ± 15.17 mmHg, moment immediately after exercise: 82.5 ± 9.42 mmHg, and after 60 Min of recovery: 76.3 ± 13.18 mmHg. Regarding the heart rate, no reactivity was noted in the post-exercise period, showing elevation, only when the moment immediately after exercise was observed. Similarly, no differences were observed between the pre and post values related to the bioavailability of nitric oxide, in both protocols. It is concluded that, in the intensity and volume evaluated, physical exercise is not able to attenuate vascular reactivity, nor does it influence nitric oxide concentrations. / O controle da pressão arterial se dá através da interação entre o sistema nervoso autônomo e substâncias secretadas por diferentes tipos de células, entre elas as endoteliais, fazendo com que alterações em algum desses mecanismos acarretem no desenvolvimento da hipertensão arterial. A prática de exercícios físicos se mostra como um tratamento não-farmacológico capaz de reduzir significativamente tanto a pressão arterial sistólica como a diastólica, atenuar a reatividade pressórica e aumentar a biodisponibilidade de óxido nítrico. O objetivo do presente estudo foi observar a resposta pressórica, da frequência cardíaca e de substância vasoativa (óxido nítrico) após uma sessão de exercício aeróbio intervalado em mulheres hipertensas. Para tanto foram realizados dois protocolos, sendo um de exercício intervalado (sequência alternando 30 seg/ 90% e 90 seg/ 60% da velocidade aeróbia máxima) e uma sessão controle (sem exercício). Como resultado, observou-se que o comportamento pressórico e da frequência cardíaca no pós-exercício mostraram-se semelhantes à sessão controle, não sendo evidenciada atenuação da reatividade pressórica pós teste de estresse (CPT) quando observadas a pressão arterial sistólica com relação ao repouso: CPT:160,9 ± 32,27 mmHg x Repouso: 122,4 ± 18,58 mmHg e após 60 minutos de recuperação: CPT:160,9 ± 32,27 mmHg x 60 min: 121,4 ± 13,87 mmHg. Bem como não foram observadas em relação à pressão arterial diastólica (CPT 99,3 ± 16,19 mmHg x Repouso 75,4± 15,17 mmHg; momento imediatamente após o exercício: 82,5± 9,42 mmHg; e, após 60 min de recuperação: 76,3± 13,18 mmHg. Com relação à frequência cardíaca, não foi apontada reatividade no pós exercício, tendo apresentado elevação, apenas, quando observado o momento imediatamente após o exercício.
