The theoretical part is concerned with the functional application of selenium and iodine supplementation methods, risks disproportionate to their income, including changes in biochemical and hematological parameters and evaluation methods of adequate saturation. The practical part analyzes the results of experiments on sheep with a defined content of selenium and iodine in the ration evaluation of selected indicators of the metabolic profile of ewes and their lambs. The results are processed statistically. It reflected the dynamics and interactions of monitored parameters.

This work deals with the content of iodine in soils, furthermore with obtaining and processing samples from four selected sampling areas, all of which are in the proximity of the Arnoštov settlement in district Prachatice in the foothills of Šumava. The soil samples were obtained from forest, meadow, pasture and fallow soil in 2009 to 2011. The iodine content in the soils was determined by using the spectrophotometric method, which had been optimized for the soil samples. Iodine concentration in lysimeter water was determined by using method inductively coupled plasma - mass spectrometry. The results obtained from each sampling areas were compared with each other and with the results from other areas. Consequently, the iodine concentration results in the soils were compared with the iodine concentration in lysimeter waters. It was found, that the highest iodine content in soils was measured in a sample obtained from Area 1 (meadow), part B in a depth of 16 - 30 cm in June of 2009, namely a content of 8,67 mg of I per kg of dry soil. The lowest content of 1,42 mg of I per kg of soil was found in the area 4 (forest), sample obtained in June of 2010, in the L horizon. By comparing iodine content with iodine concentrations in lysimeter waters, it was concluded that the concentration of iodine in lysimeter waters does not depend on iodine concentrations in soils and that it does not even represent the absolute iodine content in soil, as was determined by comparing the results from Arnoštov with data acquired from Agrovýzkum Rapotín in Jeseníky.

At the nuclear power plant accident, the radionuclides with very short half-life come to enviroment, they decay the period from several second to several months. On the other sides come to enviroment radionuclides with long half-life. Their radionuclides occur in surrounding of the power plant for hundreds of years before they decay totally. Immediately after a nuclear power plant accident, radioiodine represents the most severe health hazard for population. Because it is a radionuclide with short half-life, it causes the danger during first months after the accident. Its health hazard lies in the fact, that it gets to food chains and irradiates a man, who consumed contaminated water and food. Radiocaesium and radiostrontiom are the next important radionuclides. They have long half-lifes, it means that will decay after hundred years after the accident and they endanger population a long time after the accident yet.

Background: Although radioactive iodine 131 (RAI) has been successfully used in differentiated thyroid cancer (DTC) patients therapy for more than 70 years, thus far published data regarding RAI- induced salivary glands functional impairment have been inconsistent. Due to the trend towards using lower activities or even RRA omission, the possible RAI untoward effects have been widely discussed. Aim: To evaluate and quantify salivary gland function in thyroidectomised DTC patients before and after RRA using activity of 3.7 GBq and to compare salivary gland functional changes in DTC patients after single or repeated RAI treatment (using activities  5.5 GBq) with their age- and sex-matched RAI-naive counterparts using salivary gland scintigraphy with 99m Tc- pertechnetate. In addition, we performed subjective symptoms evaluation. Patients and Methods: Salivary gland scintigraphy (SGS) was performed in 31 RAI-naive patients (6 men, 25 women, median age 52 years) before and 4.6 months after RRA. Salivary gland function was measured also in 23 patients (7 men, 16 women, median age 60 years) with RAI administration history and compared with their age- and sex-matched counterparts. Optical frequency references based on hyperfine transitions in molecular iodine

Diese Arbeit beschäftigt sich mit der Entwicklung und Untersuchung von optischen Absolutfrequenzreferenzen basierend auf rovibronischen Übergängen in molekularen Jod. Dabei werden Methoden der Doppler-freien Sättigungsspektroskopie angewendet, um einzelne Übergänge der Hyperfeinstruktur mit Linienbreiten unterhalb von 1 MHz im B-X System von molekularem Iod bei 532 nm, der zweiten harmonischen des Nd:YAG-Laser, aufzulösen. Elektronische Regelungstechniken ermöglichen eine präzise Stabilisierung der optischen Frequenz auf die Linienmitte der Übergänge mit einer Auflösung von Teilen in 10^5. Mit dem Ziel einer weltraumtauglichen Absolutfrequenzreferenz für zukünftige Weltraummissionen, wurden zwei Spektroskopiemodule konzipiert und in quasi-monolithischen Glaskeramik-Aufbauten, als sogenanntes elegant breadboard model und engineering model, realisiert. Diese Jodfrequenzreferenzen wurden im Detail in Bezug auf ihre Frequenzstabilität und Reproduzierbarkeit untersucht und Letzteres wurde für die angestrebte Weltraumqualifizierung ersten Umwelttests, sowohl vibrations- als auch thermischen Belastungstests, unterzogen. Für die Untersuchung der Frequenzstabilität dieser Jodreferenzen wurde ein auf einen optischen Resonator hoher Güte stabilisiertes Lasersystem für direkte Frequenzvergleiche bei 1064 nm realisiert. Die Analyse der Frequenzstabilität der Jod Referenzen zeigt eine Frequenzstabilität von 6x10^−15 bei 1 s, und weniger als 2x10^−15 bei 100 s Integrationszeit, was der bis heute besten veröffentlichten Frequenzstabilität entspricht die mit Jod Referenzen erreicht wurde. Mit der erreichten Frequenzstabilität ermöglichen diese Absolutfrequenzreferenzen präzise Lasersysteme für zukünftige Weltraummissionen wie z.B. zur Detektion von Gravitationswellen, zur Vermessung des Gravitationsfelds der Erde oder für Präzisionstest fundamentaler Theorien der Physik. / This thesis deals with the development and investigation of optical absolute frequency references based on rovibronic transitions in molecular iodine. Doppler-free saturation spectroscopy methods are employed to resolve individual transitions of the hyperfine structure with linewidths below 1 MHz in the B-X system of molecular iodine at 532 nm with the second harmonic of Nd:YAG lasers. Electronic feedback control systems are employed for laser frequency stabilization to the line center of the optical transitions with a line splitting of 10^5. With the goal of a space qualified optical absolute frequency reference for future laser-interferometric space missions, two spectroscopy setups were designed and realized in quasi-monolithic, glass-ceramic setups as so called elegant bread board model and engineering model. These iodine references were characterized in detail with respect to their frequency stability and reproducibility and the engineering model was subject to environmental tests, including vibrations and thermal cycling to verify its applicability in future space missions. For the investigation of the frequency instability of these iodine references, a frequency stabilized laser system was realized based on a temperature controlled high Finesse ULE cavity for direct frequency comparisons at 1064 nm. Analysis of the frequency stability of the iodine references revealed exceptionally low fractional frequency instability of 6x10^−15 at 1 s, averaging down to less than 2×10^−15 at 100 s integration time, constituting the best reported stability achieved with iodine references to date. With the demonstrated performance, these absolute frequency references enable precision laser systems required for future space missions that are dedicated to, e.g., the detection of gravitational waves, mapping of the Earth’s gravitational field or precision test of fundamental physics.

Chemical methods to increase the reactivity  of lignin : In the context of green chemistry  and education for sustainable development

The research  concerning lignin in high value applications has increased  during the last years due to its renewability and availability in the black liquor from pulp mills. Today  the major part of kraft lignin found  in the black liquor is used as fuel to gain energy  in the recovery boiler. Lignin functions as natural  glue in plants so that the function of kraft lignin as a phenol replacement in wood adhesives has been researched. Due to lignin's low reactivity the molecule must be modified  prior to use. Demethylation is a method  to increase the phenolic content  in lignin to produce  more reactive  sites. Thiol  mediated  and iodine mediated demethylation was performed. Demethylated lignin was characterized by changes  in phenolic and hydroxylic groups,  molecular  mass, elemental  composition and other  properties  using methods including  UV, SEC and 31 P NMR.   The results showed  a decrease in the phenolic  content  contrary  to the increase  that was expected. Really low yields were also gained  which makes the results non-representative. Size evaluation showed  that the percentage of high molecular  content  in the demethylated lignin sample  had increased,  which  point towards the loss of low molecular mass fractions.  Due to demethylation lignin may have been more hydrophilic and soluble  in DMF and water. In addition  to this bond cleavage  may have produced  smaller  fragments which also increase  the solubility. The results point towards  the loss of smaller  fragments in the DMF and water phases.   The applied demethylation methods were evaluated  in context of green chemistry. Production, waste,  involving chemicals and efficiency were discussed  and analyzed. The applied demethylation methods  use DMF as solvent  which  is not a green alternative, greener solvents such as water or other energy  adding  methods  could  be used to make the process  greener.  The use ofNaOMe produces  methanol  as a byproduct  which could be eliminated by using NaOH, future studies  on the efficiency of the bases in the needs to be done.   Nature science  has a reputation  of being hard and firm. By bringing  in social  issues in science education new ways of looking  at science  opens  up. A social problem  and at the same time an environmental problem  in today's  society  is the large plastic mountain in the Pacific Ocean. An educational material  of the "Samhallsfragor med Naturvetenskapligt Innehiill",SNI, (Societal  issues with scientific content)  principle  has been evaluated with respect  to the abilities that can developed together  with whether  students  increase  their science  knowledge through  this. The study showed  that students  can develop  almost  all abilities  described  in the curriculum and their knowledge in science  by this type of material. Idag används den största delen av sulfatligninet från svartluten som bränsle for att producera energi i sodapannan. Lignin fungerar som ett naturligt lim i växter och på grund av detta undersöks funktionen av kraftlignin som fenolersättning i trälim. Med anledning av ligninmolekylens låga reaktivitet behover lignin modifieras fore användning i produkter. Demetylering är en metod för att öka fenolhalten i lignin och skapa en högre reaktivitet. I denna studie utfördes Tiolmedierad och jodidmedierad demetylering. Det demetylerade ligninet utvärderades med avseende på forändringar i  fenol-och hydroxylgrupper, molekylvikt, elementarsammansättning och andra egenskaper med hjälp av olika metoder,inklusive UV, SEC och 31 P NMR.   Resultaten visade en minskning i fenolinnehall  i motsats till den ökning som förväntades. Riktigt låga utbyten påvisades också vilket gör att resultaten inte är representativa. Storleksutvärdering  visade att andelen med högt molekylviktsinnehåll i det demetylerade ligninproven hade ökat, vilket pekar mot förlust av lågmolekylära fraktioner. På grund av demetyleringen kan ligninet ha blivit mer hydrofilt och lösligt i DMF och vatten. Utöver detta kan bindningsklyvning ha skapat mindre fragment som också ökat lösligheten. Resultaten pekar mot förlust av mindre fragment i DMF-och vattenfaserna.   De tillämpade demetyleringsmetoderna  utvärderades med avseende på grön kemi. Produktion, avfall, kemikalier och effektivitet diskuterades och analyserades. De tillämpade demetyleringsmetoderna  använder DMF som lösningsmedel, vilket inte är ett grönt alternativ. Grönare lösningsmedel såsom vatten, eller andra typer av energitillsättning, kan användas för att göra processen miljövänligare. Användandet av NaOMe i den thiolmedierade demethyleringen skapar metanol som en biprodukt vilket kan bytas ut mot vatten om NaOH istället används. Vidare studier behöver göras för att undersöka de båda baserna effektivitet.   Naturvetenskapen har ett rykte om att vara hård och fast. Genom att föra in sociala frågor i den naturvetenskapliga utbildningen kan nya sätt att se på vetenskapen skapas. Ett samhälls problem och samtidigt ett miljöproblem i dagens sämhalle är det stora plast berget i Stillahavet. Ett undervisningsmaterial för"Samhällsfrågor Med Naturvetenskapligt Innehåll", SNI, principen har utvärderats med avseende på vilka förmågor som kan tränas tillsammans med huruvida eleverna kan öka sin vetenskapliga kunskap.
