Transcriptional and post-transcriptional control of therapeutic gene expression during disease activity

Rheumatoid arthritis (RA) is a chronic inflammatory autoimmune disease which predominantly affects the synovial joints. Local gene therapy represents an approach to produce therapeuticmolecules (i.e. soluble TNF receptor (sTNFR)-Fc and interleukin-1 receptor antagonist (IL-1Ra)) directly in arthritic joints. Gene therapy could be designed to link the level of therapeutic gene expression directly to disease activity, through the use of transcriptional and posttranscriptional regulatory elements. The experiments in this thesis describe the construction of multi-responsive, composite synthetic promoters, comprised of the binding sites for an array of transcription factors activated in arthritic joints. Optimal spatial arrangements of binding sites in relation to each other and to the TATA box were determined by Assembly PCR cloning and the functionality of the resulting synthetic promoters revealed additive or synergistic induction of luciferase reporter gene expression in response to combined stimulation. Candidate synthetic promoters were cloned into a lentiviral vector between insulator elements and displayed significantly enhanced induction, in excess of 1,500 fold in response to combined stimulation. Inflammation-specific activation of lentiviral synthetic promoters was confirmed in a carrageenan-induced paw inflammation mouse model, which demonstrated the strong correlation between local luciferase gene expression and paw inflammation. Post-transcriptional gene regulation was also investigated by exploiting the differential expression of endogenous miR-23b during inflammation. Insertion of miR-23b target sites into the 3’UTR of the luciferase gene subjected luciferase mRNA to regulation by miR-23b. Experiments demonstrated that high basal gene expression driven by constitutive and inducible promoters was significantly downregulated by miR-23b without significantly impairing high gene expression upon stimulation. Overall, the experiments in this thesis have confirmed the induction of inflammation-specific gene expression, regulated by inflammationresponsive endogenous transcriptional and post-transcriptional elements.

A Coordenação de Aperfeiçoamento de Pessoal de Nível Superior - CAPES e o Deutscher Akademischer Austauschdienst - DAAD são organizações do Brasil e da Alemanha, respectivamente, que apoiam iniciativas de cooperação bilateral em ensino superior, ciência, tecnologia e inovação de forma muito positiva. Os acordos governamentais amparam a cooperação desde o início dos anos 1960 com o propósito de manter as relações de amizade e os interesses comuns em fomentar a pesquisa científica, e promover o desenvolvimento tecnológico. A idealização em favorecer oportunidades para a criação, e execução, de projetos conjuntos de pesquisa bilateral se sustenta em apoiar a formação qualificada de recursos humanos e fortalecimento de atividades concomitantes de ensino, pesquisa e gestão em ciência/tecnologia/inovação. São iniciativas que estimulam a expansão da cooperação universitária internacional com o intuito de fortalecer nosso sistema nacional de pós-graduação, mediante a concessão de investimentos para criar, ou vitalizar, redes internacionais de pesquisa. A convivência entre grupos de excelência beneficia o compartilhamento de soluções para problemas comuns, assim como busca possibilidades de retribuir à coletividade propostas de inovações. O primeiro artigo descreve: 1) a distribuição regional das instituições brasileiras envolvidas; 2) as grandes áreas de conhecimento que abrigam os projetos executados; e 3) as redes de colaboração brasileiras e alemãs com maior número de projetos executados. No que se refere ao Brasil é visível, além das interações com instituições alemãs, uma ampla interação nacional, uma assimétrica distribuição regional, o predomínio das grandes áreas Ciências Exatas e da Terra, Engenharias e Ciências Biológicas, e um crescimento do número de instituições brasileiras envolvidas e da formação de redes bilaterais. Além disso, se discute a potencial contribuição deste duradouro Programa para a Pós Graduação e Pesquisa Brasileira. O segundo artigo identifica as contribuições do PROBRAL na produção científica brasileira e a colaboração entre pesquisadores brasileiros e alemães ao longo dos anos de 1994 a 2016, e nas três grandes áreas predominantes anteriormente citadas. É um estudo exploratório–descritivo, baseado em dados e documentos publicados pelas entidades responsáveis por sua execução, a CAPES e o DAAD. A partir destes dados, trata-se de identificar, através de indicadores cientométricos (produção, coautoria e Fator de Impacto), as contribuições do programa para a produção e internacionalização da pesquisa científica brasileira. Observa-se que a área de Ciências Exatas e da Terra publicou em maior número que as duas outras áreas relacionadas. Quanto à produção dos pesquisadores brasileiros em colaboração com pesquisadores alemães, a grande área de Ciências Biológicas, se sobressai com cerca de 20% da produção. Este artigo mostra que a política de cooperação científica internacional é de extrema importância, apoiando e fomentando o intercâmbio científico entre grupos de pesquisadores brasileiros e alemães. The coexistence between groups of excellence benefits the sharing of solutions to common problems, as well as the seeking of possibilities to return to the collective proposals for innovations. This paper describes the bilateral cooperation between Brazil and Germany, with emphasis on the Program of University Cooperation between Brazil and Germany - PROjetos de Cooperação Universitária entre o BRasil e a ALemanha (PROBRAL) in some of its stages from 1994 to 2014. The first article describes: 1) the regional distribution level of the Brazilian institutions involved; 2) the main areas of knowledge that house executed projects; and 3) the Brazilian and German collaboration networks with the greatest number of executed projects. With regards to Brazil, in addition to interactions with German institutions, there is a wide national interaction, an asymmetric regional distribution, the predominance of main areas of Natural Sciences, Engineering and Biological Sciences, and a growth in the number of Brazilian institutions and the formation of bilateral networks. In addition, the potential contribution of this long-standing Program for Postgraduate and Brazilian Research is discussed. The second article identifies the contributions of PROBRAL in Brazilian scientific production and the collaboration between Brazilian and German researchers over the years 1994 to 2016 and in the three main areas mentioned above. This is an exploratory-descriptive study, based on data and documents published by the entities responsible for its execution, CAPES and DAAD. From these data, it is a question of identifying, using scientometrics (production, co-authorship and Impact Factor), the program's contributions to the production and internationalization of Brazilian scientific research. It is seen that most publications are originated from the area of Natural Sciences, in a greater number than the two other related areas. As for the production of Brazilian researchers in collaboration with German researchers, the great area of Biological Sciences stands out with about 20% of production. This article suggests that the policy of international scientific cooperation is extremely important, supporting and fostering the scientific exchange between groups of Brazilian and German researchers. Therefore, international peer mobility is significant for the increase of scientific research and the internationalization of Higher Education Institutions (IES - Instituições de Ensino Superior) as it creates a favorable environment for cooperation and networking. Lastly, one should respect the recommendation of the National Postgraduate Plan 2011-2020 regarding the search for excellence and new knowledge. Such an orientation means avoiding endogenous positions and interacting strongly with national and international institutions to promote the growth of Brazilian science and highlight the country on the world stage.

Kunskapsbildning mellan träindustri och akademi - en studie av dess förutsättningar och möjligheter / Knowledge development between the wood & timber industry and academia: A study of the prospects, basic conditions and opportunities

<p>Small and medium-sized enterprises badly need new knowledge. Their need is constantly increasing due to the market demand for custom-designed products, which means companies must be more flexible. Wood processing companies are having difficulty establishing contact with academia in order to meet this need.</p><p>In investigating the form and content of research collaboration, nine situations have been studied with the aid of participatory observation to determine what characterises situations where learning occurs. Situations where researchers collaborate with operators and production managers have been selected because of the key role that those in such positions play when research findings are to be applied in the wood & timber industry. Analytical tools of a pedagogic/didactic nature have been used when interpreting the material.</p><p>The results show what is needed if research collaboration is to succeed. Both parties must have room for reflection, and must give due consideration to the basic assumptions and problem areas they have in common. Enterprises must be sophisticated enough to collaborate with academia. In approaching their task, researchers must emphasise dialogue, equal standing and respect, and they must seek to ensure that collaboration proceeds in the right direction. In the case of small and medium-sized enterprises, a link between the researcher and the company is often required.</p> / <p>Små och medelstora företag har ett stort behov av ny kunskap. Behovet ökar kontinuerligt pga. marknadens krav på kundanpassning vilket medför krav på större flexibilitet hos företagen. De träbearbetande företagen har svårigheter att komma i kontakt med akademin för att tillgodose detta behov.</p><p>För att undersöka former och innehåll vid forskningssamverkan har nio situationer studerats med hjälp av deltagande observation för att utreda vad som kännetecknar de situationer där lärande uppstår. Situationer där forskare samverkar med operatörer och med produktionschefer har valts ut på grund av dessa yrkesgruppers nyckelposition när det gäller tillämpning av forskningsresultat i träindustrin. Analysverktyg med pedagogisk/didaktisk anknytning har använts vid tolkningen av materialet.</p><p>Resultaten visar förutsättningar och villkor för en lyckad forskningssamverkan. Det måste finnas ett utrymme för reflektion för de som samverkar liksom att gemensamma utgångspunkter och problemområden måste uppmärksammas i samverkan. Det måste finnas en mogenhet hos företag för att kunna samverka med akademin. Forskarens arbetssätt måste präglas av dialog, jämbördighet och respekt och forskaren har ett ansvar för att samverkansprocessen går i rätt riktning. För små och medelstora företag är en länk mellan forskningen och företaget ofta nödvändig.</p>

It examines which strategies managers used to overcome these problems in the case of the Brazilian Oil and Gas company Petrobras. By conducting an inductive case study it develops a contextual model based on the use of grounded theory. It argues that the most important categories were to overcome problems of (a) the interpersonal relationship, (b) power shifting inside the organization, and (c) making people more valuable to the organization and shows how managers tackled these challenges. It contributes with a deep analysis of HRM challenges in Open Innovation that is important for a better understanding of management problems that can come along with Open Innovation processes. / Este artigo leva em consideração os desafios da Gestão de Recursos Humanos (HRM) em processos de Inovação Aberta. São examinadas quais estratégias os gerentes utilizavam para superar esses problemas no caso da companhia brasileira de óleo e gás, Petrobrás. Ao conduzir um indutivo estudo de caso, é desenvolvido um modelo contextual baseado no uso da teoria fundamentada. Argumenta-se que as categorias mais importantes a serem superadas foram (a) as relações interpessoais, (b) mudança de poder dentro da organização e (c) a valorização dos funcionários pela organização, e é apresentado como os gerentes solucionaram esses problemas. Este artigo contribui com uma análise aprofundada dos desafios da HRM em Inovação Aberta, o que é importante para melhor entender os problemas gerenciais que podem surgir durante os processos de Inovação Aberta.

Kunskapsbildning mellan träindustri och akademi : en studie av dess förutsättningar och möjligheter / Knowledge development between the wood &amp; timber industry and academia : A study of the prospects, basic conditions and opportunities

Small and medium-sized enterprises badly need new knowledge. Their need is constantly increasing due to the market demand for custom-designed products, which means companies must be more flexible. Wood processing companies are having difficulty establishing contact with academia in order to meet this need. In investigating the form and content of research collaboration, nine situations have been studied with the aid of participatory observation to determine what characterises situations where learning occurs. Situations where researchers collaborate with operators and production managers have been selected because of the key role that those in such positions play when research findings are to be applied in the wood &amp; timber industry. Analytical tools of a pedagogic/didactic nature have been used when interpreting the material. The results show what is needed if research collaboration is to succeed. Both parties must have room for reflection, and must give due consideration to the basic assumptions and problem areas they have in common. Enterprises must be sophisticated enough to collaborate with academia. In approaching their task, researchers must emphasise dialogue, equal standing and respect, and they must seek to ensure that collaboration proceeds in the right direction. In the case of small and medium-sized enterprises, a link between the researcher and the company is often required. / Små och medelstora företag har ett stort behov av ny kunskap. Behovet ökar kontinuerligt pga. marknadens krav på kundanpassning vilket medför krav på större flexibilitet hos företagen. De träbearbetande företagen har svårigheter att komma i kontakt med akademin för att tillgodose detta behov. För att undersöka former och innehåll vid forskningssamverkan har nio situationer studerats med hjälp av deltagande observation för att utreda vad som kännetecknar de situationer där lärande uppstår. Situationer där forskare samverkar med operatörer och med produktionschefer har valts ut på grund av dessa yrkesgruppers nyckelposition när det gäller tillämpning av forskningsresultat i träindustrin. Analysverktyg med pedagogisk/didaktisk anknytning har använts vid tolkningen av materialet. Resultaten visar förutsättningar och villkor för en lyckad forskningssamverkan. Det måste finnas ett utrymme för reflektion för de som samverkar liksom att gemensamma utgångspunkter och problemområden måste uppmärksammas i samverkan. Det måste finnas en mogenhet hos företag för att kunna samverka med akademin. Forskarens arbetssätt måste präglas av dialog, jämbördighet och respekt och forskaren har ett ansvar för att samverkansprocessen går i rätt riktning. För små och medelstora företag är en länk mellan forskningen och företaget ofta nödvändig.

Vědecko-výzkumná spolupráce z pohledu soutěžního práva EU / Research and development cooperation from the perspective of competition law of the EU

Presented thesis is focused on research and development cooperation between companies from the perspective of the competition law of the European Union. The thesis analyses the evolution of the legislation applicable to on this kind of cooperation in the context of examined axis of laissez-faire - interventionism. The emphasis is put on convergence of the EU legislation for this category of collaboration with the regulations applicable for R&D agreements in the USA. The aim of presented thesis is to analyse and determine the direction of the European Union competition law regulation of research and development cooperation in the context of abovementioned axis, namely the evolution of block exemptions for this category of agreements. Apart from the coherent description of the applicable set of rules, the analysis aims to foresee the future evolution of these rules. .Another aim is to compare the European course of research and development legislation with the same kind of legislation in the USA. That is why presented thesis contains detailed analysis of the legislation applicable to R&D agreements in the USA. Multi-hazard analysis of steel structures subjected to fire following earthquake

Fires following earthquake (FFE) have historically produced enormous post-earthquake damage and losses in terms of lives, buildings and economic costs, like the San Francisco earthquake (1906), the Kobe earthquake (1995), the Turkey earthquake (2011), the Tohoku earthquake (2011) and the Christchurch earthquakes (2011). The structural fire performance can worsen significantly because the fire acts on a structure damaged by the seismic event. On these premises, the purpose of this work is the investigation of the experimental and numerical response of structural and non-structural components of steel structures subjected to fire following earthquake (FFE) to increase the knowledge and provide a robust framework for hybrid fire testing and hybrid fire following earthquake testing. A partitioned algorithm to test a real case study with substructuring techniques was developed. The framework is developed in MATLAB and it is also based on the implementation of nonlinear finite elements to model the effects of earthquake forces and post-earthquake effects such as fire and thermal loads on structures. These elements should be able to capture geometrical and mechanical non-linearities to deal with large displacements. Two numerical validation procedures of the partitioned algorithm simulating two virtual hybrid fire testing and one virtual hybrid seismic testing were carried out. Two sets of experimental tests in two different laboratories were performed to provide valuable data for the calibration and comparison of numerical finite element case studies reproducing the conditions used in the tests. Another goal of this thesis is to develop a fire following earthquake numerical framework based on a modified version of the OpenSees software and several scripts developed in MATLAB to perform probabilistic analyses of structures subjected to FFE. A new material class, namely SteelFFEThermal, was implemented to simulate the steel behaviour subjected to FFE events.
