Os testes aplicados foram velocidade crítica (Vcrit) e máxima fase estável de lactato (MFEL) e a amostra do estudo fundistas de alto rendimento. Estudos preliminares foram desenvolvidos para avaliar a confiabilidade e aplicabilidade da ferramenta utilizada na determinação de um parâmetro fisiológico de carga confiável (LAn). A metodologia utilizada foi descrita apresentando as técnicas e procedimentos para coletar os dados, assim como a seleção da amostra e o tratamento estatístico. O protocolo de teste de Vcrit para estimar o LAn, quando aplicado no modelo distância vs tempo e com tomada de tempo para 4 distâncias e 3 distâncias se mostrou eficaz e confiável, uma vez que não foram encontradas diferenças significativas nos valores dessas variáveis. A aplicação dos instrumentos de monitoramento de carga, percepção subjetiva de intensidade (PSI) e percepção subjetiva de cansaço (PSC) se mostraram eficazes na escolha dos métodos, meios de treinamento e vias metabólicas trabalhadas quando relacionados à dinâmica de carga do microciclo-tipo, entretanto, não foram suficientemente sensíveis para identificar diferenças na acurácia entre as equações de predição de MFELmc e MFELms. Houve boa correlação entre as melhores marcas dos atletas na Meia Maratona vs Distância Limite e Meia Maratona vs Tempo Limite ( r= -0,8 e r= -0,6, respectivamente). As intensidades de corrida em competições oficiais de Meia Maratona, a Vcrit real e a MFELms mostraram importante relação na determinação de velocidades de treinamento e competições. Os métodos e meios de treinamento de fundistas de alto rendimento foram identificados e caracterizados, concluindo que o treinamento organizado contendo intensidades de corrida preditas através de parâmetros confiáveis deve prevalecer na busca do melhor desempenho possível. Os resultados atléticos conhecidos até o presente momento indicam a relevância de estudos fundamentados em respostas fisiológicas diretas para aplicação de intensidades de corrida em fundistas de elevado rendimento / Abstract: The purpose of this study was to relate aerobic performance tests used to estimate the anaerobic threshold (AT) to running intensities in training and competition. The tests applied were critical speed (CS) and maximal lactate steady state (MLSS) and study sample high performance distance runners. Preliminary studies were developed as part of the literature review to support the tool that was used to determine a reliable load physiological parameter (AT), easily applicable and highly replicable. The methodology was described presenting the techniques and procedures for collecting data, as well as sample selection and statistical analysis. The CS test protocol to estimate the AT showed to be effective and reliable when applied to the distance vs time model by collecting the time for 4 distances and 3 distances, once no significant differences were found in the values of these variables. The application of the subjective perception of intensity (SPI) and subjective perception of fatigue (SPF) load tools for monitoring showed to be effective on the choice of methods, means training and metabolic pathways when related to microcycle-type dynamic loading, however, were not sufficiently sensitive to identify differences in accuracy between MLSScm and MLSSsm prediction equations. There was a good correlation between athletes personal bests Half Marathon vs Distance Limit and Half Marathon vs Time Limit (r = -0.8 and r = -0.6, respectively). The running intensities at Half Marathon official competitions, the real CS and MLSSsm, showed an important relation to determine the speed on training and competitions. The methods and means of training for high performance distance runners were identified and characterized, concluding that the organized training that contains predicted running intensities through reliable parameters should prevail in pursuit the best performance. Lack of Age-Related Respiratory Changes in Daphnia

Aging is a multifaceted process of accumulation of damage and waste in cells and tissues; age-related changes in mitochondria and in respiratory metabolism have the focus of aging research for decades. Studies of aging in nematodes, flies and mammals all revealed age-related decline in respiratory functions, with somewhat controversial causative role. Here we investigated age-related changes in respiration rates, lactate/pyruvate ratio, a commonly used proxy for NADH/NAD+ balance, and mitochondrial membrane potential in 4 genotypes of an emerging model organism for aging research, a cyclic parthenogen Daphnia magna. We show that total body weight-adjusted respiration rate decreased with age, although this decrease was small in magnitude and could be fully accounted for by the decrease in locomotion and feeding activity. Neither total respiration normalized by protein content, nor basal respiration rate measured in anaesthetized animals decreased with age. Lactate/pyruvate ratio and mitochondrial membrane potential (∆Ψ) showed no age-related changes, with possible exceptions of ∆Ψ in epipodites (excretory and gas exchange organs) in which ∆Ψ decreased with age and in the optical lobe of the brain, in which ∆Ψ showed a maximum at middle age. We conclude that actuarial senescence in Daphnia is not caused by a decline in respiratory metabolism and discuss possible mechanisms of maintaining mitochondrial healthspan throughout the lifespan.

Lactate Induces Vascular Permeability via Disruption of VE-Cadherin in Endothelial Cells During Sepsis

Circulating lactate levels are a critical biomarker for sepsis and are positively correlated with sepsis-associated mortality. We investigated whether lactate plays a biological role in causing endothelial barrier dysfunction in sepsis. We showed that lactate causes vascular permeability and worsens organ dysfunction in CLP sepsis. Mechanistically, lactate induces ERK-dependent activation of calpain1/2 for VE-cadherin proteolytic cleavage, leading to the enhanced endocytosis of VE-cadherin in endothelial cells. In addition, we found that ERK2 interacts with VE-cadherin and stabilizes VE-cadherin complex in resting endothelial cells. Lactate-induced ERK2 phosphorylation promotes ERK2 disassociation from VE-cadherin. In vivo suppression of lactate production or genetic depletion of lactate receptor GPR81 mitigates vascular permeability and multiple organ injury and improves survival outcome in polymicrobial sepsis. Our study reveals that metabolic cross-talk between glycolysis-derived lactate and the endothelium plays a critical role in the pathophysiology of sepsis.

Effects of biostimulators on growth and physiological reactions of vegetables

Biotische und abiotische Stressfaktoren mindern die Quantität und die Qualität landwirtschaftlicher Erzeugnisse. Sogar die kontrollierten Wachstumsbedingungen eines Gewächshauses tragen nur bedingt zur Minderung der schädlichen Auswirkungen von suboptimalen Wachstumsfaktoren bei. Die negativen Effekte, die durch die Entstehung suboptimaler Wachstumsbedingungen auftreten können, manifestieren sich oft erst nach geraumer Zeit. Aus diesem Grund ist es wichtig, Maßnahmen zu treffen, die diesen negativen Auswirkungen entgegenwirken und eine nachhaltige Produktion von Obst und Gemüse sichern. Um die Widerstandsfähigkeit der Pflanzen gegenüber den zahlreichen Stressfaktoren zu erhöhen, werden sogenannte Pflanzenstärkungsmittel verwendet. Die zahlreichen gärtnerischen Substrate, die bei der Kultivierung von Obst und Gemüse heutzutage unentbehrlich geworden sind, besitzen verschiedene chemische und physikalische Eigenschaften. Diese Eigenschaften unterliegen dem ständigen Einfluss von verschiedenen Faktoren, insbesondere dem Pflanzenwachstum. Anderseits beeinflussen gärtnerische Substrate die Entwicklung der Pflanzen. Die Fähigkeit eines Substrates zur Wasserhaltung, sowie zur Gewährleistung einer optimalen Versorgung mit pflanzlichen Nährstoffen während der kritischen Wachstumsperioden der Pflanzen kann die Produktivität der gärtnerischen Kulturen stark beeinflussen. Dynamische Veränderungen der Substrateigenschaften während des Pflanzenwachstums und Änderungen in physiologischen Reaktionen der gärtnerischen Pflanzen können als System betrachtet werden, und als System können sie, zwecks Optimierung der pflanzlichen Systeme, beeinflusst werden. Diese Einflussnahme kann durch Verwendung von Pflanzenstärkungsmitteln verschiedener Herkunft erreicht werden. In der Fachliteratur wird häufig über Pflanzenstärkungsmittel diskutiert. Dabei wird sehr oft außer Acht gelassen, dass jedes Pflanzenstärkungsmittel ein eigenes Wirkungsspektrum besitzt. Die Breite des Wirkungsspektrums eines Pflanzenstärkungsmittels ist begrenzt, weshalb die positiven Effekte des verwendeten Mittels häufig geringer ausfallen können als erwartet. Die Arbeitshypothese dieser Arbeit belegt, dass durch die Kombination verschiedener bioaktiver Stoffe mit Pflanzenstärkungsmitteln dieses Wirkungsspektrum erweitert werden kann. Aus diesen Zusammenhängen leitet sich das Interesse an Kombinationen verschiedener Pflanzenstärkungsmittel sowie anderer bioaktiver Stoffe ab. Diese Arbeit untersucht die biologischen Effekte verschiedener Kombinationen von bioaktiven Stoffen und Pflanzenstärkungsmitteln zur Stabilisierung pflanzlicher Systeme. Als bioaktive Stoffe werden in dieser Arbeit Laktate und Humate bezeichnet, wobei Mikroorganismen als Pflanzenstärkungsmittel eingesetzt wurden. Die Ergebnisse dieser Arbeit zeigen, dass ein kombinierter Einsatz von Mikroorganismen, Humaten und Laktaten zum Einen zum Erhalt der chemischen und physikalischen Eigenschaften gärtnerischer Substrate, zum Anderen zur Stabilisierung der Pflanzen gegenüber suboptimalen Wachstumsfaktoren beiträgt. Die Anwendung der Kombination von allen drei Komponenten auf Versuchsvarianten mit Gewächshausgurken als Modellpflanze zeigte, dass die Pflanzenproduktivität bei diesen Varianten am höchsten war. Die physiologischen Reaktionen der Gurkenpflanzen im Versuch mit modellierten Stressfaktoren wurden durch Chlorophyllfluoreszenz ermittelt. Die Ergebnisse zeigten, dass die Pflanzen, die mit allen drei Komponenten behandelt wurden, gegenüber suboptimalen Wachstumsbedingungen resistent sind. Die Resultate dieser Arbeit wurden in einem gärtnerischen Unternehmen approbiert. / Biotic and abiotic stress factors reduce the quality and quantity of the horticultural produce. Even controlled growing conditions of a greenhouse can not reduce all the negative influences of the suboptimal growing factors. The negative effects caused by the suboptimal growing factors manifest themselves only after substantial period of time. That is why it is important to take steps that counter these negative effects and enable sustainable production of fruits and vegetables. To improve the resistance of the plants against different stress factors, the plant strengtheners are applied. The horticultural substrates, which are used for cultivation of the fruits and vegetables, have become unalienable and possess the range of different physical and chemical properties. These properties are subjected to constant influence of different biotic and abiotic factors; especially those resulted from the plant growth itself. On the other hand, the substrates influence the growth of the horticultural plants. The water holding capacity of the substrate and its capacity to provide plants with nutrient elements during the critical vegetation periods can influence the productivity of the plant system. Dynamic changes of the substrate properties during the plant growth and changes in physiological reactions of horticultural plants can be viewed as a system, and as the system it can be influenced for the purposes of its optimization. This influence can be achieved by application of different plant strengtheners of different origin. The role of the plant strengthening agents is being discussed in the literature. The missing point in the discussion however is the fact that the plant strengtheners have their own activity spectrum. The activity spectrum of the bioactive substance is limited and expected positive effects of its application can be lesser that expected. The hypothesis of the thesis is based on the premise that combination of the different bioactive substances and plant strengthening agents can increase the activity spectrum. These interrelations cause an interest on combinations of different plant strengtheners and others bioactive substances. This thesis investigates biological effects different combinations of bioactive substances and plant strengtheners with an aim to stabilize plant systems. In this work, lactates and humates are described as bioactive compounds, whereas microorganisms are perceived as plant strengtheners. The results of this work show that the combined application of microorganisms, humates and lactates sustain chemical and physical properties of the horticultural substrates and at the same time stabilize plant systems under the suboptimal growing factors. The experiment variants with greenhouse cucumbers as a model plant treated with all three components showed the highest productivity. Physiological reactions of the cucumber plants were investigated through measurement of the chlorophyll fluorescence of the cucumber leaves in the experiments with modeled suboptimal growing factors. The results showed that the plants treated with a combination of humates lactates and microorganism were more resistant to suboptimal growing conditions than the plants on variants without treatment. The results of this work were approbated at a horticultural enterprise.

Suivi du métabolisme énergétique cérébral chez les patients victimes d'hémorragies sous-arachnoïdiennes graves : intérêt pour le pronostic individuel et le diagnostic des complications ischémiques / Monitoring of cerebral energy metabolism in patients experiencing severe subarachnoid hemorrhage : interest for the individual prognosis and for the diagnosis of ischemic complications

L'intérêt du suivi du métabolisme énergétique cérébral dans la prise en charge des patients victimes d'hémorragie sous-arachnoïdienne anévrismale (aSAH) grave reste actuellement controversé en raison de l'absence de valeurs seuils décisionnelles applicables en pratique. Ce travail avait pour objectif de réévaluer l'intérêt des paramètres biochimiques de trois techniques, la microdialyse intracérébrale (cMD), la mesure de la pression tissulaire cérébrale en oxygène (PbtO2) et le cathéter rétrograde jugulaire, pour prédire l’issue fonctionnelle de ces patients et diagnostiquer la survenue d'un infarctus. Il parait évident que ce suivi peut permettre de prédire à l'échelon individuel l'issue fonctionnelle à long terme. Le metabolic ratio (MR) ou l'association de ce MR avec des paramètres des deux autres techniques (ratio Lactate/Pyruvate >40, lactates hypoxiques) représentent des potentiels biomarqueurs pronostiques. Il est en revanche difficile de conclure sur l'intérêt de ce suivi pour diagnostiquer les complications ischémiques secondaires. Bien qu'il ait été montré que le MR peut être considéré comme un biomarqueur, il n'est pas possible de conclure actuellement sur les deux approches locales (cMD et PbtO2). Des règles d'implantation ont tout de même pu être identifiées et validées permettant leur application rapide en pratique courante. Au final, le suivi du métabolisme énergétique cérébral doit être envisagé dans la prise en charge des patients aSAH graves notamment pour prédire l'issue fonctionnelle à long terme car des valeurs seuils décisionnelles ont été identifiées et faciliteront ainsi l'utilisation de ce type de monitoring / The interest of cerebral energy metabolism monitoring in the care of patients suffering from aneurysmal subarachnoid hemorrhage (aSAH) currently remains controversial because of the absence of decision making thresholds applicable in practice. This work aimed to reassess the value of biochemical parameters from three techniques, intracerebral microdialysis (cMD), the measurement of brain tissue oxygen pressure (PbtO2), the retrograde jugular catheter to predict the functional outcome and diagnose the occurrence of secondary ischemia.It seems obvious that this monitoring can predict at the individual level the functional long-term outcome. The metabolic ratio (MR) or association of MR with the parameters of the two other techniques (lactate/pyruvate >40, hypoxic lactate) represent potential prognostic biomarkers.It is however difficult to conclude on the interest of such monitoring to diagnose secondary ischemic complications. Although it has been shown that the MR can be considered as a biomarker, it is currently not possible to conclude on the two local approaches (cMD and PbtO2). Nevertheless, implantation rules have been identified and validated for their rapid application in clinical practice.Finally, the monitoring of brain energy metabolism remains a reference technique in the care of serious aSAH patients, especially to predict functional long-term outcome because decision thresholds have been identified and thus will facilitate the use of this kind of monitoring
