The modulating effect of conjugated linoleic acid (CLA) on cancer cell survival in vitro

Magister Scientiae (Medical Bioscience) - MSc(MBS) / Conjugated linoleic acids (CLA) are geometrical and positional isomers of n-6 octadecadenoic acid (linoleic acid, LA, 18:2n-6), which form part of a family of essential polyunsaturated fatty acids (PUFA). There are 28 identified CLA isomers that mostly found in the meat and milk from ruminant animals. CLA has shown to possess a number of health benefits including; reduction in body fat and increased lean body mass, prevention of atherosclerosis, hypertension, increased immune function and in particular the prevention of cancer. The effects of CLA on cancer cell lines will be evaluated to discover the mechanisms that are employed to achieve this great phenomenon on cell growth. The aim of this study was to determine the effect of CLA on various parameters that are essential in the development of cancer cell phenotype. The objectives were to evaluate the effect of CLA on iron-induced lipid peroxidation of microsomes isolated from rat liver cells and in vitro cytotoxicity, cell proliferation and apoptosis in HepG2 hepatocarcinoma cells. The Fatty acid incorporation in HepG2 cells was also assessed.

This work deals with the optimization of the method for determination of conjugated linoleic acid (CLA) using gas chromatography. The summary about formation and occurrence of CLA in animal materials, its biological effects in human organism and methods suitable for its determination is introduced in the theoretical part of this study. The experimental part verify, if the gas chromatography is applicable method for assesment of CLA in selected biological matrices. The chosen method was introduced and verified at FCH of Brno university of technology. At the end the applicability of this method to CLA determination is discussed here.

Fatty Acid Composition of Diets, Metabolism, and Deposition in Subcutaneous Adipose Tissue of Pasture and Feedlot Finished Cattle

An experiment was conducted to evaluate the effects of pasture finishing versus high-concentrate finishing, over time, on fatty acid metabolism in Angus crossbred (n = 24) beef steers. Ruminal fluid, serum, and adipose tissue biposies were obtained on d 0, 28, 84, and 140. Pasture forages and diet ingredient samples were obtained at 14 d intervals to determine nutritive value and fatty acid composition. The high-concentrate diet consisted of corn silage, cracked corn, soybean meal, and a vitamin and mineral supplement. The pasture-finished steers grazed sequentially on triticale (Triticale hexaploide)/annual ryegrass (Lolium multiflorum), alfalfa (Medicago sativa)/orchardgrass (Dactylis glomerata), and a cool-season grass/legume mixture. The high-concentrate diet consisted of 57 % linoleic acid and 7 % linolenic acid (of total fatty acids). The pasture forages contained an average 9 % linoleic acid and 66 % linolenic acid (of total fatty acids). Adipose tissue concentrations of 18:2 cis-9, trans-11 CLA were higher (P < 0.05) in the pasture-finished steers than high-concentrate finished steers. Concentrations of 18:2 cis-9, trans-11 CLA declined in the high-concentrate finished steers (P < 0.05) from d 0 to 28 and d 28 to 84. In the pasture-finished steers concentrations peaked (P < 0.10) on d 28, and remained high throughout the duration of the study. Concentrations of linolenic acid were higher (P < 0.05) in adipose tissue, ruminal fluid, and serum of the pasture-finished steers, compared to the high-concentrate finished steers. In the pasture-finished steers linolenic acid concentrations peaked (P < 0.05) on d 28, and remained high throughout the study. Concentrations of linolenic acid gradually decreased (P < 0.05) over time within the high-concentrate finished steers. Thus, it appears that only a short time is needed to alter the omega-3 and CLA composition of adipose tissue in cattle finished on pasture. / Ph. D.

Polyunsaturated Fatty Acids and the Epidermal Growth Factor Receptor/ Mitogen- Activated Protein Kinase Signal Transduction Cascade in Mammary Cancer

Mammary cancer is one of the leading causes of death in both the human and companion animal population. There are many histological and pathophysiological similarities between human and feline mammary cancer, therefore investigating the molecular pathways of disease progression in one species may provide useful information for both. The EGFR/MAPK signal transduction pathway is upregulated in many human breast tumors, and both EGFR and MAPK have been implicated as independent prognostic indicators for decreased survival times in human breast cancer patients. We report here that active MAPK levels can be detected in both peripheral white blood cells (WBC) and mammary adipose tissue in cats. Adipose tissue levels of active MAPK were similar to those observed in peripheral WBC, suggesting that WBC MAPK might serve as a useful biomarker in the diagnosis or follow-up treatment of disease. PUFA have been reported to influence breast cancer risk in humans, and may modulate the EGFR/MAPK pathway through a variety of mechanisms. Dietary PUFA n-6-to-n-3 ratio in cats was reflected in mammary adipose tissue and resulted in altered active MAPK levels in both adipose tissue and peripheral white blood cells, suggesting that PUFA may have similar effects on the feline and human MAPK pathway. In human breast tumor cell line studies, rather than having opposing effects, as was hypothesized, it was demonstrated that n-6 and n-3 PUFA exerted similar effects on EGFR+ breast tumor cell proliferation and activity of the EGFR/MAPK pathway. Slightly more than twice the concentration of n-3 PUFA was needed to elicit the same response as n-6 in cells. These results indicate n-6 and n-3 PUFA can modulate proliferation of EGFR+ tumor cells similarly and may be exerting their effects, in part, through the EGFR/MAPK pathway. Decreasing total PUFA intake, while increasing the n-3-to-n-6 PUFA ratio, may be practical as preventative or adjuvant therapy for breast cancer. / Ph. D.

The Effect of Antioxidants on Flaxseed Stability in Yeast Bread

The antioxidants BHA, BHT, and ascorbic acid were added to flaxmeal breads to prevent rancidity. Six types of yeast leavened breads were evaluated: control (100% bread flour), flaxmeal (15%) bread, and flaxmeal (15%) bread that contained 0.01% respectively of BHA, BHT, BHA and BHT, and ascorbic acid. Vital wheat gluten was added in all the flaxmeal breads. Chemical, objective and sensory tests were used to evaluate the breads. The crumb texture of all the experimental breads was significantly softer (p¡à 0.05) than the control breads, but the control breads were significantly moister (p¡à 0.05) than the flaxmeal breads that contained BHA and BHT, separately. No significant differences (p>0.05) were found in loaf volume of the control bread and the experimental breads. The crumb color of the experimental breads was significantly darker (p<0.0001) due to the incorporation of flaxmeal. The acid value of the flaxmeal breads was significantly higher (p¡à 0.05) than the control breads. No significant differences (p>0.05) were found in peroxide values between the control breads and experimental breads after eight weeks. The QDA sensory tests showed that breads containing BHA or in combination with BHT were moister, chewier and had the least noticeable stale taste when compared to the control breads. Ascorbic acid was not as effective as BHA or a combination of BHA and BHT in preventing lipid oxidation, but produced the softest bread. This study showed that flaxmeal breads made with BHA and BHT provided the best protection against lipid oxidation and produced a moist and chewy bread. / Master of Science

Concentrations of Conjugated Linoleic Acid Isomers in Human Plasma Reflect Intake of Dairy Products with Enhanced cis-9, trans-11 or tran-10, cis-12 Isomer Content

Dairy products are a primary source of c9, t11-CLA, an anti-carcinogenic agent, in the diet of humans. The t10, c12-CLA isomer, typically in trace amounts in bovine milk fat, also may benefit human health. Four cows received abomasal infusions of c9, t11-CLA or t10, c12-CLA to obtain milk fat used to prepare butter and yogurt with enhanced c9, t11-CLA or t10, c12-CLA content. Human subjects (3 males, 3 females, ages 22 to 29) received CLA-enhanced butter and yogurt (14% of total kcal) in a crossover study with 2-wk periods. Prior to the study (2 wk) and during a 2-wk washout period between the experimental periods, subjects received butter and yogurt without enhanced CLA content. Blood samples were obtained at 0, 2, 4, 6, and 8 wk relative to the start of the first experimental period. The t10, c12- CLA isomer was detected in plasma (1.32 ug/mL) only when dairy products with enhanced t10, c12-CLA was consumed. Baseline c9, t11-CLA was 6.94 ug/mL plasma during control periods, but increased to 8.95 ug/mL when dairy products with enhanced c9, t11-CLA content were consumed. Results indicated concentrations of CLA isomers in human plasma respond to small changes in daily intake of the isomers in dietary sources. / Master of Science

Plantas transgênicas constituem interessantes alternativas para a produção de compostos com elevado valor agregado como polímeros industriais, proteínas farmacológicas e lipídios nutracêuticos. Como candidata à biofábrica, a cana-de-açúcar (Saccharum spp.) apresenta características agronômicas favoráveis, além de versatilidade de matéria-prima, que é empregada em fins tradicionais, como produção de álcool e açúcar, e até mesmo para geração de energia ou como forragem para alimentação de gado. Em nutrição animal, uma molécula bastante estudada é o ácido linoleico conjugado (CLA), que tem atividades anticarcinogênica, antidiabética, antiaterosclerose e moduladora do sistema imune e metabolismo de lipídios. O isômero t10,c12- CLA tem sido particularmente promissor na agropecuária por conta de inibição da síntese de gordura do leite e na melhoria do desempenho reprodutivo de vacas. Com o objetivo de avaliar a cana-de-açúcar como sistema biotecnológico para produção de CLA, o gene da isomerase do ácido linoleico de Propionibacterium acnes (pai) foi isolado e clonado, previamente caracterizado em Escherichia coli e, em seguida, utilizado na biolística de calos embriogênicos para regeneração de plantas transgênicas. No sistema procariótico, foi induzida a expressão de pai e a proteína heteróloga foi verificada como uma banda de 50 kDa, porém não houve alteração evidente do perfil de ácidos graxos da bactéria. Na cotransformação de cana-de-açúcar, o vetor pHA9, que contém o gene de seleção da neomicina fosfotransferase (nptII), foi bombardeado com uma das duas construções desenvolvidas com o promotor da poliubiquitina 1 de milho (pUbi1) dirigindo a expressão de pai adicionado ou não de sequência sinal da proteína de reparo de DNA recA para direcionamento para cloroplasto. Das cultivares CTC2 e IACSP9303046, foram obtidas eficiências de transformações de 2,2% e 1,7-7,1%, respectivamente. Um total de 156 plântulas foram regeneradas após regime de seleção com geneticina. Em seguida, 115 plântulas transgênicas foram identificadas por PCR para nptII e, entre essas, 29 e 48 foram PCRpositivas para pai e rec-pai, respectivamente. De 50 plantas aclimatizadas, 12 foram analisadas por Southern blot para nptII e 4 para pai, confirmando-se a transformação genética. Para comprovar a expressão de mRNA de pai, 27 plantas foram averiguadas por RT-PCR e RT-qPCR e indicaram produção de transcritos estudados. A análise de expressão das proteínas de folha por Western blot em 21 plantas selecionadas não produziu resultados conclusivos quanto à detecção de PAI. Essas mesmas foram analisadas por Cromatografia Gasosa, mas não foi detectado acúmulo de CLA. Embora seja possível a introdução e expressão do gene pai em cana-de-açúcar, é necessário avaliar em maior detalhe os fatores que possam afetar positivamente a produção de CLA, como tipo de tecido, compartimentos subcelulares para direcionamento proteico ou coexpressão de fosfolipases. / Transgenic plants are attractive alternatives for the production of high value compounds as industrial polymers, pharmacological proteins and nutraceutical lipids. As a candidate for biofactory, sugarcane presents favorable agronomic characteristics and versatility of raw material which is used for traditional purposes such as ethanol and sugar production and even for power generation or as forage feeding of cattle. In animal nutrition, a widely studied molecule is the conjugated linoleic acid (CLA), which has anticarcinogenic, antidiabetic, antiatherosclerosis and immune system and metabolism of lipids modulator activities. The isomer t10,c12-CLA has been particularly promising in agriculture because of inhibition of milk fat synthesis and improvement of the reproductive performance of cows. Aiming to evaluate the sugarcane as a system for biotechnological production of CLA, the linoleic acid isomerase gene from Propionibacterium acnes (pai) was isolated and cloned, previously characterized in Escherichia coli and then used for biolistic of embryogenic calli for regenerating transgenic plants. In the prokaryotic system, the expression of pai was induced and the heterologous protein was verified as a band of 50 kDa, but there was no obvious change in fatty acid profile of the bacterium. In cotransformation of sugarcane, pHA9 vector containing the selection gene neomycin phosphotransferase (nptII) was bombarded with one of the two constructions developed with the promoter of maize polyubiquitin 1 (pUbi1) driving the expression of pai, either added or not with the signal sequence of DNA repair protein recA for targeting to chloroplast. Cultivars CTC2 and IACSP93-3046 were obtained with transformation efficiencies of 2.2% and 1.7 a 7.1%, respectively. A total of 156 plantlets were regenerated after selection with geneticin regimen. After that, 115 transgenic plantlets were identified by PCR for nptII and among then, 29 and 48 were PCR-positive for pai and rec-pai, respectively. Of 50 acclimatized plants, 12 were analyzed by Southern blot for nptII and 4 for pai, confirming the genetic transformation. In order to prove the expression of pai mRNA, 27 plants were verified by RT-PCR and RT-qPCR and indicated the production of the studied transcripts. Expression analysis of leaf proteins by Western blot in 21 plants selected produced no conclusive results regarding the detection of PAI. Those were analyzed by gas chromatography and no accumulation of CLA was detected. Although it was possible to introduce and express the pai gene in sugarcane, it is necessary to evaluate in more detail the factors that may positively affect the production of CLA, as tissue type, subcellular compartments for protein targeting or coexpression of phospholipases.
