Spectral, spatial and temporal properties of multilayered epithelial tissue in vivo in presence of metal nanoparticles in multimodal spectroscopy / Propriétés spectrales, spatiales et temporelles de tissus épithéliaux multicouches in vivo en présence de nanoparticules métalliques en spectroscopie multimodalités

Le travail de thèse est consacré aux interactions spatio-, temporo- et spectro- résolue de laser avec des tissus biologiques. L'objectif de cette thèse était d'étudier l'influence des nanoparticules colloïdales embarqués dans les tissus biologiques multicouches sur leurs propriétés optiques afin de fournir plus profond et / ou plus précise de sondage. Pour ce faire, les paramètres spectroscopiques intégrales et durée de vie de fluorophore dans les environs de nanoparticules métalliques ont été analysées théoriquement et expérimentalement. L’autre partie de l'étude était de proposer de nouvelles solutions algorithmiques pour l'amélioration de la performance du processus d'estimation des valeurs des propriétés optiques de la résolution spatiale des mesures spectroscopiques. La dernière partie de la thèse est la modélisation expérimentale et théorique de la cinétique de fluorophores en présence des nanoparticules d'or colloïdales. La composante ultra-courte pico-seconde (environ 100 ps) a été résolue et a été corrélée à champ dipolaire forte des nanoparticules qui compense le dipôle de la molécule / The thesis work is devoted to spatially-, temporally- and spectrally- resolved laser and biological tissue interactions. The aim of the present thesis was to investigate the influence of colloidal nanoparticles embedded into multilayered biological tissues on their optical properties in order to provide deeper and/or more precise probing. To do so, the integral spectroscopic parameters and lifetime of fluorophore in vicinity of metal nanoparticles were analyzed theoretically and experimentally. Another part of the study was to propose new algorithmic solutions for improving the performance of the estimation process of the optical properties values from spatially resolved spectroscopic measurements. The last part of the thesis was the experimental and theoretical modelling of fluorophore’s kinetics in presence of colloidal gold nanoparticles. The ultra-short pico-second component (around 100 ps) was resolved and correlated to strong nanoparticles dipole field which is compensating the molecule’s dipole

Ab initio simulation of optical properties of noble-metal clusters / Modélisation des propriétés optiques de nanoparticules métalliques

L'intérêt de la recherche fondamentale pour les morceaux nanométriques de métaux nobles est principalement dû à la résonance localisée des plasmons de surface (LSPR) dans l'absorption optique. Différents aspects, liés à la compréhension théorique de la LSPR dans le cas de clusters de métaux nobles de taille dite intermédiaire, sont étudiés dans ce manuscrit. Afin d'avoir une vision plus large nous utilisons deux approches : l'approche électromagnétique classique et le formalisme ab initio en temps réel de la théorie de la fonctionnelle de la densité dépendant du temps (RT-TDDFT). Une comparaison systématique et détaillée de ces deux approches souligne et quantifie les limitations de l'approche électromagnétique lorsqu'elle est appliquée à des systèmes de taille quantique. Les différences entre les excitations plasmoniques collectives et celles impliquant les électrons d, ainsi que leurs interactions, sont étudiées grâce au comportement spatial des densités correspondantes. Ces densités sont obtenues en appliquant une transformée de Fourier dans l'espace à la densité obtenue par les simulations DFT utilisant une perturbation delta-kick. Dans ce manuscrit, des clusters de métaux nobles nus et protégés par des ligands sont étudiés. En particulier, motivé par de récents travaux sur les phénomènes d'émergence de plasmon, l'étude par TD-DFT de nano-alliages Au-Cu de taille tout juste inférieure à 2nm à fourni de subtiles connaissances sur les effets d'alliages sur la réponse optique de tels systèmes. / The fundamental research interest in nanometric pieces of noble metals is mainly due to the localized surface-plasmon resonance (LSPR) in the optical absorption. Different aspects related to the theoretical understanding of LSPRs in `intermediate-size' noble-metal clusters are studied in this thesis. To gain a broader perspective both the real-time \ai formalism of \td density-functional theory (RT-TDDFT) and the classical electromagnetics approach are employed. A systematic and detailed comparison of these two approaches highlights and quantifies the limitations of the electromagnetics approach when applied to quantum-sized systems. The differences between collective plasmonic excitations and the excitations involving $d$-electrons, as well as the interplay between them are explored in the spatial behaviour of the corresponding induced densities by performing the spatially resolved Fourier transform of the time-dependent induced density obtained from a RT-TDDFT simulation using a $\delta$-kick perturbation. In this thesis, both bare and ligand-protected noble-metal clusters were studied. In particular, motivated by recent experiments on plasmon emergence phenomena, the TDDFT study of Au-Cu nanoalloys in the size range just below 2~nm produced subtle insights into the general effects of alloying on the optical response of these systems.

Práce pojednává o vlastnostech plazmonických antén v infračervené a viditelné oblasti. Práce zahrnuje výrobu, měření a numerické modelování optických vlastností antén. Infračervené plazmonické antény na absorbujícím substrátu (SRON) jsou studovány pro jejich rezonanční a absorpční vlastnosti. Byla nalezena geometrie antény, která poskytuje maximální účinnost absorpce ve SRON vrstvě. Dále je studována možnost zesílení daného vibračního módu substrátu (obsahujícího 3-4 materiálové rezonance) pomocí plazmonické rezonance antény. Nakonec jsou prezentována měření katodoluminiscenčních spekter antén ve viditelném spektru.

Enhancement of Light Emission from Metal Nanoparticles Embedded Graphene Oxide

A fully oxidized state of graphene behaves as a pure insulating while a pristine graphene behaves as a pure conducting. The in-between oxide state in graphene which is the controlled state of oxide behaves as a semiconducting. This is the key condition for tuning optical band gap for the better light emitting property. The controlling method of oxide in graphene structure is known as reduction which is the mixed state of sp2 and sp3 hybrid state in graphene structure. sp2 hybridized domains correspond to pure carbon-carbon bond i.e. pristine graphene while sp3 hybridized domains correspond to the oxide bond with carbon i.e. defect in graphene structure. This is the uniqueness of the graphene-base material. Graphene is a gapless material i.e. having no bandgap energy and this property prevents it from switching device applications and also from the optoelectronic devices applications. The main challenge for this material is to tune as a semiconducting which can open the optical characteristics and emit light of desired color. There may be several possibilities for the modification of graphene-base material that can tune a band gap. One way is to find semiconducting property by doping the defects into pristine graphene structure. Other way is oxides functional groups in graphene structure behaves as defects. The physical properties of graphene depend on the amount of oxides present in graphene structure. So if there are more oxides in graphene structure then this material behaves as a insulating. By any means if it can be reduced then oxides amount to achieve specific proportion of sp2 and sp3 that can emit light of desired color. Further, after achieving light emission from graphene base material, there is more possibility for the study of non-linear optical property. In this work, plasmonic effect in graphene oxide has been focused. Mainly there are two kinds of plasmon effects have been studied, one is long range (surface) and short range (localized) plasmon. For long range plasmon gold thin film was deposited on partially reduced graphene oxide and for short range plasmon silver nanoparticles have used. Results show that there are 10-fold enhancement in light emission from partial graphene oxide coated with gold thin film while 4-fold enhancement from reduced graphene oxide solution with silver nanoparticles. Chemical method and photocatalytic method have been employed for the reduction of graphene oxide for the study of surface plasmon and localized plasmon. For the characterization UV-Vis spectrometer for absorption, spectrofluorophotometer for fluorescent emission, Raman spectrometer for material characterization, photoluminescence and time resolved photoluminescence have been utilized. Silver and gold nanoparticles are spherical of average size of 80 nm and 40 nm have been used as plasmons.
