CONSTRAINED VOLUME PACKING OF DEPLOYABLE WINGS FOR UNMANNED AIRCRAFT

UAVs are becoming an accepted tool for sensing. The benefits of deployable wings allow smaller transportation enclosures such as soldier back packs up to large rocket launched extraterrestrial UAVs. The packing of soft inflatable wings and Hybrid inflatable with rigid section wings is being studied at the University of Kentucky. Rigid wings are volume limited while inflatable wings are mass limited. The expected optimal wing design is a hybrid approach. Previous wing designs have been packed into different configurations in an attempt to determine the optimal stowed configurations. A comparison of rigid, hybrid, and inflatable wings will be presented. Also a method for simulating optimally packed wings with respect to geometric constraints will be presented. A code has been written to study soft wing packing and verified the soft wing packing results. This code can be used during initial wing design to help predict wing size and packing configurations. In this thesis, an over view of the packing configurations as well as packing observations will be covered such , packing inefficiencies, wing mounting limits, long term storage, and scaling of packing.

The Effect of Long Term Storage of Nonfat Dry Milk On Sensory Quality and on the Development Of Oxidative Derivatives of Sterols

Nonfat dry milk (NOM) was evaluated to determine the effect of long term storage on its sensory quality, its solubility index and on the development of oxidative derivatives of sterols. Storage variables included the form of the milk, the atmosphere in the storage container, the type of package and the storage temperature. A trained taste panel evaluated the milk for the development of cooked, metallic, and oxidized/stale flavors, mouthfeel and overall acceptability. Gas chromatography was used to determine cholesterol content and the quantity of oxidative derivatives of cholesterol present in the samples. Although long term storage of NOM did not affect the solubility index of the milk, the sensory quality of the milk was affected. Over time storage temperature and type of package had the greatest effect on sensory quality. Milk stored for 30 months at 32 C developed an unacceptable oxidized/stale flavor and at 42 months of storage, milk stored in polybags at 21 C had also developed an oxidized/stale flavor. These results substantiate the importance of controlling storage conditions during long term storage of NOM. NOM stored for four years did not develop harmful quantities of oxidative derivatives of cholesterol. However, certain expected results did occur. NOM stored in polybags developed more oxidative derivatives than samples stored in cans. Samples stored at 32 C developed more oxidative derivatives of cholesterol than any other sample analyzed and 7S-hydroxycholesterol, the only known cytotoxic derivative of cholesterol identified, was present in the samples stored at 32 C. Fresh and control samples of NOM contained similar quantities of oxidative derivatives of cholesterol.

Examination of defects and long term crack growth in rolled bars during long term storage

This work has had the purpose to examine defects and the cause of crack growth during long term storage in rolled bars. The bars in question are rolled and stored at Ovako in Hofors. The problem is approached by theoretically examining the possibilities of hydrogen damage, room temperature creep and naturally occurring porosity. Seven bars of three steel type groups and with different rolling dates were selected and examined for defects with ultrasonic testing. Four of the bars show defects. Specimens containing the defects are cut out and grinded so that the defects can be examined in the scanning electron microscope and analyzed with energy dispersive x-ray spectroscopy. One of the specimens shows some form of cavity containing iron carbide. One of the others shows a surface with inclusions. The remaining two both shows inclusions and cracks that seem to originate from inclusions. The theoretical studies show that there are two types of hydrogen damage that could be in effect in the studied case and also that room temperature creep could facilitate but not cause crack growth and that porosity could serve as crack initiation points. Steps should be taken to minimize the problems mentioned above when they are suspected causes. Further examination of inclusions in different steel types should be made if considered a problem. / Detta arbete har haft syftet att undersöka defekter och orsaken till spricktillväxt under långtidsförvaring av spårvalsade stänger. Stängerna i undersökningen är valsade och lagrade hos Ovako i Hofors. Problemet är angripet genom att teoretiskt undersöka risken för väteskador, kryp i rumstemperatur och naturligt förekommande porositet. Sju stänger indelade i tre stålsortsgrupper och med olika valsningsdatum valdes ut och undersöktes med ultraljud. Fyra av stängerna visade på defekter. Prov innehållande dessa defekter blev utkapade och slipade så att defekterna kan bli undersökta i svepelektronmikroskop och analyserade med energidispersiv röntgenanalys. Ett av proverna visar på ett hålrum innehållande en järnkarbid. Ett av de andra proverna visar på en yta med inneslutningar. De två återstående proverna visar på inneslutningar och sprickor som verkar utgå från inneslutningar. De teoretiska studierna visar att det finns två typer av väteskador som är aktuella i det studerade fallet och även att kryp vid rumstemperatur kan underlätta spricktillväxt men inte orsaka den och att porositet kan tjäna som sprickinitieringsställen. Åtgärder för att minimera de nämnda problemen bör tas i fall där de är misstänkta orsaker. Ytterligare undersökningar av inneslutningsbilden bör göras om inneslutningar anses vara ett problem.

Development of Business Models for Electrical Energy Storage in Europe - Techno-economic evaluation of combining storage services / Utveckling av affärsmodeller for lagring av el i Europa - Tekno-ekonomisk utvärdering av kombinerade lagringstjänster

Europe aims for a transition towards less greenhouse gas emission and dependency on fossil fuels. The integration of intermittent renewable energy sources, as wind or solar power, can be facilitated by, among others, temporally decoupling demand and supply of electricity. If technologies for electrical energy storage are profitable, wide implementation could support the transition. Therefore, this study assesses the revenues and costs of long-term storage technologies and evaluates the possibility to stack several services provided by a storage unit to fully utilise it. Key market characteristics influential on storage potential are outlined and used to classify and compare the demand for storage in European markets. Considering them market clusters are formed comprising of countries with similar electricity market and from these two contrasting archetypal countries are chosen for further evaluation, Sweden and Germany. Storage technologies, as pumped hydro or compressed air, are delineated by essential technical specifications and used to determine the compatibility of them to corresponding services. A process for combining multiple services for a single storage unit is designed and employed to develop five cases. The results show that providing multiple services device can improve the profitability of the designed business cases by generating multiple revenue streams and increase the value to the electricity system. The stacking also minimises the storage’s idle time. Furthermore, the results demonstrate the influence of the key characteristics on the economic viability of the electrical energy storage in European markets. / Europa eftersträvar en övergång från att släppa ut mindre växthusgaser och minska eroendet av fossila bränslen. Att implementera förnybara energikällor, som vindkraft ller solenergi, kan underlättas genom att bland annat genom att tillfälligt rikoppla efterfrågan och leveransen av el. Om tekniken för energilagring är lönsam, an genomförande därför stödjas. Denna studie beskriver därför både intäkter ch kostnader för långsiktiga lagringstekniker och utvärderar även möjligheten att mplementera flera tjänster som tillhandahålls av en lagringsenhet till att kunna tnyttja denna till fullo. Marknaden av lagring kännetecknas redan ha potentiella förutsättningar och att den an användas för att klassificera och jämföra efterfrågan för lagring på de europeiska arknaderna. En marknadsanalys har valts att genomförts av länder med likartad lmarknad och de två länder som har välts ut för en vidare utvärdering är Sverige ch Tyskland. Lagringsteknik som vattenkraft som pumpas eller tryckluft, beskrivs ärmare med tekniska förutsättningar och används för att beskriva kompatibiliteten v tekniken och tjänsterna. Ett förfarande för att leverera flera tjänster från en enda innesenhet är utformad och används för att utveckla fem beskrivande fall. Resultatet visar att tillhandahålla flera tjänster från en enda lagringsenhet förbättrar önsamheten för affärsidéer genom att generera flera intäktsströmmar och ökar ärdet till elsystemet. Stapling minimerar också lagring är ledig tid. Dessutom visar esultatet om hur de viktigaste egenskaperna påverkas av de ekonomiska affärsmodellerna.

Změny aktivit enzymů v ovoci v průběhu dlouhodobého uchovávání / Changes of enzyme activities in fruits during long-term storage

This study is focused on study of changes of enzyme and low-molecular weight antioxidants in different fruits during long-term storage. In theoretical part individual low-molecular weight antioxidants and enzymes are described. The main causes of fruit decay and some possibilities of fruit preservation and storage are summarized. As biological material some common fruits were chosen - green and red apples, peaches, plums and white grapes. The fruits were stored in laboratory, cellar, in refrigerator and in freezer. In freezing experiments some ways of fruit preparation and processing were tested and their influence on fruit antioxidant status was compared. Shortened storage experiment was applied on blueberries, cranberries, raspberries and strawberries too. In fruits some group parameters – total antioxidant status, dry mass content, ascorbate level, total flavonoids and total phenolics were analyzed spectrophotometrically. Individual flavonoids and phenolics were determined by RP-HPLC/UV-VIS and on-line LC/PDA/ESI-MS. Antioxidant enzyme activities (superoxide dismutase SOD, catalase CAT, polyphenol oxidase PPO and lipoxygenase LOX) were measured by spectrophotometry. The surface microscopy and cultivation of moulds from fruit surface were performed too. Influence of storage conditions on biological activities is dependent on fruit sort. Freezing is the most suitable procedure for long-term storage without significant changes of active substance content. Long-term storage in controlled temperature conditions and/or atmosphere is usable for fruits with longer storage period. In these fruits stabile levels of antioxidant enzymes are stored for relatively long time. Some of enzymes act synergistically. Enzyme activities differed according to storage phase; at the beginning mainly high SOD and LOX activities were observed. CAT and PPO are probably activated as defence systems in rippened and/or damaged fruits. Levels of total as well as individual low molecular weight antioxidants varied during storage in all sorts, generally, increased course with longer storage period can be observed.

Les systèmes de stockage de chaleur par sorption (SSCS) ouvrent de nouvelles perspectives dans l'exploitation de l'énergie solaire pour le chauffage des bâtiments résidentiels. En effet, ces systèmes sont très prometteurs dans la mesure où ils permettent un stockage de chaleur sur de longues périodes (le stockage est réalisé sous forme de potentiel chimique) et offrent des densités énergétiques importantes (jusqu'à 230 kWh/m3 de matériau en moyenne) en comparaison aux systèmes classiques comme le stockage par chaleur sensible (qui, pour le cas de l'eau, dispose d'une densité énergétique moyenne d'environ 81 kWh/m3 de matériau pour une variation de 70°C) et le stockage par chaleur latente (qui atteint des densités énergétiques de 90 kWh/m3 de matériau).La présente thèse vise à étudier les performances d'un système de stockage de chaleur par sorption à base de zéolithe 13X intégré à un bâtiment type basse consommation. Des modèles mathématiques de transferts couplés de masse et de chaleur des différents composants du système sont développés et validés par le biais de l'expérimentation. La simulation numérique dynamique, comme outil de dimensionnement, permet, à partir des résultats d'analyses de sensibilité paramétrique sur les différents composants du système, l'étude de son fonctionnement et les critères de sa faisabilité. / Sorption heat storage systems (SHSS) open new perspectives for solar heating of residential buildings. These systems allow long term heat storage (storage is done in the form of chemical potential) and offer high energy densities (up to 230 kWh/m3 of material on average) compared to conventional heat storage systems such as sensible heat storage (which, for the case of water, has an average energy density of approximately 81 kWh/m3 of material for a temperature change of 70 °C) and latent heat storage (nearly reaching energy densities of 90 kWh/m3 of material on average).This thesis aims to study the performance of a sorption solar heat storage system on zeolite 13X, integrated to low-energy building. Mathematical models of coupled heat and mass transfer of various components of the system are developed and validated through experimentation. Numerical dynamic simulations allow to study the functioning of the SHSS in specific conditions, and its design with the results from the parametric sensitivity analysis on its components.

Protein adsorption and denaturation in injectable devices for pharmaceutical applications / Adsorption et dénaturation des protéines dans des dispositifs injectables pour des applications pharmaceutiques

Protéines sont largement utilisés dans la formulation dans le domaine pharmaceutique et de jouer un rôle important dans les fonctions biologiques. Il est bien connu que l'adsorption de protéines sur la surface solide est toujours observé pour un stockage à long terme, ce qui entraînera une réduction de la dose de substance active ou une perte de l'activité biologique. Dans certains cas, une courte période de contact avec la surface est suffisante pour modifier fortement la conformation des protéines : par exemple, l'insuline pertes 52% de son activité biologique après 5 minutes de contact avec la surface de verre, ainsi qu'une perte de 30% d’activité biologique du cétrorélix est observé après 2 heures de contact. Parmi tous les paramètres, la dénaturation des protéines est fortement liée à sa stabilité des propriétés de surface. La compréhension de l'adsorption de protéines est devenue une question cruciale dans l'industrie pharmaceutique.Pour mieux comprendre le comportement des protéines à la surface, la quantification des protéines adsorbées et sa conformation devrait être étudiée. L'objectif de notre recherche sera de comprendre les comportements des protéines sur différents surfaces de seringue pré - remplie classique.Le principal objectif de ce projet est de comprendre le comportement de plusieurs modèles de protéines comme la sérum d’albumine bovine (BSA), le lysozyme (LSZ) et la myoglobine (MGB) en contact avec des surfaces de seringues pré-remplie comme le verre et l’élastomère. Nous proposons d'utiliser la chromatographie liquide à haute performance (HPLC) pour la quantification de protéine adsorbée sur une surface plane en déterminant la déplétion des protéines en solution. La réflexion totale atténuée infrarouge à transformée de fourier (FTIR-ATR) spectroscopie est utilisée de suivre les changements structurels des protéines adsorbées sur des surfaces solides. It is well known that protein adsorption on solid surface is always observed for a long-term storage, which will result in a reduced dose of active compound or a loss of biological activity. In some cases, only short time of contact are sufficient to drastically modify the protein conformation: for instance, insulin losses 52% of its biological activity after 5 minutes contacting with glass surface, as well as a loss of 30% of cetrorelix is observed after 2 hours. Among all parameters, the time frame of the denaturation process is strongly related to the protein stability and surface properties. The understanding of protein adsorption has therefore become a crucial issue in the pharmaceutical industry.To gain a better understanding of proteins’ behavior on the surface, adsorbed protein quantification and its conformation should be studied. The objective of our research in a first will be to understand proteins’ behaviors on various surfaces which composed a classical prefilled syringe.The main goal of this PhD project is to understand the behaviors of several model proteins like bovine serum albumin (BSA), lysozyme (LSZ) and myoglobin (MGB) in contact with the surfaces of prefilled syringes such as glass and elastomer. We propose to use the high performance liquid chromatography (HPLC) to quantify the amount of protein adsorbed on a flat surface by determining the depletion of the proteins in solution. Reduced Burst Release of Bioactive rhBMP-2 from a Three-phase Composite Scaffold

Recombinant human bone morphogenic proteins (rhBMPs) are extensively studied and employed clinically for treatment of various bone defects. Current clinical delivery vehicles suffer wasteful burst releases that mandate supra-physiological dosing driving concerns over safety and cost. It was therefore investigated whether a unique drug delivery vehicle sequestered within a composite scaffold could lower the burst release of rhBMP-2. PLGA-calcium phosphate tri-phasic composite scaffolds delivered model protein BSA with burst release of ~13% and sustained kinetics of 0.5-1.5% BSA/day up to 45 days. rhBMP-2 was delivered with zero burst release however at much lower levels, totaling 0.09% to 0.9 % release over 10 days, but had up to 6.3-fold greater bioactivity than fresh rhBMP-2 (p<0.05). In conclusion, the three-phase composite scaffold can deliver bioactive proteins with a reduced burst release and sustained secondary kinetics.
