The Genetic Basis of Phytate, Oligosaccharide Content, and Emergence in Soybean

Soybean [Glycine max (L.) Merr] is one of the U.S.'s most economically important crops due to the protein and oil content of seeds. The major storage form of phosphorus in soybean seeds is found in the form of phytate, but because of its negative nutritional and environmental impacts, seed phytate and raffinosaccharide content have been a recent focus of breeders and molecular geneticists. The soybean line CX1834 is a low phytate mutant known to have two low phytate QTLs on linkage groups (LGs) L and N. The first objective of this research was to determine the genetic basis of the low phytate trait in CX1834. By using the whole genome sequence, we identified two candidate multidrug resistance-associated (MRP) ABC transporter genes. Sequencing the genes from CX1834 and comparing them to the reference genome sequence revealed a single nucleotide polymorphism (SNP) in the MRP gene located on LG N (causing a stop codon), and a SNP mutation in the MRP gene located on LG L (causing an amino acid change from arginine to lysine). One major concern with low phytate soybeans is the low seedling emergence. The second objective was to undertake a population-wide study of emergence in the recombinant inbred population CX1834 x V99-3337, over two years and two locations. We found a positive correlation between phytate level and emergence, and that variation among year, location, genotypic class, year x genotypic class, and year x location interactions were significantly affecting emergence. V99-5089, in addition to being low phytate, has high sucrose and low raffinosaccharide content. This phenotype of V99-5089 has been previously determined to be due to a SNP mutation in its myo-inositol phosphate synthase (MIPS) gene located on LG B1. The third objective was to use the recombinant inbred population derived from CX1834 x V99-5089 to observe the combinations of all three mutations to see how the different alleles impact phytate and raffinosaccharide content. The individuals with all three mutations, as well as those with the two MRP mutations together had lower phytate than the other genotypic classes. However, these lines (all three mutations) had unexpectedly high stachyose. / Ph. D.

Therefore, many small and medium-sized enterprises (SMEs) have attempted to manage this situation by using tools such as the Q mode, which continuously reviews inventories but does not indicate when and in what quantity a company must supply its sales outlets but requires a more complex level of supplier development and supply chain management efficiency than SMEs can achieve. Likewise, with our proposal we achieved a 50% decrease in stock depletion, based on an analysis of model results, with a confidence level of 95% and a certainty level of 75.30%, projecting a profit of 59000 soles with a return cost of 1.41 and a recovery period of 2 years and 4 months, ensuring both the sustainability and profitability of the proposal.

Nenhum sistema ou lógica específica é a solução para todos os problemas de administração industrial. Desta forma as empresas estão adotando a integração entre lógicas e sistemas, no intuito de atingir melhores soluções. Entre outras miscigenações, as empresas estão buscando unir as lógicas dos MRP com os sistemas de produção enxuta. Portanto o objetivo deste trabalho é modelar a implantação de um sistema MRP (Material Requirement Planning), como parte da implantação de um ERP (Enterprise Resource Planning) em uma empresa produtora de implementos agrícolas, a qual possui o sistema de produção enxuta implantado desde 1998. Esta proposta está fundamentada na teoria da inovação, na teoria de redes, nos sistemas de produção enxuta, nos sistemas ERP (Enterprise Resource Planning) e nos sistemas híbridos de produção, os quais utilizam tanto componentes dos sistemas ERP, como o MRP neste caso, quanto conceitos de sistemas de produção enxuta. O recorte analítico de redes de inovação permite verificar as ligações e os relacionamentos entre empresas e entre os departamentos dentro da mesma corporação. A análise é realizada a partir do projeto de implantação do MRP, passando pela entrada do MRP no ar até a sua estabilização no ambiente produtivo. O ponto principal é que o sistema MRP auxilie as operações da empresa em agilidade no tempo de resposta sobre oscilações de demanda, facilite o processo de criação e controle de filias em outros paises que utilizam componentes produzidos na matriz e garanta cálculos de estoques mais precisos. Como resultado final, apresenta-se a modelagem organizacional feita com a metodologia EKD para representar os atores e os recursos que devem estar envolvidos neste processo de implantação do MRP para a nova configuração do sistema produtivo. / No system or specific logic is the solution to all the problems of industrial management. Thus companies are adopting the integration of logic and systems in order to achieve better solutions. Among other mixtures, companies are seeking to unite with the logics of MRP systems of lean production. So the goal of this paper is to shape the deployment of a system MRP (Material Requeriment Planning), as part of the deployment of an ERP (Enterprise Resource Planning) in a company producing agricultural implements, which has a system of lean production deployed since 1998. This proposal is based on the theory of innovation in the theory of networks, systems of lean production in ERP (Enterprise Resource Planning) systems and hybrids systems, which uses both components of ERP systems such as MRP in this case, as concepts of systems of lean production. The indentation test networks of innovation allows you to check the connections and relationships between companies and between departments within the same corporation. The analysis is performed from the project deployment of MRP, through entry of MRP in the air until its stabilization in the productive environment. The main point that the MRP system assists the company\'s operations in agility in response time for on fluctuations in demand, facilitates the process of creating and controlling filias in other countries that use components produced in the matrix and ensures more accurate calculations of stock. As a final result, it is the modeling organizational made to the methodology EKD to represent the actors and resources that should be involved in this process of implementation of MRP for the new configuration of the production system.

Funktion und Zusammensetzung der nukleären RNase P und RNase MRP in Pflanzen

In allen Organismen wird die genetische Information der DNA in mRNA umgeschrieben und diese im Cytosol als Vorlage für die Proteinbiosynthese benutzt. Dabei dienen tRNAs als codonspezifische Adaptermoleküle, die als Vorläufermoleküle (pre-tRNA) transkribiert werden. Eine Reaktion bei der Reifung übernimmt die Ribonuklease P (RNase P). Sie ist eine essentielle Endonuklease, die die 5\\\'-Flanke von pre-tRNAs entfernt. Eine weitere Endonuklease ist die RNase MRP (Mitochondrial RNA Processing), die mit der RNase P bis zu zehn Proteine gemeinsam hat. Ein Unterschied ist die eigene RNA-Untereinheit sowie die Funktion. RNase MRP ist bei der Reifung der mitochondrialen 5,8S rRNA beteiligt. Während die RNase P/MRP in den Eukaryoten Hefe und Mensch sehr detailliert untersucht werden, gibt es bis dato nur wenig Wissen über die RNase P/MRP in Pflanzen. In dieser Arbeit wurden vier Proteine, die homolog zu den menschlichen RNase P/MRP Proteinen sind, identifiziert und näher charakterisiert. Zwei als RNase MRP-Typ annotierte RNAs wurden in dieser Arbeit in vivo nachgewiesen. Der Fokus dieser Arbeit lag dabei in der Charakterisierung der Transkriptions- und Translationsprodukte sowie deren Beziehung zu RNase P/MRP. Dabei wurden annotierte mRNAs bestätigt und neue Spleißvarianten identifiziert. Mit spezifischen Antikörpern, die die identifizierten Proteine erkennen, konnten die Proteine im Arabidopsis thaliana Proteinextrakt sowie im Weizenkeimproteinextrakt nachgewiesen werden. Mit AtPOP1p-spezifischen Antikörpern konnte RNase P-Aktivität aus einem Proteinextrakt isoliert werden. Darin befand sich auch eine der beiden RNase MRP RNAs, die Bestandteil der RNase P Funktion sein könnte. In dieser Arbeit wurden Grundlagen geschaffen, um das RNase P/MRP-System in Pflanzen genauer zu verstehen und zu untersuchen.
