Devido à liberação de substâncias tóxicas, os cnidários apresentam poucas associações com outras espécies, entretanto as associações mais conhecidas são frequentemente descritas como obrigatórias ou comensais facultativas. Um dos grupos deste filo, Ceriantharia, é representando por membros genericamente conhecidos por ceriantários ou anêmonas-de-tubo cuja característica mais marcante é a presença de uma secreção celular única (pticocisto) que auxilia na construção de um tubo, sintetizado pelo próprio animal, que fica inserido no substrato e abriga o animal. Embora existam estudos quanto ao histórico, taxonomia e posição sistemática, análises moleculares e morfológicas e ciclo de vida, estudos a respeito de associações interespecíficas ocorrendo em tubos de ceriantários são escassos. Logo, pouco se conhece sobre essas associações bem como os métodos que as espécies utilizam para se ancorarem aos tubos desses animais. Sendo assim, esse estudo elencou espécies que ocorrem em tubos de Ceriantharia, abordando principalmente os métodos de ancoragem e hábitos alimentares envolvidos nas relações observadas. Os tubos de 8 espécies de ceriantários foram analisados, fornecendo dados quanto a associações de Annelida, Crustacea e Mollusca aos tubos. / The phylum Cnidaria is characterized by animals that carry stinging cells named cnidocytes in their tissues. Due to the release of toxic substances, cnidarians have few associations with other species, however mostly known associations are often described as mandatory or optional commensalism. One of the groups in this phylum, Ceriantharia, is represented by members generally known as ceriantharians or tube-dwelling anemones whose most remarkable feature is the presence of a peculiar cell secretion (ptychocyst) that supports the construction of a tube, synthetized by the animal itself, and inserted into the substrate, lodging the animal. Although there are some studies concerning the history, taxonomy and systematic position, molecular and morphological analyzes and life-cycle, studies regarding interspecific associations occurring with ceriantharian tubes are scarce. Thus, little is known about these associations as well as the methods that the species use in order to anchor on these tubes. Thereby, this study listed species occurring in ceriantharian tubes mainly discussing the anchoring methods and feeding habits involved in the associations observed. Avaliação ecotoxicológica do fármaco Triclosan para invertebrados marinhos / Ecotoxicological assessment of the pharmaceutical Triclosan for marine invertebrates

Triclosan é um composto orgânico de baixa solubilidade que vem sendo utilizado em formulações de cremes dentais e faciais, xampu, sabonetes, embalagens de gêneros alimentícios e diversos tipos de materiais, tais como, adesivos, brinquedos, sapatos, selantes, tintas, colchão, roupas, pisos, toldos e rejuntes. O amplo uso deste composto deve-se à grande eficácia contra bactérias Gram negativas e Gram positivas. Por seu extenso uso, evidências da presença de Triclosan têm sido frequentemente relatadas em efluentes urbanos e industriais, águas superficiais e sedimentos de ambientes dulcícolas, estuarinos e marinhos, como também em organismos aquáticos como algas, peixes e mamíferos. Neste contexto, o presente estudo avaliou a toxicidade aguda e crônica de Triclosan para diferentes invertebrados marinhos de águas tropicais. Para tanto, ensaios de toxicidade aguda foram realizados com o copépodo Nitokra sp (mortalidade) e com o ouriço-do-mar Lytechinus variegatus (taxa de fertilização). Para a avaliação do efeito crônico, ensaios de toxicidade de curta duração (desenvolvimento embriolarval) foram realizados com o ouriço-do-mar L. variegatus e Perna perna. Além desses métodos, o ensaio do Tempo de Retenção do Corante Vermelho Neutro foi empregado com a finalidade de se avaliar os efeitos do Triclosan sobre a estabilidade da membrana lisossômica de hemócitos de P. perna. Na avaliação do efeito agudo, o valor médio da CL(I)50;96h encontrada para o copépodo foi de 0,20 mg.L-1 enquanto que o valor médio da CI(I)50;1h para ouriço-do-mar foi de 0,28 mg.L-1. Já na avaliação do efeito crônico, o valor médio da CI(I)50;24h para ouriço-do-mar foi de 0,14 mg.L-1 e para o molusco bivalve a média da CI(I)50;48h, foi de 0,13 mg.L-1. O efeito na estabilidade da membrana lisossômica de hemócitos de P. perna ocorreu em concentrações a partir de 12 ng.L-1. Estes resultados evidenciam o risco ecológico da introdução contínua desse composto em ambientes marinhos, e devem ser considerados para identificação de concentrações seguras e futura regulação do bactericida Triclosan na legislação ambiental nacional e internacional. / Triclosan is a low solubility organic compound that has been used in toothpastes, face cream, shampoos, soaps, food packages, and a variety of other materials such as stickers, toys, shoes, paints, clothes, tiles, awnings and grout. The reason for its intense use as biocide is its great efficacy against Gram-negative and Grampositive bacteria. Evidences of Triclosan presence in urban and industrial effluents, superficial waters and sediments from freshwater, estuarine, and marine environments, as well as aquatic organisms (algae, fishes, mammals) have been reported in the literature. In this context, the present study assessed the acute and chronic toxicity of Triclosan to different tropical marine invertebrates. Acute toxicity bioassays using the copepod Nitokra sp (mortality) and the sea-urchin Lytechinus variegatus (fertilization rate) were performed. Short-term chronic toxicity bioassays with Lytechinus variegatus and the bivalve mussel Perna perna were carried out in order to assess Triclosan chronic effects. Besides, the Neutral Red Retention Time assay was employed to evaluate the effect of Triclosan on the stability of lysosomal membrane of hemocytes of Perna perna. In the acute toxicity assays, the mean value of LC(I)50;96h obtained for the copepod was 0.20 mg L-1, whereas the mean value of IC(I)50;1h for the sea-urchin was 0.28 mg L-1. In the chronic toxicity assays, the mean value of IC(I)50;24h recorded for the seaurchin was 0.14 mg L-1, whilst for the bivalve mollusk the mean value of IC(I)50;48h was 0.13 mg L-1. The effect on the lysosomal membrane stability of Perna perna hemocytes started to occur from 12 ng L-1. The results evidence the ecological risk associated to the continuous introduction of Triclosan into marine aquatic environments and must be considered in the identification of safety concentrations and future regulation of this bactericide compound in national and international environmental legislation.

Metabólitos secundários bioativos de invertebrados marinhos: isolamento, determinação estrutural e atividades biológicas / Bioactive secondary metabolites from marine invertebrates: isolation, structure determination and biological activities

Ao longo dos últimos 20 anos os organismos marinhos se tornaram uma das fontes mais interessantes para o isolamento de produtos naturais biologicamente ativos. Apesar disso, os organismos marinhos da costa brasileira foram muito pouco explorados como fonte de metabólitos secundários. A presente investigação se insere no âmbito de dois projetos temáticos, o primeiro desenvolvido entre 2002 e 2006 e o segundo em andamento, que têm por objetivo melhor conhecer as potencialidades de organismos marinhos da costa brasileira como fonte de metabólitos secundários biologicamente ativos. Assim, foram investigados extratos bioativos de cinco espécies de invertebrados marinhos: três ascídias, uma esponja e um octocoral, todos oriundos da costa brasileira. O fracionamento dos extratos destes animais resultou no isolamento de 15 compostos, dos quais sete inéditos na literatura. A partir do extrato bruto da ascídia Clavelina oblonga foram isolados dois compostos: a 5-[3,5-dibromo-4([2-oxo-5-oxazonidinil)]metoxifenil]2-oxazolidinona e o (2S,3R)- 2-amino-3-dodecanol. Este último apresentou uma potente atividade antifúngica contra Candida albicans. Do extrato da ascídia Didemnum ligulum foram obtidos dois compostos: a asterubina e a N,N-dimetil-O-metiletanolamina. A investigação química do extrato da ascídia Didemnum sp. resultou no isolamento de quatro dicetopiperazinas modificadas denominadas rodriguesinas A e B, e N-acetil-rodriguesinas A e B. As rodriguesinas A e B foram obtidas na forma de mistura inseparável, e puderam ser identificadas por análises espectroscópicas detalhadas, inclusive por experimentos MS/MS. A mistura contendo as rodriguesinas A e B apresentou moderada atividade antibiótica contra um isolado clínico de Streptococcus mutans, contra S. mutans UA159 e Staphylococcus aureus ATCC6538. A partir do extrato bruto da esponja Plakortis angulospiculatus foram obtidos seis policetídeos: a plakortenona, a plakortina, o plakortídeo P, o 3,6 epoxi, 4,6,8 trietil-2,4,9-dodecatrienoato de metila, a espongosoritina A e o 3,6 epoxi, 4,6,8 trietil-2,4,-dodecadienoato de metila. Os compostos puros obtidos da esponja P.angulospiculatus foram avaliados em testes de atividade antiparasitária contra Leishmania chagasi e Trypanosoma cruzi, antineuroinflamatória e citotóxica frente a quatro diferentes linhagens de células tumorais. O composto plakortídeo P apresentou potente atividade leishmanicida altamente seletiva. Finalmente, a investigação do extrato bruto do octocoral Carijoa riisei resultou no isolamento de um único constituinte, o esteróide 18-acetoxipregna-1,4,20-trien-3-ona, na qual apresentou atividades tripanomicida e leishmanicida. / Throughout the last 20 years, marine organisms have become one of the most interesting sources for the isolation of biological active natural products. However, marine organisms from the Brazilian coastline have been largely underexplored as source of secondary metabolites. The present investigation was developed in the scope of two thematic projects, the first one during 2002 and 2006 the second one, still in progress, aims the discovery of the potentialities of marine organisms of the Brazilian coast as a source of biologically active secondary metabolites. Bioactive extracts of five species of marine invertebrates have been investigated: three ascidians, one sponge and one octocoral. The fractionation of these extracts led to the isolation of 15 compounds, of which seven are unprecedented in the literature. From the crude extract of the ascidian Clavelina oblonga two compounds have been isolated: [3,5-dibromo-4-[(2-oxo-5-oxazolidinyl)]methoxyphenyl]-2- oxazolidinone and (2S,3R)-2-aminododecan-3-ol. The last one displayed potent antifungal activity against Candida albicans. Two compounds have been obtained from the crude extract of the ascidian Didemnum ligulum: asterubine and N,N-dimethyl-O-methylethanolamine. The chemical investigation of the extract of the ascidian Didemnum sp. resulted in the isolation of four modified diketopiperazines: rodriguesines A and B, was well as N-acetil-rodriguesines A and B. Rodriguesines A and B have been obtained as an unseparable mixture, and have been identified by detailed analysis of spectroscopic data including MS/MS experiments. The mixture of the rodriguesines A and B displayed moderate antibiotic activity against a clinical isolate of Streptococcus mutans, against S. aureus mutans UA159 and Staphylococcus aureus ATCC6538. The crude extract of the sponge Plakortis angulospiculatus have been investigated and six polyketides have been isolated: plakortenone, plakortin, plakortide P, methyl 3,6-epoxy-4,6,8-triethyl-2,4,9-dodecatrienoate, spongosoritin A and methyl 3,6-epoxy-4,6,8-triethyldodeca-2,4-dienoate. The polyketides isolated from the sponge P.angulospiculatus have been evaluated in tests of antiparasitic activity against Leishmania chagasi and Trypanosoma cruzi, antineuroinflammatory and cytotoxic against four different human cancer cell lines. Plakortide P exhibited potent and highly selective leishmanicidal activity. Finally, the chemical investigation of the crude extract from the octocoral Carijoa riisei resulted in the isolation of a single constituent, the known steroid 18- acetoxypregna-1,4,20-trien-3-one, which displayed cytotoxic, antitrypanosomal and antileishmanial activity.

O tecido adiposo, através da leptina, desempenha um papel permissivo sobre a maturação sexual do indivíduo. Apesar da importância do tecido adiposo para a puberdade, pouco se sabe a respeito do processo de formação deste tecido neste período da vida. Estudos em humanos têm descrito a presença de resistência à insulina puberal. Assim, frente à importância do tecido adiposo para a puberdade, o objetivo do trabalho foi investigar a resposta do tecido adiposo à insulina no período puberal. Foram utilizados os coxins adiposos subcutâneo e periepididimal. Na puberdade, paralelamente à intolerância à glicose transitória que ocorreu nas semanas iniciais, e, portanto, prejuízo da utilização da glicose pelos tecidos insulino-dependentes, o tecido adiposo (SC e PE) teve sua responsividade normal à insulina e uma melhor capacidade de incorporação de glicose em lipídeos o que leva a crer que há um desvio da utilização deste substrato energético, a glicose, para a formação do tecido adiposo. / The adipose tissue is critical to puberty. Leptin exerts a permissive role to hypothalamic-hypophysial-gonadal maturation and, thereby, adipose tissue is necessary to pubertal development. Despite its importance to puberty, little is known about the process of adipose tissue formation during this period of life. Researches have described insulin resistance at pubertal period in humans. The adipose tissue importance to puberty added to pubertal insulin resistance described in humans lead us to investigate how the adipose tissue responds to insulin at this period since it is essential at this time. We assessed two distinct fat pads: the subcutaneous fat pad and the epididymal one. At the puberty, despite the temporary glucose intolerance and the constraint for glucose utilization by insulin-dependent tissues, the adipose tissue had normal insulin responsiveness plus an improved capacity to synthesize lipids from glucose which led us to hypothesize that glucose could have been deviated towards the adipose tissue.

Biologically active natural products from South African marine invertebrates

This thesis describes the chemical and biological investigation of the extracts of six different marine invertebrate organisms collected along the South African coastline. The work on these extracts has resulted in the isolation and structural elucidation of twenty-one previously undescribed secondary metabolites; The history of marine natural product chemistry in South Africa has not previously been reviewed and so a comprehensive review covering the literature from the 1940's up until the end of 1995 is presented here. The marine ascidian Pseudodistoma species collected in the Tsitsikamma Marine Reserve was shown to contain four new unsaturated amino alcohols [47], [48], [49] and [50] which were isolated as their acetyl derivatives. These compounds exhibited strong antimicrobial activity. Four new pyrroloiminoquinone alkaloids, the tsitsikammamines A [90] to D [93],were isolated from a new genus of Latrunculid sponge collected in the Tsitsikamma Marine Reserve. These highly pigmented compounds also possessed strong antimicrobial activity. An investigation of two phenotypic colour variants of the soft coral Capnella thyrsoidea resulted in the isolation of the known steroid 5α-pregna-1, 20-dien-3-one [97] and an additional six new metabolites, 16β-hydroxy-5α-pregna-1 ,20-dien-3-one 16-acetate [98], 3α,16β-dihydroxy-5α-pregna-1, 20-diene 3,16-diacetate [99] and four xenicane diterpenes, the tsitsixenicins A [100] to D [103]. This is the first reported isolation of xenicane diterpenes from the soft coral family Nephtheiidae. Tsitsixenicin A and B showed good anti-inflammatory activity by inhibiting superoxide production in both rabbit and human cell neutrophils. A further four new metabolites were isolated from two soft corals which could only be identified to the genus level and were designated Alcyonium species A and species B. Alcyonium species A was collected in the Tsitsikamma Marine Reserve and yielded two new polyhydroxysterols, cholest-5-ene-3β, 7β, 19-triol 19-acetate [121] and cholest-5,24-diene-3β, 7β, 19-triol 19-acetate [122]. The soft coral Alcyonium species B was collected off Aliwal Shoal and was found to contain two known xenicane diterpenes, 9-deacetoxy-14, 15-deepoxyxeniculin [110] and zahavin A [16], and two new xenicane diterpenes, 7 -epoxyzahavin A [123] and xeniolide C [124]. Compounds [110], [16] and [123] exhibited strong anti-inflammatory activity and compounds [110] and [16] showed good antithrombotic activity. The endemic soft coral A/cyanium fauri collected at Riet Point near Port Alfred yielded the new sesquiterpene hydroquinone rietone [141] in high yierd, fogether with the minor compounds 8'-acetoxyrietone [142] and 8'-desoxyrietone [143]. Rietone exhibited moderate activity in the NCl's in-vitro anti-HIV bioassays.

Structural and synthetic investigations of South African marine natural products

A chemical investigation of six different marine invertebrates, collected along the South African coastline, resulted in the isolation and structural elucidation of fifteen previously undescribed secondary metabolites along with seven known compounds. The structures of the new metabolites were determined by a combination of spectroscopic and chemical methods. The endemic false limpet Siphonaria capensis was shown to contain two unusual polypropionate metabolites capensinone (162) and capensifuranone (163) as well as 2,4,6,8-tetramethyl-2-undecenoic acid (164) and the known polypropionates (E)- and (Z)siphonarienfuranone (149 and 161). Capensinone is the first example of a marine polypropionate containing a cyc1opentenone moiety. An investigation of the endemic South African soft coral Pieterfaurea unilobata yielded six new, highly oxygenated, pregnadiene sterols (180-185) and the known metabolite (169). Compounds 180-185 are the first pregnadienes obtained from the marine environment containing a C-7 substituent. An alternative procedure for the quick assignment of the absolute configuration at C-3 in this series of compounds was proposed. A companson of the pyrroloiminoquinone alkaloids of three undescribed l'}trunculid sponges resulted in the isolation of 3-dih¥drodiscorhabdin C (243), 3-dihydrodiscorhabdin B (244), discorhabdin H (197) and the previously reported alkaloids discorhabdin A (189) and discorhabdin D (192). While all three sponges were found to be morphologically different they all contained discorhabdin A as the major metabolite and discorhabdin H as one of their minor metabolites. It was found that a feature common to most of the South African latrunculid sponges is the reduction of the C-3 carbonyl gr,o up in some of the minor metabolites. The indole alkaloids, dilemmaones A-C (261-263), containing an unusual cyc1opentanone-indole skeleton, were isolated in trace amounts by bioassay guided fractionation of an extract obtained from a mixed collection of sponges collected near Cape Town. In an attempt to acquire more of these novel compounds for further investigation of their biological activity, several synthetic strategies towards their total synthesis were explored. A key feature of these approaches was the exploitation of the regioselective Gassman's artha-alkylation procedure for the introduction of an aromatic methyl substituent.

Studies in South African marine molluscan chemistry

This thesis investigates the variability occurring in the secondary metabolites produced by three South African marine molluscs. Chapter Two discusses the isolation and spectroscopic structure elucidation of the metabolites isolated from two Siphonaria species. The re-investigation of Siphonaria capensis yielded siphonarienfuranone (2.2) as the only common polypropionate isolated from both the 1998 and 2009 collections of S. capensis from the same areas suggesting possible seasonal or genetic variation in polypropionate production. The sterol cholest-7-en-3,5,7- triol (2.33) was also isolated form the 2009 collection of S. capensis and this is the first time this compound has been isolated from a Siphonaria species. The second species, Siphonaria oculus is closely related to S. capensis and the investigation into the former’s secondary metaboliteproduction revealed 2.2 as a major metabolite suggesting an inter-species overlap in polypropionate production. Three new polypropionate metabolites, 2.35, 2.36 and 2.37 were also isolated from S. oculus. An unsuccessful attempt was made to establish the absolute configuration of 2.37 using the modified Mosher’s method and the limited amount of 2.37 available prevented any further attempts at resolving the absolute configuration of this compound. The 1H NMR analysis of the defensive mucus collected directly from S. oculus revealed the presence of the acyclic polypropionate 2.37 as a minor metabolite. The absence of characteristic signals for the furanone containing compounds 2.2, 2.35 and 2.36, might suggest that these compounds cyclise from a hypothetical acyclic precursor (2.38) during standard work up of bulk acetone extracts of Siphonaria species. Chapter Three discusses the re-isolation and spectroscopic structure elucidation of the metabolites isolated from the nudibranch, Leminda millecra. Three known natural products, millecrone A (3.1), 8-hydroxycalamenene (3.6) and cubebenone (3.8) were re-isolated from our 2010 collection of L. millecra, as well as the new minor metabolite 8-acetoxycalamenene (3.16). The cytotoxic prenylated toluquinones and toluhydroquinones (3.9-3.15) initially isolated from the 1998 collection of L. millecra were not found in the 2010 collection supporting the hypothesis that these compounds may be of fungal origin. L. millecra clearly shows variability in the compounds sequestered by this species with millecrone A (3.1) being the only common metabolite in the three investigations of L. millecra to date. An unsuccessful attempt was made to establish the absolute configuration of 3.1, 3.6 and 3.8 through initial LAH reduction of the ketone moiety contained in 3.1 and 3.8 and esterification of the resultant diastereomeric alcohol mixtures and the phenol functionality in 3.6 with (1S)-camphanic chloride. Crystallisation of the (S)- camphanate esters of 3.6 and 3.8 for X-ray analysis were unsuccessful, while the unexpected conjugate addition of a hydride in 3.1 resulted in complex diastereomeric mixtures which could not be separated by HPLC.

INTERACTIVE EFFECTS OF PREDATION AND ASSEMBLY TIME ON TROPICAL BUT NOT TEMPERATE MARINE INVASIONS

Non-native species richness has been observed to peak at mid-temperate latitudes, shaping a pattern of richness and abundance that is distinct from native species patterns that peak in the tropics. Stronger species interactions, and therefore biotic resistance, may lower invasion success in the tropics and help explain the discrepancy between native and non-native richness and abundance. To test the hypothesis that strong predation and competition in the tropics could limit invasion success, we conducted a distributed experiment on sessile marine invertebrate communities in four regions spanning 47-degrees latitude of the eastern Pacific Ocean. We manipulated predator access and resource availability at 12 sites and sampled experimental communities in early and late stages of assembly. Overall, our results suggest that biogeographic location, assembly timescale, and predation interactively shape invasion success across latitude. Strong predation reduced richness of non-native species in the tropics at both assembly timescales but increased non-native species richness in the subtropics during early assembly. Predation also increased non-native abundance in the tropics by late-stage assembly and shaped non-native composition at both assembly stages. Effects of predation at higher latitudes were weak or undetectable, and increasing resource availability never had a positive impact on non-native richness or abundance at any latitude. Further, non-native richness was greater at early relative to late assembly stages at mid to low latitudes and was consistently low in our high latitude region at both timescales. In a complementary experiment, short-term predator exposure reduced non-native abundance in Panama, further confirming the influence of predation in this tropical region. Our results highlight important biogeographic differences in invasion dynamics and disentangle local mechanisms that can shape regional patterns. / Biology

A micologia marinha é uma ciência relativamente recente e pouco se conhece sobre a diversidade das suas comunidades. Assim, o isolamento, triagem e preservação de fungos derivados do mar podem levar à descoberta de novas tecnologias. O objetivo deste estudo foi conhecer a diversidade de fungos filamentosos derivados marinhos e selecionar isolados capazes de produzir enzimas celulolíticas. Para tanto, foram isolados seletivamente fungos filamentosos a partir de amostras de macro-organismos marinhos coletados em 2007 e 2008. Os resultados demonstraram uma ampla diversidade de fungos potencialmente celulolíticos, pertencentes ao filo Basidiomycota e Ascomycota. Nos experimentos de produção de celulases, 17 apresentaram resultados satisfatórios de CMCase e FPase e foram selecionados para a avaliação da Celobiase. Os experimentos de cinética enzimática apresentaram os melhores resultados de produção de celulases em meio contendo farelo de trigo. O trabalho demonstra o potencial para aplicação biotecnológica dos fungos e estimula novos estudos com as celulases. / The Marine mycology is a relatively recent and little is known about the diversity of its communities. Thus, the isolation, separation and preservation of fungi derived from the sea can lead to the discovery of new technologies. The aim of this study was the diversity of filamentous fungi isolates derived marine and select capable of producing cellulolytic enzymes. It had been selectively isolated filamentous fungi from samples of marine macro-organisms collected in 2007 and 2008. The results showed a wide range of potential cellulolytic fungi, belonging to the phylum Basidiomycota and Ascomycota. In the experiments to produce cellulases, 17 had satisfactory results of CMCase and FPase and were selected for evaluation of cellobiase. The enzyme kinetics experiments showed better results for the production of cellulases in a medium containing wheat bran. The work demonstrates the potential for biotechnological application of fungi and stimulate further research with cellulases.
