Preparation and Characterization of Mg-Cu Binary Metallic Thin Film

In this study, Mg-Cu thin film metallic thin films were fabricated via two ways, the co-deposition and post-annealing of the multilayered thin films. Amorphous Mg1-xCux, where x is from 38 to 82, thin films with nanocrystalline particles are able to be fabricated via co-sputtering. The mechanism of formation is different from the rapid quenching process. For the Mg-Cu co-sputtering system, the mechanical properties of the Mg-Cu co-sputtered films were tested via MTS nanoindenter. Mg23.5Cu76.5 exhibits a higher Young¡¦s modulus than Mg17.7Cu82.3 and Mg40.4Cu59.6 due to the partial amorphous structure. Moreover, the pop-in effects with a smaller size occurs of the Mg23.5Cu76.5 sample in a higher frequency than of the Mg17.7Cu82.3 and Mg40.4Cu59.6 samples. The small pop-in effects in the Mg23.5Cu76.5 sample approximate match the width of amorphous matrix via the HRTEM observation. Another process to form the amorphous thin film is via the post isothermal annealing process of the multilayered thin films. However, for the specimens of 20T32 consisting of 150-nm Mg and 50-nm Cu individual layers, the Mg individual layers would react to the Cu individual layers during the annealing at a temperature of 413 K owning to the slight negative heat of mixing. Due to the localized diffusion near the interfaces, Mg2Cu gradually form during the isothermal annealing since Mg2Cu is the most stable phase below 548 K [62]. Localized interdiffusion near the interfaces between Mg and Cu individual layers induced the formation of Mg2Cu rapidly. For the 40T32 specimens consisting of 15 nm Mg and 5 nm Cu individual layers, Mg2Cu rapidly form at 413 K due to the high interface energy. Then, the similar result exhibits in the 20T14 and 40T14 specimens annealed at 363 K.

Έκφραση, απομόνωση και χαρακτηρισμός των εξωκυτταρικών περιοχών των β και δ υπομονάδων του υποδοχέα της ακετυλοχολίνης

Ο νικοτινικός υποδοχέας της ακετυλοχολίνης, επιτελεί καθοριστικό ρόλο στη σωστή λειτουργία της νευρομυϊκής σύναψης. Η δυσλειτουργία του υποδοχέα είναι υπεύθυνη για την εκδήλωση της βαριάς μυασθένειας, η οποία αποτελεί αυτοάνοσο νόσημα. Η αποκάλυψη της τρισδιάστατης δομής του υποδοχέα αποτελεί βασική προϋπόθεση στην κατανόηση της λειτουργίας και του ρόλου του στη δημιουργία της βαριάς μυασθένειας. Καθώς όμως, ο υποδοχέας είναι ένα σχετικά μεγάλο μόριο, με αρκετές υδρόφοβες περιοχές, η λύση της τρισδιάστατης δομής του καθίσταται δύσκολη. Τα μόνα δεδομένα που διαθέτουμε προέρχονται από μελέτες του AChR του ελασματοβράγχιου Torpedo californica, ο οποίος παρουσιάζει ικανοποιητική συγγένεια με τον υποδοχέα της ακετυλοχολίνης, και από μελέτες κρυσταλλογραφίας μια πρωτεΐνης του σαλιγκαριού Lymnaea stragnalis, ικανής να δεσμεύει ακετυλοχολίνη (AChBP), η οποία όμως παρουσιάζει μικρή ομοιότητα με τον ανθρώπινο μυϊκό υποδοχέα. Για την αντιμετώπιση των παραπάνω δυσκολιών, εκφράστηκαν τα εξωκυτταρικά τμήματα (extracellular domain: ECD) των β και δ υπομονάδων του ανθρώπινου νικοτινικού υποδοχέα της ακετυλοχολίνης, στο ετερόλογο σύστημα έκφρασης της Pichia pastoris. Οι δύο υπομονάδες κλωνοποιήθηκαν σε κατάλληλους φορείς έκφρασης, pPIC9 και pPICZaA. Τα ανασυνδυασμένα πολυπεπτίδια που απομονώθηκαν, χρησιμοποιήθηκαν για λειτουργικές μελέτες στα πλαίσια θεραπευτικών προσεγγίσεων και την ολοκλήρωση των μελετών σχετικά με την εξωκυτταρική περιοχή του AChR. Από τα πειραματικά δεδομένα που προέκυψαν, διαπιστώθηκε ότι η εξωκυτταρική περιοχή της β υπομονάδας, εκφράζεται σε διμερή μορφή και σε μεγαλύτερες ποσότητες συγκρινόμενη με τα ECDs των υπολοίπων υπομονάδων που εκφράστηκαν στο ίδιο σύστημα. Το πολυπεπτίδιο αυτό, διαθέτει παράλληλα και την ικανότητα να απομονώνει αντισώματα από ορούς μυασθενικών, από ορισμένους εκ των οποίων τα ποσοστά έφταναν και το 98%. Η διαδικασία απομόνωσης αντισωμάτων βασίζεται στην ακινητοποίηση των ανασυνδυασμένων πολυπεπτιδίων σε στήλη σεφαρόζης και χρήση της για αφαίρεση των αυτοαντισωμάτων έναντι της αντίστοιχης υπομονάδας. Στην περίπτωση της δ υπομονάδας τα αποτελέσματα που προέκυψαν από αντίστοιχες μελέτες δεν ήταν τα αναμενόμενα. Συγκεκριμένα, αν και η πρωτεΐνη απομονώνεται σε διαλυτή μορφή, δημιουργεί συσσωματώματα, τα οποία δεν επιτρέπουν τη χρήση της για δομικές μελέτες. Παράλληλα, οι ποσότητες που προκύπτουν είναι μικρές και δεν διευκολύνουν οποιαδήποτε χρήση της πρωτεΐνης. Για την αντιμετώπιση αυτού του προβλήματος, αντικαταστάθηκε η θηλιά του εξωκυτταρικού τμήματος της πρωτεΐνης με αυτή της AChBP, η οποία όπως έχει δειχθεί και στο παρελθόν σε άλλες υπομονάδες, βελτίωνε την ποιότητα και την ποσότητα της εκφραζόμενης πρωτεΐνης. Με αυτόν τον τρόπο η διαλυτότητα, καθώς και η ποσότητα, της απομονωμένης πρωτεΐνης αυξήθηκαν. Τα πειράματα ανοσοπροσροφήσεων όμως στην περίπτωση της δ υπομονάδας, δεν επέφεραν κάποιο σημαντικό αποτέλεσμα συγκρινόμενο με τα αντίστοιχα αποτελέσματα ανοσοπροσροφήσεων με τα ECDs των υπολοίπων υπομονάδων του μυϊκού AChR, τουλάχιστον για τους ορούς που ελέγχθηκαν. Εντοπίστηκε όμως ένας ορός, ο οποίος δεν αποκλύει την πιθανότητα εύρεσης κι άλλων θετικών αντι-δ ορών. Από όλα τα παραπάνω δεδομένα, προκύπτει ότι η εξωκυτταρική περιοχή της β υπομονάδας μπορεί να χρησιμοποιηθεί στα πλαίσια θεραπευτικών προσεγγίσεων και πιθανότατα και σε δομικές μελέτες, σε αντίθεση με τη δ, η οποία στη συγκεκριμένη μορφή δεν παρουσιάζεται κατάλληλη για δομικές μελέτες, καθώς οι παραγόμενες ποσότητες αποτρέπουν κάθε δυνατή προσπάθεια, δίχως όμως να μπορεί να αποκλειστεί η συμβολή της και σε μελλοντικά θεραπευτικά συστήματα. / -

The Sintering Behaviour of Al-Mg-Si-Cu-(Sn) Powder Metallurgy Alloys

The current, commercially available, press and sinter Al-Mg-Si-Cu alloys are based on wrought or cast alloy compositions and have not been tailored for the press and sinter process. The limited development of the Al-Mg-Si-Cu alloys for the press and sinter process can be partly attributed to a poor understanding of the effects of processing conditions on the sintering behaviour. The primary objective of this work was to investigate and understand the effects of processing conditions on the sintering behaviour of Al-Mg-Si-Cu-(Sn) alloys. Dilatometry was used in conjunction with other experimental techniques to elucidate and understand the expansion and shrinkage events that occur during the liquid-phase sintering of Al-Mg-Si-Cu-(Sn) powder metallurgy alloys. Samples were uni-axially pressed from elemental metal powder blends, de-waxed, and then sintered in a horizontal push-rod dilatometer to record the dimensional changes in the pressing direction. The processing conditions examined included the alloy composition, temperature, green density and atmosphere. A liquid forms during heating due to reactions between the alloying elements and the aluminium. This liquid is initially non-wetting on the oxide layer of the aluminium particles, resulting in separation of the particles, which is manifested by expansion of the sample. The oxide is reduced as sintering progresses, alleviating the non-wetting conditions. When more liquid forms, further expansion occurs, despite the improved wetting conditions. It is proposed that atmospheric oxygen and/or nitrogen can react with the liquid, forming a solid phase (‘shell’) at the liquid-vapour interfaces. These shells prevent the liquid from wetting the particles, resulting in further expansion and preventing shrinkage. Unbalanced diffusivities (the Kirkendall effect) between the aluminium and silicon contribute to the expansion. A mechanism is proposed to account for the transition to shrinkage, whereby the shells at the liquid-vapour interface rupture when there is a rapid increase in the volume of contained liquid. The liquid then flows out and over the shells, onto the aluminium substrate, causing shrinkage. Magnesium and nitrogen delay the transition to shrinkage by facilitating nitride shell formation at the solid-liquid interface. Silicon and tin cause an earlier transition to shrinkage by increasing the liquid volume. In addition, tin promotes shrinkage by segregating to the liquid-vapour interfaces, limiting the thickness of the shells at the liquid-vapour interfaces. The two dominant liquid-phase shrinkage mechanisms during the sintering of Al-Mg-Si-Cu-(Sn) alloys are rearrangement and pore-filling. Contact-flattening is not a dominant shrinkage mechanism, but may occur concurrently with the other mechanisms. If contact flattening occurs, a decrease in the pressure of isolated pores increases the total shrinkage rate. Nitrogen increases the shrinkage rate during rearrangement by restricting grain-growth. Magnesium increases the shrinkage rate during rearrangement by reducing the solid-liquid interface energy. Magnesium and nitrogen are essential for the formation of nitride within isolated pores, which decreases the pore pressure and increases the contribution of contact-flattening on the total shrinkage rate. Silicon reduces the beneficial influence of magnesium during rearrangement by diluting the magnesium content in the liquid. Silicon increases the pore-filling rate due to an increase in the liquid volume. Magnesium increases the pore-filling rate by facilitating aluminium nitride formation within isolated pores and by increasing the pore-filling. Tin additions can decrease the pore-filling rate due to its segregation to the liquid-vapour interface, limiting the consumption of nitrogen within isolated pores.

Nesse sentido, procuramos historicizar, a partir dessas regiões, os processos de instituição do imaginário social do trabalho, enfocando particularmente as representações e práticas que presidiram, naquele momento, a invenção do trabalhador nacional. Com efeito, por meio da pesquisa empreendida interrogamos em múltiplos discursos - jornalísticos, médicos, políticos, religiosos, historiográficos, memorialistas, entre outros, o investimento feito na construção e projeção de sentidos positivos às imagens do trabalho e do trabalhador. Percebemos nesse projeto discursivo uma das condições indispensáveis para a instituição de uma sociedade ordeira, progressista e civilizada. Nessas condições, as formas de não-trabalho – ociosidade, vadiagem – foram condenadas por serem consideradas práticas que possibilitavam a homens e mulheres se desviarem de sua missão delineada como primordial: trabalhar. O conjunto da sociedade será impelido, não sem resistência, a agir material e simbolicamente com vistas à conformação de uma sociedade civilizada. _________________________________________________________________________________________ ABSTRACT / In the transition of slave labor to free work in Brazil, people from Triângulo Mineiro and Alto Paranaíba region, situated in the western Minas Gerais state, experimented in a peculiar way the changes which were propelled by Modernization Projects and Social Reform. Both characterized the Brazilian Society in the end of 19th century and the beginning of 20th century. In this direction, we tried to document through the history to start from these regions, the processes of establishment of social imaginary about work, focalizing particularly the representations and practices which led, in that moment, the invention of National Worker. In fact through the undertaken research we questioned the multiple speeches - journalistic, medical, religious, historical representation, and memories, among others, the investment done in the construction and projection of positive directions to the images of the work and of the worker. We realized, in this discursive project one of the essential condition for the institution of an orderly, progressive and civilized society. In this way, the forms of non work – idleness, vagrancy – were condemned because of being considered practices which allow men and women to deviate themselves from their traced mission like primordial: to work. The whole of society will be forced, not without resistance, on act material and symbolically for conformation of civilized society.

Leaving from there, I guided the works in the survey of information that can serve as parameters for the elaboration of strategies of action in the most diverse directed fronts of work to development of the tourism and the culture in that city. / O foco dessa dissertação é direcionado ao levantamento diagnóstico do Parque das Águas da cidade de São Lourenço - MG, mais especificamente à importância das águas minerais como bem cultural. Pesquisei em duas frentes: dados colhidos junto a secretaria de turismo (SERVTUR) e duas pesquisas quantitativas. Para aprofundar a reflexão optei também pela realização entrevistas qualitativas com pessoas de diferentes perfis de renda, cultura e profissões. Partindo daí, norteei os trabalhos no levantamento de informações que possam servir como parâmetros para a elaboração de estratégias de ação nas mais diversas frentes de trabalho voltadas ao desenvolvimento do turismo e da cultura naquela cidade.

Em cada propriedade foram abertas duas trincheiras no bananal, ao longo da linha irrigada, a 100 cm dos microaspersores, e outras duas em área adjacente sob sequeiro, freqüentemente sem utilização agrícola, nas quais foram coletadas amostras a 0-5, 5-10, 10-15, 15-20, 20-30, 30-40, 40-60 e 60-100 cm de profundidade. Procedeu-se à caracterização química das águas de irrigação, assim como das características mineralógicas e químicas do extrato de pasta saturada, e físicas do solo, além de material vegetal. As comparações do efeito de cada fonte de água foram feitas pelo teste t em cada camada amostrada contra suas respectivas testemunhas sob sequeiro. Os resultados das análises efetuadas permitiram demonstrar a hipótese de trabalho, segundo a qual o uso de águas de irrigação de qualidade marginal induz mudanças deletérias nas características químicas e físicas dos solos da região. As águas de poços e rios se diferenciaram quimicamente, tendo as águas de poços apresentado maiores viiivalores médios de condutividade elétrica e RAS, assim como maiores teores de Ca 2+ , Mg 2+ , Na + , HCO 3- , Cl - , Si o , Mn o e Zn o . As águas de poços foram consideradas de médio risco de salinidade e baixo risco de sodicidade, enquanto as águas de rio foram classificadas como de baixo risco de salinidade e risco severo de sodicidade. As águas de poços e as práticas culturais associadas provocaram alterações equivalentes a uma calagem em doses elevadas, causando elevação do pH e dos teores de Ca 2+ e Na + dos solos, sendo mais evidentes nas camadas mais superficiais. As relações Ca/Mg e Ca/K aumentaram significativamente, distanciando-se da faixa ideal para a cultura da banana. Nos solos irrigados com água do Rio Gorutuba houve aumento dos teores de P-H 2 SO 4 , resultante da precipitação do P com o cálcio proveniente da calagem. Nos solos irrigados com água de poços foram registrados aumentos ainda maiores dessa fração. O grau de floculação das argilas decresceu nos solos irrigados com água de poços, indicando dispersão promovida pela elevação do pH e por Na + , causando elevação na densidade global e diminuição na porosidade total, o que reflete em menores valores de macroporosidade e condutividade hidráulica, devendo ser ressaltado que o grau de floculação e o pH foram bons indicadores de possíveis problemas com a condutividade hidráulica. Entre os macronutrientes somente o cálcio e o magnésio foram encontrados em níveis adequados nas plantas, nas duas fontes de água de irrigação. Apesar da menor disponibilidade de P em função da precipitação como P-Ca e P-CaCO 3 , não houve diferenciação entre os teores nos tecidos vegetais dos bananais irrigados com diferentes fontes de água. A lixiviação de K + promovida pelo uso de águas de poços levou a menores teores desse elemento nas plantas, quando comparados àquelas irrigadas com água do Rio Gorutuba. Em ambos os casos, porém, os níveis foram considerados baixos. Nos solos irrigados com águas provenientes de poços tubulares os teores de Zn, Fe, Cu e B nas plantas foram baixos, o que não aconteceu para Cu e B quando a fonte de água foi o Rio Gorutuba. / This work aimed to study the effect of irrigation water from wells and Gorutuba River on soil of 24 properties, cultivated with prata-anã banana in the region of Janaúba in northern Minas Gerais. Two ditches were opened in the banana plantation in each property, along the irrigated line, at 100 cm from the micro-sprink lers, and two in an adjacent area under frequently fallow, dry land in which samples were collected at 5, 5-10, 10-15, 15-20, 20-30,30-40,40-60 and 60-100 cm depth. Chemical characterization of the irrigation water proceeded as well as the mineralogical and chemical characteristics of the saturated paste extract and physical characteristics of the soil, besides characterization of the plant material. The effect of each source of water was compared by the t test in each sampled layer to their respective controls under dry land. The analysis results allowed showing the working hypothesis, according to which the use of irrigation water of marginal quality leads to deleterious changes in the chemical and physical characteristics of the soils in the region. The well and river waters differed chemically, with the former presenting higher average values of electric conductivity and RAS, as well as higher contents of Ca 2+ , Mg 2+ , Na + , HCO 3- , CI - , xS o , Mn o , and Zn o .The well waters were considered of medium salinity risk and low sodicity risk, while the river waters were classified as of low salinity risk and severe sodicity risk. The well waters and associated culture practices caused changes equivalent to liming at high doses, increasing the pH and the Ca 2+ and Na + contents of the soils, being more evident in the more superficial layers. The Ca/Mg and Ca/K relations increased significantly, distancing from the ideal banana culture. Even higher increases of this fraction were registered in the well water irrigated soils. The degree of clay flocculation decreased in the well water-irrigated soils, showing dispersion promoted by pH and Na + increase, increasing global density and decreasing total porosity, which was reflected in the lower macro-porosity and hydraulic conducti vity levels. It may be emphasized that the flocculation degree and pH were good indicators of possible hydraulic conductivity problems. Among the macro-nutrients, only calcium and magnesium were found in adequate levels in the plants, in both sources of irrigation water. Despite the lower availability of P in function of precipitation as P-Ca and P-CaCO 3 , there was no differentiation between the plant tissue contents of banana plantations irrigated with different sources of water. K + leaching promoted by the well waters led to lower contents of this element in the plants than in plants irrigated with river water. In both cases, however, the levels were considered low. Zn, Fe, Cu, and B contents in soils irrigated with well waters were low, in contrast with Cu and B contents when the source of water was that of the Gorutuba River.

In this study we have considered the educational process as the imposure upon students of the standards of the dominant class in society through the Symbolic Violenc which marks pedagogic action, and the school as the formal and institutional milieu where relationships of domination and imposure of cultural standards of these groups or classes are established in a legitimate way through theachers, who are also legimate agents of this process. All this study has been distinguished by and anthropological approach, guined by participating observation of the group and area chosen as object of study. It has been verified that school imposes on students through its practices aspirations typical of the dominant class in society. These aspiration were expressed in students representation. It has also been observed that school, by its agents, blames students failure solely on their cultural environment (first socialization) which doesn’t reinforce the concepts learned at school. Moreover it has been pointed out that after years in the career teachers also tend to assimilate the culture of the environrnent in which they work, creating in the teaching process the possibility to the occurrence of what we call a transgression of the Symbolic Violence code. As this report is a case study, it would be convenient that the same theme be researched under other aspects , mainly if it is intendend to test the degree of generality of the characteristics evidenced in this particular study. / O presente trabalho teve por objetivos: i) examinar por meio de que mecanismos se dá a inculcação da cultura secundária na escola primária; ii) verificar os efeitos desta inculcação nas representações da clientela que frequenta a escola; iii) verificar até que ponto a escola está cumprindo sua função de reprodução das classes culturais dominates da sociedade. Neste estudo, consideramos o processo educacional como a inculcação, nos alunos, dos valores da classe dominante da sociedade por meio do exercício da Violência Simbólica, que caracteriza a ação pedagógica, e a escola como o ambiente formal e institucional onde se processam, de modo legítimo, as relações de dominação e de inculcação dos padrões culturais desses grupos ou classes, por intemédio das professoras, agentes também legítimos desse processo de inculcação do arbitrário cultural. Todo o estudo foi caracterizado por um enfoque antropológico, guiado pela observação participativa junto ao grupo e área selecionados para estudo. Verificou-se que a escola, através de suas práticas, inculca nos alunos aspirações que são típicas da classe dominante da sociedade. Estas aspirações foram manifestadas em suas representações. Observou-se, ainda, que a escola, por intermédio de seus agentes, atribui o fracasso escolar dos alunos unicamente a seu ambiente cultural (primeira socialização), que não reforça os conteúdos que a escola ensina. Além disso, observou-se que, após muitos anos de magistério, a tendência das professoras é também se aculturarem ao meio onde trabalham, gerando no processo de ensino a possibilidade da ocorrência daquilo que chamamos de transgressão do código de violência simbólica. Como este trabalho trata-se de um estudo de caso, seria conveniente que o mesmo tema fosse pesquisado em outras configurações, principalmente se se tiver como objetivo testar o grau de generalidade das características constatadas a partir deste estudo particular.
