El presente estudio tuvo por objetivo determinar el momento de infección con Mycoplasma hyopneumoniae en 240 porcinos provenientes de madres inmunizadas y no inmunizadas, en una granja tecnificada de un solo sitio, donde se maneja un sistema de Todo dentro Todo fuera, ubicada en el Valle del río Chillón, positiva a este agente. El estudio se realizó en los meses de otoño e invierno. Un grupo de marranas fue inmunizado con 2 ml de una bacterina comercial contra M. hyopneumoniae a los 85 días de gestación, mientras que otro grupo permaneció sin vacunación. Los lechones obtenidos de estas marranas fueron divididos en dos grupos de 120 porcinos, según los antecedentes inmunitarios de las madres. Se obtuvieron muestras de suero de los porcinos de ambos grupos en diferentes momentos de proceso productivo para la medición de anticuerpos mediante la prueba de ELISA indirecta. El 66.67%(74/112) y 85.59% (101/118) de animales provenientes de madres inmunizadas y no inmunizadas, respectivamente, fueron seroreactores durante el transcurso del estudio. No se encontró diferencia estadística significativa en el momento de seroconversión de ambos grupos. Los resultados obtenidos sugieren que la infección de ambos grupos se da en el mismo momento, alrededor de las 10 semanas de edad, lo cual coincide con factores estresantes tales como el paso de animales de la etapa de recría a la de acabado y su reagrupamiento. / The aim of this study was to determine the moment of infection with Mycoplasma hyopneoumoniae in 240 piglets of vaccinated and unvaccinated sows, in a positive one site swine farm with all-in/all-out (AIAO) system, located in Chillon valley. The study had been performed in autumn and winter. One group of sows was vaccinated with 2 ml of M. hyopneumoniae commercial bacterin, at 85 days of the gestation period, while the rest of sows remain unvaccinated. Two groups of 120 piglets were formed according to the sows’ status of immunity. Blood samples from the two groups of pigs in different moments of the production period were obtained. The antibodies with an indirect assay of commercial ELISA were measured. The 66.67% (74/112) and 85.59% (101/118) of the pigs from vaccinated and unvaccinated sows, respectively, were seroreactive throughout the course of the study. There were no significant differences in the moment of seroconversion between the two groups. The results suggested that the moment of infection in both groups had occurred at the same time, at nearly 10 weeks of age. This age coincided with stress factors, such as change of environment and the animals being crowded during the relocation from nursery to growth-finishing period.

Prevalencia de anticuerpos a Mycoplasma meleagridis en pavos reproductores en el departamento de Lima

Se determinó la prevalencia de anticuerpos a Mycoplasma meleagridis en lotes de pavos reproductores del Departamento de Lima durante el año 2007. Se tomaron 400 muestras de sangre al azar pertenecientes a 20 lotes de pavos reproductores mayores a 8 semanas, machos y hembras, provenientes de los distritos de Asia y Chilca. Las muestras fueron analizadas mediante el ensayo de inmunoabsorción ligado a la enzima (ELISA) a fin de determinar la presencia de anticuerpos a Mycoplasma meleagridis. Se encontraron 3 muestras positivas pertenecientes de 3 lotes diferentes, resultando una prevalencia de 0.75% ± 0.042. El porcentaje de seroreacción por lote afectado fue de 5%, muy por debajo del esperado en lotes positivos a M. meleagridis. Además, las aves pertenecientes a los lotes positivos no mostraron signos clínicos compatibles con la enfermedad, por lo que se podría interpretar que los 20 lotes examinados no fueron expuestos al microorganismo y que estos resultados se deben a reacciones falsas positivas. / --- A field study was conducted to determine the prevalence of antibodies against Mycoplasma meleagridis (MM) in turkey breeder flocks from Lima during 2007. A total of 400 blood samples were taken from 20 different turkey breeder flocks older than 8 weeks of age, males and females, in the districts of Asia and Chilca. The indirect enzyme linked immunosorbent assay (ELISA) was used to measure the serum antibodies against M. meleagridis A total of 3 positive samples were found in 3 different flocks, which represents a prevalence of 0.75% ± 0.042. The seroreactivity in each affected flock was 5%, far lower than expected in MM-infected flocks. Besides, birds from M. meleagridis positive flocks did not show clinical signs compatible to those observed in the disease. According to these findings, it’s possible to say that the 20 flocks were not exposed to M. meleagridis and that the positive results may be due to false positive seroreactions.

Se realizó un estudio serológico para determinar el momento de mayor infección con Mycoplasma hyopneumoniae en porcinos de crianza intensiva de la provincia de Lima, en el periodo abril - agosto del 2002. Se realizó un muestreo quincenal en 30 animales, desde el nacimiento hasta las 16 semanas de edad. Se utilizó la prueba comercial de ELISA CHEKITâ-Hyoptest-II, para la detección de anticuerpos. El 66.7% (20/30) de los animales presentaron anticuerpos contra M.hyopeumoniae, de los cuales el 55 % (11/20), seroconvirtió a las 12 semanas, en tanto que el 5 % (1/20), 30 % (6/20), 10 % (2/20), seroconvirtieron a las 10, 14 y 16 semanas de edad, respectivamente. Se concluye que existió un periodo critico entre las 9 y las 10 semanas de edad, en el cual M. hyopneumoniae inició la infección, que condujo a la seroconversión. Se demostró también, que no existe efecto del sexo y número de partos de la madre de los porcinos en el momento de infección. / A serological study was conducted to identify the age of highest infection with Mycoplasma hyopneumoniae, in swine under intensive rearing system in the Lima province, during the period April – august, 2002. Blood samples were collected fortnightly from 30 piglets from birth till 16 months of age. The commercial test ELISA CHEKITâ-Hyoptest-II, was used for the detection of antibodies. The 66.7 % (20/30) of the animals showed antibodies against M. hyopneumoniae, where 55 % (11/20) seroconverted at 12 weeks of age whereas 5 % (1/20), 30 % (6/20) and 10 % (2/20) seroconverted at 10, 14 and 16 weeks of age, respectively. It is concluded that exist a critical period between 9 and 10 weeks of age, in which M.hyopnuemoniae, began the infection, conducing to the seroconversion. Análise proteômica de proteínas de membrana de Mycoplasma hyopneumoniae e Mycoplasma synoviae

No. of bitstreams: 1 278419.pdf: 1162994 bytes, checksum: e114d7b57ee534fdab87f9214a2de45e (MD5) / As espécies Mycoplasma hyopneumoniae e Mycoplasma synoviae causam pneumonia em porcos e sinovite em frangos e perus, respectivamente, e em decorrência à perda de peso dos animais e susceptibilidade a infecções secundárias, essas bactérias têm causado prejuízos à suinocultura e avicultura catarinenses. Assim, o objetivo principal deste projeto foi submeter à análise proteômica amostras bacterianas ricas em proteínas de membrana, para identificação das mesmas por espectrometria de massa e construção de mapas proteômicos para as duas espécies. A identificação e caracterização de proteínas de membrana mostram-se úteis, já que essas proteínas estão envolvidas em funções celulares essenciais como transdução de sinais, osmoregulação, metabolismo e interação com hospedeiro. Os dados obtidos servirão de base para a identificação de possíveis alvos para fármacos, anticorpos monoclonais e também no auxílio em diagnósticos. Foram analisados no total, seis géis bidimensionais para cada espécie de Mycoplasma na faixa de pH 3-10, tendo duplicatas para os três detergentes utilizados: ASB14, CHAPS e SB3-10. Em relação à resolução em número de spots e número de identificações, o detergente ASB14, mostrou-se mais eficiente para M. hyopneumoniae e no total foram identificadas por espectrometria de massas 24 proteínas de membrana (10 proteínas distintas). Para M. synoviae o detergente mais eficiente foi CHAPS e foram identificadas por espectrometria de massas 36 proteínas de membrana (10 proteínas distintas). / Mycoplasma hyopneumoniae and Mycoplasma synoviae are causative agents of pneumonia in pigs and synovitis in chickens and turkeys, respectively. Due to the loss of animal weight and susceptibility to secondary infections, these bacteria have caused damage to pigs and poultry in Santa Catarina. Thus, the main objective of this project was to perform proteomic analysis of samples rich in bacterial membrane proteins, in order to identify them by mass spectrometry and to build proteomics maps for both species. The identification and characterization of membrane proteins is useful, since these proteins are involved in essential cellular functions such as signal transduction, osmoregulation, metabolism and interaction with host. The results obtained will help the identification of possible targets for drugs, monoclonal antibodies, and also in diagnosis. We analyzed a total of six two-dimensional gels for each species of mycoplasma in the pH range 3-10, with duplicates for the three detergents used: ASB14, CHAPS and SB3-10. Regarding gel resolution by the number of spots and the number of identifications, the detergent ASB14 was more efficient for M. hyopneumoniae and a total of 24 membrane proteins were identified by mass spectrometry (10 different proteins). For M. synoviae the detergent CHAPS was most effective and there were identified 36 membrane proteins by mass spectrometry (10 different proteins).
