Possible Avian Influences in the Distribution of Dwarf MistletoeZilka, Paul James 01 July 1973 (has links)
The goal of this investigation was to determine the potential of birds as vectors for the movement of dwarf mistletoe seeds and the means by which such movement could occur. Birds would greatly speed the rate at which these parasites move horizontally or vertically through the forest. Five areas in Oregon, heavily infected by six species of Arceuthobium were studied. Observations of avian behavior point to 13 species whose general behavior make them potential vectors in the distribution of these parasites. More specifically, the roosting and subsequent bathing behavior of most forest birds proves a very important means of potential seed transport. Additionally, an examination of nesting materials revealed that birds bring viable dwarf mistletoe seeds to infectible portions of host trees during the construction of their nests. Nesting, however, seems the least important of the vector mechanisms discussed. I observed no evidence of birds feeding on the seeds of dwarf mistletoe or of seeds being present in their droppings. The more probably means of seed transport is one of epizoochory.

Este estudo teve como objetivo investigar os componentes quantitativos e qualitativos na dispersão de sementes de Struthanthus flexicaulis (Loranthaceae), uma erva-de-passarinho dióica, hemiparasita generalista e de ampla distribuição no cerrado do Brasil central. O estudo foi realizado entre janeiro de 2004 e abril de 2005 em um afloramento rochoso na Serra do Cipó, Minas Gerais. Foram realizadas observações sobre a fenologia dos indivíduos femininos, observações focais para a avaliação dos parâmetros comportamentais dos dispersores, censos para avaliação da abundância relativa, sazonalidade e parâmetros comportamentais dos dispersores, observação direta e amostragem por procura ativa para avaliar a deposição das sementes de S. flexicaulis. Observou-se uma variação individual em S. flexicaulis com relação à duração do período e quantidade de produção de flores e frutos. A população estudada, no entanto, apresentou reprodução contínua ao longo do ano, porém de baixa intensidade na maioria dos indivíduos. Struthanthus flexicaulis teve seus frutos consumidos por um número bastante restrito de frugívoros: Elaenia cristata e Elaenia obscura (Tyrannidae), Mimus saturninus (Mimidae) e Schistochlamys ruficapillus (Thraupidae). No entanto, E. cristata foi o visitante mais freqüente e abundante ao longo do ano, responsável por mais de 96% das sementes dispersadas. Na área de estudo E. cristata apresentou uma dieta predominantemente frugívora, complementada pelo consumo de artrópodes... . / Quantity and quality components of seed dispersal in Struthanthus flexicaulis (Loranthaceae) in a rupestrian field in southeastern Brazil. The investigation of interactions between plants and their seed dispersers has implications for the understanding of the biological diversity, its conservation and is an important issue in evolutionary ecology. The goal of this study is to investigate the components of quality and quantity in the seed dispersal of Struthanthus flexicaulis (Loranthaceae), a generalist dioceous mistletoe with wide distribution in central Brazil. The study was conducted between January 2004 and April 2005 in a rocky outcrop in Serra do Cipó, Minas Gerais, southeastern Brazil. I conducted observations on female plant reproductive phenology, focal observations on fruiting plants to record behavioral parameters of seed dispersers, censuses to record the relative abundance, seasonality and behavioral parameters of seed dispersers, a direct observations and search on random established plots to evaluate seed deposition. Individual variation in the duration and quantity of flower and fruit production was detected. At the population level, reproduction was continuous, but in low intensity for most of the individuals. The fruits of S. flexicaulis were consumed by a small set of avian frugivores: Elaenia cristata and Elaenia obscura (Tyrannidae), Mimus saturninus (Mimidae) and Schistochlamys ruficapillus (Thraupidae), but E. cristata was the most frequent and abundant visitor throughout the year, responsible for more than 96% of the dispersed seeds. Human Vγ9Vδ2 T cell immune responses towards congenital Toxoplasma gondii infection and mistletoe extract drug stimulationMa, Ling 21 October 2020 (has links) (PDF)
Vγ9Vδ2 T cells are the main circulating γδ T cells in human adult blood. They are known for their T cell receptor (TCR)-dependent recognition of microbe and endogenous-derived non-peptide pyrophosphate antigens (phosphoantigens, PAg). With the intrinsically biased type 1 immune responses, Vγ9Vδ2 T cells are an important force in the defense of infections and tumors. However, the immune responses of Vγ9Vδ2 T cells in early life infections and in immunotherapies are not clear yet. In this thesis, we explored Vγ9Vδ2 T cell immune responses in both aspects. Vγ9Vδ2 T cells are abundant in human fetal peripheral blood, but compared to their adult counterparts they have a distinct developmental origin, are hyporesponsive towards in vitro phosphoantigen exposure and they do not possess a cytotoxic effector phenotype. In order to obtain insight into the role of Vγ9Vδ2 T cells in the human fetus, we investigated in the first part of this thesis their responses upon in utero infection with the phosphoantigen-producing parasite Toxoplasma gondii (T. gondii). Most congenital infections are caused by viruses, T. gondii is one of the exceptions. The organelle apicoplast present in T. gondii can generate the most potent Vγ9Vδ2 T cell activator. Thus infection in utero with T. gondii makes it a good model to observe Vγ9Vδ2 T cell immune responses in early life. By comparing to age-matched controls, we found that fetal Vγ9Vδ2 T cells were highly expanded in congenital T. gondii infected newborns, and these expanded cells were highly differentiated towards potent cytotoxic effector cells. While the impact of congenital infection on Vγ9Vδ2 T cell expansion and function waned after birth, the Vγ9Vδ2 TCR repertoire of infected infants possessed a clear fetal footprint with public clonotypes, reflecting the Vγ9Vδ2 T cell response in utero. Indeed, verification of the antigen recognition related complementarity-determining region 3 (CDR3) of the TCR for γ and δ chain by high-throughput sequencing revealed the enrichment of three Vδ2 sequences in congenitally-infected infants that are already generated at 8 weeks of gestation. Vγ9Vδ2 T cells possess several characteristics, including MHC-independent recognition of tumor cells and potent killing potential, that make them attractive candidates for cancer immunotherapeutic approaches. In the second part of this thesis we investigated Vγ9Vδ2 T cell responses towards two kinds of hemiparasite plant Viscum album L. (European mistletoe) extract drugs in vitro. Mistletoe therapy is the most used complementary cancer therapy in European countries. Mistletoe extract drugs are considered to benefit for increasing the quality of life of cancer patients and modulate immune cells, but the mechanism of action is not clear. Here, we investigated in-depth the in vitro response of human T cells towards mistletoe extract drugs by analyzing their functional and TCR responses using flow cytometry and high-throughput sequencing respectively. Non-fermented mistletoe-extract drugs (AbnobaViscum), but not their fermented counterparts (Iscador), induced specific expansion of Vγ9Vδ2 T cells among T cells. Furthermore, AbnobaViscum rapidly induced the release of cytotoxic granules and the production of the cytokines IFNγ and TNFα in Vγ9Vδ2 T cells. This stimulation of anti-cancer Vγ9Vδ2 T cells was mediated by the butyrophilin BTN3A, did not depend on the accumulation of endogenous phosphoantigens and involved the same Vγ9Vδ2 TCR repertoire as those of phosphoantigen-reactive Vγ9Vδ2 T cells.In summary, in the first part of this thesis we showed that the human fetus intrinsically possesses a group of Vγ9Vδ2 T cells that are responding to congenital parasite infections that provide potential protective effects to the fetus. Reciprocal Transplant and Machine Learning Study of Oak Mistletoe on Three Host Oak Species in Santa Margarita, CaliforniaAbelli-Amen, Ella 01 June 2021 (has links) (PDF)
At Santa Margarita Ranch, California, oak mistletoe (Phoradendron villosum) parasitizes valley oak and blue oak but cannot be found growing on coast live oak despite its abundance and ability to parasitize coast live oak in other areas. It seems as though this species of mistletoe is specializing on certain host oak trees, but the mechanisms of this specialization are unknown. In order to investigate this pattern, we utilized a type of machine learning in GIS called supervised classification as well as a reciprocal transplant study in the field. The three species of oak trees were classified with 87% accuracy using drone imagery and 95% accuracy using open source NAIP imagery. This classification technique could be applied to the whole state of California as long as ground truth points for each species were collected. This could be extremely useful for large scale forest management projects and ecological questions. Unfortunately, the classifier was unsuccessful at distinguishing mistletoe from host and so the number of mistletoe on each host could not be quantified using this technique. The reciprocal transplant study involved collecting mistletoe fruit from individuals growing on each of the three hosts and experimentally applying them back onto all three hosts. This allowed us to test whether there are host races of mistletoe that specialize at growing on certain hosts. We found that seeds from each host origin germinated equally well regardless of where they were dispersed, and seeds survived best on coast live oak, regardless of where they originated from. Based on these results, there must be some mechanism, other than host races, that explains the lack of mistletoe on coast live oaks at Santa Margarita Ranch. Future projects should investigate whether evidence for host races can be found at a later stage of seedling development and the roll of bird dispersers in creating the pattern.

No / In 1834 in a tumulus at Gristhorpe, North Yorkshire, UK, an intact coffin fashioned from the hollowed-out trunk of an oak tree was found to contain a well-preserved skeleton stained black from the oak tannins, wrapped in an animal skin and buried with a range of grave artefacts, including a bronze dagger, flints and a bark vessel. The remains were deposited in the Rotunda Museum at Scarborough, where closure due to refurbishment in 2005–2008 provided an opportunity for the scientific investigation of the skeletal remains and artefacts using a wide range of techniques. Dendrochronological and radiocarbon dating has established the age of the skeleton as 2140–1940 BC at 95% confidence, in the Early Bronze Age. As part of this project, Raman spectra of several mysterious small spherical objects discovered in the coffin underneath the skeleton and initially believed to be ‘mistletoe berries’ associated with ancient burial customs have been recorded non-destructively. The interpretation of the Raman spectral data, microscopic analysis and comparison with modern specimens has led to the conclusion that the small spheres are phosphatic urinary stones, which reflect the archaeological dietary evidence and stable isotope analysis of bone collagen of Gristhorpe Man.

Les progrès réalisés en immunologie ont orienté les recherches vers des approches et des stratégies de plus en plus prometteuses et innovantes afin de mieux manipuler la réponse immunitaire. Le but de nos recherches est la prévention et le traitement des maladies liées aux dysfonctionnements du système immunitaire, telles que les maladies auto-immunes, inflammatoires et malignes. Bien que l’inflammation constitue un processus physiologique indispensable au maintien de l’homéostasie suite à une infection ou à une lésion, elle est également associée à des pathologies infectieuses, auto-immunes et tumorales. Les stratégies thérapeutiques les plus utilisées pour traiter l’inflammation sont basées sur la neutralisation des médiateurs inflammatoires par des anticorps, des antagonistes moléculaires, des immunoglobulines intraveineuses, des corticostéroïdes, des médicaments anti-inflammatoires non stéroïdiens. En plus des traitements mentionnés, des produits issus de la phytothérapie ont été largement utilisés afin d’atténuer l'inflammation et la douleur dans plusieurs maladies inflammatoires et dans le cancer. Depuis des décennies, les préparations de Viscum album, connu sous le nom de « gui européen », sont largement utilisées dans le traitement du cancer comme thérapie auxiliaire. Bien que les mécanismes d’action soient partiellement connus, plusieurs hypothèses ont été proposées. En effet, les mécanismes anti-tumoraux du Viscum album impliquent des propriétés induisant une cytotoxicité, l'apoptose, l'inhibition de l'angiogenèse et plusieurs autres mécanismes immunomodulateurs. Ce travail décrit un nouveau mécanisme anti-inflammatoire de Viscum album, qui participe à l’effet thérapeutique de ces préparations. De plus, l’effet bénéfique anti-inflammatoire observé est associé à l’inhibition des voies pro- inflammatoires de COX2 et PGE2 dans les cellules épithéliales issues d’adénocarcinome du poumon. Ce travail a identifié un des mécanismes moléculaires de Viscum album associé à son effet anti-inflammatoire participant à ses bénéfices thérapeutiques. Ainsi, ces préparations pourraient être utilisées en combinaison avec d’autres traitements dans des maladies inflammatoires et dans le cancer. / Recent advances in immunology research have led us towards more promising approaches and strategies to manipulate the immune response to prevent or treat the diseases related to immune dysfunction such as autoimmune, inflammatory pathologies and malignant diseases. Although, immuno inflammation is a basal physiological phenomenon required to eliminate the causative agent and to initiate the healing process, it is a physiopathological symptom in a diverse conditions of infectious, autoimmune and tumoral origin. Various therapeutic strategies have been developed in order to reduce inflammation and pain, including the treatment with cytokine neutralizing antibodies, molecular antagonists, intravenous immunoglobulins, corticosteroids, non-steroid anti-inflammatory drugs (NSAID) and several others. In addition to these well known anti-inflammatory therapeutic strategies, treatment with various phytotherapeutics has also contributed enormously to control inflammation and pain, associated with various severe inflammatory disorders and cancer. Viscum album (VA) preparations, commonly known as European mistletoe, are extensively used as complementary therapy in cancer for decades. However the mechanisms of action have been partially understood. Several mutually non-exclusive mechanisms have been proposed such as anti-tumor properties which involve the cytotoxic properties, induction of apoptosis, inhibition of angiogenesis and several other immunomodulatory mechanisms. This study reveals anti-inflammatory mechanism as another important mechanism of action of these phytotherapeutics, which is responsible for their therapeutic benefit and addresses the molecular mechanisms in the pro-inflammatory axis of COX-2 and PGE2 using in vitro experimental model of human lung adenocarcinoma. The present work contributes for a better understanding of mechanisms of action of Viscum album preparations underlying their therapeutic benefit and allows us to revitalize the therapeutic strategies used in treatment of inflammatory disorders and cancer.

The impacts of possum herbivory and possum control on threatened palatable species (Pittosporum patulum, Alepis flavida and Peraxilla tetrapetala) in the Lake Ohau Catchment, South Island, New ZealandHead, Nicholas January 2005 (has links)
The impacts of possum herbivory were assessed on 713 individual plants of the nationally endangered Pittosporum patulum and 115 individual plants of the threatened ('Gradual Decline') mistletoes (Alepis flavida & Peraxilla tetrapetala) over three years in the Temple and Huxley river valleys in the Ohau catchment, South Island New Zealand. Statistical models were used to test the influence of several explanatory variables on the probability of survival and growth rates of these rare palatable plants. Increasing defoliation levels were the most significant predictors of mortality for both P. patulum and mistletoe, and reduced growth rates in P. patulum. Mortality of P. patulum increased with plant size although smaller plants were also affected by mortality. P. patulum mortality was greater for non-forest habitats, and growth rates were greatest on warm aspects. Mistletoe mortality increased with warm aspect, steeper slopes and at lower altitudes. Plant survival and growth increased in the Temple valley following possum control. Although indicative of a positive treatment effect, other factors may have also influenced this result. Possums are the major herbivore responsible for the widespread decline of P. patulum and beech forest mistletoes throughout New Zealand, although the autecology of P. patulum predisposes it toward extinction more so than mistletoe. Monitoring in conjunction with possum control operations provide good opportunities for understanding the impacts of possums on palatable plants and ecosystems. Assessing defoliation levels on indicator species like P. patulum and mistletoes can serve as a guide for managers to assess ecosystem stress from herbivory.
