This thesis is dedicated to the production of aluminum casting alloys. It maps the course of changes in the morphology of eutectic and its influence on mechanical properties during the long-term meltdown. In particular, the use of image analysis methods contributes to describing these processes. The presented results show that the eutectic shape does not change even during the meltdown for 110 minutes. There is only a roughing and an increase in grain size. There was also no influence on the mechanical properties that fit the practical application of the alloy examined

Optimalizace povrchových úprav polymerů pro mikroskopická pozorování / Optimizing of Polymer Surface Treatment for Microscopic Examination

Supramolecular structure of eight commercial types of iPP, its blends with poly(L-lactide) (PP-PLLA) and copolymers with ethylene-propylene rubber (PP-EPR) together with four types of polyethylene (LDPE, HDPE) was uncovered and observed in this work. The spherulitic structure of iPP and PE homopolymers and PP copolymers was uncovered by etching of six different solutions of mineral acids with KMnO4 and by dissolution in four selected solvents. The latter was found to be ineffective. The structure of PP-PLLA blends was uncovered only after recrystallization (the change of structure by annealing and cooling rate). The uncovered crystalline structure was observed directly by confocal laser scanning microscope (CLSM) and SEM. Crystallinity together with a size distribution of crystallites was determined by DSC. It was proved that spherulitic supramolecular structure was easy to uncover by chemical etching only for molded samples and for fracture surfaces of injected samples. The efficiency of etchants varied but, generally, the samples with high degree of crystallinity were etched earlier. The etchants containing nitric acid and high content of sulphuric acid uncovered fine details of shperulites with respect to mixtures containing orthophosporic acid. It was also observed that not only the etchant (especially with nitric acid) itself but also its vapors were effective. The structure was revealed later but the appearance was plastic (3D) and the structure was very fine. The supramolecular structure of PP-PLLA blends differed with amount of each component. The ethylene-rubber phase came forth with increasing time of etching while PP spherulites disappeared in PP-EPR samples. CLSM was shown to be very good tool for observing supramolecular structure of studied samples with respect to SEM, which was proved to be entirely unsuitable for polyolefines. DSC is recommended to be performe before uncovering supramolecular structure for basic information about proportion of amorphous/crystalline phase and size of crystallites.

Vliv nanočástic na rychlost izotermické krystalizace isotaktického polypropylénu / The influence of nanoparticles on isothermal crystallization rate of isotactic polypropylene

This diploma thesis deals with the study of isothermal crystallization of nanocomposite materials based on isotactic polypropylene (iPP) and nanosilica, depending on the volume fraction (0, 2, 4 and 6 %) and type of silica and the crystallization temperature. Fumed silica and four types of silica with different surface treatment were used as filler. The crystallization performed at temperatures 136, 138, 140 and 142 °C has been studied in-situ using polarized optical microscope and the crystallization rate was evaluated from the growing radius of spherulites. It can be stated that particles of silica have been inbuilt into the spherulites due to the linear growth of spherulites of all composite materials. The most significant increase of the crystallization rate of iPP was caused by fumed silica, the most significant decrease was caused by silica TS-720 at volume fraction 4 %. The crystallization rate (G) has been slightly increased with the increasing volume fraction of filler at 136 °C, the G slightly decreased at 138 °C and there was no trend of G for two higher temperatures. Also, it was not possible to exactly evaluate the influence of the surface treatments. The reason is the most probably uneven (non-homogenous) distribution of nanosilica as revealed thermogravimetric analysis. The crystallization kinetics was evaluated according to the Lauritzen-Hoffmann theory. The morphological part of the study showed that iPP was in ? and ß-structure and spherulites of ? phase were of the type I, II and mixed.

Catheter ablation is a unique method for cardiac arrhythmias treatment based on creating spot lesions of radiofrequency energy using intracardiac ablation catheter. Transmurality of these lesions should be achieved for long-term therapeutic treatment effect. The degree of myocardial damage depth is reflected in the parameters of sensed intracardiac ECG. From research studies is evident that the change in morphology and deflection of ablation signal play a key role in the assessment of transmurality. This thesis consists of theoretical introduction to electrophysiology, proposal and realization of methods for objective assessment of transmurality by processing signals in time, frequency and time – frequency domain. The end of this diploma thesis includes the statistical analysis of these clinical dat and comparison of methods.

Pokročilé metody detekce kontury srdečních buněk / Advanced methods for cardiac cells contour detection

This thesis focuses on advanced methods of detecting contours of the cardiac cells and measuring their contraction. The theoretical section describes the types of confocal microscopes, which are used for capturing biological samples. The following chapter is devoted to the methods of cardiac cells segmentation, where we are introduced to the generally applied approaches. The most widely spread methods of segmentation are active contours and mathematical morphology, which are the crucial topics of this thesis. Thanks to the those methods we are able in the visual data to accurately detect required elements and measure their surface chnage in time. Acquired theoretical knowledge leads us to the practical realization of the methods in MATLAB.

Morfologie polyamidu 12 a polyamidu 11 vzniklá za zvýšeného tlaku v kalorimetru / Morphology of Polyamide 12 and Polyamide 11 formed at overpressure and underpressure

In this master thesis it was studied morphology of polyamide 12 (PA12), polyamide 11 (PA11) and their blends in the percent ratios 95/5, 75/25, 50/50, 25/75 and 5/95 formed at pressure 7 MPa in a calorimeter both after continual heating and cooling and after annealing and isothermal crystallization. Neat polyamides and blends were further characterized by thermogravimetric analysis, differential scanning calorimetry at atmospheric pressure, Fourier-transform infrared spectroscopy in attenuated total refection, and X-ray diffraction analysis. The increased pressure improved level of polyamide chains ordering in crystal lamellae. Thermal annealing improved especially ordering of PA12, isothermal crystallization led to considerable increase of crystallinity, whereas PA11-rich blends supported perfection of PA12 crystals. Partial transformation of - to -structure occurred in case of neat PA12 at high pressure, the change of the chain conformation into the closer one occurred in neat PA11 without transformation of the type of crystal structure.

Yeast Saccharomyces cerevisiae as an unicellular organism is one of the best-studied experimental organisms. It is an important model organism for the study of intracellular processes of eukaryotic cells. Yeasts are also social organisms with cell-to-cell communication able to form organized multicellular structures (colonies and biofilms). Yeast and other microorganisms in nature prefer to form colonies on solid substrates rather than to grow as "planktonic" single cells (Palková, 2004; Wimpenny, 2009). The yeast S. cerevisiae typically forms colonies, biofilms were described only rarely. Yeast colonies exhibit an organized morphological pattern characteristic of each particular yeast strain (Kocková-Kratochvílová, 1982). This work is focusing on morphology and differentiation of the S. cerevisiae colonies of common laboratory strains forming less structured colonies, and strains of the Σ1278b genetic background forming highly structured "fluffy" colonies. It shows that polarized budding pattern and especially cell ability to form aggregates enable development of structured morphology. During development of "fluffy" colonies two differently regulated events of dimorphic switch from yeast form to filamentous growth occur. (in English) Combs are used as toilet objects for adjustment of hair and beard, or as a decoration of hairstyle all over the world. Combs have also a number of symbolical and magical significances. The aim of this work is a comprehensive view on combs as a one part of medieval material culture. The first part deals with the manufacture of medieval combs, particularly materials used for comb making, manufacturing processes, decoration, the degree of specialization of bone and antler objects manufacturing and manufactures in which bone and antler objects were produced. The second part is applied to the function of combs, evolution of hairstyles in the Middle Ages, symbolical significance and the associated magical significances of human hair and beard and different rituals connected with their cutting and combing. The core of this thesis is analysis, synthesis and interpretation of selected finds of medieval combs from what now is the territory of the Czech Republic. The aim of the analysis, synthesis and interpretation is the comparison of morphology and decoration of combs from different periods of Middle Ages. Chapters, which are devoted to chronology, morphology and typology of medieval combs and social-representational aspects of using combs are largery based on this part.

Diverzita řas z červeného sněhu v Evropě: kombinace molekulárních a morfologických dat / The diversity of algae from red snow in Europe: combination of molecular and morphological data

We can find a lot of microorganisms living in snow including psychrophilic snow algae from the order Chlamydomonadales (Chlorophyta). They are adapted to the extreme conditions in this habitat and can cause the phenomenon of coloured snow. The species Chlamydomonas nivalis (Bauer) Wille is the most commonly associated with red snow in alpine and polar regions during summer season worldwide. In the field material, we can find red spherical cells without flagella and any morphological characteristics suitable for species determination. Until now, this species has not been isolated into laboratory culture and its life cycle is unclear. Furthermore it has been shown that red coloured snow can be caused by more species which used to be determined as Chlamydomonas nivalis. The aim of this study was to collect samples of red snow from different parts of Europe, to describe the morphological variability of Chlamydomonas nivalis-like snow algae in relation to region of origin, to try to isolate laboratory strain of this species and to describe its position and distribution by phylogenetic analysis of laboratory strains and field samples.
