The naphthoquinone α-lapachone has been shown to be suitable as a prototype for the development of substances with anticancer properties, as reported by our group to tetrahydropyran derivative (THP), which demonstrated significant cytotoxicity and selectivity against MDA-MB-435 melanoma line. The aim of the present work was to evaluate the mechanism of action involved in the cytotoxicity of α-lapachone and its THP derivative against MDA-MB-435 melanoma cells. Initially, we evaluated the cytotoxicity of α-lapachone and its THP derivative against 8 cell lines, by the MTT assay, showing IC50 of 1.37 and 8.18 µM to breast and melanoma lines, respectively, after 72 hours of incubation. The selectivity of the THP derivative in Alamar Blue assay, demonstrated that THP is 2.6 times less cytotoxic to normal cells as compared to tumor cells. Studies on the mechanism of cell death in MDA-MB-435 tumor line showed that the THP derivative caused a reduction on viable cells associated with an increase of non-viable cells by inducing loss of membrane integrity in concentrations of 5 and 10 µM. The cytotoxic activity of THP was independent of cell cycle, activation of effector caspases and formation of reactive oxygen species, suggesting the occurrence of a necrotic process after 6 hours of treatment, demonstrated by evaluation of membrane integrity. Thus, the data suggest that a tetrahydropyran group introduction in α-lapachone molecule enhances the cytotoxicity of MDA-MB-435 in melanoma cells, via necrosis, which reinforces the importance of naphthoquinones as prototypes for the development of new synthetic compounds with antitumor activity. / As quinonas são metabólitos de ampla distribuição na natureza que possuem diversas atividades farmacológicas de importância clínica. A naftoquinona α-lapachona demonstrou potencial como protótipo para o desenvolvimento de substâncias com propriedades anticâncer, como relatado pelo nosso grupo de pesquisa, em que o seu derivado tetrahidropirano (THP) apresentou citotoxicidade e seletividade significante contra a linhagem de melanoma MDA-MB-435. Assim, o objetivo deste trabalho foi avaliar o mecanismo de ação envolvido na citotoxicidade da α-lapachona e derivado THP em células de melanoma MDA-MB-435. Inicialmente, avaliou-se a citotoxicidade da alfa-lapachona e derivado THP em 8 linhagens tumorais de mama e melanoma, através do ensaio do MTT, mostrando CI50 de 1,37 e 8,18 µM, respectivamente, após 72 horas de incubação. A seletividade do derivado THP no ensaio de Alamar Blue, demonstrou que este se apresentou 2,6 vezes menos citotóxico para células normais quando comparado às células tumorais. Estudos do mecanismo de morte celular na linhagem tumoral MDA-MB-435 indicaram que o derivado THP causou redução de células viáveis associado com o aumento de células não-viáveis por indução da perda de integridade da membrana plasmática nas concentrações de 5 e 10 µM após 24 horas de incubação. A atividade citotóxica do derivado THP não está relacionada a uma fase específica do ciclo celular, ativação de caspases efetoras e formação de espécies reativas de oxigênio, sugerindo a ocorrência de um processo necrótico a partir de 6 horas de tratamento, demonstrado pela avaliação da integridade de membrana. Assim, os resultados exibidos sugerem que a introdução do radical tetrahidropirano na molécula da α-lapachona aumenta a citotoxicidade em células de melanoma MDA-MB-435, via necrose, o que reforça a importância de naftoquinonas, como protótipo para o desenvolvimento de novos compostos sintéticos com atividade antitumoral.

Bothrops species are responsible for more than 20,000 accidents per year in Brazil—90% of all recorded snakebites. The Bothrops leucurus is an important venomous snake that inhabits the northeast of Brazil. The biological effects due envenomation have similar profile to that observed in other Bothrops, ie coagulant activity, hemorrhagic, fibrinolytic, and renal failure in vivo. The aim of this study was to investigate the effects of the Bothrops leucurus venom in the renal perfusion system and in cultured renal tubular cells of the type MDCK (Madin–Darby Canine kidney). Isolated kidneys from Wistar rats weighing 250 to 300g (n=5) were perfused with Krebs-Henseleit solution containing 6% of bovine serum albumin previously dialyzed for 120 minutes. The effects of Bothrops leucurus venom (VBl) (10μg/mL) were studied on the Perfusion Pressure (PP), Renal Vascular Resistance (RVR), Urinary Flow (UF), Glomerular Filtration Rate (GFR) Percentage of Sodium (%TNa+), Potassium (%TK+) and Chloride (%TCl-) Tubular Transport. B. leucurus venom (10 μg/mL) reduction the PP at 90 and 120 min and UF at 60 and 90 min. The glomerular filtration rate decreased at 60 and 90 min. The renal vascular resistance decreased at 120 min. It was also observed a decrease on percentual tubular transport of sodium (%TNa+) at 120 min and of chloride (%TCl-) at 60 and 90 min. MDCK cells were grown in plastic flasks at 37° C in a humidified atmosphere of 5% CO2 – air, with RPMI 1640 medium supplemented with 10% fetal calf serum. The treatment with VBl caused decrease in cell viability to the lowest concentration tested with an CI50 of 1.25 μg/mL. Flow cytometry with annexin V and propidium iodide showed that cell death occurred predominantly by necrosis. When apoptosis was analyzed by nuclear staining, it was observed significant increase in the percentage of cell death, VBl 2,5 μg/mL induced 6,8% apoptosis and VBl 1,25 μg/mL caused 5,8% of apoptosis after 24 h of treatment. VBl treatment (1,25 μg/mL) led to significant depolarization of the mitochondrial membrane potential. We found increased expression of genes involved in cell death by apoptosis in the lower concentrations tested. The ion Ca2+ signaling participates in cell death induced by the venom of B. leucurus. These results demonstrate that the venom of B. leucurus altered all the renal parameters assessed in isolated kidney perfusion and was cytotoxic to MDCK cell culture. / Venenos de Bothrops consistem em uma mistura complexa de substâncias ativas, principalmente peptídeos e proteínas, capazes de interferir com muitos processos fisiológicos. Serpentes do gênero Bothrops são responsáveis por mais de 20 mil acidentes por ano no Brasil, que totalizam 90% de todos os acidentes ofídicos registrados. A Bothrops leucurus é uma serpente peçonhenta importante que habita a região nordeste do Brasil. Os efeitos biológicos devido ao envenenamento têm perfil semelhante ao observado em outras Bothrops, ou seja, a atividade coagulante, hemorrágica, fibrinolítica e insuficiência renal in vivo. O objetivo deste estudo foi investigar os efeitos do veneno de Bothrops leucurus no sistema de perfusão renal e em culturas de células tubulares renais do tipo MDCK (Madin-Darby Canine kidney). Rins isolados de ratos Wistar pesando 250 a 300g (n = 5) foram perfundidos com Krebs-Henseleit contendo 6% de albumina sérica bovina previamente dialisadas por 120 minutos. Os efeitos do veneno de Bothrops leucurus (VBl) (10μg/mL) foram estudados sobre o Ritmo de Filtração Glomerular (RFG), Fluxo Urinário (FU), Pressão de Perfusão (PP), Resistência Vascular Renal (RVR), Percentual de Transporte Tubular de Sódio (%TNa+), de Potássio (%TK+) e de Cloreto (%TCl-). O veneno de B. leucurus (10 μg / mL) reduziu a PP aos 90 e 120 min e o FU aos 60 e 90 min. O ritmo de filtração glomerular diminuiu aos 60 e 90 min. A resistência vascular renal diminuiu aos 120 min. Observou-se também uma diminuição no percentual de transporte tubular de sódio (% TNA+) aos 120 min e de cloreto (% TCL-) aos 60 e 90 min. Células MDCK foram cultivadas em garrafas plásticas a 37°C em atmosfera de 5% de CO2, com meio RPMI 1640 suplementado com soro bovino fetal a 10%. O tratamento com VBl causou diminuição da viabilidade celular até na menor concentração testada com um IC50 de 1,25 μg /mL. Citometria de fluxo com anexina V e iodeto de propídio mostrou que a morte celular ocorreu predominantemente por necrose de forma concentração dependente. Quando a apoptose foi analisada através da coloração nuclear, foi observado aumento significativo na porcentagem de morte celular, VBl nas concentrações de 2,5 e 1,5 μg/mL induziu 6,8% e 5,8% de apoptose após 24 horas de tratamento, respectivamente. O tratamento com VBl (1,25 μg/mL) levou a despolarização significativa do potencial de membrana mitocondrial. Nas menores concentrações avaliadas, verificamos aumento de expressão de genes envolvidos na morte celular por apoptose. O íon Ca2+ participa da sinalização da morte celular induzida pelo veneno de B. leucurus. Esses resultados demonstram que o veneno de B. leucurus alterou todos os parâmetros renais avaliados na perfusão de rim isolado e foi citotóxico para cultura de células MDCK.

Snake venoms display a wide array of pharmacological properties due to their complex combinations of active substances, especially peptides and proteins, capable of interfering with physiological processes. Snakes of the genera Bothrops and Bothropoides accounted for 70% of snake bites reported in Brazil. The Bothropoides insularis (golden lancehead) is endemic to Queimada Grande, an island off the state of São Paulo. Like the venom of other Bothrops species, the venom of B. insularis has coagulant, hemorrhagic and fibrinolytic properties and is known to cause kidney failure in vivo. The objective of the study was to evaluate of whole venom of B. insularis and its phospholipase A2 fraction in Madin-Darby canine kidney (MDCK) cell culture tubes. The cells were grown in RPMI 1640 medium supplemented with 10% fetal bovine serum in plastic vials with an atmosphere containing 5% CO2 at 37°C. Treatment with whole venom reduced cell viability at all the concentrations tested (200, 100, 50, 25, 12.5 and 6.25 mg/mL) and yielded an IC50 of 9 μg/mL. Cell membrane integrity during the process of cell death from whole venom was evaluated by LDH enzyme assay, indicating enzyme release at all concentrations. Flow cytometry with Annexin V-FITC and propidium iodide revealed that cell death at the lowest concentrations (18 and 36 μg/mL) of whole venom occurred predominantly by apoptosis. However, at the highest concentration (72 μg/mL), in addition to death by apoptosis, an increased number of cells in late apoptosis was observed.In the Rhodamine 123 assay the treatment with whole venom (36 μg/mL) induced significant depolarization of the mitochondrial membrane potential. The expression of genes involved in cell death by apoptosis did not show any significant results, when tested with whole venom in 24h. The phospholipase A2 fraction was also submitted to cell viability and membrane integrity assays at several concentrations (200, 100, 50, 25, 12.5 and 6.25 mg/mL), with no significant results in any of the experiments. Taken together, our results indicate that the venom of B. insularis is cytotoxic in MDCK cell culture and that cell death occurs predominantly by apoptosis. / Os produtos naturais e seus derivados têm sido as fontes mais comuns na obtenção de agentes terapêuticas inovadores, além de fornecerem ferramentas para estudos fisiopatológicos e farmacológicos. Os venenos de serpentes exibem uma infinidade de atividades farmacológicas, por serem formados por uma mistura complexa de substâncias ativas, principalmente peptídeos e proteínas, capazes de interferir com muitos processos fisiológicos. As serpentes Bothrops e Bothropoides são responsáveis por 70% dos acidentes ofídicos ocorridos no Brasil. A serpente Bothropoides insularis (jarara ilhoa), é uma espécie endêmica nativa da ilha de Queimada Grande na costa Sul do Estado de São Paulo, seu veneno apresenta características semelhantes aos das outras serpentes do gênero Bothrops, como atividade coagulante, hemorrágica, fibrinolítica e insuficiência renal in vivo. O objetivo deste estudo foi investigar os efeitos do veneno total da Bothropoides insularis e sua fração fosfolipase A2 em culturas de células tubulares renais do tipo MDCK (Madin-Darby canine kidney). Células MDCK foram cultivadas em garrafas plásticas a 37 °C em atmosfera de 5% de CO2, com meio RPMI 1640 suplementado com soro bovino fetal a 10%. O tratamento com veneno total da B.insularis causou diminuição da viabilidade celular em todas as concentrações (200; 100; 50; 25; 12,5 e 6,25 mg/mL) estudadas com um IC50 de 9 μg /mL. A integridade da membrana celular foi avaliada pelo método de determinação da enzima lactato desidrogenase, e após a exposição das células ao veneno houve um aumento significativo da enzima. Nos ensaios de citometria de fluxo com anexina V-FITC e iodeto de propídio foi observada a morte celular nas menores concentrações (18 e 36 μg/mL) estudadas com o veneno total e ocorreu predominantemente por apoptose. No entanto na maior concentração (72μg/mL) estudada, foi observado além da morte por apoptose, um aumento de células em apoptose tardia. Nos ensaios com rodamina 123, o tratamento com o veneno total da B.insularis (36 μg/ mL) levou a despolarização significativa do potencial de membrana mitocondrial. Não houve expressão significativa de genes envolvidos na morte celular por apoptose. Ensaios de viabilidade celular e integridade de membrana foram realizados com a fração fosfolipase A2 do veneno da B.insularis, em várias concentrações (200; 100; 50; 25; 12,5 e 6,25 mg/mL), não apresentado resultados significativos em nenhum dos experimentos. Esses resultados demonstram a citotoxicidade do veneno de B. insularis em culturas de células tubulares renais (MDCK), e sugerem morte celular predominantemente por apoptose.

Papel da autofagia na senescência celular

Autofagia é o processo de degradação de componentes próprios celulares e parece modular senescência e apoptose induzidas por diferentes tipos de injúria, incluindo dano ao DNA. Populacionalmente, autofagia ocorre logo após o dano ao genoma, enquanto senescência é a resposta crônica das células que resistem à morte celular. Até o momento, nenhum trabalho demonstrou o papel da autofagia na senescência de fato, principalmente considerando uma análise integrada com morte celular e a ocorrência destes mecanismos a nível de células únicas. Inicialmente, desenvolvemos uma metodologia para avaliação do tamanho e forma de núcleos de células eucarióticas em cultura, a Análise Morfométrica Nuclear, a qual foi validada para análise de apoptose, senescência e irregularidades nucleares. O segundo objetivo foi avaliar o papel da autofagia na senescência e morte celular induzidas por dano ao DNA em células de glioblastoma, e os mecanismos moleculares por trás desta relação. Para isso, células expressando estavelmente o marcador de autofagia GFP-LC3 foram tratadas com o indutor de dano ao DNA Temozolomida (TMZ), droga de escolha contra glioblastomas, por 3h seguido do replaqueamento em meio livre de droga. TMZ induziu autofagia transiente e ativação de AMPK-ULK1 e p38 MAPK, acompanhado da supressão crônica da via PI3K/Akt/mTOR. O tratamento com TMZ induziu aumento de vários marcadores de senescência celular a longo prazo, cuja cinética foi analisada de maneira integrada e correlativa, bem como aumentou níveis de ROS, os quais mediaram, ao menos parcialmente, a indução de autofagia e senescência. Considerando a relação entre autofagia e senescência, observamos uma correlação negativa forte entre os mecanismos a nível populacional. Por outro lado, a nível de células únicas esta correlação não foi observada. Através do acompanhamento de células únicas nós observamos também uma redução orquestrada da autofagia independente da aquisição de fenótipo senescente. Esta redução ocorreu antes, durante ou após o aumento da área celular que caracteriza senescência celular, mostrando que a redução da autofagia não é necessária para aquisição do fenótipo senescente. Finalmente, a ativação da autofagia aumentou o efeito pró-senescente da TMZ, enquanto a supressão da autofagia induziu apoptose e reduziu a senescência, sugerindo que a autofagia protege as células da morte celular e permite a entrada das células em estado senescente após tratamento com TMZ. Neste sentido, também discutimos aqui a importância da análise completa e integrada das alterações em mecanismos de proliferação e morte celular induzidas pela inibição da autofagia. Em conclusão, autofagia e senescência parecem ser repostas induzidas por um mesmo sinal mas com cinéticas diferentes nas células expostas a dano ao DNA, estabelecendo uma correlação negativa a nível populacional que não se confirma a nível de células únicas. / Autophagy is the process of degradation of own cellular components and appears to modulate senescence and apoptosis induced by different types of injury, including DNA damage. At a population level, autophagy occurs soon after the damage to the genome, while senescence is chronic response of cells that resist to cell death. To date , no study has demonstrated the role of autophagy in senescence indeed, especially considering an integrated analysis with cell death and the occurrence of these mechanisms at the level of single cells. This thesis consists of two main objectives. Initially, we developed a method to evaluate the shape and size of nuclei of eukaryotic cells in culture, called Morphometric Analysis Nuclear, which was validated for the analysis of apoptosis, senescence and nuclear irregularities. The second objective was to evaluate the role of autophagy in senescence and cell death induced by DNA damage in glioblastoma cells, and the molecular mechanisms behind this relationship. For this, cells stably expressing the autophagy marker GFP- LC3 were treated for 3h with the DNA alkylating agent Temozolomide (TMZ), the drug of choice against glioblastomas, followed by replating in drug-free medium. TMZ induced autophagy and transient activation of AMPK-ULK1 axis and p38 MAPK, accompanied by chronic suppression of the PI3K/Akt/mTOR pathway. The treatment with TMZ induced an increase of several markers of cellular senescence at long term, whose kinetics was analyzed and integrated correlative manner. TMZ also increased ROS levels which mediated, at least in part, the induction of senescence and autophagy . Considering the relationship between autophagy and senescence, we observed a strong negative correlation between the mechanisms at the population level. On the other hand, at a single cell level this correlation was not observed. Through the monitoring of single cells we also observed an orchestrated reduction of autophagy, independently of the senescent phenotype acquisition. This reduction occured before, during or after increasing the cell area featuring cellular senescence, showing that the reduction of autophagy is not required for the acquisition of the senescent phenotype. Finally , activation of the autophagy increased pro- senescent effect of TMZ, while autophagy inhibition triggered apoptosis and reduced senescence, suggesting that autophagy protects cells from cell death and allows senescence entry after treatment with TMZ. In this sense, we also discuss here the importance of comprehensive and integrated analysis of changes in mechanisms of proliferation and cell death induced by autophagy inhibition. In conclusion , autophagy and senescence responses appear to be induced by the same signal but with different kinetics after DNA damage, establishing a negative correlation at a population level that is not confirmed at the level of single cells.

TMZ induziu autofagia transiente e ativação de AMPK-ULK1 e p38 MAPK, acompanhado da supressão crônica da via PI3K/Akt/mTOR. O tratamento com TMZ induziu aumento de vários marcadores de senescência celular a longo prazo, cuja cinética foi analisada de maneira integrada e correlativa, bem como aumentou níveis de ROS, os quais mediaram, ao menos parcialmente, a indução de autofagia e senescência. Considerando a relação entre autofagia e senescência, observamos uma correlação negativa forte entre os mecanismos a nível populacional. Por outro lado, a nível de células únicas esta correlação não foi observada. Através do acompanhamento de células únicas nós observamos também uma redução orquestrada da autofagia independente da aquisição de fenótipo senescente. Esta redução ocorreu antes, durante ou após o aumento da área celular que caracteriza senescência celular, mostrando que a redução da autofagia não é necessária para aquisição do fenótipo senescente. Finalmente, a ativação da autofagia aumentou o efeito pró-senescente da TMZ, enquanto a supressão da autofagia induziu apoptose e reduziu a senescência, sugerindo que a autofagia protege as células da morte celular e permite a entrada das células em estado senescente após tratamento com TMZ. Neste sentido, também discutimos aqui a importância da análise completa e integrada das alterações em mecanismos de proliferação e morte celular induzidas pela inibição da autofagia. For this, cells stably expressing the autophagy marker GFP- LC3 were treated for 3h with the DNA alkylating agent Temozolomide (TMZ), the drug of choice against glioblastomas, followed by replating in drug-free medium. TMZ induced autophagy and transient activation of AMPK-ULK1 axis and p38 MAPK, accompanied by chronic suppression of the PI3K/Akt/mTOR pathway. The treatment with TMZ induced an increase of several markers of cellular senescence at long term, whose kinetics was analyzed and integrated correlative manner. On the other hand, at a single cell level this correlation was not observed. Through the monitoring of single cells we also observed an orchestrated reduction of autophagy, independently of the senescent phenotype acquisition. This reduction occured before, during or after increasing the cell area featuring cellular senescence, showing that the reduction of autophagy is not required for the acquisition of the senescent phenotype. Finally , activation of the autophagy increased pro- senescent effect of TMZ, while autophagy inhibition triggered apoptosis and reduced senescence, suggesting that autophagy protects cells from cell death and allows senescence entry after treatment with TMZ. In this sense, we also discuss here the importance of comprehensive and integrated analysis of changes in mechanisms of proliferation and cell death induced by autophagy inhibition. In conclusion , autophagy and senescence responses appear to be induced by the same signal but with different kinetics after DNA damage, establishing a negative correlation at a population level that is not confirmed at the level of single cells.

Autofagia é o processo de degradação de componentes próprios celulares e parece modular senescência e apoptose induzidas por diferentes tipos de injúria, incluindo dano ao DNA. Populacionalmente, autofagia ocorre logo após o dano ao genoma, enquanto senescência é a resposta crônica das células que resistem à morte celular. Até o momento, nenhum trabalho demonstrou o papel da autofagia na senescência de fato, principalmente considerando uma análise integrada com morte celular e a ocorrência destes mecanismos a nível de células únicas. Inicialmente, desenvolvemos uma metodologia para avaliação do tamanho e forma de núcleos de células eucarióticas em cultura, a Análise Morfométrica Nuclear, a qual foi validada para análise de apoptose, senescência e irregularidades nucleares. O segundo objetivo foi avaliar o papel da autofagia na senescência e morte celular induzidas por dano ao DNA em células de glioblastoma, e os mecanismos moleculares por trás desta relação. Para isso, células expressando estavelmente o marcador de autofagia GFP-LC3 foram tratadas com o indutor de dano ao DNA Temozolomida (TMZ), droga de escolha contra glioblastomas, por 3h seguido do replaqueamento em meio livre de droga. In this context, the events related to the endosperm formation and the accumulation of toxic compounds in the seeds could help in the production of varieties that produce more oil and are less toxic. Thus, anatomical, proteomic and gene expression analysis were made with J. curcas seeds to contribute with the knowledge about physiologic events related with the formation of endosperm and the processes of synthesis and accumulation of phorbol esters. The anatomical analysis showed the maternal tissue responsible for the nutrients transfer to the endosperm and contributed to fill the gaps present in the literature about the anatomy of J. curcas seeds. The inner integument is the maternal tissue that presents the vasculature of the mother plant and is the responsible for the nutrition of the endosperm through the process of programmed cell death (PCD). Based on this result proteomic analysis of two anatomically different regions of the inner integument, named as proximal and distal, were performed. The identified proteins indicated that the inner integument pass through PCD in different degrees in the proximal and distal regions and that in this tissue there is a spatial distribution between catabolism and anabolism. In the proximal region especially hydrolases were identified and in the distal region storage proteins and proteins of biosynthetic pathways were prevalent. The gene expression analysis had two purposes: first, verify wether the transcripts levels of the genes likely involved in PCD, whose proteins were identified and quantified in the proteomic analysis, were related to the protein abundance and second, identify the site of synthesis of the committed enzyme in the phorbol esters biosynthesis, the casbene synthase, through a study with root, leaf, embryo, inner integument and endosperm and 14 putative genes of the mentioned enzyme. The results indicated that the proteins related to PCD must be controlled at transcript level. Nine putative genes from casbene synthase were expressed in roots and one of them was expressed in the leaf and embryo. This work denotes the role of PCD in the inner integument to the development of the seed and also demonstrates the transitory nature of storage of this tissue. In addition gives notation that the seed, despite of accumulates phorbol esters, do not synthesizes these compounds, but probably import them from roots. / A importância econômica da espécie J. curcas deve-se a grande quantidade de óleo acumulada em suas sementes, especificamente no endosperma. Os eventos relacionados à formação do endosperma e ao acúmulo de compostos tóxicos nas sementes podem auxiliar na produção de variedades com maior produção de óleo e menos tóxicas. Visando isso, foram realizadas análises anatômicas, proteômicas e de expressão de genes nas sementes de J. curcas para contribuir com o entendimento sobre os eventos fisiológicos associados à formação do endosperma e aos processos de síntese e acúmulo dos ésteres de forbol. A análise anatômica mostrou o tecido maternal responsável pela transferência de nutrientes para o endosperma e contribuiu para resolver lacunas existentes na literatura sobre a anatomia de sementes de J. curcas. O integumento interno é o tecido maternal que apresenta a vascularização da planta-mãe e é o responsável pela nutrição do endosperma através do processo de morte celular programada (PCD). Com base nesse resultado, realizou-se uma análise proteômica do integumento interno na região proximal e distal ao endosperma, uma vez que as características celulares indicativas de PCD nesse tecido se apresentavam diferentes nessas regiões. As proteínas identificadas indicaram que o integumento interno sofre PCD em diferentes graus nas diferentes regiões e que nesse tecido há uma distribuição espacial entre catabolismo e anabolismo, pois na região proximal identificaram-se principalmente hidrolases e na região distal proteínas de reserva e de vias biossintéticas. A análise de expressão de genes teve dois propósitos: primeiro, verificar se os níveis de transcritos de genes possivelmente relacionados à PCD, cujas proteínas foram identificadas e quantificadas na análise proteômica, estavam relacionados à abundância de proteínas e o segundo foi identificar o local de síntese da enzima comprometida na biossíntese dos ésteres de forbol, a sintase do casbeno, através de um estudo com raiz, folha, embrião, integumento interno e endosperma e 14 genes candidatos à enzima mencionada. Os resultados indicaram que as proteínas relacionadas à PCD devem ser controladas no âmbito transcricional. Nove genes candidatos à sintase do casbeno foram expressos em raiz e um deles foi expresso na folha e no embrião. Este trabalho evidencia o papel da PCD no integumento interno para o desenvolvimento da semente, além de demonstrar a natureza transitória de reserva desse tecido. Adicionalmente, dá indícios de que a semente, apesar de acumular ésteres de forbol, não sintetiza esses compostos, mas possivelmente os importa das raízes.

O papel da autofagia na resistência de gliomas ao tratamento com temozolomida e inibidor de histonas desacetilases

Glioblastoma multiforme é o tipo mais frequente de tumor cerebral primário, sendo caracterizado por uma alta agressividade e um prognóstico bastante limitado. O ácido hidroxâmico suberoilanilida (SAHA) é um inibidor específico de histonas desacetilases aprovado para o tratamento de linfoma cutâneo de células T, e em fase de crescente investigação clínica e pré-clínica em tumores sólidos. Neste estudo, avaliamos a eficácia do ácido hidroxâmico suberoilanilida em tratamento combinado com temozolomida, o agente alquilante já utilizado em glioblastomas. Através de testes de viabilidade e analises por citometria de fluxo em células tumorais das linhagens U251MG e C6, observamos que não houve sinergismo de potenciação entre temozolomida e ácido hidroxâmico suberoilanilida, apenas efeito sinergismo de adição O tratamento combinado inicialmente promoveu parada do ciclo celular em fase G2/M (≥48 h) ao passo que a apoptose foi detectada apenas em exposição prolongada (≥96 h) aos fármacos em estudo. Ainda, as células tratadas com TMZ/SAHA apresentaram fenótipo autofágico, como determinado por citometria de fluxo e imunodetecção de proteínas marcadoras de autofagia como LC3 e o p62/SQSTM1. A autofagia temporalmente precedeu a apoptose e exerceu função citoprotetora, uma vez que o bloqueio da terminação autofágica com cloroquina promoveu uma significante redução na viabilidade celular, a qual foi associada a um aumento de apoptose em células de glioma tratadas com TMZ/SAHA. Portanto, os dados apresentados neste trabalho demonstram que a autofagia é um processo que diminui a eficácia antiglioma do temozolomida e do ácido hidroxâmico suberoilanilida, e a inibição deste fenômeno pode ser uma estratégia para aperfeiçoar a terapia com esses fármacos. / Glioblastoma multiforme (GBM) is the most frequent and aggressive type of primary brain tumor which has been associated with a dismal prognosis. In this study, we tested the efficacy of combining temozolomide (TMZ) with suberoylanilide hydroxamic acid (SAHA) - an inhibitor of HDACs 1, 2, 3, and 6 approved for the treatment of cutaneous T-cell lymphoma - in the viability of tumor cells. The data showed that potentiation synergism between TMZ e SAHA was not achieved due to activation of protective autophagy in vitro. The SAHA/TMZ treatment promoted arrest in the G2/M phase of the cell cycle as soon as 48 h after drug exposure whereas apoptosis was only detected after long-lasting exposure (≥96 h). In addition, SAHA and TMZ induced autophagy as detected by flow cytometry of acridine orange stained cells and immunodetection of the lipidated form of LC3 as well as decreases in p62/SQSTM1. Autophagy preceded apoptosis, and by blocking the termination step of autophagy with chloroquine promoted a significant reduction in the viability of glioma cells which was accompanied by increased apoptosis in SAHA/TMZ treatment. Overall, the herein presented data demonstrate that autophagy impairs the efficacy of combined TMZ/SAHA, and inhibiting this phenomenon could provide novel opportunities to improve the therapeutic potential of these compounds.

It is known that the prognosis of this tumor is directly related to its staging, with more advanced stages responsible for lower survival and impairment in the patient's quality of life, mainly due to pain complaints. Opioids are widely used to minimize pain at the oncology clinic. In the literature there are conflicting results regarding the use of opioid drugs and their action on tumor cells and the process of angiogenesis in the tumor micro-environment. Therefore, this study aimed to evaluate the expression of mu (μ), Kappa (κ) opioid receptors, correlating with prognostic factors such as markers of angiogenesis (VEGF), cell proliferation (Ki-67) and cell death (TUNEL) in Biopsies of tumors with T4 staging, with or without metastatic process, stratified for the Luminal A and Luminal B molecular subtype, diagnosed and operated at Haroldo Juaçaba Hospital (Ceará Cancer Institute) from 2011 to 2016. A total of 38 biopsy specimens from Luminal A and Luminal B breast cancer patients with T4 tumor size, with and without metastatic process, diagnosed, treated and operated at the abovementioned Hospital were included in the study. Demographic data and tumor characteristics, including the use of opioids and the presence of disease recurrence, were also a reason for evaluation and comparison. In addition, in the present study, the expression, by immunohistochemistry, of the μ and κ opioid markers, VEGF, Ki-67 and TUNEL in the breast biopsy samples by the Tissue Microarray (TMA) technique was evaluated. Our results demonstrated that: 1. It was found that there is a significant predominance of ki67 immunoblotting in lymph nodes in patients who used opioids when compared to patients who did not use opioids in the clinical follow-up (p = 0.011); 2. It was evidenced that patients of the Luminal A molecular subtype present a significant increase in the chances of presenting a 51-75% T cell count in the tumor when compared to patients defined as Luminal B subtype (p = 0.020, OR = 21.000; CI = 1,613-273,340); 3. It was possible to show that patients with T4 staging who presented recurrence (7.2 ± 3.7) demonstrated a decrease in the histoscore expression of the Opioid κ in the lymph node location when related to the patients who did not present clinical evolution associated with relapse (10.8 ± 2.2) (p = 0.033) and 4. The correlation analyzes between the opioid μ and κ markers, VEGF, Ki-67 and TUNEL demonstrate that these findings probably act on the same cascade of events in the CA of T4 stage breast. Our study demonstrated that the use of opioids is directly associated with the increase in ki-67 expression and the prevalence of relapses, which showed lower expression for the opioid kappa receptor. In addition, there was no significant difference in the biological behavior of luminal A and luminal B tumors and the immunoexpression of opioid receptors were independent of angiogenesis receptors. In summary, in luminal A and luminal B tumors the use of opioids is directly associated with a worse prognosis, due to reduction of opioid kappa receptor expression. / O câncer de mama representa uma doença de grande preocupação, devido sua alta incidência e elevada mortalidade sendo, atualmente, considerado como a segunda maior causa de morte no mundo. Sabe-se que o prognóstico deste tumor está diretamente relacionado ao seu estadiamento, sendo estágios mais avançados responsáveis pela menor sobrevida e prejuízo na qualidade de vida da paciente, devido principalmente a dor. Os opióides são bastante utilizados para minimizar a dor na clínica oncológica. Contudo, na literatura observam-se resultados conflitantes em relação a utilização de fármacos opióides e sua ação nas células tumorais e no processo de angiogênese no microambiente tumoral, ainda há muitas controvérsias. Portanto, esse estudo propôs avaliar a expressão dos receptores opióides Mu (μ), Kappa (κ), correlacionando com fatores prognósticos tais como os marcadores de angiogênese (VEGF), de proliferação celular (Ki-67) e morte celular (TUNEL) em biópsias de tumores mamários com estadiamento T4, com ou sem processo metastático, estratificados quanto ao subtipo molecular Luminal A e Luminal B, diagnosticados e operados no Hospital Haroldo Juaçaba (Instituto do Câncer do Ceará) no período de 2011 a 2016. Um total de 39 amostras de biópsias de pacientes com câncer de mama padrões Luminal A e Luminal B, com tamanho do tumor T4, com e sem processo metastático, diagnosticados, tratados e operados no Hospital supracitado foram inseridos no estudo. Dados demográficos e características tumorais, incluindo o uso de opióides e a presença de recidiva de doença, foram também motivo de avaliação e de comparação. Além disso, no presente estudo, foi avaliada a expressão, através de imunohistoquímica, dos marcadores μ e κ opióides, VEGF, Ki-67 e TUNEL nas amostras de biópsia de mama através da técnica de Tissue Microarray (TMA). Nossos resultados demonstraram que: 1. verificou-se que há um predomínio significativo de imunomarcações de ki67 nos linfonodos em pacientes que utilizaram opióides quando comparados às pacientes que não utilizaram opióides no seguimento clínico (p=0,011); 2. foi evidenciado que pacientes do subtipo molecular Luminal A apresentam um aumento significativo de chances de apresentar 51-75% de marcação de morte celular tecidual (TUNEL) no tumor quando comparadas as pacientes definidas como subtipo Luminal B (p=0.020; OR=21.000; IC=1.613-273.340); 3. foi possível evidenciar que as pacientes com estadiamento do tipo T4 que apresentaram recidiva (7,2±3,7) demonstraram uma diminuição de expressão do histoscore do κ Opióide, na localização linfonodal, quando relacionado às pacientes que não apresentaram evolução clínica associada à recidiva (10,8±2,2) (p=0.033) e 4. as análises de correlação entre os marcadores μ e κ opióides, VEGF, Ki-67 e TUNEL demonstram que estes achados, provavelmente, atuam em uma mesma cascata de eventos no CA de mama com estadiamento T4. O presente estudo demonstrou que o uso de opioides está diretamente associado ao aumento da expressão de Ki-67 e da prevalência de não recidivar, as quais mostraram menor expressão para o receptor kappa opioide. Adicionalmente, não houve diferença significante entre os tumores luminal A e luminal B e a imunoexpressão dos receptores opioides foram independentes dos receptores de angiogênese. Em síntese, nos tumores luminal A e luminal B o uso de opioides está diretamente associado a um pior prognóstico, por redução da expressão do receptor kappa opioide.

O papel da autofagia na resistência de gliomas ao tratamento com temozolomida e inibidor de histonas desacetilases

Glioblastoma multiforme é o tipo mais frequente de tumor cerebral primário, sendo caracterizado por uma alta agressividade e um prognóstico bastante limitado. O ácido hidroxâmico suberoilanilida (SAHA) é um inibidor específico de histonas desacetilases aprovado para o tratamento de linfoma cutâneo de células T, e em fase de crescente investigação clínica e pré-clínica em tumores sólidos. Neste estudo, avaliamos a eficácia do ácido hidroxâmico suberoilanilida em tratamento combinado com temozolomida, o agente alquilante já utilizado em glioblastomas. Através de testes de viabilidade e analises por citometria de fluxo em células tumorais das linhagens U251MG e C6, observamos que não houve sinergismo de potenciação entre temozolomida e ácido hidroxâmico suberoilanilida, apenas efeito sinergismo de adição O tratamento combinado inicialmente promoveu parada do ciclo celular em fase G2/M (≥48 h) ao passo que a apoptose foi detectada apenas em exposição prolongada (≥96 h) aos fármacos em estudo. Ainda, as células tratadas com TMZ/SAHA apresentaram fenótipo autofágico, como determinado por citometria de fluxo e imunodetecção de proteínas marcadoras de autofagia como LC3 e o p62/SQSTM1. A autofagia temporalmente precedeu a apoptose e exerceu função citoprotetora, uma vez que o bloqueio da terminação autofágica com cloroquina promoveu uma significante redução na viabilidade celular, a qual foi associada a um aumento de apoptose em células de glioma tratadas com TMZ/SAHA. Portanto, os dados apresentados neste trabalho demonstram que a autofagia é um processo que diminui a eficácia antiglioma do temozolomida e do ácido hidroxâmico suberoilanilida, e a inibição deste fenômeno pode ser uma estratégia para aperfeiçoar a terapia com esses fármacos. / Glioblastoma multiforme (GBM) is the most frequent and aggressive type of primary brain tumor which has been associated with a dismal prognosis. In this study, we tested the efficacy of combining temozolomide (TMZ) with suberoylanilide hydroxamic acid (SAHA) - an inhibitor of HDACs 1, 2, 3, and 6 approved for the treatment of cutaneous T-cell lymphoma - in the viability of tumor cells. The data showed that potentiation synergism between TMZ e SAHA was not achieved due to activation of protective autophagy in vitro. The SAHA/TMZ treatment promoted arrest in the G2/M phase of the cell cycle as soon as 48 h after drug exposure whereas apoptosis was only detected after long-lasting exposure (≥96 h). In addition, SAHA and TMZ induced autophagy as detected by flow cytometry of acridine orange stained cells and immunodetection of the lipidated form of LC3 as well as decreases in p62/SQSTM1. Autophagy preceded apoptosis, and by blocking the termination step of autophagy with chloroquine promoted a significant reduction in the viability of glioma cells which was accompanied by increased apoptosis in SAHA/TMZ treatment. Overall, the herein presented data demonstrate that autophagy impairs the efficacy of combined TMZ/SAHA, and inhibiting this phenomenon could provide novel opportunities to improve the therapeutic potential of these compounds.
