Propriedades físicas de filmes à base de biopolímeros reforçados com laponita / Physical properties of films based in biopolymers reinforced with laponite

Os problemas ambientais provocados pelas embalagens à base de materiais sintéticos não biodegradáveis têm provocado um importante aumento nos estudos sobre filmes à base de biopolímeros. Entretanto, esses filmes têm limitações em suas propriedades, devido, sobretudo à sensibilidade a umidade relativa ambiente. Dentre as alternativas estudadas para melhorar as características desses materiais está o uso de nanopartículas, com destaque para a montmorilonita, que tem problemas de dispersão em água. Outra nanopartícula pouco usada em estudos à base de biopolímeros é a laponita, que é uma nanoargila sintética. Assim, o objetivo geral desta tese foi o desenvolvimento de filmes à base de biopolímeros (colágeno, gelatina e fécula de mandioca), reforçados com uma nanoargila (laponita). Foi estudado o efeito da concentração do biopolímero e da laponita, assim como o método de produção dos filmes (casting e espalhamento mecânico), além da qualidade da dispersão da nanopartícula, sobre as principais propriedades físicas dos filmes nanocompósitos, com especial interesse nas propriedades de superfície. Os filmes foram preparados pela desidratação de soluções formadoras de filmes (SFF), com 2, 4 ou 8 g de biopolímero/100 g SFF; 25 ou 30 g glicerol/100 g de biopolímero; e 0, 1,5; 3; 4,5 e 6 g laponita/100 g de biopolímero. A laponita foi dispersa em água destilada, utilizando-se ultraturrax com velocidade de agitação de 20.000 rpm, por 30 minutos. As partículas de laponita em água tiveram tamanhos menores que 50 nm. Não houve efeito da concentração do biopolímero, nem do método de produção (casting ou espalhamento mecânico) sob as propriedades de topografia superficial e físico-químicas estudadas nos filmes nanocompósitos. As análises de raios X e espectroscopia de infravermelho por transformada de Fourier revelaram que as plaquetas de laponita estiveram esfoliadas e/ou intercaladas nos filmes, e que não houve nenhuma formação de ligação química entre as plaquetas de laponita e os biopolímeros em estudo. A presença de laponita incrementou a irregularidade superficial dos filmes, especialmente naqueles produzidos com colágeno e fécula de mandioca. Outras propriedades dos filmes nanocompósitos, tais como densidade, umidade, cor, opacidade, propriedades térmicas, propriedades mecânicas, solubilidade em água, ângulo de contato à água, isotermas de sorção e permeabilidade ao vapor de água não sofreram alterações com a presença de laponita. / The environmental problems caused by packaging based on non-biodegradable synthetic materials have lead to a significant increase in studies about biopolymer films. However, these films have limited physicochemical properties due mainly to its sensitivity to ambient relative humidity. Among the alternatives studied to improve the physicochemical properties of these materials is the use of nanoparticles, especially the montmorillonite, which has problems of dispersion in water. Another nanoparticle no so much studied in films based on biopolymers is laponite, which is a synthetic nanoparticle. Thus, this these aims to development and characterize films based on biopolymers (collagen, gelatin and cassava starch), with a nanoparticle (laponite). The effects of biopolymer and laponite concentrations were studied, as well as, the film production method (casting and spreading), besides the quality of laponite dispersion and its relationship with the physicochemical properties of the films were investigated, with special interest on the surface properties. The films were produced by the dehydration of filmogenic-forming solutions (FFS), with 2, 4 or 8 g of biopolymer/100 g FFS; 25 or 30 g glycerol/100g of biopolymer; and 0, 1.5, 3, 4.5 and 6 g of laponite/100g of biopolymer. The laponite was dispersed in water using ultraturrax, at 20,000 rpm, for 30 minutes. The laponite particles in water had sizes smaller than 50 nm. There was not effect of biopolymer concentration and film production method (casting or spreading) on the surface and physicochemical properties studied in the nanocomposite films. X-ray analysis and Fourier transform infrared spectroscopy revealed that laponite platelets were exfoliated and/or intercalated in the films, and that there were no formed chemical bonds between laponite platelets and the biopolymers studied. The presence of laponite increased the surface irregularity of the films, especially in those produced with collagen and cassava starch. Other properties in the nanocomposite films, such density, moisture content, color, opacity, thermal properties, mechanical properties, water solubility, water contact angle, sorption isotherms and water vapor permeability were not altered by the presence of laponite.

Fast-paced lifestyles result in consumers replacing traditional meals with on-the-go snack foods. In general, snacks are higher in saturated fats and simple sugars, and pose health concerns for consumers, which prompts the need for healthy nutritious alternatives to common snacks. Proso millet is a nutritious, and fast growing gluten free cereal. Spent grain (SG), the main by-product of brewing and distilling, contains high amount of protein and insoluble fibers. This study utilized proso millet and spent grain in the production of an extruded expanded snack, demonstrating their appropriateness as an ingredient in food production. The first objective of this study was to determine the effect of extrusion processing conditions (moisture, barrel temperature, screw speed) on the physical, chemical, and functional properties of proso millet based extruded snack. The second objective of this research was to determine the effect of distiller’s spent grain (DSG) addition level and particle size on physicochemical and functional properties of extruded snacks. The third objective of this research was to evaluate the effect of spent grain type and particle size on the physicochemical and functional properties of extruded snacks. Samples were produced using a co-rotating twin-screw 25:1 L/D laboratory scale extruder.

Nous proposons une approche pour ajouter des propriétés non-fonctionnelles (traitement d'exceptions, atomicité, sécurité, persistance) à des coordinations de services. L'approche est basée sur un Modèle de Politiques Actives (AP Model) pour représenter les coordinations de services avec des propriétés non-fonctionnelles comme une collection de types. Dans notre modèle, une coordination de services est représentée comme un workflow compose d'un ensemble ordonné d'activité. Chaque activité est en charge d'implante un appel à l'opération d'un service. Nous utilisons le type Activité pour représenter le workflow et ses composants (c-à-d, les activités du workflow et l'ordre entre eux). Une propriété non-fonctionnelle est représentée comme un ou plusieurs types de politiques actives, chaque politique est compose d'un ensemble de règles événement-condition-action qui implantent un aspect d'un propriété. Les instances des entités du modèle, politique active et activité peuvent être exécutées. Nous utilisons le type unité d'exécution pour les représenter comme des entités dont l'exécution passe par des différents états d'exécution en exécution. Lorsqu'une politique active est associée à une ou plusieurs unités d'exécution, les règles vérifient si l'unité d'exécution respecte la propriété non-fonctionnelle implantée en évaluant leurs conditions sur leurs états d'exécution. Lorsqu'une propriété n'est pas vérifiée, les règles exécutant leurs actions pour renforcer les propriétés en cours d'exécution. Nous avons aussi proposé un Moteur d'exécution de politiques actives pour exécuter un workflow orientés politiques actives modélisé en utilisant notre AP Model. Le moteur implante un modèle d'exécution qui détermine comment les instances d'une AP, une règle et une activité interagissent entre elles pour ajouter des propriétés non-fonctionnelles (NFP) à un workflow en cours d'exécution. Nous avons validé le modèle AP et le moteur d'exécution de politiques actives en définissant des types de politiques actives pour adresser le traitement d'exceptions, l'atomicité, le traitement d'état, la persistance et l'authentification. Ces types de politiques actives ont été utilisés pour implanter des applications à base de services fiables, et pour intégrer les données fournies par des services à travers des mashups.

From Models to Code and Back : A Round-trip Approach for Model-driven Engineering of Embedded Systems

The complexity of modern systems is continuously growing, thus demanding novel powerful development approaches.In this direction, model-driven and component-based software engineering have reached the status of promising paradigms for the development of complex systems. Moreover, in the embedded domain, their combination is believed to be helpful in handling the ever-increasing complexity of such systems.However, in order for them and their combination to definitively break through at industrial level, code generated from models through model transformations should preserve system properties modelled at design level. This research work focuses on aiding the preservation of system properties throughout the entire development process across different abstraction levels. Towards this goal, we provide the possibility of analysing and preserving system properties through a development chain constituted of three steps: (i) generation of code from system models, (ii) execution and analysis of generated code, and (iii) back-propagation of analysis results to system models.With the introduction of steps (ii) and (iii), properties that are hard to predict at modelling level are compared with runtime values and this consequently allows the developer to work exclusively at modelling level thus focusing on optimising system models with the help of those values. / Denna doktorsavhandling presenterar nya och förbättrade tekniker för modelldriven och komponentbaserad utveckling av programvara. Syftet är att bevara systemegenskaper, som specificerats i modeller, genom de olika stadierna av utvecklingen och när modeller översätts mellan olika abstraktionsnivåer och till kod. Vi introducerar möjligheter att studera och bevara systemets egenskaper genom att skapa en kedja i tre steg som: (i) genererar kod från systemmodellen, (ii) exekverar och analyserar den genererade koden och (iii) slutligen återkopplar analysvärden till systemmodellen. Introduktionen av steg (ii) och (iii) gör det möjligt att genomföra en detaljerad analys av egenskaper som är svåra, eller till och med omöjliga, att studera med hjälp av endast systemmodeller. Fördelen med det här tillvägagångssättet är att det förenklar för utvecklaren som slipper arbeta direkt med kod för att ändra systemegenskaper. Istället kan utvecklaren arbeta helt och hållet med modeller och fokusera på optimering av systemmodeller med hjälp av analysvärden från testkörningar av systemet. Vi är övertygade om att denna typ av teknik är nödvändig att utveckla för att stödja modelldriven utveckling av programvara eftersom dagens tekniker inte möjliggör för systemutvecklare att specificera, analysera och optimera systemegenskaper på modellnivå. / La continua crescita in complessitá dei sistemi software moderni porta alla necessitá di definire nuovi e piú efficaci approcci di sviluppo. In questa direzione, metodi basati su modelli (model-driven engineering) e componenti (component-based software engineering) sono stati riconosciuti come promettenti nuove alternative per lo sviluppo di sistemi complessi. Inoltre l'interazione tra loro é ritenuta particolarmente vantaggiosa nella gestione nello sviluppo di sistemi integrati. Affinché questi approcci, cosí come la loro interazione, possano definitivamente prendere piede in campo industriale, il codice generato dai modelli tramite apposite transformazioni deve essere in grado di preservare le proprietá di sistema, sia funzionali che extra-funzionali, definite nei modelli. Il lavoro di ricerca presentato in questa tesi di dottorato si focalizza sul preservamento delle proprietá di sistema nell'intero processo di sviluppo e attraverso i diversi livelli di astrazione. Il risultato principale é rappresentato da un approccio automatico di round-trip engineering in grado di sostenere il preservamento delle proprietá di sistema attraverso: 1) generazione automatica di codice, 2) monitoraggio e analisi dell'esecuzione del codice generate su piattaforme specifiche, e 3) offrendo la possibilitá di propagare verticalmente i risultati da runtime al livello di modellazione. In questo modo, quelle proprietá che possono essere stimate staticamente solo in maniera approssimativa, vengono valutate in rapporto ai valori ottenuti a runtime. Ció permette di ottimizzare il sistema a livello di design attraverso i modelli, piuttosto che manualmente a livello di codice, per assicurare il preservamento degli proprietá di sistema d'interesse.

Protein functionality in turkey meat

Turkey with pale, soft, exudative (PSE)-like condition is one of the growing concerns in the poultry industry as it affects meat quality due to low ultimate pH at 24 h post mortem (pH24). Hence, there is a need for better utilization of PSE-like meat for the preparation of further processed products. In the first two studies, the biochemical, functional, rheological, and textural properties of proteins in turkey breast meat with different pH24 in fresh and frozen conditions were investigated. These studies revealed that low and normal pH meat had similar properties indicating similar extent of protein denaturation, except for lower water holding capacity (WHC) in low pH meat. High pH meat had similar or better functional properties than normal pH meat. In the third study, improvements in WHC, protein solubility, and gel forming ability of low pH meat was achieved by the application of high pressure processing (HPP). / Food Science and Technology

The search for new sources starch is growing, causing the production sector is seeking new technologies, as well as natural starches with different chemical and physical characteristics. The objective of this study was to extract and characterize starch from jackfruit seeds, varieties of soft and hard, as their chemical, structural and morphological, thermo-economic and technological. In proxi-mate analysis of starch extracted from jackfruit seeds had a content of 92.81% to 94.49% range for soft and hard variety, thus verifying a high purity in the extraction process. The ob-servation in scanning electron microscopy and optical microscopy showed starch granules with rounded shape, the bell-shaped and some go-presenting regular cuts in its surface, diam-eters ranging from 6 to 11μm-jackfruit soft and lasts for 6-13 μm. The pattern of crystallinity shown by X-ray diffraction for the two varieties was type A. The swelling power and solubili-ty of starches studied increased with increasing temperature, being significantly above the temperature of 75 ˚ C (gelatinisation). Absorption capacity for water and oil, starch of the present study showed a high absorption, and the soft jackfruit obtained the highest values, 123.18 g 100g-1 and 99.58 g 100g-1. The starch gel showed low transmittance values, with folders for the kid-opaque. The maximum viscosity of starch from jackfruit seed of the varie-ty found hard was higher for the variety of soft starch, and starch of the variety you lasts less resistant to mechanical agitation at high temperature (higher breakdown). In Differential Scanning Calorimetry (DSC) showed an initial gelatinization temperature of 36 ˚C and a final temperature of 65 ˚ C for soft jackfruit and jackfruit was last of 40 ˚C and 70 ˚C respectively. / O amido é um polissacarídeo utilizado como fonte de carboidratos na dieta humana, com grande aplicação na indústria de alimentos, têxtil e farmacêutica. A procura por novas fontes amilacéas vem crescendo, fazendo com que o setor produtivo esteja em busca de novas tecno-logias, bem como de amidos naturais com características físico-quimicas diferenciadas. O objetivo deste trabalho foi extrair e caracterizar o amido de semente de jaca, das variedades mole e dura, quanto a sua composição química, estrutural e morfológica, propriedades térmi-cas e tecnológicas. Na analise centesimal o amido extraído da semente de jaca teve um teor de 92,81% para variedade mole e 94,49% para variedade dura, verificando assim uma alta pureza no processo de extração. A observação em microscopia eletrônica de varredura e microscopia óptica mostrou grânulos de amidos com formato arredondados, na forma de sino e alguns ir-regulares apresentando cortes na sua superfície, diâmetros variando de 6-11μm para jaca mole e 6-13 μm para dura. O padrão de cristalinidade mostrado por difratometria de raios-X para as duas variedades foi do tipo A. O poder de intumescimento e solubilidade dos amidos estuda-dos aumentou com o aumento da temperatura, sendo significativo acima da temperatura de 75˚C (gelatinização). Para capacidade de absorção de água e de óleo, o amido do presente estudo mostrou uma alta absorção, sendo que a jaca mole obteve os maiores valores, 123,18 g·100 g-1 e 99,58 g·100 g-1. O gel do amido mostrou valores de transmitância baixos, com pastas de amido opacas. A viscosidade máxima do amido de semente de jaca variedade dura foi maior à encontrada para o amido da variedade mole, sendo o amido da variedade dura me-nos resistente à agitação mecânica a quente (maior quebra). Na Calorimetria Exploratória Di-ferencial (DSC) verificou uma temperatura inicial de gelatinização de 36˚C e uma temperatu-ra final de 65˚C para jaca mole e para jaca dura foi de 40˚C e 70˚C respectivamente.

As condições para o preparo do IP foram definidas a partir da curva de solubilidade em água x pH e temperatura de 25 l 3 ºC. Tais condições permitiram obter isolados protéicos com rendimento de extração de 45,2 l 0,5% (pH10,0) e 66,2 l 0,5% (pH11,5) e elevado conteúdo protéico 96,4 l 0,5% e 93,5 l 0,4% respectivamente. A capacidade de absorção de água e óleo foram baixas, apresentando 1,05 l 0,07 e 2,3 l 0,01 mL/g proteína respectivamente para IP 10,0 e 1,65 l 0,07 e 1,70 l 0,07 mL/g pr oteína respectivamente para IP 11,5. A maior capacidade de emulsificação, foi observado para o IP 11,5, 140 l 8 g óleo/g prot., comparado com o IP 10,0, 37 l 2 g óleo/g prot. A formação de gel foi observada em pH neutro e ausência de sal, apresentando as concentrações de 8% para IP 10,0 e 10% para IP 11,5. A cromatografia revelou a presença de dois picos para ambos isolados com sete frações de proteínas de diferentes pesos moleculares. As curvas TG-DTG / DSC revelaram maior quantidade de água para o IP 10,0 e elevada temperatura de estabilidade térmica 200 oC para ambos isolados. / From the guava seed flour (Psidium guajava), whose centesimal composition belongs to about 6,17 ??0,04% of moisture, 8,43 ??0,12% of protein and high content of fibers, 60,88 ??0,9%, the protein isolate (PI) was obtained through the isoelectric precipitation (Ip 4,5) whose majority belongs to glutelins class proteins. The chromatography shows the presence of two peaks for both protein isolated with seven fractions of proteins with different molecular weights. The curves of TG-DTG / DSC revealed high water quantity for PI 10,0 and high temperature for thermal stability 200 °C for both isolates.

CoordenaÃÃo de AperfeiÃoamento de Pessoal de NÃvel Superior / Amburana cearensis (cumaru) Ã uma leguminosa subutilizada da Caatinga. O alto teor proteico das sementes faz delas uma possÃvel nova fonte de proteÃnas alimentares. Foram determinadas as melhores condiÃÃes de extraÃÃo proteica para posterior produÃÃo do isolado proteico de cumaru (IPAc). As anÃlises tambÃm foram conduzidas com a farinha delipidada (FDAc) e com um isolado proteico de soja comercial (IPS). IPAc apresentou teor proteico > 90% e composiÃÃo de aminoÃcidos comparÃvel a IPS, com notÃvel deficiÃncia em metionina. Foram determinadas algumas propriedades funcionais para predizer sua adequaÃÃo como ingrediente na indÃstria alimentÃcia. Os resultados para FDAc, IPAc e IPS foram, respectivamente: solubilidade 20-79; 9-99 e 3-76%; capacidade de retenÃÃo de Ãgua 1,7; 3,9 e 7,5 g/g; capacidade de retenÃÃo de Ãleo 1,5; 6,3 e 1,6 g/g; capacidade emulsificante / estabilidade 52 / 46%; 55 / 51% e 53 / 21%; capacidade espumante / estabilidade 47 / 32%; 65 / 53% e 33 / 8 e menor concentraÃÃo geleificante 18, 16 e 12%. Para investigar se IPAc e IPS apresentam indÃcios de toxicidade, cÃlulas de adenocarcinoma humano foram expostas por 24 h aos isolados. Para comparaÃÃo, tambÃm foi realizada exposiÃÃo com os isolados de soja (IPGm), feijÃo comum (IPPv), feijÃo de porco (IPCe) e mamona (IPRc), a fraÃÃo proteica de mamona (FPRc), as lectinas PHA-E e ConA e o controle quÃmico (Crtl_Quim). O RNA das cÃlulas foi hibridizado em microarranjos contendo o genoma humano completo. Os perfis de expressÃo gÃnica apÃs exposiÃÃo Ãs proteÃnas foram comparados aos de PBS e Crtl_Quim e agrupados hierarquicamente. Crtl_Quim, IPAc e IPGm se mostraram inadequados para a continuaÃÃo das anÃlises por provocar um efeito superestimado. A anÃlise de vias e processos biolÃgicos afetados apontou que genes envolvidos com a biossÃntese de colesterol, estresse do RE e resposta imune foram superexpressos e genes envolvidos com o ciclo celular e replicaÃÃo do DNA foram subexpressos apÃs exposiÃÃo a IPPv, IPCe, PHA-E e ConA. As amostras IPRc e FPRc regularam para cima genes envolvidos com a apoptose e resposta imune e para baixo genes envolvidos com o ciclo celular e sinalizaÃÃo do cAMP. AnÃlise do mapa conectivo mostrou alta correlaÃÃo de ConA, PHA-E e IPCe com drogas bloqueadoras de canais de Ca2+ e de K+; enquanto IPRc e FPRc apresentam funÃÃo biolÃgica semelhante a pelo menos seis drogas inibitÃrias da sÃntese proteica. Conclui-se que IPAc tem grande potencialidade como novo alimento proteico com grande aplicabilidade na indÃstria de alimentos, devendo, no entanto, sofrer modificaÃÃes no mÃtodo de obtenÃÃo visando a melhora na solubilidade e a eliminaÃÃo do sal residual. / Amburana cearensis (cumaru) is an underutilized legume from Caatinga. Because of their high protein content its seeds are a possible novel source of food protein. Were determined the best conditions for protein extraction and preparation of protein isolate (IPAc).Analysis were carried out also with the defatted flour (FDAc) and with a marketed soybean protein isolate (IPS). IPAc showed protein content of 90% and amino acid composition compatible with the IPSâs composition, with remarkable low methionine content. Some functional properties were measured in order to predict the adequacy of the isolates as an ingredient for food industry. The results for FDAc, IPAc and IPS were, respectively: solubility 20-79; 9-99 e 3-76%; water holding capacity 1.7; 3.9 e 7.5 g/g; oil holding capacity 1.5; 6.3 e 1.6 g/g; emulsion formation / stability 52 / 46%; 55 / 51% e 53 / 21%; foamability / stability 47 / 32%; 65 / 53% e 33 / 8; least gelling concentration (LGC) 18, 16 and 12%. In order to investigate whether IPAc e IPS exhibit signs of toxicity, two cell lines of human adenocarcinoma (MCF-7 e Caco-2) were exposed to these isolates as well as to others prepared from soyabean (IPGm), white bean (IPPv), Jack bean (IPCe) and castor bean (IPRc), to the protein fraction from castor bean (FPRc), to the lectins PHA-E and ConA (50 Âg/mL) and to the chemical control (Crtl_Quim). Subcitotoxic doses were determined by viability test to later exposure assay for 24 h. The MCF-7 cellâs RNA were extracted and hybridized to a microarray containing the complete human genome. The expression profiles after the exposition to the proteins were compared to that ones generated after PBS and Crtl_Quim and hierarchicaly clustered. Crtl_Quim, IPAc and IPGm were unsuitable for further analysis by their overestimated effect. Analysis of biological pathways and process showed that genes involved with the cholesterol biosynthesis, ER stress and immune response were upregulated and genes involved with cell cycle and DNA replication were downregulated after exposure of cells to IPPv, IPCe, PHA-E and ConA. The samples IPRc and FPRc upregulated genes involved with apoptosis and immune response and downregulated genes involved with cell cycle and cAMP signaling. Connectivity Map analysis showed high correlation of ConA, PHA-E and IPCe with drugs that are Ca2+ and K+ channel blockers, while IPRc and FPRc showed biological function similar to at least six drugs inhibitors of protein synthesis. Itâs possible to conclude that the IPAc has a high potentiality as a novel protein food with wide applicability in the food industry and should, however, be modified in the method of obtaining aiming the improvement in its solubility and eliminating the residual salt.
