Mecanismos moleculares da ação dos glicocorticóides endógenos e da anexina-A1 sobre o tráfego de neutrófilos: caracterização da ação sobre os  eixos SDF-1α/CXCR4 e IL-17/IL-23/G-CSF / Molecular mechanisms of endogenous glucocorticoid and annexin-a1 actions on neutrophil traffic: characterization of this action on the SDF-1α/CXCR4 e IL-17/IL23/G-CSF axis

Isabel Daufenback Machado 17 December 2013 (has links)
The traffic leukocytes is a complex process dependent on the action of severals chemical mediators, in addition to perfect cell interaction. Therefore, this study aimed to evaluate the effect of GCe and ANXA1 on SDF-1α/CXCR4 and IL-17/IL-23/G-CSF and on the expression of adhesion molecules CD18, CD49d and CD62L. Balb/C wild type and ANXA1-/- male mice were employed. The analysis were performed at physiological conditions, in the presence of high concentrations of GCe and during of inflammatory process induced by ACTH administration (5 µg/animal, i.p.) or LPS injection (100 µg/kg, i.p.), respectively or in the absence of GCe action, by the action of RU 38486 (RU, 10 mg/kg , i.p.). The involvement of the receptor FPR2 and ANXA1 was assessed by pre-treatment with Ac2-26 (1 mg/kg, i.p.) or BOC2 (10 µg/animal, i.p.) for 4 days, once a day. The quantification of total and differential cell was performed in a Neubauer chamber and stained smears by May-Grunwald and flow cytometry. Quantification of expression of CXCR2, CXCR4, FPR2, CD18, CD49d, CD62L and granulocytic maturation (CD11b/Ly6G) in the bone marrow and circulation were performed by flow cytometry. The expression of ANXA1 on tissues was performed by western blotting and on cells from bone marrow and blood by immunocytochemistry. Quantification of IL-17, IL-23, G-CSF, SDF-1α and corticosterone were performed by ELISA. The chemotaxis of neutrophils from the bone marrow and blood was tested in the chemotaxis chamber with filter pore of 8 microns. The phagocytosis of apoptotic neutrophils by bone marrow macrophages was assessed by in vitro assay. To investigate the effects of ACTH in the migration of neutrophils in the inflammatory process, the model employed was air pouch (100 µg/ ml, LPS), and the behavior of circulating leukocytes from animals treated with ACTH were evaluated by intravital microscopy. The results obtained, which are presented in three sections, showed that: 1) neutrophils from the bone marrow and blood expressed ANXA1 in the cytoplasm and membrane, as well as FPR2, constitutively and the expression of both is regulated by GCe. The ANXA1 via FPR2 receptor expressed in bone marrow cells, controls the neutrophilic maturation and traffic of these cells from the bone marrow into the blood. The ANXA1 via interaction to FPR2 controls the clearance of neutrophils from the blood to the bone marrow by modulating the SDF-1α/CXCR4 axis; 2) the administration of ACTH induces neutrophilia and the circulating neutrophils are ANXA1+, CD18+, CD49d+ and CD62L+, showing that the injection of ACTH in vivo alters the phenotype of these cells in the blood. These modifications alter the behavior of neutrophils in the blood, as well as the migration to the air pouch in the presence of inflammation and to the tissue clearance, and these effects may be dependent, at least in part, on inhibition of migration oriented events, as chemotaxis in response to fMLP or SDF-1α were reduced. Further, the clearance of neutrophils is reduced in animals treated with ACTH due to the lower phagocytic and secretory activity of medullary macrophages; 3) Animals treated with RU 38486 and ANXA1-/- mobilize granulocytes from bone marrow into the blood, and from this compartment to the focus of inflammation with higher intensity than that observed in the control group. The axis IL-17/IL-23/G-CSF seems to be involved in granulopoiesis and mobilization of neutrophils into the blood during inflammation, but it is not the target of action of ANXA1 and GCe at this step of inflammatory process. Additionally, it was observed that in the presence of peritonitis, the adhesion molecules, CD49d and CD62L are involved in the migration of neutrophils from the bone marrow into the blood. The results obtained allow concluding that the GCe and ANXA1 are relevant to the granulopoiesis and the traffic of neutrophils from bone marrow under physiological conditions and in the presence of inflammation. Furthermore, together with literature data, the data presented here may suggest the involvement of ANXA1 the GCe in phenotypic plasticity of neutrophils according to the stimuli that are submitted, and may support to understand the new concepts of production, half-life, location and function of neutrophils.

Modélisation des systèmes éoliens verticaux intégrés aux bâtiments : modélisation du couple production / Bâtiment / Modeling of vertical axis wind systems integrated into buildings : modeling of coupling between Production / Buildings

Jaohindy, Placide 20 August 2012 (has links)
The building integration of the vertical axis wind turbine (VAWT) to supply the individual, collective and tertiary residences consumption is an interesting approach that can help architects and the actors of the energy control to promote a rational use of renewable energy in the in homes. The choice of the location of the urban wind turbine type is determined by building height, wind speed and turbulence intensity of the site. The severe conditions of wind at low altitude are favorable for a VAWT installation. In some cities, the average buildings height is low, in these places, the VAWTs must be appreciable compared to the HAWTs. The modelling of the air flow through the wind turbine and the couple building-wind turbine involves the computation fluid dynamics (CFD). A problem modeled with a suitable turbulence model will give results that approach the physical reality and the experiment results. In this study, the standard k-" and SST k-! models were used. After analyzing the possibilities of VAWT integration, the roof is the most interesting integration area. In addition to CFD method, we have started to study the electrical model of the VAWT. The work was conducted to determine the electrical power generated by the wind turbine using Matlab/Simulink software. To complete the study, a VAWT model coupled with a building where the building is considered as a consumption model is presented.

Constellation Optimization using Genetic Algorithm : Combining SAR & Optical Sensors with AI Requirements / Konstellationsoptimering med hjälp av genetiska algoritmer : Med kombinering av SAR- och optiska sensorer med AI-krav

Pellnäs, Adrian January 2023 (has links)
With increasing world tensions and improvements of satellites and their sensors, the interest and possibility of using space and satellites for defensive purposes has increased greatly. However, not much research has been conducted into the needs and possibilities of satellite constellations over Sweden, especially using SAR and optical sensors combined with AI object detection. This thesis provides insight in to the needs and requirements to achieve certain coverage and gap times and explores different constellation design methods to do so. This is done by combining large scale tests performed with genetic algorithm and a dual-axis propagator with theoretical and analytical methods. Results show that for micro-satellites under 100 kg based on current commercial technology, it is found that between 24 to 63 satellites are needed for 1 hour gap times depending on what combination of SAR and optical satellites are used. The genetic algorithm was found to not generate optimal constellations as the number of satellites increased beyond 12. It was however useful in mapping out possibilities and finding certain optimal parameters such as the inclination. The dual-axis propagator tested for its low processing load was found to be good for coverage analysis and estimating the shapes of the orbits. It was noted to have large positional errors however, limiting its use to analysis and not full constellation design.

Personality and the HPA-axis in Association with Postpartum Depression

Iliadis, Stavros I January 2016 (has links)
Postpartum depression is a psychiatric disorder affecting a substantial proportion of newly delivered women, and remains a significant cause of childbirth-related morbidity. The aim of the present thesis was to examine psychological, endocrine and genetic aspects of postpartum depression in a large, population-based sample of women in Uppsala, Sweden. All included studies were undertaken as parts of the BASIC-project, a longitudinal study on psychological wellbeing during pregnancy and the postpartum period. Study participants were screened for depressive symptoms in pregnancy week 17 and 32 as well as at six weeks and six months postpartum, mainly by use of the Swedish version of the Edinburgh Postnatal Depression Scale (EPDS). Furthermore, personality was assessed with the Swedish universities Scale of Personality (SSP) in pregnancy week 32. Evening cortisol levels in saliva were measured in pregnancy week 36 and at six weeks postpartum. Blood samples were obtained to measure corticotropin-releasing hormone levels (CRH) and to perform genetic analyses. The results of this thesis demonstrate that neuroticism is a strong and independent predictive factor of depressive symptoms at six weeks and six months postpartum, and has a significant mediatory role in the association between a single nucleotide polymorphism in the hydroxysteroid (11-beta) dehydrogenase 1 gene (HSD11B1) and postpartum depression. Furthermore, women with postpartum depressive symptoms present with a dysregulated hypothalamic-pituitary-adrenal axis activity in terms of elevated cortisol levels postpartum, as well as elevated CRH levels in mid-gestation. In conclusion, this thesis develops current knowledge on several attributes of postpartum depression. Further studies are required to replicate and expand on these results, which would further contribute to early identification of women at risk of postpartum depression and adoption of proper interventions that may moderate the short- and long-term consequences of the disorder.

Consistent Automation Solutions for Electrohydraulic Drives in Times of Industry 4.0

Köckemann, Albert, Birke, Benno 02 May 2016 (has links) (PDF)
Electrohydraulic drives are primarily used whenever a low power/weight ratio, a compact build and/or large forces are required for individual applications. These drives are often used together with electric drive technology in machines. However, in terms of automation, unlike electric drives, electrohydraulic drives are still largely connected via analog interfaces and centralized closed control loops today. To compensate for this competitive disadvantage of hydraulic drive technology and, at the same time, significantly enhance its performance and diagnostics capability, a consistent automation solution has been developed that can be configured for both centralized and decentralized solutions. This contribution firstly gives an overview over this complete solution already available and its classification in the automation world. In a second step, the subset of decentralized drive solutions contained therein is presented in more detail and their benefits are explained on the basis of some exemplary applications.

Design of an aerodynamic attitude control system for a CubeSat

Auret, Jacoba 03 1900 (has links)
Thesis (MscEng)--Stellenbosch University, 2012. / ENGLISH ABSTRACT: The Cape Peninsula University of Technology, in collaboration with Stellenbosch University, is developing a 3-unit CubeSat for a low earth polar orbit. The two main payloads are a camera and a radio frequency beacon. This beacon will be used to calibrate the radar antenna patterns of an antenna of the Hermanus Magnetic Observatory at their base in Antarctica. This thesis describes the development of an aerodynamic attitude determination and control system needed to achieve three-axis stabilisation of the satellite and to perform accurate pointing of the camera. The satellite structure is designed to utilise aerodynamic means of control. It includes four feather antennae for passive pitch-yaw stabilisation and two active aerodynamic roll control paddles. The sensors used are a three-axis magnetometer, ne sun sensor and nadir sensor. Three attitude determination methods are investigated, namely the Triad, Rate Kalman Filter and Extended Kalman Filter algorithm. Apart from the aerodynamic control elements of the satellite, three magnetic torque rods and three nano-reaction wheels are also included in the design. Three control modes for the satellite are identi ed and various control methods are investigated for these control modes. The various attitude determination and control methods are evaluated through simulations and the results are compared to determine the nal methods to be used by the satellite. The magnetic Rate Kalman Filter is chosen as attitude determination method to be used when the satellite is tumbling and a combination of the sun Rate Kalman Filter and the Triad algorithm is to be used when the satellite experiences low angular rates. The B-dot and Y-spin controller is chosen for the detumbling control mode, the aerodynamic and cross-product control method for the three-axis stabilisation control mode and the quaternion feedback control method for the pointing control mode of the satellite. The combination of magnetic and aerodynamic control proved to be su cient for the initial stabilisation of the satellite, but the three nano-reaction wheels are required for the pointing control of the imaging process.

Studies of the regulation of serine protease activity in the establishment of the dorsal-ventral axis of the Drosophila embryo

Cho, Yong Suk, 1970- 05 October 2010 (has links)
Dorsal-ventral (DV) polarity in the Drosophila embryo is defined by spatially regulated activation of the transmembrane receptor Toll, which is uniformly distributed throughout the early embryo's plasma membrane. Ventral activation of Toll is accomplished through the local production of its activating ligand, a processed C-terminal fragment of the Spätzle protein, which is generated in the last step of a proteolytic cascade involving the sequentially-acting proteases Gastrulation Defective (GD), Snake and Easter. Pipe protein, a homologue of vertebrate glycosaminoglycan modifying enzymes, which is expressed during oogenesis in ventral follicle cells adjacent to the developing oocyte, is believed to control the ventrally restricted processing of Spätzle. pipe expression and the sulfation of its enzymatic target in the ventral follicle cells leads to the formation of a stable ventral cue, embedded in the eggshell. Recently the Pipe enzymatic target has been identified as several protein components of the vitelline membrane, the inner layer of the eggshell. Prior to this work, an important piece of information missing from our understanding of Drosophila DV patterning was the identity of the initial step in the protease cascade that requires Pipe activity. Here, I show that the processing of Snake is independent of Pipe activity, while the processing of Easter requires Pipe function, indicating that Easter processing by Snake is the key proteolytic step that is controlled by Pipe activity and presumably the first cleavage event that is spatially regulated. A second key gap in our understanding of Drosophila embryonic DV patterning concerned the role of GD in the protease cascade. While GD is the protease that cleaves and activates Snake, the existence of two distinct classes of complementing gd alleles has suggested that GD provides another, distinct function. Investigations described here indicate that the second function of GD is to promote the ability of activated Snake to process Easter, independent of its Snake-processing function. Finally, I provide evidence for the formation of protein complexes containing various components of the serine protease cascade, which suggest that conformational changes in the complexes, which act to promote productive interactions between the proteins, are an important aspect of their activation. / text

Discrete topology and geometry algorithms for quantitative human airway trees analysis based on computed tomography images

Postolski, Michal 18 December 2013 (has links) (PDF)
Computed tomography is a very useful technic which allow non-invasive diagnosis in many applications for example is used with success in industry and medicine. However, manual analysis of the interesting structures can be tedious and extremely time consuming, or even impossible due its complexity. Therefore in this thesis we study and develop discrete geometry and topology algorithms suitable for use in many practical applications, especially, in the problem of automatic quantitative analysis of the human airway trees based on computed tomography images. In the first part, we define basic notions used in discrete topology and geometry then we showed that several class of discrete methods like skeletonisation algorithms, medial axes, tunnels closing algorithms and tangent estimators, are widely used in several different practical application. The second part consist of a proposition and theory of a new methods for solving particular problems. We introduced two new medial axis filtering method. The hierarchical scale medial axis which is based on previously proposed scale axis transform, however, is free of drawbacks introduced in the previously proposed method and the discrete adaptive medial axis where the filtering parameter is dynamically adapted to the local size of the object. In this part we also introduced an efficient and parameter less new tangent estimators along three-dimensional discrete curves, called 3D maximal segment tangent direction. Finally, we showed that discrete geometry and topology algorithms can be useful in the problem of quantitative analysis of the human airway trees based on computed tomography images. According to proposed in the literature design of such system we applied discrete topology and geometry algorithms to solve particular problems at each step of the quantitative analysis process. First, we propose a robust method for segmenting airway tree from CT datasets. The method is based on the tunnel closing algorithm and is used as a tool to repair, damaged by acquisition errors, CT images. We also proposed an algorithm for creation of an artificial model of the bronchial tree and we used such model to validate algorithms presented in this work. Then, we compare the quality of different algorithms using set of experiments conducted on computer phantoms and real CT dataset. We show that recently proposed methods which works in cubical complex framework, together with methods introduced in this work can overcome problems reported in the literature and can be a good basis for the further implementation of the system for automatic quantification of bronchial tree properties

Gender differences in a prototypical analysis of psychopathy.

Hazelwood, Lisa L. 08 1900 (has links)
Psychopathy research has focused primarily on characteristics of male offenders. With little empirical knowledge on psychopathy in women, gender differences within psychopathy are not well understood. To gain a better understanding of these differences, the current study used prototypical analysis to compare ratings of 242 forensic experts when considering their most representative case of male or female psychopathy. The present study investigated gender differences for psychopathic traits and Cluster B personality disorder criteria. Most aspects of psychopathy were less prototypical of female psychopaths than males. In particular, the antisocial behavior facet does not appear to apply to males and females equally. The distinction between Cluster B disorders and psychopathy was more ambiguous in females than males; however, the affective deficits facet differentiated psychopathy from Cluster B disorders across genders. Current research also raises the question of potential diagnostic gender bias in the assessment of psychopathy.

Att förebygga stressrelaterad psykisk ohälsa med yoga : En systematisk litteraturstudie om yoga och stresshormonet kortisol / To prevent stress-related mental illness with yoga : A systematic literature study on yoga and the stress hormone cortisol

Nielsen, Annica January 2019 (has links)
Artiklarna söktes upp i PubMed samt Web of Science. / Background: Stress and stress-related mental illness is increasing in our society and it is important to find options to slow down the development. Stress is a normal physical condition but for a long time without recovery stress can lead to mental illness. Yoga studies have shown a stress-reducing effect on mood and yoga has become a popular option to reduce perceived stress symptoms. Therefore it can be interesting to study underlying mechanisms for perceived stress reduction. The hormone cortisol increases during stress. Perhaps yoga has a stress-reducing effect through decreasing the hormone. Aims: The aim of the study is to exam whether perceived stress reduction in yoga practice is related to lowered cortisol levels in healthy adults. Methods: A systematic literature study was carried out on articles published in 2009-2018. The articles were searched in PubMed and Web of Science. Results: Nine studies were reviewed (seven experimental and two observations studies). Six studies showed complete or partial reduction of cortisol levels. An experimental and two observation studies did not show significantly lower cortisol levels. Partially improved mood and cognitive ability were seen. Conclusions: Decreased cortisol levels in yoga practice were observed. However, there were studies that did not show decreased cortisol levels and the studies have shortcomings that lower the strength of evidence. Better studies are needed to establish a relationship.
