Modulation of Neurodevelopmental Outcomes using Lactobacillus in a Model of Maternal Microbiome Dysbiosis

Neurodevelopmental disorders, such as autism spectrum disorders, schizophrenia, and attention deficit hyperactivity disorder, are a heterogeneous set of developmental disorders affecting the central nervous system. Studies into their etiology remain challenging, as neurodevelopmental disorders frequently present with a wide range of biological, behavioral, and comorbid symptomologies. Increasing epidemiological reports of antibiotic use during pregnancy as a significant correlate of subsequent mental disorder diagnosis in children suggest a mechanism of influence via the maternal gut-fetal brain axis. Importantly, antibiotics cause dysbiosis of the gut microbiome and disrupt the delicate composition of the microbial inoculum transferred from mother to child, which is critical for development of the immune system and holds implications for long-term health outcomes. The research objective of this dissertation is to reveal a causal mechanism of maternal microbial influence on neurodevelopment by examining the brain's resident immune cells, microglia, and corresponding behavioral outcomes in a mouse model of antibiotics-driven maternal microbiome dysbiosis (MMD). We identify early gross motor deficits and social behavior impairments in offspring born to MMD dams, which paralleled hyperactivated microglia in brain regions specific to cognition and social reward. The MMD microglia also exhibited altered transcriptomic signatures reflective of premature cellular senescence that support evidence of impaired synaptic modeling found in MMD brains. We report that these deficits are rescued in the absence of Cx3cr1, a chemokine receptor expressed ubiquitously on microglia, to highlight a pathway in which maternal microbiota may signal to neonatal microglia to undergo appropriate neurodevelopmental actions. Finally, we characterize Lactobacillus murinus HU-1, a novel strain of an important gut bacterium found in native rodent microbiota, and demonstrate its use as a probiotic to restore microglial and behavioral dysfunction in MMD offspring. / Ph. D. / Population studies on neurodevelopmental disorders, such as autism spectrum disorders, schizophrenia, and attention deficit hyperactivity disorder, highlight antibiotic use during pregnancy as a major correlate of subsequent diagnoses in children. These findings support a growing body of evidence from animal and human studies that the microbial ecosystems (“microbiome”) found in and on our bodies play significant roles in mental health, including mood, cognition, and brain function. Importantly, antibiotics during pregnancy create an imbalance of the gut microbiome (“dysbiosis”) and disrupt the microbial inoculum transferred from mother to child, which is critical for maturation of the infant immune system and holds implications for long-term health outcomes. Thus, the research objective of this dissertation is to identify a mechanism of influence from the mother’s gut to the neonate’s brain by examining the brain’s resident immune cells (“microglia”) in a mouse model of antibiotics-driven maternal microbiome dysbiosis (MMD). We uncover autism-like behavioral deficits and dysfunctional microglia in MMD offspring, and characterize signaling cues specific to microglia by which improper neurodevelopment may be taking place. We also reveal that the detrimental effects of MMD are reversed in mice born to mothers pretreated with a probiotic candidate, Lactobacillus murinus HU-1, to suggest maternally-derived Lactobacillus may help to mediate proper neurodevelopment.

Strength and Stiffness of Weak-Axis Moment End-Plate Connections

Three full-scale experimental tests were conducted to investigate the strength and stiffness of weak-axis moment end-plate connections. Each test consisted of two girders connected to a column web with four-bolt extended moment end-plates. Two tests were conducted with bare steel. One test included a composite concrete slab that confined the top extension of the end-plate. Finite element models of the tests were created with the commercial software SAP2000. A simplified modeling procedure was developed to overcome the contact problems between the end-plates and column web, and between the bolts and holes in the end-plates and web. The simplified modeling procedure accurately predicted the experimental elastic stiffness, in the form of column web rotations, of the connections. Yield line theory was used to investigate the plastic strength of the column web. Several yield line patterns were examined. Analytical plastic moment strengths were very conservative when compared to the observed behavior of the column web. The experimental stiffness of the test with the concrete slab confining the top extension of the end-plate was compared to the stiffness of a similar test without a slab. The slab increased the elastic stiffness of the connection; however, after the concrete began cracking and crushing around the connection, the stiffness was greatly decreased. / Master of Science

Experimental Investigation on Heat Transfer and Pressure Loss Characteristics of Rotating Rectangular and Annular Ducts

In a gas turbine, a small portion of air is bled from the compressor to provide cooling to keep the turbine at a safe operating temperature. The air flows through several passages in between where the components of the turbine are assembled. In this study, the heat transfer and pressure loss characteristics of two of these passages are investigated experimentally. The first of the two passages investigated is the passage in between the turbine blade root and disc. This passage has a unique geometry resembling an S-shape. The heat transfer and pressure loss characteristic of this passages in not well documented. For this study, a model of the realistic S-shaped passage has been made. In addition, a simplified rectangular duct with hydraulic diameter similar to that of the realistic S-shaped passage was constructed along with three other rectangular passages at aspect ratios, 17.33, 8.81, 3.93, and 2.02. This study aims to determine if rectangular duct correlations are valid for the realistic S-shaped model. Specifically, flow in low Reynolds number ranges of less than 3000 are of interest. With the effect or rotation and aspect ratio being of primary concern in the study, an experimental rig was constructed to measure the heat transfer and pressure loss in these models. The experiments were conducted with both clockwise and counterclockwise rotation to account for the passage on the pressure side and suction side of the passage. The centerline Nusselt number distribution measured to check if the flow was fully developed. The effect of rotation caused swirling, increasing the entrance length in the duct and also enhanced heat transfer. The rotation also enhanced the heat transfer in the fully developed region. The fully developed experimental data for the simplified rectangular ducts showed good correlation with that of literature. However, the realistic S-shaped duct showed lower heat transfer values than the simplified rectangular ducts. Still, the effect of rotation is seen enhancing the rotation inf the realistic S-shaped duct. Additionally, the friction factor which was measured using the cross-sectional average static pressure showed similar results for the realistic S-shaped duct and the simplified rectangular duct. The passage between turbine disc bore and shaft is modeled as an annular duct with inner surface rotation. Flow in the turbulent region is studied and the test sections are made to have short length to hydraulic dimeter ratios. Along the centerline, the onset of Taylor vortices can be seen enhancing the Nusselt number in regions where the flow should be fully developed. This effect can also be seen enhancing the heat transfer in the fully developed region. The presence of Taylor vortices also adds resistance increasing the pressure loss across the duct. / Master of Science / Industrial gas turbines are designed to have an optimum overall pressure ratio for target temperatures rise. The demand for higher efficiency and power continues to push the operating pressure and temperature. Air systems is the flow network to provide necessary cooling to keep the machinery at a safe operating temperature. In this study, two passages of the air system in the turbine are of interest. The passage between turbine blade root and disc, and the passage between the turbine disc and shaft. The effect of rotation on the flow through the two passages are of primary interest with focus on heat transfer and pressure loss characteristics. This experimental study presents unique results as a realistic model of the passage which resembles an S-shape was constructed and tested. The passage in between the turbine disc and shaft forms a rotating annular passage. There is limited data available representing the realistic geometrical shape of the annular passage under rotation. Therefore, the present study aims to present data for more realistic geometry and operating conditions. In addition, simplified rectangular ducts and annular ducts are also tested for comparison purpose. The results of the study showed that the rotation does provide a significant increase in heat transfer and pressure loss in experiment modeling the passage between the turbine blade root and disc. Comparing the realistic S-shape passage and the rectangular passage with similar aspect ratio, the realistic S-shape passage showed less heat transfer and less sensitivity to the effect of rotation. The pressure loss characteristics on the other hand proved to be very similar. For the experiments modeling the passage between turbine disc and shaft, the effect of rotation once again showed to increase the heat transfer and pressure loss. The effect is more prominent when there is less axial flow.

It Takes T-Cells to Tango: Host Adaptive Immunity Orchestrates Microbiome-Gut-Brain Axis Development

The gut-brain axis describes a paradigm wherein the trillions of microorganisms inhabiting the gastrointestinal tract engage in bidirectional communication with the host central nervous system. Adaptive immunity represents an important intermediate in this dynamic crosstalk; previous work in our lab has demonstrated that T-lymphocytes, a main class of immune effector cells, contribute to neurodevelopmental processes and behavioral outcomes across the lifespan. Parallels between the phenotype of T-cell deficient and germ free mice led us to hypothesize that bidirectional T-cell-microbe communication is critical for normal neurodevelopment, and that T-cell deficiency impacts the neural circuitry underpinning behavior via disruption of the gut-brain axis. The main objective of this thesis was to elucidate the mechanisms by which T-cells mediate developmental gut-brain signalling. The first installation examined the gut microbiome, gut metabolome, and neurochemical profile in wild-type and T-cell deficient mice from adolescence to adulthood, demonstrating that absence of T-cells impacts the developmental trajectory of functional microbiome output and levels of neuroactive molecules in the brain. Experiment two investigated the impact of T-cell deficiency on gut-brain communication through the lens of host gene expression in the parenchyma and the intestine. T-cell deficient mice showed significant changes in genes related to intestinal immunity and barrier function, in addition to decreases in microglia-related genes in the prefrontal cortex during early life. The final experiment transitioned into a wild-type model to measure the co-evolution of T-cell subsets in mucosal and central immune compartments with composition and diversity of the microbiota. We demonstrated a parallel diversification of the gut microbiome and the functional T-cell repertoire, whereby emergence and proliferation of specific T-cell subsets is linked to compositional shifts in dominant microbial communities across development. Together, our results demonstrate the importance of T-cells for normal development of the holo-organism, with implications for the developmental wiring of functional brain circuitry. / Thesis / Doctor of Philosophy (PhD) / Modern medicine has increasingly placed emphasis on the mind-body connection. This has been exemplified by a series of recent discoveries surrounding the importance of the gut microbiome in maintaining our physical and mental health. One of the key channels through which the microbiome communicates with the host is through the immune system, an equally complex network of cells and proteins that protect the body against invading pathogens. Indeed, these systems evolve alongside each other and engage in constant crosstalk throughout the lifespan, with downstream impacts on the developing brain. This thesis sought to further explore the role of T-cells, a key component of the adaptive immune system, in coordinating gut-microbiome-brain interactions across development. The first experiment examined the microbiome as well as small molecules in the gut and brain of normal mice and mice lacking T-cells. The second experiment built on this work to examine how T-cells influence the expression of different genes in the gut and brain. Finally, the third experiment mapped different populations of T-cells and microbiome composition from the first week of life to adulthood, to better understand how they interact at different stages of development. This work will offer insight into how T-cells talk to the microbiome and how they transmit signals from the gut to the brain, with implications for understanding neurodevelopmental disorders and how they arise.

This thesis attempts to explore approaches to design decision making, as a response to a project's site, its history, and the larger cultural context. The above issues are investigated through the design of a multipurpose building at the east end of the Drillfield on the Virginia Tech campus in Blacksburg, Virginia. The building will form a "stage" to accommodate the daily life of the campus. In addition, the proposal was conceived to: a. emphasize both axial and radial geometric features of the site; b. redefine the edge of the oval Drillfield; c. create magnetic force between the proposed building and the existing War Memorial Chapel; d. translate the idea into tectonics. / Master of Architecture

The Substantive

In order to understand a sentence fully, one must understand its parts and the way they work. A complete sentence, in its most simple form, is a substantive with a verb in agreement. Additions to this sentence may include other substantives and verbs, but most additions are modifiers, either adjectival or adverbial. These modifiers should enhance the substantive and its verb, elevating their existence within the sentence. However, modifiers may also detract from the substantive and verb. This occurs when the substantive and verb become lost among the additions. A simple, easily understood complete sentence, i.e. a sentence containing only a substantive with a verb in agreement, becomes with the addition of too many modifiers that are not really necessary a foudryant expression that is difficult, although not impossible to diagram for the purpose of grasping the relationships between words that are, in their present state, convoluded by the addition of modifiers within and without clauses, to decipher. In other words, too many modifiers make a sentence difficult to understand. While the simple complete sentence is concise by its very nature, a concise sentence may contain much more than a substantive with verb. A concise sentence contains the essential and whatever else, if anything, to elevate the essential. Modifiers are not bad per se, but modifiers must remain just that: modifiers. They must be chosen carefully and remain secondary to the essential (provided that their primacy is not the intent). Modifiers and their use within a sentence are distinguishing factors between carefully crafted prose or poetry and misleading verbosity. The beauty of the concise sentence is its ability to express the most while using the least. The same can be true for architecture. Association entre l'utilisation de Facebook et les marqueurs de stress psychologiques et physiologiques chez les adolescents

La création de Facebook est venue changer la façon dont les gens interagissent, mais nous en savons peu sur les impacts de Facebook sur la santé et le bien-être. À l’heure actuelle, environ 90 % des adolescents sont actifs sur Facebook et la majorité l’utilise tous les jours. Sachant que l’adolescence est une période critique du développement et que lors de cette période les adolescents sont particulièrement vulnérables aux effets du stress, il importe de comprendre les facteurs pouvant entrainer une augmentation des hormones de stress chez les adolescents. Le but du présent mémoire était donc d’étudier la relation entre l’utilisation de Facebook chez les adolescents et des marqueurs de stress psychologique et physiologique. Pour ce faire, nous avons mesuré les hormones de stress chez 88 adolescents (41 garçons, 47 filles) âgés entre 12 et 17 ans. Les adolescents devaient remplir le ‘Social Network Survey’, un questionnaire mesurant différents facteurs associés à l’utilisation de Facebook et le ‘Child Depression Inventory’, qui mesure les symptômes de dépression. Les résultats suggèrent que ce n’est pas le temps passé sur Facebook qui est en lien avec le stress psychologique et physiologique, mais plutôt la nature de l’utilisation de Facebook. Pour les filles, c’est le nombre d’amis sur Facebook qui est relié à des hauts niveaux de cortisol, tandis que pour les garçons c’est le fait de s’exposer sur Facebook. Cette étude est la première à démontrer une association entre la nature de l’utilisation de Facebook et les niveaux d’hormones de stress chez des adolescents. / The creation of Facebook can change the way people interact, but we know little about the impact of Facebook on health and well-being. As of today, 90 % of adolescents are active on Facebook and most of them connect everyday. Adolescence is a critical period in childrens’ development and during this period they are particularly vulnerable to stress hormones. It is therefore important to understand factors that may cause an increase in stress hormones in adolescents. The goal of this Masters thesis was to determine the relation between Facebook utilization in adolescents and psychological and physiological markers of stress. In order to do so, we measured stress hormones in 88 adolescents (41 boys, 47 girls) aged between 12 and 17 years old. Adolescents where asked to fill out the Social Network Survey, a questionnaire measuring different factors related to Facebook utilization and the Child depression Inventory, measuring symptoms of depression. Results suggest that it is not the time spent on Facebook that is related to psychological and physiological stress but the nature of Facebook utilization. For adolescent girls, it is the number of Facebook friends that is associated with high levels of cortisol, whereas in adolescent boys it is the fact of exposing one-self on Facebook that is associated with high levels of cortisol. This study is the first to show an association between Facebook utilization and stress hormones in adolescents.

O tráfego de leucócitos é um processo complexo, dependente da ação de inúmeras substâncias químicas, além da perfeita interação celular. Desta forma, este estudo teve como objetivo avaliar a ação dos GCe e da ANXA1 sobre o eixo SDF-1α/CXCR4 e IL-17/IL-23/G-CSF e sobre a expressão de moléculas de adesão CD18, CD49d e CD62L. Foram utilizados camundongos machos Balb/C selvagens (WT) ou ANXA1-/-. As avaliações foram realizadas em condições basais, na presença de altas concentrações de GCe e na vigência de processo inflamatório, induzidos pela administração de ACTH (5 µg/animal, i.p.) ou pela injeção de LPS (100 µg/kg, i.p.), respectivamente, ou na ausência da ação dos GCe, pela ação do RU 38486 (RU, 10 mg/kg, i.p.). A participação da ANXA1 e do receptor FPR2 foi avaliada pelo pré-tratamento com Ac2-26 (1 mg/Kg, i.p.) ou com BOC2 (10 µg/animal, i.p.) durante 4 dias, 1 vez ao dia. A quantificação total e diferencial das células foi realizada em câmara de Neubauer e em esfregaços corados por May-Grunwald ou citometria de fluxo. As quantificações de CXCR2, CXCR4, FPR2, CD18, CD49d, CD62L e maturação granulocítica (CD11b/Ly6G) em células da medula e da circulação foram realizadas por citometria de fluxo. A expressão de ANXA1 nos tecidos do estomago e do baço foi realizada por western blotting e nas células da medula óssea e sangue circulante foi realizada por imunofluorescência. As quantificações de IL-17, IL-23, G-CSF, SDF-1α e corticosterona foram realizadas por ELISA. A quimiotaxia de neutrófilos da medula óssea e sangue periférico foi ensaiada na placa de quimiotaxia com filtro de poro de 8 µm. A fagocitose de neutrófilos apoptóticos por macrófagos da medula óssea foi avaliada por ensaio in vitro. Para verificar os efeitos do ACTH na migração de neutrófilos no processo inflamatório, foi empregado o modelo de bolsa de ar (100 µg/mL; LPS); e o comportamento dos leucócitos circulantes de animais tratados com ACTH foi avaliado pela técnica de microscopia intravital. Os resultados obtidos, que estão apresentados em quatro temáticas, mostraram que: 1) neutrófilos da medula óssea e sangue periférico expressam ANXA1 no citoplasma e membrana, bem como o receptor FPR2, constitutivamente, e a expressão de ambos é regulada pelos GCe. A ANXA1, via receptor FPR2 expresso em células da medula óssea, controlam a maturação neutrofílica e o tráfego destas células da medula óssea para o sangue. A ANXA1, via interação ao FPR2, controla o clearance de neutrófilos do sangue para a medula óssea, modulando o eixo SDF-1α/CXCR4; 2) A administração do ACTH causa neutrofilia e os neutrófilos circulantes são ANXA1+, CD18+, CD49d+, CD62L+, mostrando que injeção do ACTH in vivo altera o fenótipo destas células na circulação. Estas modificações alteram o comportamento dos neutrófilos na circulação, bem como a migração para a bolsa de ar na vigência de inflamação e para os tecidos de clearance. Estes efeitos podem ser dependentes, pelo menos em parte, da inibição de migração orientada, já que quimiotaxia frente ao fMLP ou ao SDF-1α estavam reduzidas. Ainda, o clearance de neutrófilos é reduzido em animais tratados com o ACTH pela menor atividade fagocítica e secretora dos macrófagos medulares; 3) Animais tratados com RU 38486 e ANXA1-/- mobilizam granulócitos da medula óssea para o sangue circulante e, deste compartimento para o foco de inflamação com maior intensidade que o observado em animais controles. O eixo IL-17/IL-23/G-CSF parece estar envolvido na granulopoiese e na mobilização de neutrófilos para o sangue durante a inflamação, mas não é alvo de ação da ANXA1 e o GCe nesta etapa do processo inflamatório. Adicionalmente, foi observado que na vigência de peritonite, as moléculas de adesão, CD49d e CD62L estão envolvidas no processo de migração de neutrófilos da medula óssea para o sangue. Os resultados aqui obtidos permitem concluir que os GCe e a ANXA1 são relevantes para granulopoiese e tráfego dos neutrófilos da medula óssea em condições fisiológicas e na vigência de processo inflamatório. Ainda, em conjunto com os dados da literatura, os nossos resultados podem sugerir a participação da ANXA1 dos GCe na plasticidade fenotípica dos neutrófilos de acordo com os estímulos a que são submetidos, e podem auxiliar na compreensão dos novos conceitos sobre a produção, tempo de vida, localização e funções de neutrófilos. / The traffic leukocytes is a complex process dependent on the action of severals chemical mediators, in addition to perfect cell interaction. Therefore, this study aimed to evaluate the effect of GCe and ANXA1 on SDF-1α/CXCR4 and IL-17/IL-23/G-CSF and on the expression of adhesion molecules CD18, CD49d and CD62L. Balb/C wild type and ANXA1-/- male mice were employed. The analysis were performed at physiological conditions, in the presence of high concentrations of GCe and during of inflammatory process induced by ACTH administration (5 µg/animal, i.p.) or LPS injection (100 µg/kg, i.p.), respectively or in the absence of GCe action, by the action of RU 38486 (RU, 10 mg/kg , i.p.). The involvement of the receptor FPR2 and ANXA1 was assessed by pre-treatment with Ac2-26 (1 mg/kg, i.p.) or BOC2 (10 µg/animal, i.p.) for 4 days, once a day. The quantification of total and differential cell was performed in a Neubauer chamber and stained smears by May-Grunwald and flow cytometry. Quantification of expression of CXCR2, CXCR4, FPR2, CD18, CD49d, CD62L and granulocytic maturation (CD11b/Ly6G) in the bone marrow and circulation were performed by flow cytometry. The expression of ANXA1 on tissues was performed by western blotting and on cells from bone marrow and blood by immunocytochemistry. Quantification of IL-17, IL-23, G-CSF, SDF-1α and corticosterone were performed by ELISA. The chemotaxis of neutrophils from the bone marrow and blood was tested in the chemotaxis chamber with filter pore of 8 microns. The phagocytosis of apoptotic neutrophils by bone marrow macrophages was assessed by in vitro assay. To investigate the effects of ACTH in the migration of neutrophils in the inflammatory process, the model employed was air pouch (100 µg/ ml, LPS), and the behavior of circulating leukocytes from animals treated with ACTH were evaluated by intravital microscopy. The results obtained, which are presented in three sections, showed that: 1) neutrophils from the bone marrow and blood expressed ANXA1 in the cytoplasm and membrane, as well as FPR2, constitutively and the expression of both is regulated by GCe. The ANXA1 via FPR2 receptor expressed in bone marrow cells, controls the neutrophilic maturation and traffic of these cells from the bone marrow into the blood. The ANXA1 via interaction to FPR2 controls the clearance of neutrophils from the blood to the bone marrow by modulating the SDF-1α/CXCR4 axis; 2) the administration of ACTH induces neutrophilia and the circulating neutrophils are ANXA1+, CD18+, CD49d+ and CD62L+, showing that the injection of ACTH in vivo alters the phenotype of these cells in the blood. These modifications alter the behavior of neutrophils in the blood, as well as the migration to the air pouch in the presence of inflammation and to the tissue clearance, and these effects may be dependent, at least in part, on inhibition of migration oriented events, as chemotaxis in response to fMLP or SDF-1α were reduced. Further, the clearance of neutrophils is reduced in animals treated with ACTH due to the lower phagocytic and secretory activity of medullary macrophages; 3) Animals treated with RU 38486 and ANXA1-/- mobilize granulocytes from bone marrow into the blood, and from this compartment to the focus of inflammation with higher intensity than that observed in the control group. The axis IL-17/IL-23/G-CSF seems to be involved in granulopoiesis and mobilization of neutrophils into the blood during inflammation, but it is not the target of action of ANXA1 and GCe at this step of inflammatory process. Additionally, it was observed that in the presence of peritonitis, the adhesion molecules, CD49d and CD62L are involved in the migration of neutrophils from the bone marrow into the blood. The results obtained allow concluding that the GCe and ANXA1 are relevant to the granulopoiesis and the traffic of neutrophils from bone marrow under physiological conditions and in the presence of inflammation. Furthermore, together with literature data, the data presented here may suggest the involvement of ANXA1 the GCe in phenotypic plasticity of neutrophils according to the stimuli that are submitted, and may support to understand the new concepts of production, half-life, location and function of neutrophils.

As methodological course, it was a kind of interpretative-explanatory study of a qualitative nature. The educational projects of the six study target schools were obtained by means of a documentary collection, and subsequently were treated through a documentary analysis. From an agreement between researchers from the Post-Graduation Program in the UFSCar Clinic Management was built a matrix as a methodological tool containing two axes: Curricular Axis and Professional Profile Axis and, from these, the analysis categories were defined: C1-Curricular organization: most courses are organized according to the rules of DCN, however, traditional curriculum models are still present; C2- Learning conceptions: more than half of the courses guided their curricula in teachinglearning process proposed by the DCN, and some of these still have more innovative experiences. Other courses present traditional learning conceptions and distant from the DCN; C3- Student assessment procedures: the student assessment envisaged by the DCN is not well defined, as in the case of half of the courses. P1 General Profile; most courses include somehow the professional profile recommended by DCN; P2- Attention and Health Care: two courses are distant from health care benchmarks and model of care proposed by the DCN, and, two stand out due to their larger focus. P3- Health Management: most of the courses, as well as DCN, don’t have a clear delineation of management processes; P4- Health education: the DCN and the courses do not explicitly show this category, except in one of these. Data analysis shows the similarities and differences of pedagogical projects of the courses by DCN about the categories, as well as their intentions. The results indicate the current state of the curricula and training of Psychology for health, and the need for changes both in DCN as in the courses curricular development and reform. / Por considerar as DCN uma importante política pública para formação em saúde do Psicólogo, e para estruturação dos currículos, o presente estudo teve como objetivo analisar os projetos pedagógicos dos cursos de graduação em Psicologia das Instituições de Ensino Superior públicas do estado de São Paulo à luz das DCN. Enquanto percurso metodológico tratou-se de um tipo de estudo interpretativo- explicativo, de natureza qualitativa. Os projetos pedagógicos dos seis cursos alvo do estudo foram obtidos por meio de uma coleta documental, e posteriormente, foram tratados através de uma análise documental. A partir de um acordo entre pesquisadores do Programa de Pós- Graduação em Gestão da Clínica da UFSCar foi construída uma matriz como ferramenta metodológica contendo dois eixos: Eixo Curricular e Eixo Perfil Profissional e, a partir destes, foram definidas as categorias de análise: C1- Organização curricular: a maioria dos cursos organiza-se de acordo com as normas das DCN, no entanto, modelos curriculares tradicionais ainda estão presentes; C2- Concepções de aprendizagem: mais da metade dos cursos pautam seus currículos em processos de ensino- aprendizagem propostos pelas DCN, e alguns destes ainda apresentam experiências mais inovadoras. Outros cursos apresentam concepções de aprendizagem tradicionais e distantes das DCN; C3- Processos de avaliação do estudante: a avaliação do aluno prevista pelas DCN não é bem definida, assim como no caso de metade dos cursos. P1- Perfil Geral; a maioria dos cursos contempla de alguma forma o perfil profissional preconizado pelas DCN; P2- Atenção e Cuidado em Saúde: dois cursos distanciam-se dos referenciais de cuidado em saúde e modelo de atenção proposto pelas DCN, e, outros dois destacam-se por apresentarem maior foco. P3- Gestão em Saúde: a maioria dos cursos, assim como DCN, não possuem um delineamento claro sobre processos de gestão; P4- Educação em saúde: as DCN e os cursos não apresentam explicitamente esta categoria, exceto em um destes. A análise dos dados apresenta as aproximações e distanciamentos dos projetos pedagógicos dos cursos à luz das DCN quanto às categorias, assim como as suas intencionalidades. Os resultados indicam a situação atual dos currículos e da formação dos cursos de Psicologia para a área da saúde, e a necessidade de reformulação e atualização dos projetos pedagógicos.
