FreeIPA - správa přístupu dle URI / FreeIPA - URI Based Access Management

Cílem práce je navržení a implementace řízení přístupu na základě URI požadovaného zdroje. Pro implementaci bylo jako základ použito rozšíření Host Based Access Control v nástroji pro správu identit FreeIPA. Zároveň bylo třeba rozšířit související infrastrukturu, především program SSSD. Jako příklad aplikace využívající HBAC na základě URI byl implementován autorizační modul pro Apache HTTP Server. Zásadním řešeným problémem byl návrh infrastruktury pro komunikaci nezbytných parametrů a návrh strategie vyhodnocení HBAC pravidel definujících přístupová práva. Kompletní řešení bylo předvedeno na příkladu zabezpečení instance webové aplikace Wordpress.

El siguiente trabajo de tesis presenta una propuesta de transformación digital alineada al plan estratégico institucional de una entidad de control gubernamental del Estado peruano luego de realizar la evaluación de los procesos con el uso de COBIT 5 PAM. En este documento, se encontrará como primer capítulo, la definición del proyecto, donde se detalla la entidad de objeto de estudio, la visión, misión, objetivos estratégicos y una explicación breve de los mismos. El segundo capítulo, nos muestra el cumplimiento de los Outcomes – ABET. En el tercer capítulo, se presenta el marco teórico utilizado para la tesis. En el cuarto capítulo encontraremos el desarrollo del proyecto, que está dividido en dos secciones, la primera relacionada al Gobierno Corporativo de TI donde hemos utilizado como instrumento de evaluación COBIT 5 PAM, aquí se detallan los procesos de TI, se realiza un contraste con los procesos de TI de COBIT 5 PAM, se evalúan los niveles actuales de cada proceso, la brecha encontrada y finalmente se plantea un plan de mejora. Como segunda sección, encontramos el tema de Transformación Digital, en el cual se detallan las tecnologías a utilizar, el proceso que se desea transformar y cómo las herramientas tecnológicas planteadas brindan soporte al mismo, así también, se determina una factibilidad técnica y económica de la propuesta. Como quinto capítulo, se detallan los resultados del proyecto de tesis, y finalmente, se presenta las herramientas para la gestión del proyecto y anexos. / The following thesis work presents a proposal for digital transformation aligned with the institutional strategic plan and after a self-evaluation has been carried out in relation to the use of COBIT 5 PAM on a government control entity of the Peruvian State. In this document, you will find as the first chapter, the definition of the project, which details the entity under study, the vision, mission, strategic objectives, process diagram and a brief explanation of them. In the third chapter, the theoretical framework used for the realization of this thesis is presented. In the fourth chapter we will find the development of the project, which is divided into two large sections, the first related to the Corporate Governance of IT where we have used as an evaluation instrument COBIT 5 PAM, here the IT processes are detailed, a contrast is made With the IT processes proposed by COBIT 5 PAM, the current levels of each process, the expected levels, the gap found are evaluated and finally an improvement plan is proposed. Durante los últimos 10 años la necesidad de contar con un efectivo gobierno de TI se hace indispensable para las empresas que quieren lograr una ventaja competitiva en el mercado. El plan estratégico de las empresas debe contener su plan a futuro y las acciones que debe realizar para lograrlo. Asimismo, sus objetivos estratégicos deben estar alineados con los procesos de TI. Bajo este escenario, en el que se presenta un constante cambio, muchas empresas se enfrentan a problemas de bajo nivel de control y uso inadecuado de recursos, causados por la baja concientización de un Gobierno de TI, que posteriormente desencadenan en una insuficiente definición de procesos, en la carencia de la evaluación de capacidades de procesos y en la falta de iniciativa para la mejora continua. Asimismo, a través de la tecnología se busca transformar el proceso de negocio de operación de pesca porque está asociado a los objetivos estratégicos de la empresa. Por ende, cualquier impacto negativo se ve reflejado en la rentabilidad y costos Ante esta situación, se desarrolla la propuesta para el caso de estudio de una empresa del sector pesquero, en el que se propone mejorar los procesos de TI mediante un marco de trabajo basado en COBIT PAM y transformar el proceso de operación de pesca utilizando la tecnología basada en internet de las cosas para consolidar este proceso obteniendo rentabilidad y reduciendo costos. / During the past 10 years the need for effective IT governance has become indispensable for companies that want to gain a competitive advantage in the market. The strategic business plan must contain its future plan and the actions it must take to achieve it. At the same time, its strategic objectives should be aligned with the IT processes. Under this scenario, in which there is constant change, many companies face problems of low level of control and inaddecuate use of resources, caused by the lack of awareness of an IT Government, which subsequently trigger an insufficient process definition, in the carence of evaluation of process capacities and in the lack of initiative for continuous improvement. Likewise, technology seeks to transform the fishing operation business process because it is associated with the company's strategic objectives. Therefore, any negative impact is reflected in profitability and costs. Given this situation, the proposal is developed for the case study of a company in the fishery sector, in which it is proposed to improve IT processes through a framework based on COBIT PAM and transform the fishing operation process using technology based on the internet of things to consolidate this process obtaining profitability and reducing costs. / Tesis

The effect of maternal nicotine exposure on rat lung tissue morphology. ' a light and electron microscopic study

Masters of Science / The infants of women who smoke during pregnancy have a lower birth mass than those born of women who abstain. Animal studies reveal that reduced growth due to maternal nicotine exposure during gestation is accompanied by lung hypoplasia. Biochemical analysis suggests that these lungs contain more cells which implies that lung damage occurs. In this study we examined the in vivo effects of maternal nicotine exposure (lmg/Kg/day), the equivalent of 32 cigarettes per day, on the following parameters of fetal and neonatal Wistar rat lung:(i) the content and distribution of glycogen in fetal and neonatal lung (ii) the status of connective tissue in neonatal lung (iii) the cell composition of the alveoli in neonatal lung. Fetal rat lungs of ages 17, 18, 19 and 20 days and neonatal lungs of 1, 7, 14 and 21 day old pups were used. Light microscope techniques and special stains were used to investigate glycogen, connective tissue, macrophage numbers and morphological status of the lungs. Transmission electron microscope (TEM) techniques were employed to investigate the characteristics and composition of the alveolus The results show clearly that maternal nicotine exposure elevates pulmonary alveolar macrophage numbers'(PAM's) and lung glycogen levels. The quantity of elastic fibres in 1 day old neonates was significantly reduced but no changes in the quantity of reticulin and collagen fibres was observed. As a result of this change in connective tissue status, emphysema-like lesions and alveolar collapse was evident in the lungs of nicotine-exposed pups. TEM investigations revealed that changes to the composition of alveoli occurred. These included increased numbers of type II pneumocytes with high numbers of lamellar bodies with degenerative changes. Thickening of the blood-air barrier was also observed. The effect of maternal nicotine exposure has been documented in this study. However, it has not been possible to pinpoint the mechanisms involved but explanations have been proposed. Further research is required to elucidate the mechanisms by which nicotine produces these effects. Information thus obtained could help prevent the harmful effects to the fetus and neonate caused by smoking during pregnancy.

A PAM Decomposition of Weak CPM

The Enhanced Flight Termination System uses weak CPM as its modulation scheme and a limiter-discriminator as its demodulation scheme. A PAM representation of weak CPM was developed which representation provided the necessary componenents to build a simplified PAM-based receiver that outperformed the EFTS limiter-discriminator, even in the presence of phase noise. The PAM representation also provided a new perspective into the negative characteristics of weak CPM.

Estimating distributions of two declining aerial insectivorous Nightjars species using passive acoustic monitoring in southern Illinois

Nightjars are a group of nocturnal and aerial insectivorous birds that have experienced long-term decline likely driven primarily by habitat loss and declines in prey populations. Eastern Whip-poor-will (Antrostomus vociferus) and Chuck-will’s-widow (Antrostomus carolinensis), two nightjar species native to Illinois, declined 69% and 58% since 1966, respectively. Although previous survey efforts have documented presence of Chuck-will’s-widow and Whip-poor-will, their current distribution in the state is not well known. Using Autonomous Recording Units (ARUs) deployed in a uniform, systematic grid, I surveyed 142 locations from May – July 2022 on public and private lands across the southern eleven counties of Illinois to assess Whip-poor-will and Chuck-will’s-widow distribution and estimate species occupancy. I estimated species relationships with proportion of landcover types, forest patch configuration, and proximity to other landcover types. Additionally, I quantified disturbances from the past 15 years to estimate species relationships to the severity and duration of disturbances. I deployed ARUs for 710 survey days collecting 170,400 minutes or 3,000 hours of recordings. Acoustic bird call identification software, BirdNet, was highly accurate at detecting focal species and greatly reduced the time spent manually reviewing acoustic data. BirdNet identified 43,922 calls of Whip-poor-will and 31,447 calls of Chuck-will’s-widow. I detected Whip-poor-will on 78 surveys with 100% accuracy and Chuck-will’s-widow on 75 surveys with 76% accuracy. Whip-poor-will were positively associated with forest patches with large core areas that neighbored pastures. Additionally, Whip-poor-will were likely to occupy landscapes that had experienced low to moderate disturbance within the previous 15 years. Covariates used to model Chuck-will’s-widow occupancy explained little variation in detection or occupancy and there were no significant relationships with any covariate. However, examining non-significant trends suggest similar relationships as Whip-poor-will in the area. Results highlight the efficiency of passive acoustic monitoring for these birds and the need for further investigation into Chuck-will’s-widow species-environmental relationships. In southern Illinois, Chuck-will’s-widow populations appears to be consistent with previous estimates from the 1990s while Whip-poor-will follow the broader trend of decline.

Identification of novel active Cas9 orthologs from metagenomic data

CRISPR-Cas is the state-of-the-art biological tool that allows precise and fast manipulation of the genetic information of cellular genomes. The translation of the CRISPR-Cas technology from in vitro studies into clinical applications highlighted a variety of limitations: the currently available systems are limited by their off-target activity, the availability of a Cas-specific PAM sequence next to the target and the size of the Cas protein. In particular, despite high levels of activity, the size of the CRISPR-SpCas9 editing machinery is not compatible with an all-in-one AAV delivery system and the genomic sequences that can be targeted are limited by the 3-NGG PAM-dependency of the SpCas9 protein. To further expand the CRISPR tools repertoire we turned to metagenomic data of the human microbiome to search for uncharacterized CRISPR-Cas9 systems and we identified a set of novel small Cas9 orthologs derived from the analysis of reconstructed bacterial metagenomes. In this thesis study, ten candidates were chosen according to their size (less than 1100aa). The PAM preference of all the ten orthologs was established exploiting a bacterial-based and an in vitro platform. We demonstrated that three of them are active nucleases in human cells and two out of the three showed robust editing levels at endogenous loci, outperforming SpCas9 at particular targets. We expect these new variants to be very useful in expanding the available genome editing tools both in vitro and in vivo. Knock-out-based Cas9 applications are very efficient but many times a precise control of the repair outcome through HDR-mediated gene targeting is required. To address this issue, we also developed an MS2-based reporter platform to measure the frequency of HDR events and evaluate novel HDR-modulating factors. The platform was validated and could allow the screening of libraries of proteins to assess their influence on the HDR pathway.
