Bioaccumulative contaminants in marine mammals: uptake and effects

This thesis provides insights into the transport and fate of contaminants of concern (polychlorinated biphenyls (PCBs), polybrominated diphenyl ethers (PBDEs) and mercury (Hg)), as well as results on the impacts of these compounds on marine mammal health. Atmospheric transport is known to be a significant pathway for the delivery of contaminants to remote food webs. Air and rain samples were collected from one remote site on the west coast of Vancouver Island, British Columbia (BC), Canada, and from one near-urban site in the Strait of Georgia, BC. While global atmospheric dispersion was observed for the legacy PCBs, 40% of PBDEs detected in BC air appeared to be originating from trans-Pacific transport. It was estimated that 3kg of PCBs and 17kg of PBDEs were deposited every year in the Strait of Georgia. Once deposited, PCBs, PBDEs and Hg biomagnify up the food chain. Harbour seals are non-migratory and can be used to provide signals of local contaminant sources. They have been extensively used as indicators of PCB and PBDE food web contamination in the BC coastal environment. The collection of over 200 harbour seal fur samples from various locations around Vancouver Island, BC and Puget Sound, WA, USA helped us pinpoint three sites where Hg levels were significantly higher than our reference site, Bella Bella (Queen Charlotte Strait, Port Renfrew and central Puget Sound). A combination of anthropogenic sources and marine food web processes appeared to influence the delivery of methylmercury (MeHg) to the top of this coastal marine food chain. Our results also suggested that these Hg levels (1.6-46.9 µg/g) could be a concern for the health of these harbour seals. Genomic techniques were used to generate insights into the implications of contaminant exposure on the health of marine mammals inhabiting industrialized regions (harbour seals from the Northeastern Pacific and Northwestern Atlantic) and remote, supposedly pristine, environment (Arctic beluga whales). In harbour seal blubber, there were positive correlations between the mRNA levels of several genes, including estrogen receptor alpha (Esr1), thyroid hormone receptor alpha (Thra), and glucocorticoid receptor (Nr3c1), and PCB levels. In beluga blubber, aryl hydrocarbon receptor (Ahr) and cytochrome P450 (Cyp1a1) mRNA levels increased with PCBs, consistent with their role in toxicity.While PCB-related toxic responses were observed in both species, additional factors appeared to be affecting the expression of important genes in beluga. Our results suggested that a shift in beluga diet during periods of low sea ice extent, as evidenced by changes in δ13C isotope ratios, had a significant impact on mRNA levels coding for genes involved in growth, metabolism and development. The use of a dual study design to evaluate the long range versus local sources of contaminants highlighted the importance of trans-Pacific transport in the delivery of PBDEs to coastal BC and the occurrence of local Hg sources in this marine environment. However, consistent with previous studies, our results suggested that PCBs remain the top contaminant of concern for marine mammal health. Evaluation of EDA tools for electronic development and a study of PLM for future development businesses

Electronic Design Automation (EDA) tools are today very capable computer programs supporting electronic engineers with the design of printed circuit board (PCB). All tools have their strengths and weaknesses; when choosing the right tool many factors needs to be taken into consideration aside from the tools themselves. Companies need to focus on the product and revenues for a business to be viable. Depending on the knowledge and strengths of the company, the choice of tools varies. The decision should be based on the efficiency of the tools and the functions necessity for the company rather than the price tags. The quality and availability of support for the tools, training costs, how long will it take to put the tool in operation and present or future collaboration partners is equally important factors when deciding the right tool. The absence of experience and knowledge of the current tool within a company is a factor which could affect important operation; therefore it is important to provide training and education on how to use the tool to increase its efficiency. Providing training and education can be a large expense, but avoids changes within and makes the business competitive. The choice of EDA tool should be based on the employed engineer’s current knowledge and experience of the preferred tool. If the employed engineer’s knowledge and experience varies too much, it might be preferable to make a transition to one of the tool by training and education. Product lifecycle management (PLM) is a data management system and business activity management system which focuses on the lifecycle of a product. To manage the lifecycle of a product it is necessary to split the lifecycle into stages and phases for a more manageable and transparent workflow. By overseeing a product’s entire lifecycle there are benefits which affects many areas. PLM greatest benefits for EDA are collaboration across separate groups and companies by working together through a PLM platform, companies can forge strong design chains that combine their best capabilities to deliver the product to the customers. This report is a study on evaluating which EDA suits the company with consideration of the employed engineer’s demands, requests and competence. The interests in PLM made the company suggest a short theory study on PLM and EDA benefits.

Biochemical Monitoring Of Toxic And Carcinogenic Organic Pollutants Along The Izmir Bay After The Great Canal Project And Possible Health Effects

The induction of hepatic cytochrome P4501A1 and its monooxygenase activity 7-ethoxyresorufin O- deethylase, (EROD) in fish by PAHs, PCBs and dioxins has been suggested as an early warning system &ldquo / most sensitive biochemical response&rdquo / for assessing environmental contamination conditions. In this study, the degree of induction of cytochrome P4501A1 protein as determined immunochemically and CYP1A1 associated EROD activity in fish were utilized as biomarkers of exposure to PAHs, PCBs and related organic pollutants along the izmir Bay on the Aegean Sea Coast after the Great Canal Project. Three different fish species were used throughout this study, namely leaping mullet (Liza saliens), annular seabream (Diplodus annularis) and common sole (Solea vulgaris) which were representatives of pelagic, benthopelagic and benthic fish, respectively. Fish were sampled in November 2002 and October 2003 from different sites of the Bay. The mullet caught from Harbor, &Uuml / &ccedil / kuyular port site, and Pasaport region displayed highly elevated EROD activities which were 2258&plusmn / 840 (n=15), 2011&plusmn / 490 (n=4), 1813&plusmn / 287 (n=11) pmole /min/mg protein, respectively and were 104, 80 and 79 fold higher than that of fish obtained from the reference point (25&plusmn / 9 pmole/min/mg protein / n=4). Mullet caught along the pollutant gradient at three other sites (Hekim Island, inciralti, and Zeytinalani) exhibited less but highly significant induced EROD activity. EROD activities of common sole sampled from Fo&ccedil / a open sites (107&plusmn / 20 pmol/min/mg protein, n=5) and site16A (80&plusmn / 12 pmol/min/mg protein, n=9) were found to be very low and the latter was accepted as reference site. The highest EROD activity were seen in fish captured from inciralti which was about 6.3 times higher than those obtained from reference site. Common sole caught from the mouth of Gediz River and Hekim Island exhibited also highly elevated EROD activities. Annular seabream was tested to monitor CYP1A inducing chemicals for the first time in this study. The highest EROD activity (1376&plusmn / 279 pmol/min/mg protein, n=8) were detected in fish samples collected from Harbor region. An inverse relationship was found between distance to the harbor region and EROD activities of annular seabream captured from other sampling sites. In this study for the first time, major cytochrome P450 dependent mixed function oxidase activities such as benzphetamine N- demethylase, ethylmorphine N-demethylase and aniline 4- hydroxylase, were characterized in annular seabream. Changes in the P450 1A1 protein level were determined by immunochemical analysis to monitor the pollutant based induction in all fish species and good correlation was obtained between EROD activity and CYP1A protein content. Fish from polluted sites had both highly induced EROD activity and cytochrome P450 1A content. Chemical analysis of total PAH concentration in sediment and liver tissues of some fish sample were also carried out. Although, izmir Great Canal Project has been active since 2000 to treat and protect the izmir Bay from the contamination of domestic and industrial wastes this study clearly demonstrated that the level of PAH, PCB and dioxin type persistent organic contaminants are still very high especially in the Inner and Middle Bay. This has implications for human fish consumption from contaminated areas, as well as for the health status of aquatic organisms.

FPGA-based Instrumentation for Advanced Physics Experiments

Modern physical experiments often demand advanced instrumentation based on advances in  technology. This work describes four instrumentation physics projects that are based on modern, high-capacity Field-Programmable Gate Arrays, making use of their versatility, programmability, high bandwidth communication interfaces and signal processing capabilities. In the first project, a jet-finding algorithm for the ATLAS detector at the LHC experiment at CERN was developed and implemented, and different verification methods were created to validate the functionality and reliability. The experiment uses a three level trigger system, where the first level uses custom FPGA-based hardware for analysis of collision events in real-time. The second project was an advanced timing and triggering distribution system for the new European X-Ray Free Electron Laser (XFEL) facility at DESY in Hamburg. XFEL will enable scientists to study nano structures on the atomic scale. Its laser pulses will have the strongest peak power in the world with extremely short duration and a high repetition rate, which will even allow filming of chemical reactions. The timing system uses modern FPGAs to distribute high-speed signals over optical fibers and to deliver clocks and triggers with high accuracy. The third project was a new data acquisition board based on high-speed ADCs combined with high-performance FPGAs, to process data from segmented Ge-detectors in real-time. The aim was to improve system performance by greatly oversampling and filtering the analog signals to achieve greater effective resolution. Finally, an innovative solution was developed to replace an aging system used at CERN and Stockholm University to test vital electronics in the Tile Calorimeters of the ATLAS detector system. The new system is entirely based on a commercial FPGA development board, where all necessary custom communication protocols were implemented in firmware to emulate obsolete hardware. / Inom området instrumenteringsfysik bedrivs forskning och utveckling av avancerade instrument, som används inom moderna fysikexperiment. Denna avhandling beskriver fyra projekt där programmerbara kretsar (FPGA) har nyckelfunktioner för att lösa krävande instrumenteringsuppgifter. Den första projektet beskriver utveckling och implementering av en algoritm för detektering av partikelskurar efter partikelkollisioner i LHC-experimentets ATLAS-detektor. Experimentet genererar 40 miljoner händelser per sekund, som måste analyseras i real-tid med hjälp av snabba parallella algoritmer. Resultatet avgör vilka händelser som är tillräckligt intressanta för fortsatt noggrannare analys. Den andra projektet beskriver utvecklingen av ett system som distribuerar klock- och trigger-signaler över ett 3 kilometers experimentområde med extrem precision, i den nya röntgenlaseracceleratorn XFEL vid DESY i Hamburg. Vid XFEL kommer man utforska nanostrukturer och till och med filma molekylers kemiska reaktioner. I den tredje projektet beskrivs utvecklingen av ett höghastighets datainsamlingssystem, för segmenterade Ge-detektorer. Genom att översampla signalen med hög hastighet kan man uppnå en bättre noggrannhet i mätningen än vad AD-omvandlarens egna upplösning medger. Detta leder i sin tur  till förbättrade systemprestanda. Slutligen beskrivs en innovativ lösning till ett test system för den elektronik, som Stockholms universitet har levererat till ATLAS detektorn. Det nya systemet ersätter det föregående testsystemet, som är baserad på föråldrade inte längre tillgängliga komponenter. A relativa estagnação econômica e os investimentos massivos na indústria bélica, a ampliação da contestação nos países do leste-europeu, dentre outros, eram sintomas da crise de hegemonia da burocracia socialista, que tentou uma última cartada com a Perestroika. A crise iniciada na Europa oriental, que tem como marco simbólico a queda do muro de Berlim, culminou com a desintegração da União Soviética. Visceralmente ligados à URSS, os partidos comunistas de todo o mundo sentiram o impacto dessa crise terminal. O Partido Comunista Brasileiro (PCB), que assistia à saída de Luis Carlos Prestes do partido, também sentiu a crise e foi diretamente envolvido por ela, sendo que, em 1992, no seu X Congresso, consumou a sua própria derrocada. A ideia deste projeto é analisar como o PCB entra em crise no contexto de suas interpretações ilusórias das relações internacionais, deixando sua legitimidade minada, tendo em vista a queda do sistema socialista adotado na URSS e na Europa Oriental. / Since the early 80´s, the crisis of the called “real socialism” was an obvious fact. The relative economic stagnation and the massive investments in the war industry, the expansion of contestation in the Eastern European Countries, among others, were crisis symptoms of the socialist bureaucracy hegemony, which tried a last attempt with the Perestroika. The crisis, started in the Eastern Europe, whose symbolic mark is the fall of the Berlin Wall, culminated with the Soviet Union disintegration. Viscerally connected to the USSR, the communist parties around the world felt the impact of that terminal crisis. The Brazilian Communist Party (PCB), that was watching Luis Carlos Prestes leaving the party, also felt the crisis and was directly involved in it, being that, in 1992 during its X Congress, consummated its own collapse. The project idea is to analyze how PCB entered into the crisis in the context of its international relations illusory interpretations, making its legitimacy undermined, in view of the fall of the USSR and Eastern European socialist system .

Ambos nascidos em 1922, os partidos comunistas do Brasil e do Chile, ao longo do século XX, exerceram um importante papel nos debates de esquerda em seus respectivos países. A comparação entre essas esquerdas visa resgatar, enquanto objeto de estudo e de reflexão, elaborações teóricas e linhas de atuação que buscavam alcançar o poder de maneira distinta daquilo que, a partir de 1959, devido ao impacto da Revolução Cubana, ficaria consagrado como o “modelo revolucionário” a ser adotado em toda a América Latina. A enorme influência da Revolução Cubana no conjunto das organizações de esquerda na América Latina acabaria por encobrir a existência de outras formulações e propostas de acesso ao poder por parte da esquerda latino-americana, que estavam em curso antes da sua eclosão e que permaneceram nas décadas posteriores. Ademais, o exercício comparativo aqui pretendido parte do pressuposto de que houve uma inversão nos caminhos estratégicos desses dois partidos. Tendo como principal referência o ano de 1958, a análise em torno das estratégias políticas desses dois Partidos ilustra os distintos posicionamentos acerca dos processos de modernização em curso no Brasil e no Chile. Desse modo, enquanto o PCB deu início a uma transformação em sua cultura política, passando a legitimar a relação entre desenvolvimento capitalista e democratização social, o PCCh, juntamente com outros partidos da esquerda chilena, elaborou um projeto político que objetivava a hegemonia na condução de um projeto socialista de sociedade. No decorrer dos anos 1980, quando o Movimento Comunista Internacional se deteriorava, os sentidos dessas estratégias definiram o papel de cada Partido nos processos de redemocratização no Brasil e no Chile. / This paper makes a comparative analysis between the political strategies developed by two Latin American Communist parties, the Brazilian Communist Party (PCB) and the Communist Party of Chile (PCCh). Both of them were bom in 1922 and throughout the twentieth century, they played an important role on the left discussions in their countries. Comparisons between left parties aims to rescue, as an object of study and reflection, theoretical elaborations and areas of activity that were used to achieve the power differently from the " revolutionary model "which was adopted by Latin America. The big influence of the Cuban Revolution into ali Left organizations in Latin America would hide the existence of other formulations and proposals for access to power by the Latin American left, which were in progress before their outbreak and they have remained after decades. Furthermore, there was a reversai in both patties' strategies. Toe analysis about the political strategies of them illustrates the different positions on the processes of modernization in Brazil and Chile. Thus, while the PCB has begun a change in its political culture, legitimizing the relationship between capitalist development and social democratization, the PCCh and other Chilean left parties, drew up a political project that aimed hegemony in conduction of a socialist society project. At the end of 1970s and during the 1980s, while the International Communist Movement was being deteriorating, the meanings of these strategies have been defining the role of each Patty on the redemocratization processes in Brazil and Chile. / El presente trabajo realiza un análisis comparativo entre las estrategias políticas elaboradas por dos partidos comunistas latinoamericanos, el Partido Comunista Brasileiro (PCB) y el Partido Comunista de Chile (PCCh). Ambos nacidos en 1922, los partidos comunistas de Brasil y de Chile, a lo largo dei sigla XX, ejercieron un importante papel en los debates de izquierda en sus países. La comparación entre esas izquierdas tiene como objetivo rescatar, mientras objeto de estudio y reflexión, elaboraciones teóricas y líneas de actuación que buscaban alcanzar el poder de manera distinta de aquello que, a partir de 1959, debido ai impacto de la Revolución Cubana, quedaría consagrado como el "modelo revolucionaria" a ser adaptado en toda América. La enorme influencia de la Revolución Cubana en el conjunto de las organizaciones de izquierda en Latinoamérica acabaría por ocultar la existencia de otras formulaciones y propuestas de acceso ai poder por parte de la izquierda latinoamericana, que estaban en curso antes de su eclosión y que permanecieron en las décadas posteriores. Monitores de LCD : caracterização dos materiais e processamento mecânico das placas de circuito impresso

Dentre os setores mais promissores da indústria está o setor de equipamentos eletroeletrônicos. Parte destes equipamentos eletroeletrônicos apresentam componentes tóxicos podendo causar danos ao meio ambiente e a população e apresentam também materiais nobres e raros que, caso não reciclados correm o risco de desaparecer nas próximas gerações. Alguns desses equipamentos já possuem rotas de reciclagem definidas, no entanto novas tecnologias como a tela de cristal líquido (LCD) estão começando a sofrer os primeiros descartes. Os monitores de LCD são compostos por uma tela de LCD, partes poliméricas e placas de circuito impresso (PCI) que são componentes de composição muito heterogênea, sendo 70% da sua massa composta de frações não metálicas e 30% correspondem a metais como cobre, chumbo, ferro, níquel, ouro e prata. Neste trabalho, monitores e telas de LCD danificados foram coletados em assistências técnicas. Estes foram desmontados e caracterizados quanto a sua estrutura, composição e periculosidade. As placas de circuito impresso foram cominuídas a granulometrias específicas e caracterizadas quanto a quantidades de metais presentes. Posteriormente, estas placas foram submetidas a ensaios de separação gravimétrica com líquidos densos e com um concentrador Mozley. Os monitores têm suas partes poliméricas compostas principalmente por termoplásticos como PC, PET, ABS e acrílico exceto nas placas de circuito impresso, onde a fração polimérica é composta por resina epóxi. A tela de vidro, fração cerâmica, foi caracterizada por FRX e MEV/EDS, sendo observada a presença de metais como índio, estanho, cobre e alumínio, todos na forma de óxido. As PCI após serem cominuídas e classificadas foram submetidas a ensaio em meio denso a fim de determinar qual granulometria apresenta melhor grau de liberação, sendo escolhido o tamanho de partícula -1,0+0,25mm. As frações afundado e flutuado foram caracterizadas através de lixiviação com água régia e posterior análise por FAAS e ICP-AES. Nos ensaios realizados com o concentrador Mozley, variando parâmetros como inclinação da bandeja, vazão de água e tempo, os resultados obtidos mostram que é possível concentrar metais nobres como ouro (92%), prata (94%) e cobre (88%) em uma fração com um baixo teor de não metais. / Among the most promising sectors of the industry is that of electronic equipment. Part of these electronic equipments has toxic components, which can cause damage to the environment and population and also have rare and precious materials which, if not recycled they are at risk of disappearing in the next generations. Some of these devices already have recycling routes defined; however, new technologies such as liquid crystal display (LCD) are starting to suffer the first discart. The LCD monitors are composed of an LCD screen, polymeric parts and printed circuit boards (PCB) which are components with a very heterogeneous composition, being 70% of its mass consisting of non-metallic fractions and 30% are metals such as copper, lead, iron, nickel, gold and silver. In this work, LCD monitors and screens were collected in technical assistance. These were dismantled and characterized for their structure, composition and dangerousness. The printed circuit boards were comminuted to specific particle sizes and characterized for their amounts of metals present. Later, these boards were analyzed by gravimetric separation with dense liquids and Mozley concentrator. The monitors have their polymeric parts mainly composed of thermoplastics such as PC, PET, ABS and acrylic except for printed circuit boards, where the polymeric fraction is composed of epoxy resin. The screen glass, ceramic fraction, was characterized by XRF and SEM / EDS, showing the presence of metals such as indium, tin, copper and aluminum, all in oxide form. The PCI after being comminuted and classified were realized assays in dense medium to determine which granulometry provides a better degree of particle release, being chosen particle size -1.0 +0.25 mm. The sunk and floated fractions were characterized by leaching with aqua regia and subsequent analysis by FAAS and ICP-AES. For assays realized with the concentrator Mozley, varying parameters such as tilt tray, water flow and time, the results show that it is possible to concentrate noble metals like gold (92%), silver (94%) and copper (88%) in a fraction with a low content of non-metals.

Hodnocení kontaminace ryb ve významných rybářských revírech / Assessment of fish contamination in important fishing grounds

The object of this thesis is the assessment of fish contamination by selected foreign substances as toxic metals, polychlorinated biphenyls and pesticides in 31 selected fishing grounds. The monitoring was realized between years 2006 and 2010. Fish were caught by electric device and fishing rods. Concentrations of Hg, Cd, Pb, PCB, DDT, HCH and HCB in fish muscle tissues were monitored. Common bream (Abramis brama) was selected as a reference species for a comparison of the monitored sites. The results of analysis were compared with valid hygienic limits. Overrun of hygienic limit for some of the monitored pollutants were observed in 18 localities. We found mercury as a major pollutant in the aquatic environment.

Assim sendo, será realizada uma reflexão crítica sobre os pressupostos teóricos e metodológicos utilizados em seu pensamento dedicado à historiografia econômica. Embora o referido autor afirmasse sempre utilizar o materialismo histórico, coube a este trabalho questionar a corroboração de uma análise integralmente marxista em seus escritos. Nesse sentido, através da investigação de alguns aspectos recorrentes em sua obra, será demonstrada a subordinação conceitual ao pensamento de Roberto Simonsen, principal líder da burguesia industrial do século XX. A fim de elucidar a constituição e o sentido da obra de Lima, buscar-se-á compor o ambiente intelectual em que o autor estava inserido. Para tanto, além dos livros de Lima, serão analisados seus artigos mais relevantes publicados na Revista Brasiliense, juntamente com os de Caio Prado Jr., outro intelectual de esquerda cuja visão sobre a formação social brasileira também apresenta aspectos análogos aos argumentos de Simonsen. Portanto, por meio da observação de evidências que comprovam uma análise mais economicista, do que efetivamente marxista, considera-se que seu pensamento fez parte do arcabouço ideológico da burguesia industrial em prol do nacional-desenvolvimentismo. / The present study analyzes the trajectory and work of Heitor Ferreira Lima, left intellectual formed in the PCB, linked the industrial bourgeoisie and defender the consolidation of the industrialization process in Brazil. Therefore, a critical reflection about the theoretical and methodological assumptions present in his thinking devoted to economic history will be held. Although this author always claimed to use historical materialism, this study questions the corroboration of a fully Marxist analysis in his writings. In this sense, through the investigation of some recurrent aspects in his work, it will be demonstrated conceptual subordination to the thought of Roberto Simonsen, principal leader of the industrial bourgeoisie of the twentieth century. In order to elucidate the formation and direction of the work of Lima, will be composed the intellectual environment that the author was inserted. To this end, in addition to Lima books, his most important articles published in Revista Brasiliense will be analyzed, along with the Caio Prado Jr., another left intellectual whose vision about the Brazilian society also has similar aspects to Simonsen‟s arguments. Therefore, by observing the evidence that support more economistic analysis, than effectively Marxist, consider that his thinking was part of the ideological framework of the industrial bourgeoisie in favor of developmentalism national.
