Metabolic diversity and synthesis of medium chain length polyhydroxyalkanoates by Pseudomonas putida LS46 cultured with biodiesel-derived by-products

The metabolism and physiology of Pseudomonas putida strain LS46 was investigated using biodiesel-derived waste streams as potential low cost substrates for production of medium chain length polyhydroxyalkanoates (mcl-PHA). Proteomic and trranscriptomic analyses were used to correlate specific gene and gene product expression patterns with differences in phenotypes of mcl-PHA biosynthesis by P. putida LS46. Growth and mcl-PHA content of P. putida LS46 were similar in cultures containing biodiesel-derived waste glycerol versus pure glycerol, and mcl-PHA synthesis occurred during stationary phase after nitrogen concentrations in the medium were exhausted. Waste glycerol cultures contained elevated concentrations of heavy metal ions, such as copper, which induced significant changes in gene expression levels related to heavy metal resistance. Several membrane-bound proteins, such as CusABC efflux and CopAB were identified and putatively play a role in regulating cellular copper concentrations. Cultures containing waste free fatty acids synthesized mcl-PHA throughout the exponential growth phase. Protein expression levels of two mcl-PHA synthases were suppressed during exponential phase growth in waste glycerol cultures, putatively via post-transcriptional regulation. Culture specific expression of monomer supplying proteins (PhaJ1 and PhaG), and sets of fatty acid oxidation enzymes were observed, and may have contributed to differences in the composition of polymers synthesized by P. putida LS46 cultured on the two substrates. Expression levels of the majority of mcl-PHA biosynthesis pathway genes were stable during active polymer synthesis in waste glycerol cultures. However, variations in protein expression levels, and in some cases their corresponding mRNAs, were observed in a number of other metabolic patheays, such as glycerol transportation, partial glycolysis, pyruvate metabolism, the TCA cycle, and fatty acid biosynthesis. These data suggest potential regulatory points that may determine carbon flux during mcl-PHA biosynthesis. Evaluation of identified genetic targets in P. putida LS46 that putatively influence mcl-PHA biosynthesis and monomer composition merit further studies. / February 2016

Optimising properties of poly(lactic acid) blends through co-continuous phase morphology

This PhD project investigated the effects of a co-continuous phase structure on the ductility of poly(lactic acid) (PLA) blends. An empirical model was used to predict the phase inversion point of the blends. The co-continuous phase morphology was first observed in polybutylene succinate(PBS)/PLA blends. With as little as 10 wt% of PBS, PBS/PLA blends achieved a dramatic improvement in ductility, over 250% elongation-to-break. Clay additives were incorporated in PBS/PLA blends. Clay was found out to affect the compatibility and co-continuous phase morphology of PBS/PLA blends. The clay dispersion was found to have an intercalated and exfoliated structure at a PBS/PLA composition ratio of 20/80. The clay particles increased the mixing conditions between the polymers by producing a finer structure, but also destroyed the co-continuous phase morphology, resulting in a substantial decrease in elongation-at-break. PLA was then blended with Poly(butylene adipate-co-terephthalate) (PBAT) to examine whether the co-continuous phase model could also be applied to other PLA-polymer blends. From the melt viscosity ratio of PLA and PBAT in the processing regime used in the study, the predicted phase inversion value was 19 wt% of PBAT. This value was verified by the results of mechanical properties, where results for elongation-to-break show a dramatic rise from around 10% up to 300% in the composition range between 10 and 20wt% of PBAT. Polyhydroxyalkanoate (PHA) was also blended with PLA and this project investigated how co-continuous phase morphology affects the blends of two brittle polymers. It was found that when PHA content ranged from 10 to 20wt%, the brittle-brittle polymer blends showed ductile behaviour due to a plane stress effect.

Students’ Perspectives about the American Pharmacists Association Career Pathway Evaluation Program for Pharmacy Professionals

Class of 2008 Abstract / Objectives: The APhA Pathway Program provides Pharm.D. candidates with direction in refining their career choices. The purpose of this study was to collect data about pharmacy student perspectives regarding which year of pharmacy school is the optimal time for students to complete the APhA Career Pathways Briefing Document. Methods: The subjects of the study were Pharm.D. candidates currently enrolled in their 1st, 2nd, or 3rd year of didactic courses at the University of Arizona College of Pharmacy. Students were provided a copy of the Pathway Program Briefing Document and asked to complete a 19-item questionnaire regarding their opinions about this information. The independent variable in the study was the class year. The dependent variable was the usefulness of the Pathway Program. Results: The overall response rate was 121 of 219 (55%) distributed questionnaires. Baseline characteristics of the three classes were similar. Statistically significant differences in responses existed between the 1st and 3rd year classes. The first year pharmacy students found the Pathway Program the most useful. Students with less work experience found the Pathway Program more useful than students with more work experience. Conclusions: First year pharmacy students had the strongest level of agreement that the Pathway Program was a useful tool. Students with less pharmacy work experience found the Pathway Program more useful than students with more extensive work experience. Based on these results, it appears that the first year of pharmacy school is the most appropriate time to utilize the program.

Effects of Phytohemagglutinin-P (PHA-P) on Bone of the Growing Rat

The effects of phytohemagglutinin-P, (PHA-P), a mitogen known to selectively stimulate cells of hematogenous or lymphoid monocytic origin, 25 and 50 mg/kg/day administered for 15 days on proximal tibiae of growing male Sprague-Dawley rats, were studied. The general effect of PHA-P was to decrease the amount of cartilage, hard tissue, and longtitudinal growth in the proximal tibial metaphysis. A decrease in longitudinal bone growth, in the number of chondrocytes, in the thickness of cartilage plate, in the metaphyseal mass of hard tissue, in the percentage of calcified cartilage core, and in the number of osteoblasts per mm of bone surface was observed. Additionally, PHA-P increased the number of osteoclasts, the number of labeled osteoclastic nuclei, and the average number of nuclei per osteoclast. There was a significant decrease in the time to the first appearance of labeled osteoclastic nuclei as the dose of PHA-P increased. Thus, PHA-P treatment leads to the dominance of osteoclastic over chondroblastic and osteoblastic activity and results in a hard tissue deficit in a growing skeleton. The data indicate that PHA-P administration selectively increases osteoclast numbers by elevating osteoclastic progenitor cell proliferation and enhancing their fusion and differentiation to osteoclasts.

The Intradermal Skin Test in the Horse: Value as a Diagnostic Modality in Equine Allergies

Recent studies have provided conflicting results in regards to equine intradermal skin testing and its use in defining causative antigens in IgE mediated diseases such as equine recurrent airway obstruction (RAO). This study was divided into two experiments. In the first experiment of this study, the hypothesis tested was normal horses would have minimal variability in the wheals formed by intradermal injection of positive control stimulants. This was evaluated by examining the repeatability of skin test wheals created by 5 concentrations of histamine, compound 48/80, and phaseolus vulgaris (PHA) within a normal horse and between 12 normal horses at 0.5 hours, 4 hours, and 24 hours post injection. Minimal variability was detected within individual horses and between 12 horses for histamine and compound 48/80 at 0.5 hours and for PHA at 4 hours. This information suggests that the intradermal injection of positive control substances is a repeatable test in normal horses. In the second experiment of this study, the hypothesis tested was normal horses react differently to intradermal injection of positive control stimulants (histamine, compound 48/80, PHA) and/or an environmental antigen (Aspergillus) in comparison to horses affected with RAO. This was evaluated by identifying differences in wheal responses between normal horses and RAO affected horses. Concentration response curves were created in normal and RAO affected horses to the aforementioned stimulants at 0.5 hours, 4 hours, and 24 hours post injection. No statistically significant differences were noted in concentration response between normal and ROA affected horses when compound 48/80 and PHA were evaluated. RAO affected horses demonstrated a greater slope at the 0.5 hour time when compared to normal horses suggesting that RAO affected horses are hypersensitive to intradermal injection of histamine. Injection of Aspergillus mix at 4000 protein nitrogen units/ml caused an intradermal wheal reaction at the 24-hour time in 4/5 RAO horses. This reaction was not noted in normal horses. This information suggests that there may be a positive relationship between causative antigens (i.e. Aspergillus) that may induce clinical RAO and positive intradermal skin test results. An additional aspect that was evaluated in both experiments involved histologic examination of skin biopsies taken from wheals created by intadermal injection of histamine, compound 48/80, PHA, and Aspergillus at various times post injection. In the first experiment, intradermal injection of histamine caused severe dermal edema and margination of neutrophils and eosinophils at 0.5 hours. Compound 48/80 demonstrated mild to modest dermal edema at 0.5 hours while PHA demonstrated severe dermal edema, hemorrhage, and lymphactic ectasia at 4 and 24 hours. PHA also demonstrated a neutophilic inflammation at 4 hours that progressed to a mixed lymphohistiocytic and neutrophilic inflammation at 24 hours. In the second experiment, no edema and modest to moderate neutrophilic inflammation was noted in normal horses after intradermal injection of Aspergillus at 24 hours. In contrast, RAO affected horses demonstrated mild to modest edema and a mild to moderate mixed inflammatory response (lympho-histocytic, neutrophilic, eosinophilic) after intradermal injection of Aspergillus at 24 hours suggesting a delayed type response. / Master of Science

A área tegmental ventral (VTA) contém neurônios dopaminérgicos do grupamento A10 e envia projeções muito densas para o estriado ventral. Esta circuitaria está crucialmente envolvida em mecanismos de recompensa. Recentemente, a organização destas projeções foi reexaminada por Ikemoto S. (Brain Res. Rev., 56:27-78, 2007), em um estudo de rastreamento retrógrado minucioso, sendo proposto a subdivisão destas projeções em um sistema dopaminérgico mesoestriatal ventromedial que inerva a concha medial do accumbens e o tubérculo olfatório medial, e um sistema dopaminérgico mesoestriatal ventrolateral que inerva o cerne e a concha lateral do accumbens e o tubéculo olfatório lateral. Afim de complementar o conhecimento destas projeções, no presente estudo elas foram examinadas com a técnica anterógrada da leucoaglutinina do Phaseolus vulgaris. Nossos resultados indicam que há um extenso embricamento dos campos terminais estriatais inervados por diferentes setores/núcleos da VTA e reforçam a noção de que as eferências da VTA podem ser subdivididas em um sistema mesoestriatal ventromedial e um sistema mesoestriatal ventrolateral. Eles revelam ainda que as projeções da VTA para o estriado ventral têm uma organização topográfica médio-lateral mais complexa do que previamente reconhecido, a faixa médio-lateral do estriado ventral inervada depende de uma combinação da região médio-lateral e dorsoventral da VTA. Assim, as regiões mais ventrais e mediais da VTA (correspondendo ao núcleo interfascicular) inervam os distritos mais mediais do estriado ventral (a concha dorsomedial do accumbens e a extremidade medial do tubérculo olfatório), e as regiões mais dorsais e laterais da VTA (correspondendo à região dorsolateral do núcleo parabraquial pigmentoso) se projetam para os distritos mais laterais do estriado ventral (o cerne lateral e a concha lateral do accumbens, o caudado-putâmen ventral e o tubérculo olfatório lateral). Por outro lado, as projeções da VTA para o estriado ventral não possuem uma organização topográfica rostrocaudal. Outro fato a ser destacado é que a organização das projeções mesoestriatais da VTA lembra o padrão das projeções córticoestriatais, sendo observado no estriado, além de um campo terminal principal, pequenos focos isolados de marcação. / The ventral tegmental area (VTA) contains dopaminergic neurons of the A10 group and sends dense projections to the ventral striatum. This circuitry is critically involved in reward mechanisms. Recently, the organization of these projections was reexamined by Ikemoto S. (Brain Res. Rev., 56:27-78, 2007) in a detailed retrograde tracing study, being proposed that these projections can be subdivided into two main systems, a ventromedial mesostriatal dopaminergic system that innervates the medial shell of the accumbens and medial olfactory tubercle, and a ventrolateral mesostriatal dopaminergic system that targets the core and lateral shell of the accumbens and lateral olfactory tubercle. In order to complement these data, in the present study the VTA mesostriatal projections were examined with a sensitive anterograde tracing technique using the Phaseolus vulgaris leucoaglutinin. Our results indicate that there is an extensive overlap of terminal fields innervated by different sectors / nuclei of the VTA and reinforce the notion that VTA efferents can be subdivided into a ventromedial and a ventrolateral mesostriatal system. They also show that the VTA projections to the ventral striatum have a mediolateral topographical organization more complex than previously acknowledged. In fact, projections along the mediolateral dimension of the ventral striatum depends on a combination of the mediolateral and dorsoventral axis of the VTA. In other words, the most ventral and medial parts of the VTA (corresponding to the interfascicular nucleus) innervates the most medial districts of the ventral striatum (corresponding to the dorsomedial shell of the accumbens and medial tip of the olfactory tubercle), and the most dorsal and lateral parts of the VTA (corresponding to the dorsolateral region of the parabrachial pigmented nucleus) project to the most lateral districts of the ventral striatum (lateral core and lateral shell of the accumbens, ventral caudate-putamen and lateral olfactory tubercle). Moreover, VTA projections to the ventral striatum do not seem to have a rostrocaudal topographical organization. It is also of note that the organization of the VTA mesostriatal projections shares features with cortico-striatal projections, in the sense that both fiber systems have a main terminal field and also give rise to small, scattered isolated foci of terminal labeling.

Potentiels des poly-hydroxyalcanoates (PHAs) bactériens pour l'encapsulation de molécules à visée thérapeutique / Potentials of bacterial Poly-HydroxyAlkanoates (PHA) for the encapsulation of therapeutic molecules

Les Poly(HydroxyAlcanoates) (PHA) sont des polymères naturels, biodégradables et biocompatibles, synthétisés par de nombreux organismes, et plus particulièrement des procaryotes. Il existe à ce jour plus de 150 types de monomères de PHA différents, accumulés chez différents genres bactériens, en tant que source d’énergie et de carbone. En effet, les granules de PHA intracellulaires sont produites en réponse à un apport en excès de sources de carbone dans l’environnement (glucides, acides gras…), couplé à une carence en éléments azotés nécessaires à la division cellulaire. De par leur caractère biodégradable et biocompatible, les PHA sont employés depuis plus de 20 ans comme biomatériaux dans les domaines pharmaceutiques et biomédicaux, notamment comme micro/nanovecteurs à visée thérapeutique. Ce doctorat met en évidence des méthodes de criblage moléculaire par PCR pour la sélection de bactéries productrices de PHA, isolées de sites hydrothermaux des océans Atlantique et Pacifique au cours de campagnes océanographiques Ifremer. Selon des protocoles de fermentation standardisés et optimisés, des polymères de poly(3-hydroxybutyrate-4-hydroxybutyrate) P(3HB4HB) d’intérêt biomédical ont été produits, puis des études taxonomiques et phylogénétiques ont été menées pour explorer la biodiversité microbienne associée aux environnements marins profonds. Ensuite, des PHA ont été modifiés par réaction thiol-ène photoactivée afin d’obtenir des copolymères hydrosolubles, adaptés pour l’enrobage de nanoparticules poreuses de type Metal-Organic Frameworks (MOF). La caractérisation physicochimique a été réalisée par différentes techniques, et notamment par SEM et STEM-EDX. Les systèmes hybrides poreux MOF-PHA ont ensuite été évalués quant à leur biocompatibilité vis-à-vis de cellules immunitaires (macrophages), par des tests de cytotoxicité et de prolifération cellulaire. Cette étude met en lumière les potentialités de cette nouvelle génération de nanovecteurs, synthétisés pour augmenter le bénéfice thérapeutique tout en minimisant les effets secondaires sur l’organisme humain. / Poly(HydroxyAlkanoates) (PHA) are natural polymers, biodegradable and biocompatible, synthesized by many organisms, especially prokaryotes. There are over 150 kinds of these polyesters, accumulated in a wide variety of bacteria as carbon and energy storage material. PHA granules are deposited intracellularly when microorganisms are cultivated in the presence of an excess of carbon source (glucids, fatty acids...) together with a nitrogenous nutrient deficiency. Due to their biodegradability and biocompatibility, PHA can be used as biomaterials in medical or pharmaceutical fields, and numerous therapeutic micro/nanovectors have already been developed over the past two decades.The present PhD research project highlighted molecular screening methods by PCR for the PHA producing Bacteria selection, isolated during Ifremer cruises from hydrothermal vents in Atlantic and Pacific oceans.According to standardized and optimized fermentation protocols, poly(3-hydroxybutyrate-4-hydroxybutyrate) P(3HB4HB) polymers of biomedical interest were produced, then taxonomic and phylogenetic studies were performed to explore microbial biodiversity associated with deep-sea environments. Next, PHA were modified by ‘click chemistry’ to obtain hydrosoluble copolymers, suitable for coating high porous Metal-Organic Frameworks (MOF) therapeutic nanoparticles. Physico-chemical characterization was performed using different techniques, and more particularly by SEM and STEM-EDX. MOF-PHA hybrid porous systems were then evaluated for their biocompatibility against immune cells (macrophages), by cytotoxicity and cellular proliferation tests. This study highlights potentials of these new generations of nanovectors, synthesized to increase the therapeutic benefit while minimizing side effects on the human body.

O trabalho é proposto visando melhorar o consumo de xilose na bactéria Burkholderia sacchari utilizando o acúmulo de PHB como modelo de produção Foi desenvolvido um processo de evolução por meio da aplicação de feast and famine e Cultivos sequenciais em fase exponencial. Foi obtida uma linhagem mutante com uma velocidade especifica de crescimento de 0,24 h -1. Foi feita uma análise de fluxos metabólicos da qual foi possível concluir que o metabolismo da xilose acontecia em sua maioria pela VP junto com a ED. Foi feito um ensaio de acumulo com carbono marcado utilizando uma solução de xilose, de 20:80 de xilose marcada 13C em todos os carbonos e xilose não marcado, para determinar quais seriam as possíveis vias metabólicas no uso da xilose por parte de B. sacharia LFM 101 e da linhagem evoluída BSEV11. Foi determinado que houve embaralhamento de carbonos, fato que só acontece quando o metabolismo da xilose e feito pela VP junto com a via ED, assim foi possível conferir a via ED como principal via para o metabolismo da xilose em B. sacchari LFM 101. / To evaluate the possibilities of improving the productivity of PHA production from xylose, evolutionary engineering techniques were applied to B. sacchari to select cells with maximum specific growth rates (max) higher than the wild type. Metabolic flux analysis was also performed to evaluate the fluxes through central pathways and the possibility of further improvements by modifying fluxes rates. The evolved strain reached a max of 0.24 ± 0.01 h-1 at the end of the evolutionary process. Strains were submitted to bioreactor experiments. A metabolic network of the strain was usedn to determine the possible distribution of metabolic fluxes. A total of 19 elementary modes were obtained. It was concluded that the metabolism of xylose occurred mostly by VP along with the ED. The ED pathway has the major activity going on in a cyclic way. It was also performed a 13C labeled xylose assay, in which it was possibly to confirm the obtained results from the metabolic flux analyses.

Construção de biblioteca metagenômica e prospecção de genes para a síntese de polihidroxalcanoatos / Metagenomic library construction for PHA synthase screening

Os microrganismos constituem dois terços da diversidade biológica na Terra, no entanto, muitos deles não podem ser cultivados por técnicas tradicionais. Portanto, o acesso a esta diversidade tem sido feita através da utilização de técnicas independentes de cultivo. Diante deste panorama, a metagenômica apresenta-se como uma alternativa, pois dispensa a necessidade de cultivo. Tal técnica possibilita inclusive a identificação e utilização do potencial metabólico destes organismos para o desenvolvimento de novos processos e produtos. Os polihidroxialcanoatos (PHAs) são poliésteres bacterianos, acumulados intracelularmente em forma de grânulos, cujas propriedades são similares a de alguns plásticos de origem petroquímica. O objetivo deste trabalho foi identificar e avaliar a diversidade de genes relacionados à produção de PHAs em bibliotecas metagenômicas de solo. A prospecção realizada resultou na identificação de clones contendo o gene phaC. De uma forma geral, pôde-se concluir que ainda há uma grande diversidade deste gene a ser descoberta no ambiente estudado. / Microorganisms constitute two third of the Earth\'s biological diversity, however, many of them cannot be cultured by standard techniques. Therefore, access to this diversity has been achieved through the use of culture-independent techniques. Facing this scenario, the metagenomic presents itself as an alternative, since it eliminates the need for cultivation. This technique also allows the identification and use of the metabolic pathways of these organisms to develop new processes and products. The polyhydroxyalkanoates (PHAs) are bacterial polyesters accumulated as granules, whose properties are similar to some plastics of petrochemical origin. The aim of this work was to identify and access the diversity of genes related to PHAs production in soil metagenomic libraries. The screening resulted in the identification of clones containing the phaC gene. In a general way, it was concluded that there is still a considerable diversity of this gene to be discovered in the study environment.

Assim, as regiões mais ventrais e mediais da VTA (correspondendo ao núcleo interfascicular) inervam os distritos mais mediais do estriado ventral (a concha dorsomedial do accumbens e a extremidade medial do tubérculo olfatório), e as regiões mais dorsais e laterais da VTA (correspondendo à região dorsolateral do núcleo parabraquial pigmentoso) se projetam para os distritos mais laterais do estriado ventral (o cerne lateral e a concha lateral do accumbens, o caudado-putâmen ventral e o tubérculo olfatório lateral). Por outro lado, as projeções da VTA para o estriado ventral não possuem uma organização topográfica rostrocaudal. Outro fato a ser destacado é que a organização das projeções mesoestriatais da VTA lembra o padrão das projeções córticoestriatais, sendo observado no estriado, além de um campo terminal principal, pequenos focos isolados de marcação. / The ventral tegmental area (VTA) contains dopaminergic neurons of the A10 group and sends dense projections to the ventral striatum. This circuitry is critically involved in reward mechanisms. Recently, the organization of these projections was reexamined by Ikemoto S. (Brain Res. Rev., 56:27-78, 2007) in a detailed retrograde tracing study, being proposed that these projections can be subdivided into two main systems, a ventromedial mesostriatal dopaminergic system that innervates the medial shell of the accumbens and medial olfactory tubercle, and a ventrolateral mesostriatal dopaminergic system that targets the core and lateral shell of the accumbens and lateral olfactory tubercle. In order to complement these data, in the present study the VTA mesostriatal projections were examined with a sensitive anterograde tracing technique using the Phaseolus vulgaris leucoaglutinin. Our results indicate that there is an extensive overlap of terminal fields innervated by different sectors / nuclei of the VTA and reinforce the notion that VTA efferents can be subdivided into a ventromedial and a ventrolateral mesostriatal system. They also show that the VTA projections to the ventral striatum have a mediolateral topographical organization more complex than previously acknowledged. In fact, projections along the mediolateral dimension of the ventral striatum depends on a combination of the mediolateral and dorsoventral axis of the VTA. In other words, the most ventral and medial parts of the VTA (corresponding to the interfascicular nucleus) innervates the most medial districts of the ventral striatum (corresponding to the dorsomedial shell of the accumbens and medial tip of the olfactory tubercle), and the most dorsal and lateral parts of the VTA (corresponding to the dorsolateral region of the parabrachial pigmented nucleus) project to the most lateral districts of the ventral striatum (lateral core and lateral shell of the accumbens, ventral caudate-putamen and lateral olfactory tubercle). Moreover, VTA projections to the ventral striatum do not seem to have a rostrocaudal topographical organization. It is also of note that the organization of the VTA mesostriatal projections shares features with cortico-striatal projections, in the sense that both fiber systems have a main terminal field and also give rise to small, scattered isolated foci of terminal labeling.
