Investigating the role of Integrin Linked Kinase in mammary epithelial cell differentiationRooney, Nicholas January 2014 (has links)
Epithelial cell adhesion to the surrounding extracellular matrix (ECM) is necessary for their proper behaviour and function. During pregnancy and lactation mammary epithelial cells (MECs) require signals imparted by specific β1 integrin-laminin interactions for their functional differentiation in response to Prolactin (Prl) and for the correct formation of polarised secretory acini. Downstream of β1 integrin (β1Itg), the scaffold protein Integrin Linked Kinase (ILK) has been identified as the key signal transducer that is required for both Prl driven lactational differentiation and the establishment of apico-basal polarity in MECs. ILK is a multifunctional adaptor protein that links integrins to the actin cytoskeleton and Rho GTPases such as Rac1. ILK forms a ternary IPP (ILK-PINCH-Parvin) complex with PINCH and Parvins, which are central to its adaptor functions. However, it is not known which of ILKs interacting partners are important for controlling tissue-specific gene expression, or what acts downstream of the IPP complex. In this thesis I have now established that inducible ILK deletion in MECs from ILKfl/flCreER mice, prevents phosphorylation of Stat5 leading to a failure of Prl induced milk expression. In addition I have established a 3-dimensional culture model using the EpH4 mammary epithelial cell line, which respond to Prl treatment and form polarised acini similar to primary cells. In these cells knocking down β1Itg and ILK by lentiviral shRNA delivery was confirmed to have a profound effect on β-Casein production. Expression of ILK mutants that disrupt its protein-protein interactions, showed that mutation of K220 and E359 in the kinase domain also reduced milk production. This means that ILKs kinase domain is important for MEC differentiation, and suggests that Parvin binding (which is disrupted by these mutations) is key in mediating ILKs differentiation functions. Using a complimentary shRNA approach, knockdown of the βParvin binding Rac guanine nucleotide exchange factor αPix also prevented MEC differentiation. This identified for the first time that αPix is required for differentiation and suggests a route by which ILK, via it’s interaction with Parvin, can link integrins to αPix and Rac activity. Interestingly, αPix depletion did not disrupt the IPP complex or polarity, suggesting that αPix represents a differentiation specific bifurcation point in β1Itg-ILK adhesive signalling. Together, this work has helped to establish how ILK is involved in MEC differentiation and has identified a new role for the downstream Rac GEF αPix. In addition, this work contributes to our understanding of the molecular mechanisms by which cell adhesion regulates fundamental cell biological behaviours.

The other equipments of Cisco, which are improving the level of security, are also mentioned. The practical part of this diploma thesis is composed of networks connections and configuration of system consisting of router, PIX firewall and switch. It also includes the detailed procedure and description of configuration of network equipments. The focus is put on minimalization of threats and elimination of DoS attacks.

This thesis is about the creation of a new game framework named Pix and a comparison between it and the game engine Godot. The purpose is to create a game framework that focuses on making the workflow streamlined for the pro- grammer and at the same time giving the programmer a lot of creative power. Pix uses C# as programming language which runs on the .NET Core platform. Pix is built with Entity Component System (ECS) design which means that the design is data-oriented and that it focuses on composition. Thus the ECS design brings with it a clear separation between logic and data. The ECS design also makes it easy to write multithreaded code and to serialize the game world state. The comparison against Godot shows clear differences between how you use them and the philosophy about how the users become familiar with them. The obvious difference between them is that the main time spent working with Godot will be in the graphical user interface. In Pix on the other hand you will only work in a text-editor or an IDE if you do not create your own editor. The coding in Godot is done primarily with the script language GDScript. It is made for creating common game functionality but can’t be used to extend the engine with new advanced functionality. Thus to integrate new more advanced features Godot will need to be recompiled. Being forced to recompile the engine to add new advanced features results in that extending the engine can both be time consuming and challenging. In Pix there is no difference between coding game functionality and extending the framework. This equality in Pix between coding game functionality and extending the framework brings with it that when you can make a game in Pix you can also extend the engine with more functionality. The differences results in that Godot is simpler to learn but also gives the regu- lar user less power. Pix is the opposite of that. I you look at the workflow of us- ing Pix and combine that with the technologies Pix is built upon you can see that Pix differentiates itself from the already existing alternatives. The work- flow Pix provides and the technologies it is built upon shows that Pix can be a good alternative for creating complex games that doesn't need the most ad- vanced graphical features. / Rapporten handlar om skapelsen av ett nytt spelramverk namngivet Pix som jämförs mot den existerande spelmotorn Godot. Syftet är att skapa ett spelram- verk som fokuserar på att göra arbetssättet smidigt för programmeraren samtidigt som det ger mycket kreativ makt till programmeraren. Pix använder C# som programmeringsspråk vilket körs på .NET Core plattformen. Det är upp- byggt med en Entity Component System (ECS) design vilket betyder att det är en dataorienterad design med ett fokus på komposition. Det medför en klar separation mellan logik och data. ECS designen medför också att serialisering samt använda sig av flera trådar blir väldigt enkelt. Jämförelsen mot spelmotorn Godot visar tydliga skillnader i hur man använder spelmotorn/spelramverket samt tydliga skillnader i filosofin kring hur en användare blir bekant med spelmotorn/spelramverket. De tydliga skillnader är hur större delen av arbetstiden i Godot spenderas i Godots grafiska gränssnitt medans Pix kan kontrolleras endast från kod om man inte skapar en editor själv. Programmering i Godot görs med skriptspråket GDScript. Det är gjort för att skapa vanlig spel funktionalitet och inte utöka spelmotorn med ny funktionalitet. Det betyder att hela motorn måste kompileras om för att lägga till mer avancerad funktionalitet vilket kan vara tidskrävande. I Pix är är det samma sak att programmera spel funktionalitet och utöka ramverket med ny funktionalitet. Det medför att så fort som man kan programmera ett spel i Pix kan man också utöka ramverket med mer funktionalitet. Det resulterar i att Godot är enklare att lära sig men samtidigt ger mindre makt till den vanlige användaren medans Pix är tvärtom. Kollar man på hur man använder sig av Pix samt de teknologierna Pix är uppbyggt med så kan man se att det skiljer sig från redan existerande alternativ. Det leder till att Pix kan vara ett gott alternativ för komplexa spel som inte kräver den mest avancerade grafiken.

Nagrinėjama skirtingais medicinos informatikos standartais ar jų skirtingomis versijomis sąveikaujančių asmens sveikatos priežiūros įstaigos informacinių sistemių integracija. Pagal Kauno Dainavos poliklinikos informacinių sistemų integracijos uždavinius pasirinktas artimiausias teorinis integracijos modelis. Jo pagrindu parengti tarpinio (integracinio) IS sluoksnio/modulio funkciniai reikalavimai, užtikrinantys vienareikšmius paciento sąryšius su jo sveikatos duomenimis, esančiais skirtingose sistemose. Panaudojant medicinos informatikos standarto HL7 bibliotekas 2-ai ir 3-iai versijoms, sukurtas ir sėkmingai išbandytas tarpinio (integracinio) IS sluoksnio/modulio prototipas, leidžiantis automatiškai formuoti procedūrų užsakymo identifikatorius ir, jų pagrindu, nuorodas į duomenų archyvą. Integralumas tarp 2-os ir 3-os versijos sutampančių komponentų užtikrinamas trigeriais: įterpiant, keičiant bei trinant įrašus iš vietos versijos komponento, jis bus atitinkamai modifikuotas ir kitoje versijoje. Toks duomenų sluoksnio realizavimas leidžia sumažinti verslo logikos sluoksnio sudėtingumą, kai naudojama trijų lygių architektūra, bei pačių duomenų transformacijų skaičių. / The research is devoted to an integration of health information systems, when the interoperability should be utilised using different health informatics standards or their versions. To meet the integration goals, the theoretical model, closest to a particular situation of the Dainava outpatient clinic in Kaunas, had been chosen. Based on the model, the functional requirements for the middleware (integration) layer had been defined in order to ensure unambiguous links between patient data, stored in different systems. The middleware software prototype had been developed using messaging libraries for the HL7 versions v2 and v3. The middleware was successfully tested for automatic generation of orders for procedures and links to the procedure result data in the archive. The integration of the HL7 v2 and v3 coincidental components is achieved using trigger events for synchronous data manipulations. Such rules, implemented at the data layer, leads to less complex logics at business process layer and to lower number of data transformations. After automatic orders generation from referrals model implementation, we had to produce Worklist management system prototype to ensure full control of coming patient examinations. In this scenario DICOM based data models and terms had been chosen to use.

A Novel Role for Calpain 4 in Podosome AssemblyDowler, THOMAS 27 September 2008 (has links)
Podosomes are adhesive and invasive structures which may play an important role in numerous physiological and pathological conditions including angiogenesis, atherosclerosis, and cancer metastasis. Recently, the cysteine protease m-calpain (m-Capn) has been shown to cleave cortactin, an integral component of the podosomal F-actin core, as well as various proteins found in the peripheral adhesive region leading to the disassembly of these dynamic structures. In this study, I investigated whether Capn plays a role in the formation of podosomes downstream of c-Src. I show that: 1) phorbol-12, 13-dibutyrate (PDBu) as well as c-Src-Y527F expression induces podosome formation in mouse embryonic fibroblasts; 2) PDBu- and constitutively active c-Src-induced podosome formation is inhibited by the knockout of the m- and µ-Capn small regulatory subunit Capn4 in mouse embryonic fibroblasts (Capn4-/-), but is partially restored by re-expression of Capn4; 3) Capn4 localizes to podosomes; and 4) Inhibition of m- and µ-Capn proteolytic activity by the cell permeable calpain inhibitors has little effect on the formation of podosomes downstream of active c-Src. I conclude that Capn4 may play a role in the assembly phase of podosomes independent of calpain proteolytic activity. Work done in collaboration to determine a possible mechanism of action for the role of Capn4 in podosome assembly indicates that a possible binding partner of Capn4, β-PIX, co-localizes with, and shows in vivo association with Capn4. Furthermore, β-PIX and Capn4 bind directly in vitro in the presence of Ca2+. Case study of a contract system : considering pulp prices from 1996-2006Janstad, Tobias January 2007 (has links)
Södra Cell sells 1 900 000 ton pulp every year. Of this 490 000 tonne is sold with a contract system based on a pricing index called PIX NBSK. This index was started in 1996 and reflects the price of pulp from conferious forest. We study the NBSK PIX value of softwood from October 1996 to December 2006. People working in this branch known that there is strong periodicity in the prices. We use predictive analysis to see if clients can benefit from the periodicity and use the options in the contract system Södra offers today. We conclude that a drawback for the current contract system is that there are too many contracts in proportion to the duration time that is one year for all contracts. Using a time series model called ARMA we make successfull predictions the price difference between two contracts. Based on this prediction we change between these contracts, reducing the price with 0.81% in mean during 1997-2006. Due to the total turnover, if all clients would used such predictions during 1997-2006 Södra's income would have been reduced with 2.77 million USD a year in mean. The prices used before PIX are called list prices. The list prices seem to behave like the PIX index. Supposing that the same contract system we see in PIX today was used 1975-2006 with the list price as the base index I made a prediction of the list prices from 1986-2006. Thanks to my predictions, if I had been a client during this period and under mentioned considerations I would have been buying pulp to a price reduced with 0.57%. If clients had known the PIX between 1996-2006 in say 1995 Södra's contract system based on PIX would give them a price reduction that were 1.5% in mean during 1996-2006. Price reduction is not possible all years, but when it occurs it can be as big as 3% of the price. Suppose the clients always choose the contract with the lowest price and thereby get a reduced price over time. Then with 95% probability over a long period the price reduction is somewhere in between 0.4-2.7%. To strangle this price reduction possibility for the clients there are two ways to go: either reduce the number of contracts or extend the duration time of the contracts. To find a suitable duration time, we do spectral density estimation to get indications of which periods that are most important. From this we see that PIX index has a period of five years, wavelet approximated PIX index has 3.4 years and the list prices has a period of 5.6 years. This indicates that current duration time one year is too short. Therefore if it wouldn't effect Södra's clients, an extension of the duration time from one to five years would be good. If Södra don't extend the duration time of the contracts my recommendation is to have fewer contracts. The possibility to change between the contracts ''average last three months'' and ''average current month'' every other year is the weakest point of today's system. Therefore I recommend stop selling pulp to the contract ''average PIX last three months''. We can't prove any longterm difference between the contracts. If Södra chooses to have just one contract from this point of view it does not matter which one they choose. However, it seems like a good idea to follow the global market and therefore I recommend to choose ''average PIX current month'' rather than ''average PIX last three months'' which lags behind the market front.
