Estudo comparativo do efeito da adição de proteases fungica e bacteriana nas caracteristicas reologicas da massa e na qualidade do biscoito tipo cracker

O enfraquecimento do glúten resultante, torna a massa adequada às etapas de laminação, formação e cozimento. Esta pesquisa objetivou estudar os efeitos da adição de proteases comerciais, fúngica e bacteriana, nas variáveis do processamento de crackers, nas propriedades da massa, e na qualidade do produto final. Para a obtenção do cracker foram desenvolvidas a formulação e as condições de processo em laboratório, usando-se o procedimento convencional esponja e massa, no qual os tempos de fermentação da esponja e da massa foram estabelecidos em 18 e 6 horas, respectivamente. Com relação as variáveis do processo, a adição das proteases fúngica (400 e 500ppm) e bacteriana (200 e 250ppm), diminuíram em 20% o tempo de mistura da massa, estabelecido no processo convencional, sem adição de proteases. Os tempos de fermentação da esponja e da massa foram também reduzidos em 33,3 e 50%, respectivamente, o que representou a redução de 9 horas no tempo total de produção de crackers (24 horas). A adição de proteases provocou mudanças nas massas fermentadas, medidas por parâmetros determinados no extensógrafo Brabender, pH, viscosidade e solubilidade das proteínas. As modificações reológicas sofridas, principalmente na elasticidade da massa, durante 18 horas de fermentação da esponja no processo convencional (sem adição de proteases) foram similares às obtidas nas massas com adição de protease fúngica (500ppm) e bacteriana (250ppm), durante 12 horas de fermentação da esponja. A presença das proteases na esponja, nas concentrações estudadas, causaram o abaixamento gradual do pH após 4, 8 e 12 horas de fermentação da esponja a valores próximos, porém superiores aos encontrados no processo convencional (sem adição de enzimas),comparando-se os mesmos tempos de fermentação. Com relação ao processo convencional, os resultados mostraram que a adição de proteases na esponja reduziu a viscosidade e aumentou a solubilidade das proteínas nas massas, a medida que o tempo de fermentação aumentou. Comparando-se a ação das proteases após 12 horas de fermentação da esponja, concluiu-se que a enzima bacteriana (250ppm) foi a mais eficaz na redução da viscosidade dos extratos de proteínas, enquanto que a enzima fúngica (500ppm) foi a mais efetiva no aumento da solubilidade das mesmas. A qualidade dos crackers foi avaliada por medidas físicas (peso, comprimento, largura, espessura, volume e densidade) e por análises instrumentais (cor e textura). As médias das medidas físicas avaliadas nos crackers elaborados nos processos T3 (com protease fúngica, 500ppm) e T5 (com protease bacteriana, 250ppm), não diferiram entre si ao nível de 5% de significância, pelo teste de Tukey, com exceção do volume e densidade. Em relação ao processo TI (processo convencional), as médias dos pesos dos crackers não diferiram dos valores obtidos nos processos T3 e T5. Entretanto, as demais médias das medidas físicas apresentaram diferenças significativas. As massas contendo 500ppm de protease fúngica (T3) produziram crackers com maior volume e menor densidade, em relação aos do processo TI e T5. A análise das medidas dos parâmetros de cor no sistema L, a*, b* dos crackers processados com protease fúngica (T3) foram similares as obtidas no processo convencional (TI). No entanto, foi observado que os biscoitos obtidos nos três tratamentos apresentaram aspecto e coloração atraentes. A avaliação instrumental da textura dos biscoitos, obtida no analisador de textura T A-XT2 mostrou que as médias dos valores de dureza, nos tratamentos com proteases (T3 e T5), não diferiram entre si, ao nível de 5% de significância, pelo teste de Tukey, sendo porém diferentes (menores) das médias obtidas no processo convencional (T I). Portanto a adição de protease fúngica e bacteriana melhorou a textura dos biscoitos, considerando-se que a menor dureza é um atributo de qualidade desejável nos crackers / Abstract: During the long period of sponge fermentation, on the crackers' processing, the occurrence of several transformations causes the falling or the pH and the resistance to the extension of the dough. The resulting gluten weakness turns the dough appropriate for the steps of lamination, formation and baking. This research aimed to study the effects of the addition of commercial, fungal, and bacterial proteases in variables of the crackers' processing, in dough's properties and in the quality of the final product. In order to obtain the crackers, formulation and appropriate conditions of the process were developed in laboratory, using the conventional procedure of sponge and dough, allowing fermentation to take place on sponge and dough during 18 and 6 hours, respectively. In relation to the variables of the process, the addition of fungal (400 and 500ppm) and bacterial (200 and 250ppm) proteases reduced in 20% the time of the dough mixture, as established in the conventional process without the proteases addition. The period of the sponge and dough fermentation was reduced in 33.3 and 50%, respectively, representing a reduction of 9 hours in the total period of production of crackers (24 hours). The addition of proteases provoked changes in the fermented dough, measured by the rheological characteristics determined on the Brabender extensograph, pH, viscosity and solubility of proteins. The rheological changes occurred, mainly in the elasticity of the dough during 18 hours of sponge fermentation in the conventional process (without addition of proteases) were similar to those obtained in dough with addition of fungal (500ppm) and bacterial (250ppm) proteases during 12 hours of sponge fermentation. The presence of both types of proteases (fungal and bacterial) in the sponge, in the studied concentrations, caused the gradual decline of pH after 4.8 and 12 hours of the sponge fermentation to values close to but higher than the ones found in the conventional process (without addition of enzymes), comparing the same times of fermentation. In relation to the conventional process, the results showed that the addition of proteases to the sponge reduced the viscosity and increased the solubility of the proteins in the dough, as the time of fermentation increased. Comparing the action of the proteases after 12 hours of the sponge fermentation, it was concluded that the bacterial enzyme (250ppm) was the most effective in the reduction of the protein viscosity, and the fungal enzyme (500ppm) was the most effective in the increase of their solubility. The quality of the crackers was evaluated by physical measurements (weight, length, width, thickness, volume and density) and by instrumental analysis (colour and texture). The average of the physical measurements of the crackers made in the processes T3 (with fungal protease, 500ppm) and T5 (with bacterial protease, 250ppm) did not differ among each other at the 0.05 significance level, using the Tukey test, with exception to volume and density. In relation to the process TI (conventional process) the average of the variables of weight of the crackers did not differ from the values obtained in the processes T3 and T5. Nevertheless, the remaining averages of the physical measurements presented significant differences. The dough which contains 500ppm of fungal protease (T3) produced crackers with more volume and less density in relation to the processes TI and T5. The analysis of colour parameter measurements in the system L, a *, b* of the crackers processed with fungal protease (T3) were similar to the ones obtained in the conventional process (T I), but the biscuits obtained by the three treatments presented attractive colour and appearance. The instrumental evaluation of the texture of the biscuits through the values of hardness obtained by the T AXT2 Texture Analyzer, indicated that the averages of the values of hardness in treatments with proteases (T3 and T5) did not differ among each other, at the 0.05 significance level, using the Tukey test, but they indicate differences in the averages obtained in the conventional process (T I). O conhecimento a respeito dos fatores que de fato definem a patogenicidade de Acanthamoeba é limitado. A atividade proteolítica apresentada por isolados de Acanthamoeba tem sido associada a funções importantes na patogênese de doenças como ceratite e encefalite granulomatosa amebiana. O presente trabalho avaliou os perfis de atividade proteolítica de meios condicionados de Acanthamoeba isoladas de casos clínicos e do ambiente em gel de zimograma, com o objetivo de caracterizar as proteases secretadas e relacioná-las à capacidade patogênica apresentada pelos isolados nos testes in vivo e in vitro. Os isolados avaliados secretaram quantidades e tipos de proteases diferentes entre si, as quais não foram associadas à origem do isolado, genótipo ou espécie e nem à patogenicidade in vivo de Acanthamoeba. Os modelos in vivo aplicados foram úteis para avaliar a capacidade do mesmo isolado em estabelecer infecções uculares olhos e sistêmicas e diferenciar os isolados quanto ao grau de virulência. Além disso, demonstramos que cepas de origem ambiental são capazes de estabelecer infecções oculares e sistêmicas em animais, destacando o risco que estes protozoários podem oferecer à saúde humana. Em conjunto, nossos dados indicam que, apesar de importantes, testes fenotípicos e genotípicos para verificar diferenças entre isolados não podem prever a virulência dos mesmos, sendo necessário associar estes dados a testes in vivo. / The knowledge about the factors that determine the pathogenicity of Acanthamoeba is limited. The proteolytic activity presented by Acanthamoeba isolates has been associated with important roles in the pathogenesis of diseases such as keratitis and granulomatous amoebic encephalitis. This study evaluated the profiles of proteolytic activity of Acanthamoeba-conditioned medium from clinical and environmental isolates in zymogram gels, in order to characterize the proteases secreted and relate them to the pathogenic capacity demonstrated by the isolated in in vitro and in vivo tests. The isolates secreted different amounts and types of proteases, which were not associated with the source of the isolate genotype or species, and in vivo pathogenicity of the Acanthamoeba. The in vivo models applied were useful to evaluate the ability of the same strain to establish systemic and eye infections, and to differentiate them in the degree of virulence. Furthermore, we demonstrated that isolates of environmental origin are able to establish ocular and systemic infections in animals, highlighting the risk that this protozoan can offer to human health. Furthermore, our data indicate that, despite important, phenotypic and genotypic tests for detecting differences between isolates cannot predict the virulence of the Acanthamoeba, being necessary to link these data to in vivo tests.

Nosso grupo ja purificou e caracterizou inibidores de calicreina plasmatica humana (CeKI), elastase (CeEI) e catepsina B (CeCBI) presentes nas sementes de Caesalpinia echinata. Depois dos inibidores, nos interessamos em estudar as peptidases. As proteinas das sementes foram extraidas em tampao Tris 0,02 M pH 7,5. A peptidase foi purificada do extrato por cromatografias de troca ionica, filtracao em gel e interacao hidrofobica. Alguns substratos cromogenicos foram testados e essa enzima apresentou alta atividade na hidrolise de H-D-Pro-Phe-Arg-pNan (Km = 55,7 μM, Vmax = 0,319 μmol min-1 mg proteina-1) e Bz- Ile-Glu-Gly-Arg-pNan (Km = 297 μM, Vmax = 0,544 μmol min-1 mg proteina-1). A enzima foi inativada por inibidores de serino-endopeptidases do tipo da tripsina (TLCK e benzamidina), mas nao por inibidores de cisteino (E64), metalo (EDTA) e aspartilendopeptidases (pepstatina A). A peptidase apresentou pH otimo entre 7,1 e 9,2, e se manteve estavel entre 30 e 45oC, por 30 min, perdendo totalmente a atividade acima de 55oC. As caracteristicas da enzima purificada permitem a classifica-la como uma serino-endopeptidase / Some proteases found in different longevity stages of the leguminous seeds play an important role on their viability. Caesalpinia echinata seeds contain high amounts of proteins, including enzymes and their inhibitors. When stored in laboratory environmental for one month or at 4ºC for 18 months, these seeds lose their viability. We had already identified kallikrein (CeKI) and elastase (CeEI) inhibitors on stored and freshly harvested C. echinata seeds. Now, we are interested in studying different proteolytic activities in these seeds, in several longevity stages, and purifying and characterizing one of them. Proteins from green seeds, freshly harvested or 18-months-stored were extracted on 20 mM Tris buffer pH 7.5. The best amydolytic activity was found in the older seeds.The peptidase was purified from the extracts by ion exchange, gel filtration and hydrophobic interaction chromatographies. Several chromogenic substrates were tested and this protein, presented high enzyme activity on the hydrolysis of Bz-Ile-Glu-Gly-Arg-pNan (Km=0.268 mM, Vmax=0.334 nmol/min). The enzyme was inactivated by serine peptidase inhibitors (AEBSF, SBTI and CeKI), but not by cystein (E-64) and metallo peptidase (EDTA) nor by elastase (CeEI) inhibitors. Esses aditivos são normalmente usados para enfraquecer o glúten, tornando a massa adequada ás etapas de laminação, formação e cozimento. No Brasil, esses problemas vêm sendo solucionados através da adição de agentes redutores como os sulfitos e derivados, especialmente o bissulfito de sódio. Entretanto, o uso desses aditivos em produtos de panificação não é permitido pela legislação brasileira de alimentos. O objetivo deste trabalho foi estudar a possibi1idade de se utilizarem proteases comerciais de origem fungica ou bacteriana como aditivos naturais em substituição ao bissulfito de sódio. Foram selecionados os parâmetros reológicos resistência à extensão e resistência máxima, determinados no extensógrafo Brabender, como indicadores da proteólise na massa. A protease bacteriana estudada apresentou uma ação mais eficaz do que a fungica na hidrólise do glúten nas condições analisadas. A influência da concentração da enzima, tempo de ação, pH e temperatura na atividade da protease bacteriana nas massas de biscoito foi avaliada através de testes extensográficos. Messe estudo, foram estabelecidas as condições de processamento de bicoitos em laboratório, que foram posteriormente adaptadas às condições industriais para a produção de biscoitos tipo "Maizena". Os resultados dos testes de laboratório e industriais mostraram ser tecnologicamente viável a produção de biscoitos com protease bacteriana com caracteristicas físicas e sensoriais similares àquelas tradicionalmente obtidas com bissulfito de sódio. / Abstract: The necessity of the use of dough conditioners in the industrial production of hard semi sweet cookies is a. consequence of the uti1ization of wheat flours with technological characteristics inadequate to the processing. These additives are normally used to weaken the gluten, rendering the dough adequate to the laminating, forming and baking stages. In Brazil, these problems have been solved through the addition of reducing agents such as sulfites and derivates, especial1y sodium bi sulfate. However, the use of these additives in baking products is not permitted by Brazilian food legislation. The aim of this work was to study the possibility of using commercial proteases from fungal or bacterial sources as natural additives in substitution for sodium bisulfite. The rheological parameters resistance to extension and maximum resistance, measured with the aid of the Brabender extensigraph, were selected as indicators of dough proteolysis. The bacterial protease studied showed a more effective- action than the fungal one in the hydrolysis of the gluten under the analyzed conditions. The influence of enzyme concentration, duration of action, pH and temperature on the activity of the bacterial protease in cookie doughs was determined by means of extensigraphic tests. In this study, laboratory processing conditions were established, which were afterwards adapted to the industrial conditions of production of "Maizena" type cookies. The results of the laboratory and industrial tests have shown it to foe technological1y feasible to produce cookies with the use of bacterial proteases presenting physical and sensory characteristics similar to those traditionally obtained with the sodium bisulfite. / Mestrado / Mestre em Tecnologia de Alimentos

No queijo Prato este processo corresponde a um período de aproximadamente 40 dias. Este produto tipicamente brasileiro vem sofrendo uma descaracterização progressiva devido principalmente a alterações no processo de cura, visando diminuir os custos. Diante de tal problema a alternativa mais viável consiste na aceleração da maturação, e dentre os métodos empregados, a adição de enzimas proteolíticas é o de mais fácil aplicação prática. Neste trabalho, foram avaliados os parâmetros tecnológicos de fabricação e as variáveis envolvidas na cura do queijo tipo Prato obtido pela adição de enzimas proteolíticas. Foram estudados paralelamente queijos fabricados pelo método tradicional e obtidos com três concentrações de enzima (25, 15 e 10 mg/kg leite) adicionadas à massa antes da pré prensagem. A avaliação da proteólise foi efetuada pelos teores de Nitrogênio (N) solúvel a pH 4,6 e em ácido tricloroacético (TCA) 12%, pela determinação da concentração de tiro sina e triptofano, por reação colorimétrica com ácido trinitrobenzeno sulfônico (TNBS) e ninidrina-cádmio e por eletroforese nos queijos com 1, 7, 14,21,28 e 40 dias de maturação. Os atributos de textura e sensoriais também foram avaliados. O uso da enzima Neutrase® acelerou a proteólise do queijo Prato. Os queijos fabricados pelo método modificado apresentaram já no primeiro dia de maturação e durante todo período da mesma, um índice de extensão (% NS-pH 4,6/NT) e de profundidade (% NS TCA 12%/NT) significativamente maiores do que aqueles fabricados pelo método tradicional, nos processos com adição de 25, 15 e 10 mg enzima/kg leite. Os queijos modificados do processo A (25mg enzima/kg leite) apresentaram um índice de extensão aos 7 dias (17,32%) mais elevado do que o correspondente com 30 dias conforme descrito na literatura (14,0 - 14,5%), ao passo que no processo B (15mg enzima/kg leite) o valor obtido para os queijos com um dia (16,02%) já foi maior. No processo C (10mg enzima/kg leite) um valor próximo (15,61%) àquele correspondente a 30 dias na literatura foi alcançado com 14 dias de maturação. A adição de enzima Neutrase® à massa dos queijos modificados não acarretou perdas no rendimento. Nos processos A e B houve um decréscimo do índice de firmeza até o 210 dia de maturação, correspondendo à primeira fase da maturação onde a quebra da ?S1-caseína promoveu um amolecimento da massa. A região analisada (periferia e centro) influenciou a textura dos queijos. O índice de firmeza nas fatias retiradas da região mediana foi menor do que aquele obtido nas fatias da ponta do queijo. O perfil eletroforético dos queijos mostrou que a adição de enzimas levou a diferenças quantitativas nas frações protéicas dos queijos modificados quando comparados aos tradicionais. Houve uma maior degradação das frações ?S1 e ?-caseínas, com acúmulo maior de alguns produtos de degradação destas duas frações nos queijos modificados. A degradação da fração ?-caseína nos queijos modificados foi maior do que a da ?S1-caseina / Abstract: The ripening process of cheese is a very complex process that involves the development in many different ways of a great variety of flavor compounds that affect the aroma and taste of the finished product. Throughout the ripening process, the protein content undergoes a considerable number of changes that influence its physical and structural properties. The ripening of Prato cheese is completed after about 40 days. In recent times, however, this typically Brazilian product has gradually lost some of its main characteristics due to changes introduced in the ageing procedure by many cheese plants to reduce production costs. In view of the need to incorporate the economic aspects of cheesemaking, the most viable alterative consists in accelerating the ripening process. From a technological standpoint, the addition of proteolytic enzymes is the technique that offers the greatest facility to be implemented in practice. The present research project aimed at evaluating the technological manufacturing parameters and the variables involved in the ageing of Prato cheese obtained with the incorporation of a proteolytic enzyme (Neutrase®). Parallel experiments were conducted to study cheese made without enzyme (traditional method) and cheeses made with the incorporation of three different enzyme concentrations (25, 15 and 10 mg enzyme/kg milk) in the raw curd prior to the pre-pressing stage (modified method). Proteolysis after 1, 7, 14, 21, 28 and 40 days ripening was evaluated by determining: 1) the content of soluble Nitrogen (N) at pH 4,6; 2) the content of soluble nitrogen in a 12% trichloroacetic acid solution; 3) the concentration of tyrosin and tryptophane. Additional methods to determine proteolysis included colorimetric reactions with trinitrobenzene sulphonic acid (TNBS) and ninidrin-cadmium, respectively. Finally, proteolysis was also determined by electrophoresis. The texture and sensorial attributes were also evaluated. The use of Neutrase® accelerated proteolysis in Prato cheese. As for the modified experiments (25, 15 and 10 mg enzyme/kg milk), the cheeses showed, from the very first day of the ripening process on and throughout the rest of the ripening period, extension (% NS-pH 4,6/NT) and depth (% NS TCA 12%/NT) indexes significantly higher in comparison to the values produced by the traditional cheeses manufactured without the addition of enzyme. The modified cheeses of process A (25 mg enzyme/kg milk) presented after 7 days ripening an extension index (17,32%) which was higher than the corresponding index figure obtained for cheese after 30 days ripening described in literature (14,0 - 14,5%). On the other band, the cheeses of process B (15 mg enzyme/kg milk) exceeded (16,02%) the figure referred to in literature (14,0 - 14,5 % after 30 days) already after only 1 day ripening. As for Process C (10 mg enzyme/kg milk), a value quite close (15,61%) to the one referred to in literature for 30 days (14,0 - 14,5%) was reached after 14 days. The incorporation of the enzyme Neutrase® in the curd of modified cheeses did not bring about any losses in terms of cheese yield. Processes A and B showed a decrease ofthe firmness index up to the 21st day ofthe ripening process. This time span of 21 days corresponds to the first stage ofthe breakdown process of ?S1-casein which causes softening of the curd. The texture of the cheeses varied according to the areas submitted to analysis (peripheral and core). The firmness values produced by slices taken from the central part of the cheeses were smaller than those produced by slices taken from the edges. The electrophoretic profile of the cheeses clearly showed that the incorporation of enzymes brought about quantitative differences relative to the protein fractions of the modified cheeses in comparison to the values obtained for the traditional product. A more intense breakdown of the ?S1 and ?-casein fractions is observed, evidenced by higher concentrations of some degradation products of these two fractions found in modified cheeses.
