Targeted Killing of Bacteria by Conjugation of a Soluble Photosensitizer to an Antimicrobial Peptide: Priniciples and Mechanisms

Antimicrobial peptides (AMPs) and photosensitizers (PS) have gained attention as potential alternatives to traditional antibiotics for the treatment of microbial infection due to the decreased likelihood for acquired resistance. However, many AMPs and PS suffer from insufficient activity, specificity, or a combination thereof. AMPs can require high concentrations for effective activity, leading to non-specific side effects and increased costs. PS, on the other hand, are quite active, but are typically hydrophobic and suffer from non-specific binding and damage to host tissues. To solve these problems, we report a novel PS-AMP construct of the soluble PS eosin Y conjugated to the selective AMP (KLAKLAK)_(2). Eosin Y has a high singlet oxygen quantum yield, which is suitable for photodynamic activity, although the solubility of eosin Y results in poor binding and activity toward membranes on its own. On the other hand, the specificity of (KLAKLAK)_(2) is high for an AMP, but could still benefit from enhanced activity at lower concentrations. The killing activity and binding specificity of eosin-(KLAKLAK)_(2) toward both bacteria and mammalian cells was assessed using microbiology, biochemistry, and fluorescence microscopy techniques. Additionally, the mechanism of eosin-(KLAKLAK)_(2) activity was investigated using liposome models to determine factors involved in binding and membrane disruption. Furthermore, novel applications of transmission electron microscopy (TEM) methods were employed to observe the photodynamic effects of eosin-(KLAKLAK)_(2) against bacteria. The PS-AMP conjugate eosin-(KLAKLAK)_(2) displays synergistic activity between PS and AMP in model liposome systems, and is capable of killing several clinically relevant bacteria, including the multi-drug resistant Acinetobacter baumannii AYE strain. Furthermore, bacterial killing is achieved in the presence of red blood cells (RBCs) and other mammalian cell lines without significant toxicity. Liposome models reveal that the lipid composition of bacteria is a potential factor responsible for the observed binding specificity and corresponding activity. Additionally, TEM methods show that eosin-(KLAKLAK)_(2) causes extensive membrane damage to both Gram positive Staph aureus and Gram negative Escherichia coli, indicating a primary cause of cell death. A model is proposed where the activities of the PS and AMP, respectively, facilitate the activity of one another, leading to enhanced membrane disruption, and effective antibacterial activity while maintaining cell selectivity.

Inativação fotodinâmica aplicada a biofilmes de S. aureus formados em tubo endotraqueal / Photodymanic inactivation applying at S. aureus biofilm developed at endotracheal tube

A capacidade dos micro-organismos formarem biofilme em superfícies pode afetar diretamente a saúde do homem. A formação de biofilme microbiano pode ocorrer em superfícies como em tubos de água, ar condicionado, dispositivos médicos como endoscópios, cateteres e tubos endotraquais (TE). Pacientes debilitados por causas diversas como traumas e insuficiência respiratória podem ser portadores do TE. Este tubo é instalado na traqueia e facilita a troca gasosa no organismo debilitado. Na superfície deste tubo endotraqueal, células bacterianas, gram positivas ou negativas, podem se anexar e formar colônias, desenvolvendo assim, um biofilme. Essas células formam uma matriz extra polimérica (EPS) que protege as colônias contra agentes externos. A Pneumonia Associada a Ventilaçao Mecanica (PAV) pode ocorrer entre 48 a 72h após a instalação do TE no paciente. Staphylococcus aureus é a principal bactéria que causa esta doença, a qual apresenta linhagens resistentes a antibióticos. A resistência microbiana faz parte do processo evolutivo de adaptação destas células. Uma das estratégias para combater as infecções hospitalares é a inativação fotodinâmica. A terapia utiliza uma molécula fotossensível, luz e oxigênio celular. O fotossensibilizador, na presença da luz e oxigênio pode formar espécies reativas de oxigênio que levam a célula a morte. Não existe relatos que esta terapia cause resistência microbiana, sendo assim uma alternativa ao insucesso de alguns antibióticos. A curcumina, derivada da Curcuma longa, é uma molécula natural, hidrofóbica e que apresenta propriedades antioxidantes, antimicrobianas, antiinflamatórias e antitumorais com absorção de luz na região de 430 nm. O presente estudo combina o uso da inativação fotodinâmica associada a molécula de curcumina para a eliminação de biofilme de S. aureus desenvolvidos em TE. Métodos diversos foram testados para inativar o biofilme. Formulações com hidrogel, filme, nanoskin foram sintetizados com curcumina e testados no biofilme. Formulações utilizando curcumina natural são mais eficazes para inativar biofilme de E. coli e S. aureus. A inativação fotodinâmica foi otimizada utilizando solução de curcumina sintética, com auxílio de um planejamento experimental. A curcumina sintética em solução se mostrou mais estável e com melhores resultados de inativação comparada com curcumina natural em solução. Uma nova forma de entrega da curcumina sintética foi testada, a nebulização, e esta se mostrou eficaz e com resultados satisfatórios de morte do biofilme de S. aureus. Reações de funcionalização entre FS e o TEH foram testadas utilizando curucmina e porfirinas substituídas. As reações foram possíveis e foram otimizadas. Os resultados microbiológicos destes tubos funcionalizados indicam que os mesmos podem evitar a anexação das células microbianas. Este estudo mostra que a curcumina natural e as formulações são estáveis e podem inativar biofilmes bacterianos. A solução de curcumina sintética é eficaz quando associada a técnicas que também desestruturam o biofilme alcançando assim uma redução total de células. A nebulização é uma forma eficaz de transportar a curcumina sem alterar suas propridades físico-químicas e pode ser usada para a entrega da curcumina. As reações de funcionalização foram otimizadas, não indicando a deformação na estrutura do tubo e este tubo funcionalizado pode ser uma alternativa futura em uma aplicação clínica, evitando a adesão de células microbianas com a luz a ação fotodinâmica pode ocorrer, levando as células a morte. / The capacity of the bacteria develop a biofilm at surfaces can direct affect the humans health. Microbial biofilm formation can occurs on surfaces such as water pipes, air conditioning, medical devices as endoscope, catheters and endotracheal tubes (ET). Patients debilitated by causes such as traumas and respiratory insufficiency can be the cause for the patient can be carried by ET. This tube is installed in the trachea and facilitates gas exchange in the weakened organism. Bacterial cells, gram positive or negative, can attach and form colonies at the tube surface, developing a biofilm. This cells form an extra polymeric matrix (EPS) that protect the colonies against external agents. The Ventilator Associated Pneumonia (VAP) can occurs between 48-72 hours after the tube installation. Staphylococcus aureus is the main bacteria it can cause this disease and is an antibiotic resistant. The microbial resistance is a part of the microbial evolution and adaptation of the cells. The strategy designed to combat nosocomial infection is the photodynamic inactivation. The therapy combines the photosensitive molecule, light and cellular oxygen. The photosensitizer, in the presence of light can produce oxygen reactive species and this species can leave the microbial cells to death. There are no relates that this treatment cause microbial resistance, therefore is an alternative to the failure of some antibiotics. The curcumin, derivate of the Curcuma longa, is a natural molecule, hydrophobic molecule with antioxidant, anti-inflammatory and antitumor properties, with absorbance in the 430 nm region. This study associate the curcumin application in the photodynamic inactivation to eliminate S. aureus biofilm formed at endotracheal tubes. Different methods were tested to inactivate the biofilm. Formulations with hydrogel, a film, nanoskin were synthesized with curcumin and tested at the biofilm. Formulation prepared with natural curcumin were more efficient to E. coli e S. aureus biofilm inactivation. Photodynamic inactivation was optimized testing synthetic curcumin in solution with the aid of the experimental design. The synthetic curcumin in solution showed to be more stable and with the best inactivation results compared to the natural curcumin in solution. A new form of delivery the synthetic curcumin was tested, the nebulization, and this prove efficient and with satisfactory results of S. aureus biofilm death. Functionalization reactions between FS and TE were tested using curcumin and substituted porphyrins. The reactions were possible and optimized. The microbiological results of these immobilized tubes indicate that these tubes can avoid the bacterial cells attached. This studies show that the natural curcumin and the formulations were stables and inactivate bacterial biofilm. The synthetic curcumin in solution is effective when associated with techniques that also helps to de-structure the biofilm thus achieving a total reduction of cells. The nebulization is an effective form to transport without alters the physical-chemical properties of the curcumin. The functionalization reactions were optimized not indicating the deformation in the tube structure and this functionalized tube may be a future alternative in a clinical application, avoiding the adhesion of microbial cells with the light the photodynamic action can occur, leading the cells to death.

Viabilização da curcumina natural nanoencapsulada para inativação fotodinâmica / Viability of natural curcumin nanoencapsulated for photodynamic inactivation

Devido ao uso excessivo de antibióticos houve e ainda há um crescimento no número de cepas resistentes aos medicamentos existentes. Por causa desse crescimento de bactérias multirresistentes, o número de pesquisas que procuram alternativas terapêuticas antibacterianas tem aumentado, e dentre elas está a terapia fotodinâmica antimicrobiana ou inativação fotodinâmica (IFD). A inativação fotodinâmica, utilizada no controle biológico de microrganismos, envolve a ação de um fotossensibilizador (FS), ativado por um comprimento de onda específico, no intuito de oxidar substratos biológicos, resultando em efeito citotóxico. A cúrcuma ou curcumina natural, conhecida como açafrão da terra, consiste em mistura de três curcuminóides: curcumina, demetoxicurcumina e bis-demetoxicurcumina. A curcumina apresenta várias propriedades farmacológicas, no entanto, possui solubilidade extremamente baixa em soluções aquosas, que dificulta a sua utilização como agente terapêutico. O presente estudo propôs desenvolver nanopartículas poliméricas de PLGA contendo curcumina natural a fim de melhorar sua solubilidade e estabilidade, e também verificar sua eficácia na inativação fotodinâmica de microrganismos. As nanopartículas PLGA-CURC sintetizadas através da nanoprecipitação resultaram em três sistemas diferentes, com tamanho médio e eficiência de encapsulamento de 172 nm e 70% para PLGA-CURC1, 215 nm e 80% para PLGA-CURC2, e 242 nm e 80% para PLGA-CURC3. Testes de estabilidade mostraram proteção do polímero contra a degradação precoce da curcumina natural. Os ensaios microbiológicos in vitro com a solução de curcumina natural, as PLGA-CURC1 e PLGA-CURC2 foram eficientes na inativação da bactéria Gram-positiva Staphylococcus aureus, e do fungo Candida albicans. Porém, a solução apresentou toxicidade no escuro em altas concentrações, ao contrario das nanopartículas. O sistema PLGA-CURC2 com sua carga superficial modificada, gerou o sistema PLGA-CURC3, que inativou efetivamente a bactéria Gram-negativa Escherichia coli. Assim, concluiu-se que foi possível deixar a curcumina natural solúvel em água através do encapsulamento em nanopartículas de PLGA, certificar a melhora na estabilidade em meio aquoso (estocagem), além de inativar bactérias e fungo. / Due to excessive over use of antibiotics there was and still are growth in the number of bacterial strains resistant to existing drugs. Because of the growth of multiresistant bacteria, the number of searches looking for alternatives antibacterial therapeutic has increased, and among them is the antimicrobial photodynamic therapy or photodynamic inactivation (PDI). The photodynamic inactivation used in biological control of microorganisms, involves the action of a photosensitizer (PS), activated by a specific wavelength in order to oxidize organic substrates, resulting in cytotoxic effect. Turmeric or natural curcumin, consists of a mixture of three curcuminoids: curcumin, demethoxycurcumin and bis-demethoxycurcumin. Curcumin has various pharmacological properties, however, has extremely low solubility in aqueous solutions, which makes the use as therapeutic agent harder. The present study aims to develop polymeric PLGA nanoparticles containing natural curcumin in order to improve their solubility and stability, and also verify its efficacy in photodynamic inactivation of microorganisms. The PLGA-CURC nanoparticles was synthesized by nanoprecipitation, resulting in three different systems, with an average size of 172 nm and 70% encapsulation efficiency for PLGA-CURC1, 215 nm and 80% for PLGA-CURC2, and 242 nm and 80 % for PLGA-CURC3. Stability tests showed the polymer protected the natural curcumin against premature degradation. Microbiological tests in vitro with the natural curcumin solution, the PLGA-CURC1 and PLGA-CURC2 were efficient in the inactivation of Gram-positive bacterium Staphylococcus aureus, and fungus Candida albicans. However, the solution presented dark toxicity at high concentrations, unlike the nanoparticles. The PLGA-CURC2 system with a modified surface charge, gave the PLGA-CURC3 system, which effectively inactivated Gram-negative bacterium Escherichia coli. Thus, it was concluded that it was possible to let curcumin water soluble by encapsulation in PLGA nanoparticles, to ensure improved stability in aqueous medium (storage), and to inactivate bacteria and fungus.

Photodynamic Inactivation (PDI) is emerging as a promising approach, as effects are based on oxidative stress, preventing the emergence of resistant microorganism strains. In previous studies, the in vitro inactivation of Streptococcus pneumoniae using indocyanine green (ICG) and infrared light source was successful, and achieved reduction of 5 log10 colony-forming units (CFU/mL) with concentration as low as 10 μM ICG. In the present study, a proof-of-principle protocol was designed to treat lung infections by PDI using extracorporeal illumination with a 780 nm laser device and ICG as photosensitizer. In a first row of experiments, hairless mice were infected with S. pneumoniae and PDI was performed two days after infection. For control groups, CFU recovery ranged between 103-104 CFU/mL/mouse. For PDT group, however, no bacteria were recovered in 80% of the animals. Animal survival was evaluated separately over 50 days. No deaths occurred in PDT group, whereas 60% of the control did not survive. Lung injury analyses were performed in BALB/c mice, the bacteria reduction were 2 and 4 log10 in 2 mice (5 in total) and the wet-to-dry ratio showed that PDI did not increase the edema in lungs. The bronchoalveolar lavage data indicated a larger absolute number of cells (mononuclear and polymorphonuclear) in the PDI group in contrast to control group, meaning that the technique could increase the immune system response. In vitro results showed the irradiated ICG could generate aggregates or photoproducts that help PDI to inactivate the bacteria. Our results indicate that extracorporeal PDI has potential for pneumonia treatment, and pulmonary decontamination with PDI may be used as a single therapy or as an antibiotics adjuvant. / A pneumonia infecciosa é uma das principais causas de morbidade/mortalidade no mundo, principalmente devido à taxa crescente de micro-organismos resistentes a antibióticos. A inativação fotodinâmica (IFD) está emergindo como uma abordagem promissora, cujos efeitos são baseados no estresse oxidativo, impedindo o surgimento de cepas de micro-organismos resistentes. Em estudos anteriores, a inativação in vitro de Streptococcus pneumoniae utilizando indocianina verde (ICV) e fonte de luz de infravermelho foi efetiva, inativando 5 log10 (UFC/mL) com apenas 10 μM ICV. Neste estudo, foi avaliado protocolo de prova de princípio para tratar infecções pulmonares por IFD usando irradiação extracorpórea com dispositivo a laser emitindo em 780 nm e ICV como fotossensibilizador (FS). Em uma primeira série de experimentos, camundongos hairless SKH-1 foram infectados com S. pneumoniae e a IFD foi realizada dois dias após a infecção. Para os grupos de controles, a recuperação de UFC variou entre 103-104 UFC/mL/animal. Para o grupo IFD, no entanto, nenhuma bactéria foi recuperada em 80% dos animais. A sobrevivência animal foi avaliada durante 50 dias. Não ocorreram mortes no grupo IFD, enquanto 60% do grupo controle foi a óbito. Foram realizadas análises de lesões pulmonares em camundongos BALB/c, onde a redução bacteriana foi de 2 e 4 log10 em dois animais (total 5) em comparação com o grupo controle, e a proporção de peso úmido e seco mostrou que a IFD não aumentou o edema nos pulmões. Os dados de lavagem broncoalveolar indicaram um aumento no número absoluto de células (mononucleares e polimorfonucleares) no grupo IFD, o que indica que a técnica pode aumentar a resposta do sistema imunológico. Os resultados in vitro mostraram que a ICV irradiada pode gerar agregados ou fotoprodutos que auxiliam a IFD a inativar a bactéria. Os resultados deste estudo indicam que a IFD com irradiação extracorpórea tem potencial para o tratamento de pneumonia, e a descontaminação pulmonar com IFD pode ser usada como terapia ou como adjuvante aos antibióticos. / CAPES: 99999.003154/2015-07

Assim, o objetivo deste estudo foi verificar os efeitos da inativação fotodinâmica sobre biofilmes de Candida albicans, avaliando seus efeitos sobre a expressão dos genes TEC1 (fator de transcrição), HWP1 (proteína de parede celular das hifas), EFG1 (regulador transcricional relacionado com a morfogênese), BCR1 (regulador da formação de biofilme e da parede celular), CPH1 (regulador transcricional envolvido na morfogênese) e ALS3 (adesina) de C. albicans. Foram avaliadas 30 amostras isoladas de pacientes portadores de HIV e 30 amostras de pacientes com estomatite protética, quanto a produção de biofilme, peso seco e filamentação. Destas, foram selecionadas as amostras mais virulentas de cada grupo que apresentaram melhor capacidade de formação de biofilme e filamentação. Assim, foi utilizada uma amostra clínica de C. albicans isolada de paciente portador de HIV, uma amostra clínica de C. albicans isolada de paciente com estomatite protética e uma cepa padrão ATCC 18804. The objective of this study was verify the effects of aPDI on C. albicans biofilms, evaluating its effects on genes expression: TEC1 (transcription factor), HWP1 (cell wall protein hyphae), EFG1 (transcriptional regulator related to morphogenesis), BCR1 (regulator of biofilm formation and cell wall), CPH1 (transcriptional regulator involved in morphogenesis) and ALS3 (adhesin) of C. albicans. Were evaluated 30 samples isolated from patients with HIV and 30 samples from patients with denture stomatitis, as the production of biofilm, dry weight and filamentation. Of these, the most virulent strains of each group that presented better biofilm formation capacity and filamentation were selected. Therefore, were used a clinical sample of C. albicans isolated from HIV positive patient, a clinical sample of C. albicans isolated from patient with denture stomatitis and a standard strain ATCC 18804. Among numerous alternatives to combat vector, we can cite the use of photosensitizers (PSs) as inactivating of Aedes aegypti larvae. These compounds are able to interact with the light in a specific wavelength, so the cause highly reactive cytotoxic oxygen, resulting in the oxidation of biological targets. The present study had as main objective to analyze the phototoxic effects of PSs: Photogem® and turmeric, in three molecular variations (curcumin, curcuminoids pigment mixture (curcumin, desmethoxycurcumin and bisdemethoxycurcumin) and curcumin in sucrose), against Aedes aegypti larvae under different conditions of delivery of the PSs and lighting. The experiments of this study were divided into two stages: i) Photogem® and the three compositions of turmeric were applied in trials of photodynamic inactivation (PDI) in solution. The groups that received the Photogem®, were also fed with two different types of food, in order to verify the influence of these foods on larval mortality. ii) in a second step the three compositions of turmeric were incorporated in pet food, where it obtained a lyophilized powder, which was subsequently offered to the larvae for scanning. After the receipt of the PSs in solution as in the form of freeze-dried powder, the larvae were irradiated with two sources of light: artificial and natural. In addition to the trials of mortality, other studies were performed, such as checking the time of degradation of PSs when exposed to light, the anatomical location of accumulation of FSs in the larvae, and characterization studies of the product obtained in the form of lyophilized powder. On PDI with the Photogem® comparing the foods, exposed to artificial lighting, one of the foods introduced a higher mortality, indicating the importance of this on effectiveness of PDI. Already on PDI with variations of turmeric, mortality rates varied according to the molecular forms of this compound, showing high mortality for curcumin and curcumin in sucrose. In conditions involving sunlight, high mortality rates were obtained for all FSs delivered solution, featuring in various conditions 100% mortality in 8 hours from exposure to light. For IFD using the freeze-dried powder of three variations of turmeric and sunlight, mortalities in the order of 80% were achieved in lighting 16 hours. The analysis of the fluorescence image obtained by confocal microscope showed that both the three variations of turmeric, such as Photogem®, after an incubation period of 12 hours, accumulated throughout the alimentary canal of the larvae. The Photogem®, as well as turmeric showed potent photodynamic activity being more effective in conditions with higher light intensities. Regarding the delivery conditions of the PS, we observe that the highest values of mortality were obtained through the application of PSs in solution, when compared with the mortality studies using the freeze-dried powder. These results indicate that PDI can be a promising technique in the control of vectors as the Aedes aegypti. / O mosquito Aedes aegypti é o principal vetor responsável pela transmissão da dengue. Dentre as inúmeras alternativas de combate ao vetor, podemos citar o uso de fotossensibilizadores (FSs) como inativadores de larvas do Ae. aegypti. Esses compostos são capazes de interagir com a luz, num comprimento de onda específico, de modo a originar espécies altamente reativas de oxigênio citotóxico, resultando na oxidação de alvos biológicos. O presente estudo teve como principal objetivo analisar os efeitos fototóxicos dos FSs: Photogem® e cúrcuma, em três variações moleculares (curcumina, mistura de pigmentos curcuminóides (curcumina, demetoxicurcumina e bis-demetoxicurcumina) e curcumina em sacarose), contra larvas do Ae. aegypti sob diferentes condições de entrega dos FSs e de iluminação. Os experimentos deste estudo foram divididos em duas etapas: i) tanto o Photogem® quanto as três composições de cúrcuma foram aplicados nos ensaios de inativação fotodinâmica (IFD) em solução. Os grupos que receberam o Photogem® foram também alimentados com dois diferentes tipos de ração, com a finalidade de verificar a influência desses alimentos na mortalidade larval. ii) em uma segunda etapa as três composições de cúrcuma foram incorporadas nas rações, onde obteve-se um pó liofilizado, que foi posteriormente ofertado às larvas para a verificação da mortalidade. Após o recebimento dos FSs, tanto em solução como na forma de pó liofilizado, as larvas foram irradiadas com duas fontes de luz: artificial e natural. Além dos ensaios de mortalidade, outros estudos foram realizados, como a verificação do tempo de degradação dos FSs quando expostos à luz, o local anatômico de acumulação dos FSs nas larvas, e estudos de caracterização do produto obtido na forma de pó liofilizado. Na IFD com o Photogem® comparando as rações, expostas à iluminação artificial, uma delas apresentou uma mortalidade superior, indicando a importância desta na efetividade da IFD. Já na IFD com as variações de cúrcuma, as porcentagens de mortalidade variaram de acordo com as formas moleculares deste composto. Nas condições envolvendo luz solar, foram obtidas altos valores de mortalidade para todos os FSs entregues em solução, apresentando em diversas condições 100% de mortalidade em 8 horas de exposição à luz. Nos ensaios de IFD utilizando o pó liofilizado das três variações de cúrcuma e luz solar, mortalidades da ordem de 80% foram atingidas em 16 horas de iluminação. As análises das imagens de fluorescência obtidas por microscópio confocal mostraram que tanto as três variações de cúrcuma, como o Photogem®, após um período de incubação de 12 horas, acumularam-se em todo o canal alimentar das larvas. O Photogem®, assim como a cúrcuma apresentaram atividade fotodinâmica potente, sendo mais efetivos em condições com maiores intensidades de luz. Com relação às condições de entrega do FS, observamos que as maiores mortalidades foram obtidas através da aplicação dos FSs em solução, quando comparadas com os estudos de mortalidade utilizando o pó liofilizado. Esses resultados indicam que IFD pode ser uma técnica promissora no controle de vetores como o Ae. aegypti.

Viabilização da curcumina natural nanoencapsulada para inativação fotodinâmica / Viability of natural curcumin nanoencapsulated for photodynamic inactivation

Devido ao uso excessivo de antibióticos houve e ainda há um crescimento no número de cepas resistentes aos medicamentos existentes. Por causa desse crescimento de bactérias multirresistentes, o número de pesquisas que procuram alternativas terapêuticas antibacterianas tem aumentado, e dentre elas está a terapia fotodinâmica antimicrobiana ou inativação fotodinâmica (IFD). A inativação fotodinâmica, utilizada no controle biológico de microrganismos, envolve a ação de um fotossensibilizador (FS), ativado por um comprimento de onda específico, no intuito de oxidar substratos biológicos, resultando em efeito citotóxico. A cúrcuma ou curcumina natural, conhecida como açafrão da terra, consiste em mistura de três curcuminóides: curcumina, demetoxicurcumina e bis-demetoxicurcumina. A curcumina apresenta várias propriedades farmacológicas, no entanto, possui solubilidade extremamente baixa em soluções aquosas, que dificulta a sua utilização como agente terapêutico. O presente estudo propôs desenvolver nanopartículas poliméricas de PLGA contendo curcumina natural a fim de melhorar sua solubilidade e estabilidade, e também verificar sua eficácia na inativação fotodinâmica de microrganismos. As nanopartículas PLGA-CURC sintetizadas através da nanoprecipitação resultaram em três sistemas diferentes, com tamanho médio e eficiência de encapsulamento de 172 nm e 70% para PLGA-CURC1, 215 nm e 80% para PLGA-CURC2, e 242 nm e 80% para PLGA-CURC3. Testes de estabilidade mostraram proteção do polímero contra a degradação precoce da curcumina natural. Os ensaios microbiológicos in vitro com a solução de curcumina natural, as PLGA-CURC1 e PLGA-CURC2 foram eficientes na inativação da bactéria Gram-positiva Staphylococcus aureus, e do fungo Candida albicans. Porém, a solução apresentou toxicidade no escuro em altas concentrações, ao contrario das nanopartículas. O sistema PLGA-CURC2 com sua carga superficial modificada, gerou o sistema PLGA-CURC3, que inativou efetivamente a bactéria Gram-negativa Escherichia coli. Assim, concluiu-se que foi possível deixar a curcumina natural solúvel em água através do encapsulamento em nanopartículas de PLGA, certificar a melhora na estabilidade em meio aquoso (estocagem), além de inativar bactérias e fungo. / Due to excessive over use of antibiotics there was and still are growth in the number of bacterial strains resistant to existing drugs. Because of the growth of multiresistant bacteria, the number of searches looking for alternatives antibacterial therapeutic has increased, and among them is the antimicrobial photodynamic therapy or photodynamic inactivation (PDI). The photodynamic inactivation used in biological control of microorganisms, involves the action of a photosensitizer (PS), activated by a specific wavelength in order to oxidize organic substrates, resulting in cytotoxic effect. Turmeric or natural curcumin, consists of a mixture of three curcuminoids: curcumin, demethoxycurcumin and bis-demethoxycurcumin. Curcumin has various pharmacological properties, however, has extremely low solubility in aqueous solutions, which makes the use as therapeutic agent harder. The present study aims to develop polymeric PLGA nanoparticles containing natural curcumin in order to improve their solubility and stability, and also verify its efficacy in photodynamic inactivation of microorganisms. The PLGA-CURC nanoparticles was synthesized by nanoprecipitation, resulting in three different systems, with an average size of 172 nm and 70% encapsulation efficiency for PLGA-CURC1, 215 nm and 80% for PLGA-CURC2, and 242 nm and 80 % for PLGA-CURC3. Stability tests showed the polymer protected the natural curcumin against premature degradation. Microbiological tests in vitro with the natural curcumin solution, the PLGA-CURC1 and PLGA-CURC2 were efficient in the inactivation of Gram-positive bacterium Staphylococcus aureus, and fungus Candida albicans. However, the solution presented dark toxicity at high concentrations, unlike the nanoparticles. The PLGA-CURC2 system with a modified surface charge, gave the PLGA-CURC3 system, which effectively inactivated Gram-negative bacterium Escherichia coli. Thus, it was concluded that it was possible to let curcumin water soluble by encapsulation in PLGA nanoparticles, to ensure improved stability in aqueous medium (storage), and to inactivate bacteria and fungus.

Transmissive spongiform encephalopathies, also called prion disorders, are fatal neurodegenerative diseases affecting mammals. In patients, the pathological prion protein (PrPTSE ) accumulates in CNS and causes death. Prions possess high binding affinity to surfaces. Moreover, they are highly resistant to conventional sterilization procedures which rise the risk of nosocomial transmission from patients in subclinical stage of prion disease through medical tools. In the thesis, we evaluate the efficiency of photodynamic inactivation (PDI) for prion decontamination. The PDI is induced by photoactivation of phthalocyanine (Pc) derivates AlPcOH(SO3)2, SiPc(OH)2(SO3)1-3 or ZnPc(SO3)1-3. Pc exposed to light generate reactive oxygen species, mainly singlet oxygen (O2(1 ∆g)). Production of O2(1 ∆g) in aqueous solution was confirmed by iodide method, quenching by NaN3 and oxidative degradation of uric acid. The photoactivation of Pc in infectious brain homogenate led to elimination of PrPres signal (= proteinase K-resistant PrPTSE fragment) below the detection limit of western blot by using nanomolar AlPcOH(SO3)2 concentration. Síntese e caracterização de nanopartículas emissoras de luz por conversão ascendente de energia (UCNPS) funcionalizadas para uso em aplicações biológicas / Synthesis and Characterization of Upconversion Nanoparticles Functionalized for Biological Applications

As nanopartículas emissoras de luz por conversão ascendente de energia (UCNPs) têm atraído grande interesse nos últimos anos devido à sua versatilidade e capacidade de converter radiação menos energética em fótons de maior energia. Quando combinadas com outras moléculas, as UCNPs podem ser empregadas em um grande número de bioaplicações e, apesar de encontrar um de seus maiores usos na terapia fotodinâmica (PDT) de células cancerígenas, os mesmos princípios podem ser empregados na fotoinativação de bactérias (PDI). Nos últimos 20 anos a resistência bacteriana têm se tornado um dos maiores problemas de saúde pública e novas estratégias precisam ser desenvolvidas para superar esse desafio. Nesse trabalho, nós apresentamos a síntese e caracterização de nanopartículas de NaYF4 dopadas com Yb3+ (20%) e Er3+ (2%) e funcionalizadas para uso em PDI. As UCNPs foram sintetizadas utilizando-se o método de co-precipitação e foram revestidas com uma camada de sílica mesoporosa (@mSiO2) visando o aumento da dispersibilidade em água, o incremento da área superficial e a possibilidade de funcionalizações, e a diminuição da citotoxicidade do material. As UCNP@mSiO2 foram decoradas com moléculas orgânicas e com o fotossensibilizador (PS) ftalocianina de silício (SiPc) para estudo de sua interação e efeito bactericida contra cepas bacterianas Gram positivas e negativas. Diferentes abordagens foram utilizadas em busca de nanopartículas com as melhores propriedades: tamanho, forma, geração de oxigênio singleto e estabilidade em solução aquosa. Verificou-se que a transferência de energia entre as UCNPs e as SiPc em sua superfície é muito dependente do tipo de funcionalização realizada e da extensão com que as moléculas de PS adentram os poros da sílica. O material final apresentou toxicidade na ausência de luz contra a bactéria E. coli, enquanto a viabilidade das cepas de S. aureus não foi afetada no escuro. A excitação direta da SiPc nas nanopartículas levou a completa erradicação da E. coli e um drástico decréscimo no número de unidades formadoras de colônia (CFU) da S. aureus de até sete ordens de magnitude. Com esse estudo demonstramos diferentes estratégias para sintetizar e potencializar o uso das UCNPs como agentes teranósticos, principalmente para uso em PDI. / Lanthanide-doped upconverting nanoparticles (UCNPs) that emit high-energy photons upon excitation by low-energy near infrared (NIR) light, have garnered great interest in recent years due to their versatility. These nanoparticles combined to other molecules can be used for a broad range of bio-applications as theranostic agents, for bioimaging, drug and gene delivery and photodynamic therapy (PDT) applications. Although phototherapy is commonly used in the treatment of cancer, the same principles can be applied for the inactivation of bacteria (PDI). In the last twenty years bacterial resistance against antibiotics has become a critical public health issue and new strategies are needed to overcome this challenge. Herein, we describe the synthesis, detailed characterization and application of functionalized NaYF4: Yb (20%), Er (2%) UCNPs for use in PDT and especially for PDI. The UCNPs are produced using the co-precipitation method and covered with a mesoporous silica shell to enhance the dispersibility in water, increase the surface area and grant functionalization possibility and to decrease cytotoxicity. The nanoparticles were decorated with different molecules and functionalized with the photosensitizer (PS) silicon phthalocyanine (SiPc) to study interaction and enhance the bactericidal effect against Gram positive (+) and Gram negative (-) bacteria. Different approaches were used to identify the best particles concerning the shape, size, photophysical properties, generation of singlet oxygen and stability in water. It was verified that the energy transfer (FRET) efficiency from the UCNP towards SiPc depended on the surface functionalization and resulting encapsulation depths of the PS into the mesoporous silica. Functionalized UCNPs displayed dark toxicity towards Gram (-) E. coli while Gram (+) S. aureus viability was not decreased in the dark. Directly exciting the PS on the UNCP led to complete eradication of E. coli and a drastic decrease of colony forming units of S. aureus of up to seven orders of magnitude. Through this study we demonstrate different strategies to synthesize and potentialize the use of UCNPs as theranostic agents, mainly for use in PDI.
