This thesis provides a deep study of the Reach Control Problem (RCP) for affine systems defined on simplices. Necessary conditions for solvability of the problem by open loop control are presented, improving
upon the results in the literature which are for continuous state feedback only. So-called reach control indices are introduced and developed
which inform on the structural properties of the system which cause continuous state feedbacks to fail. A novel synthesis method is presented consisting of a subdivision algorithm based on these indices
and an associated piecewise affine feedback. The method is shown to solve RCP for all cases in the literature where continuous state feedback fails, provided it is solvable
by open loop control. Textbook examples of existing synthesis methods for RCP are provided. The motivation for studying RCP and its relevance to complex control
specifications is illustrated using a biomedical application.

Inference of regulatory networks has received attention of researchers from many fields. The challenge offered by this problem is its being a typical modeling problem under insufficient information about the process. Hence, we need to derive the apriori unavailable information from the empirical observations. Modeling by inference consists of selecting or defining the most appropriate model structure and inferring the parameters. An appropriate model structure should have the following properties. The model parameters should be inferable. Given the observation and the model class, all parameters used in the model should have a unique solution restriction of the solution space). The forward model should be accurately computable (restriction of the solution space). The model should be capable of exhibiting the essential qualitative features of the system (limit of the restriction). The model should be relevant with the process (limit of the restriction). A piecewise linear formulation, described by a switching state transition matrix and a switching state transition vector with a Boolean function indicating the switching conditions is proposed for the inference of gene regulatory networks. This thesis mainly concerns using a formulation of switching networks obeying all the above mentioned requirements and developing an inference algorithm for estimating the parameters of the formulation. The methodologies used or developed during this study are applicable to various fields of science and engineering.

Intramyocellular Lipids and the Progression of Muscular Insulin ResistanceJanuary 2017 (has links)
abstract: Diabetes is a disease characterized by reduced insulin action and secretion, leading to elevated blood glucose. In the 1990s, studies showed that intravenous injection of fatty acids led to a sharp negative response in insulin action that subsided hours after the injection. The molecule associated with diminished insulin signalling response was a byproduct of fatty acids, diacylglycerol. This dissertation is focused on the formulation of a model built around the known mechanisms of glucose and fatty acid storage and metabolism within myocytes, as well as downstream effects of diacylglycerol on insulin action. Data from euglycemic-hyperinsulinemic clamp with fatty acid infusion studies are used to validate the qualitative behavior of the model and estimate parameters. The model closely matches clinical data and suggests a new metric to determine quantitative measurements of insulin action downregulation. Analysis and numerical simulation of the long term, piecewise smooth system of ordinary differential equations demonstrates a discontinuous bifurcation implicating nutrient excess as a driver of muscular insulin resistance. / Dissertation/Thesis / Doctoral Dissertation Applied Mathematics for the Life and Social Sciences 2017
