Diet enrichment with arachidonic and docosahexaenoic acid during the lactation period attenuates the effects of intrauterine growth restriction from birth to maturity in the guinea pig and improves maternal bone mass

Burr, Laura Lynn. January 2008 (has links)
Intrauterine growth restriction (IUGR) reduces bone mass by 10-30% and impairs arachidonic (AA) and docosahexaenoic (DHA) acid status in infants. Because AA and DHA enhance neonatal bone mass, the aim of this study was to determine the effects of dietary 0.5% AA and 0.2% DHA (w/w) prior to weaning on bone and growth. 40 guinea pigs were randomized to either a control (C) or low-protein diet (LP) during pregnancy and the C diet or the C diet with AA+DHA during lactation. Measurements included bone mass, metabolism, and strength, and erythrocyte lipid of sows and offspring from birth to 16 wk post-partum. The LP diet induced IUGR, while the AA+DHA increased bone mass by 5-20% in sows and offspring and corrected growth and bone mass in IUGR pups. Thus, AA+DHA provided in lactation rescues the growth trajectory in an IUGR state and is beneficial to maternal and neonatal bone mass.

STANDARDIZED ILEAL DIGESTIBLE TRYPTOPHAN TO LYSINE RATIOS IN GROWING PIGS FED U.S. TYPE AND NON U.S. TYPE FEEDSTUFFS

Quant, Anthony David 01 January 2008 (has links)
The objective of these studies was to determine the optimum standardized ileal digestible tryptophan to lysine (SID Trp:Lys) ratio for growing pigs fed both U.S.–type (corn–soybean meal) and non–U.S.–type (wheat–barley) diets. When feeding U.S.–type diets, average daily gain (ADG) and plasma urea nitrogen (PUN) concentrations improved as SID Trp:Lys ratios increased, yielding optimum ratios of 15.32% (P<0.001) and 15.25% (P<0.001), respectively. Averaging these resulted in an optimum SID Trp:Lys ratio of 15.29%. In non–U.S.–type diets, feeding increasing SID Trp:Lys ratios improved ADG and PUN, displaying optima of 15.99% (P=0.048) and 15.29% (P=0.054), respectively. This produced an average optimum SID Trp:Lys ratio of 15.64%. The final study included the other dietary essential amino acids (EAA) at a higher level, and ADG and PUN improved as SID Trp:Lys ratios increased, yielding optima of 17.93% (P<0.001) and 16.17% (P=0.009). This averaged to an optimum SID Trp:Lys ratio of 17.05%. Overall, diet composition did not affect the Trp:Lys requirement of growing pigs, however, feeding the other EAA at higher levels resulted in a higher estimated optimum SID Trp:Lys ratio. The optimum SID Trp:Lys ratio is estimated to lie between 15.29% and 17.05%.

THE EFFECTS OF ACTIGEN® AND THREONINE SUPPLEMENTATION ON GROWTH PARAMETERS, IMMUNE FUNCTION, AND INTESTINAL HEALTH IN MONOGASTRICS

Good, Lindsay 01 January 2013 (has links)
The objectives of these experiments were to evaluate the main effects and interactive effects between dietary supplementation of a mannan oligosaccharide based product, Actigen® (ACT), and different levels of threonine in monogastrics, as evidenced by growth parameters, immune function, and intestinal health characteristics. In nursery piglets, ACT supplementation decreased average daily feed intake (P=0.04), but had no effect on body weight or feed conversion ratio. There were no noted differences between threonine levels on performance traits. There were no consistent differences in complete blood count or cytokine gene expression profiles in the blood. The highest level of threonine, 77% true ileal digestible threonine:lysine (Thr:Lys), increased villus height (P=0.007) and villus height:crypt depth (P=0.01). The lowest level, 57% Thr:Lys, decreased villus surface area (P=0.04) and goblet cell density (P=0.04). Supplementation with ACT increased total goblet cell area (P=0.02) and density (P=0.05). There were no interactions observed between ACT and Thr:Lys levels. In broiler chicks, feeding a diet containing 0.56% threonine decreased body weight (PPPP=0.07). On d 7, birds supplemented with ACT had heavier spleens as a percentage of body weight (P=0.01) compared to no ACT. When adjusted for body weight, the 0.56% threonine fed birds had smaller spleens (P=0.05) on d 7 when compared to the other threonine levels. Humerus (PPP=0.04) and birds fed 0.56% threonine had a higher concentration of potassium (PP=0.03) and few goblet cells (P=0.04) on d 7 when compared to the other threonine levels. In d 21 jejunum, supplementation with ACT reduced apical width (P=0.03) and surface area (P=0.02). An interaction was observed between ACT and threonine level in the jejunum on d 21 on basal width (P=0.03) and surface area (P=0.02), indicating that in diets lacking ACT, excess threonine increased villus size. Overall, ACT and threonine acted primarily independently to modulate the intestinal architecture of both nursery piglets and broiler chicks. However, in broiler chicks ACT and threonine interacted to alter villus size. These results indicate that ACT and threonine have direct effects on the intestines of monogastrics.

Postnatal vitamin D supplementation normalizes neonatal bone mass following maternal dietary vitamin D deficiency in the guinea pig

Finch, Sarah L. January 2007 (has links)
Since vitamin D deficiency is common at birth, the objective of this study was to test if postnatal vitamin D supplementation would normalize bone mineralization. Forty guinea pigs were randomized to receive a diet with or without vitamin D3 during pregnancy. Newborn pups were randomized to receive 10 IU of vitamin D3 or a placebo daily until d28. Measurements at birth and d28 included whole body and regional bone mass, osteocalcin and deoxypyridinoline, plus biomechanical testing of excised tibias and femurs. Offspring from deficient sows had lower body weight, whole body and tibia bone mineral content (BMC) and lower osteocalcin and biomechanical integrity. By d28 this group had lower whole body bone density and femur BMC, unless supplemented. Interactions with gender showed males continued to have low 25(OH)D despite supplementation. Therefore, neonates born to sows with dietary vitamin D deficiency require supplemental vitamin D to support normal bone mineral accretion.
