A soja transgênica resistente ao herbicida glifosato (RG) tem sido amplamente utilizada devido a proporcionar o eficiente controle de plantas daninhas. O herbicida glifosato é o composto mais utilizado em soja RR, e possui características de baixa toxicidade e rápida degradação. Porém, este herbicida pode causar amarelecimento das folhas logo após a aplicação e resultar na imobilização de micronutrientes, principalmente Fe e Mn, no solo e nas plantas. Os objetivos deste estudo foram avaliar os efeitos do herbicida glifosato em soja RG em função da dose do herbicida, da variabilidade genética de cultivares e da disponibilidade de micronutrientes sobre parâmetros fisiológicos, nutricionais, e de crescimento da cultura e no rendimento de grãos. O primeiro experimentos a campo consistiu da avaliação de 8 cultivares transgênicas e suas respectivas linhas isogênicas com aplicação de glifosato na dose de 900 g ha-1. O segundo experimento a campo avaliou o efeito do adubo foliar com micronutrientes em interação com duas cultivares de soja tratadas com glifosato. Os experimentos em casa de vegetação foram conduzidos em sistema hidropônico com a presença e ausência de Fe e Mn na solução nutritiva com 3 cultivares e diferentes doses de glifosato. O herbicida glifosato na dose de 900 g ha-1 diminuiu o rendimento de grãos das cultivares BRS Estância RR, BRS Taura RR e Fundacep 59 RR. A aplicação de adubo foliar não resultou em incremento no rendimento de grãos nas cultivares avaliadas. O efeito do herbicida glifosato não alterou a dinâmica dos micronutrientes Fe e Mn em 2 cultivares em comparação com as respectivas cultivares isogênicas próximas. Apenas a cultivar BRS Taura RR apresentou menor acúmulo de Fe foliar em comparação com a sua linha isogênica BRS 154. A fluorescência da clorofila, apesar de ser uma medida rápida e não destrutiva, apresentou grande variabilidade, e necessita de aprimoramentos para a determinação dos efeitos secundários do herbicida glifosato em soja. As generalizações sobre o efeito do herbicida glifosato em soja RG e da resposta da adubação foliar com micronutrientes não foram evidenciadas no presente estudo. Assim, isto indica a necessidade de estudos adicionais em relação ao diagnóstico dos fatores envolvidos no desenvolvimento de cultivares nas recomendações de nutrição mineral em soja RG. / The transgenic soybean resistant to glyphosate (RG) has been widely used due to provide effective weed control. However, this herbicide can cause a symptom called yellow flashing immediately after application and may result in the immobilization of micronutrients, especially Fe and Mn, in the soil and plants. The objectives of this study was to evaluate the effects of the herbicide glyphosate in soybeans RR in function of the herbicide rates, the genetic variability of cultivars and the availability of micronutrients through physiological and nutritional parameters, crop growth and grain yield analysis. The first field experiments consisted of evaluating 8 transgenic cultivars and their respective isogenic lines with application of glyphosate at doses of 900 g ha-1. The second field experiment evaluated the effect of foliar fertilization with micronutrients applied in interaction with two soybean cultivars treated with glyphosate. The greenhouse experiments were conducted in a hydroponic system in the presence and absence of Fe and Mn in the nutrient solution with 2 cultivars and different doses of glyphosate. The herbicide glyphosate at a dose of 900 g ha-1 decreased the grain yield of the cultivars BRS Estância RR, BRS Taura RR and Fundacep 59 RR. The application of foliar fertilizer did not increased the grain yield in the cultivars tested. Eficácia da soja geneticamente modificada MON 87701 × MON 89788 com a expressão da proteína Cry1Ac no controle de Anticarsia gemmatalis (Hübner) e Pseudoplusia includens (Walker) (Lepidoptera: Noctuidae)

Adotou-se o delineamento de blocos casualizados em parcelas subdividas com 4 repetições, sendo que os níveis de infestação corresponderam aos tratamentos primários, enquanto os genótipos de soja corresponderam aos tratamentos secundários. A soja foi cultivada em vasos plásticos sob gaiola telada. Os tratamentos primários para o ensaio com A. gemmatalis foram a) sem infestação; b) baixa infestação com liberação de 64,75+66,75 (fêmea+macho) adultos/parcela e c) alta infestação com liberação de 317,25+302,5 (fêmea+macho) adultos/parcela; enquanto para P. includens foram a) baixa infestação com liberação de 55,25+58,55 (fêmea+macho) adultos/parcela e b) alta infestação com liberação de 213,25+196,5 (fêmea+macho) adultos/parcela. Os tratamentos secundários foram soja MON 87701 × MON 89788; soja isogênica não transgênica; soja RR. Os resultados indicaram que a soja MON 87701 × MON 89788 foi altamente eficiente no controle da lagarta-da-soja e da falsa-medideira. A soja isogênica não transgênica (M 8329) e soja RR (M 8360 RR) não tiveram qualquer efeito nas populações de ambas as lagartas. Os adultos não discriminaram a soja geneticamente modificada e portanto não tiveram preferêncica para oviposição / The transgenic soybean MON 87701 × MON 89788 with Cry1Ac was compared to the isogenic non-transgenic soybeans (M 8329) and RR soybean (M 8360 RR) resistant to glyphosate in the control of the these pests, under greenhouse conditions. Split plot design with four randomized blocks (replications) and infestation levels corresponded to the primary treatment, together with the soybean genotypes corresponded to the secondary treatments was adopted in this study. Soybeans were planted in plastic pots under screened cage. For A. gemmatalis, the primary treatments were a) non-infested plants b) low infestation in which 64.75 +66.75 (female + male) adult/plot were releases c) high infestation in which 317.25 +302.5 (female + male) adult/plot were released; whereas, for P. includens the treatments were a) low infestation with 55.25 +58.55 (female + male) adult/plot and b) high infestation with 213.25 +196.5 (female + male) adult/plot released. The secondary treatments were soybean MON 87701 × MON 89788; isogenic non-transgenic soybean and RR soybean. The results indicated that soybean MON 87701 × MON 89788 was highly effective in controlling both velvetbean caterpillar and soybean looper. Adults of both insects were not able to discriminate the genetically modified soybean, therefore they did not show oviposition preference

Aspectos biológicos de Spodoptera cosmioides Walker, 1858 (Lepidoptera: Noctuidae) nas cultivares de algodoeiro DeltaOPAL e NuOPAL (Bollgard I)

Neste trabalho, estudou-se a biologia comparada de S. cosmioides sobre a cultivar transgênica comercial NuOPAL (Bollgard I, Evento 531) e sobre a cultivar convencional DeltaOPAL, procurando-se detectar a influência da α -endotoxina CryIAc no desenvolvimento biológico dessa espécie. O experimento foi desenvolvido em condições de laboratório (26 ± 1ºC; UR: 70 ± 10%; fotofase: 12h), a partir de lagartas recém-eclodidas e individualizadas. Foram avaliadas a duração e a viabilidade das fases imaturas, duração do ciclo biológico, peso de pupas, razão sexual, porcentagem de deformação de adultos e de adultos não liberados dos invólucros pupais, longevidade dos adultos, fecundidade e viabilidade de ovos. Os parâmetros biológicos observados não diferiram significativamente em relação às duas variedades. Na cultivar NuOPAL verificou-se maior porcentagem de ovos inviáveis, de adultos deformados e de adultos não emergidos, o que indica que essa cultivar provavelmente afeta a fase adulta de S. cosmioides. / Spodoptera cosmioides Walker, 1858 is a polyphagous species, presents resistance to insecticides in various regions of Brazil, and although it occurs at low densities, it’s considered a potencial pest in the crop of cotton, soybeans and beans in the Brazilian cerrado. In this work was studied the comparative biology of S. cosmioides on the comercial transgenic NuOPAL (Bollgard I, Evento 351) and the conventional cultivar DeltaOPAL to detect the influence of endotoxin-α CryIAc on the biological development of this species. The experiment was conducted under specific conditions in a laboratory (26 ± 1º C; RH: 70 ± 10%; photoperiod: 12h), from newlyhatched and individualized larvae. The duration and viability of immature phases and biological cycle, weight of pupae, sex ratio, percentage of deformed adults and adults not released the pupal beg, longevity of adults, fecundity and viability of eggs were evaluated. The biological parameters observed showed that there weren’t significant difference between two varieties studied. In NuOPAL cultivar there was a greater percentage of eggs unviable, deformed adults and adults not emerged, which indicates that this cultivar probably affects the adult phase of S. cosmioides.

A lagarta Pseudoplusia includens (Walker) mais encontrada na soja, tem se destacado atualmente na região Centro-Oeste do Brasil, devido aos novos sistemas de cultivos sucessivos, soja-algodão. Um dos principais avanços tecnológicos é a disponibilidade de cultivares transgênicas, resistentes para algumas espécies desfolhadoras de insetos-pragas, porém, algumas espécies, consideradas alvo da tecnologia, tem sobrevivido e preocupado produtores. Este trabalho teve como objetivo avaliar os principais aspectos biológicos de P. includens alimentadas com folhas de quatro cultivares de algodoeiro, bem como estudar o efeito da cultivar transgênica NuOPAL, sobre o comportamento, sobrevivência e desenvolvimento do inseto. Além disso, avaliou-se o consumo foliar de lagartas e a atratividade a estas cultivares. Avaliou-se também a distribuição vertical de lagartas de P. includens na cultivar transgênica WideStrike e na sua isolinha comercial FM 993. O delineamento experimental utilizado foi inteiramente casualizado, com quatro tratamentos (cultivares DeltaOPAL, NuOPAL, FM 993 e FM 910), com 30 ou 60 repetições, conforme o experimento. No ensaio de não preferência para alimentação de lagartas, foi utilizado o delineamento inteiramente casualizado, com 10 repetições, com as mesma cultivares. Para a distribuição vertical de lagartas nas plantas das cultivares FM 993 e FM 975 WS foi utilizado delineamento em blocos casualizados em esquema de parcelas subdivididas, com 06 tratamentos (3 terços da planta x 2 faces foliares), e 40 repetições. The caterpillar Pseudoplusia includens (Walker) most commonly found in soybean, has been highlighted today in the Midwest region of Brazil, due to new systems of crop rotation, soybean-cotton. One of the major technological advances is the availability of transgenic cultivars resistant to some species of defoliating insect pests, but some species considered target technology, has survived and concerned farmers. This study aimed to evaluate the main biological aspects P. includens feed by leaves of four cotton cultivars, as well as studying the effect of transgenic NUOPAL cultivar, on behavior, survival and development of the insect. Furthermore, were evaluated the attractiveness and foliar consumption by larvae to the cultivars. Another objective was to evaluate the vertical distribution of larvae of P. includens in transgenic cultivar WideStrike and its isoline commercial FM 993. Were used a completely randomized design, using treatments with cultivars DeltaOPAL, NUOPAL, FM 993 and FM 910, with 60 replicates for the antibiosis experiment. In the test of non-preference for feeding caterpillars, was used completely randomized design with 10 replicate with the same cultivars. For the vertical distribution was used as a randomized block design in a split-plot, with 06 treatments (3 parts of plants x 2 leaf surface), and 40 replicate. The data were transformed into (x +0.5)1/2 and subjected to analysis of variance and treatments means were compared by Tukey test at 5% probability. The Bt cultivar Bt (Bollgard I) exerts the sort of antibiosis to half of the larval population, extending the larval stage of the survivors. O objetivo do trabalho foi estudar uma forma alternativa para o controle do endurecimento dos frutos do maracujazeiro, pela produção de plantas transgênicas contendo o gene da proteína capsidial do Passionfruit woodness virus - PWV. O vetor de expressão foi construído utilizando-se os plasmídeos pCambia 2300 e pCambia 2301, que contêm o gene de seleção nptII, para resistência ao antibiótico canamicina. O plasmídeo pCambia 2301 contém também o gene repórter uidA (GUS). Os plasmídeos foram introduzidos em Agrobacterium tumefaciens, estirpes EHA 105 e LBA 4404, pelo método do choque térmico. Os explantes para transformação genética constituíram-se de discos de folhas jovens (6 mm de diâmetro), das variedades IAC 275 e IAC 277, coletados de plantas mantidas em sob fotoperíodo de 16 h luz, a 27 °C. Os explantes foram inoculados com suspensão bacteriana (5x108 UFC/mL) por 20 min e transferidos para placa de Petri contendo o meio de cultura MS + thidiazuron (TDZ - 0,25 mg/L) + nitrato de prata (AgNO3 - 4 mg/L) + acetoseringona (1 µM/L). O co-cultivo foi realizado à temperatura de 24 °C, em ausência de luz, por um período de 3 dias. Para seleção e regeneração de plantas os explantes foram transferidos para meio de cultura de seleção MS + TDZ (0,25 mg/L) + AgNO3 (4 mg/L) + canamicina (100 mg/L) + cefotaxime (500 mg/L). A incubação foi realizada a 27 °C, em ausência de luz, por um período de 4 - 6 semanas. As gemas adventícias desenvolvidas foram transferidas para o meio de cultura MSM + 10% de água de coco e incubadas sob fotoperíodo de 16 h de luz. A transformação genética foi identificada pelo teste histoquímico GUS e por PCR. Obteve-se um total de 22 plantas PCR positivas. Destas, 8 foram analisadas por Southern blot para confirmação da integração do transgene. A transcrição e expressão do transgene foram analisadas por Northern e Western blot, respectivamente. As plantas transgênicas avaliadas foram multiplicadas e inoculadas com 3 diferentes estirpes do PWV. A linhagem T2 apresentou resistência a infecção dos três isolados utilizados. / The main purpose of this work was to study an alternative way to control the Passionfruit woodiness virus - PWV through the production of transgenic plants which contained the Passionfruit woodness virus coat protein gene. The binary vector was built by using pCambia 2300 and pCambia 2301 plasmids, which contain the selection gene nptII. The pCambia 2301 plasmid also contains the reporter gene uidA (GUS). The plasmids were introduced into Agrobacterium tumefaciens, EHA 105 and LBA 4404 strains, via thermal shock method. The explants for the genetic transformation were young leaf disks (6 mm of diameter) of IAC 275 and IAC 277 varietys, extracted from plants kept under 16 h photoperiod, at 27 °C. The explants were inoculated with a bacterial suspension (5x108 UFC/mL) for 20 min and then transferred to Petri dishes containing cocolture medium MS + thidiazuron (TDZ - 0,25 mg/L) + silver nitrate (AgNO3 - 4 mg/L) + acetosyringone (1 µM/L). The co-culture was performed at 24 °C t, in the dark, for a three-day period. For the selection and regeneration of plants, the explants were transferred to the selection culture medium MS + TDZ (0,25 mg/L) + AgNO3 (4 mg/L) + kanamycin (100 mg/L) + cefotaxime (500 mg/L). The incubation was performed at 27 °C, in dark, for 4 - 6 weeks. The adventitious buds developed were then transferred to the culture medium MSM + 10% coconut water and kept incubated under 16 h photoperiod. The genetic transformation was identified through GUS and PCR tests. There were 22 PCR positive plants. Out of those, 8 were Southern blot analyzed for the confirmation of transgenc integration. The transgene transcription and expression were determined by Northern and Western blot respectively. The transgenic plants were then multiplied and inoculated with 3 different strains of PWV, and the line 2 showed resistance to the three strains used.

A produção de papel e celulose representa um importante componente do setor industrial brasileiro, com perspectivas a expandir-se nos próximos anos, sendo o Brasil o principal produtor de celulose a partir de eucalipto. A técnica de transformação genética pode contribuir significativamente para introduzir caracteres de interesse econômico que permitam: aumentar a produtividade da cultura, resistência a doenças e melhorar a qualidade da madeira. Os sistemas de transformação genética requerem um sistema de regeneração de plantasin vitro eficiente associado a um sistema de integração estável do transgene ao genoma das mesmas. Por este motivo o trabalho foi dividido em duas etapas. A primeira parte foi orientada para o desenvolvimento de um sistema de regeneração eficiente e reproduzível por organogênese indireta tanto para E. grandis como para o híbrido E. grandis x E. urophylla. Foram realizados experimentos para avaliar-se o efeito dos diferentes tipos de explantes, genótipos, concentrações de reguladores de crescimento e taxa regenerativa de diferentes clones. A regeneração de cotilédones e folhas de plântulas apresentou uma eficiência de 30 % e 25 respectivamente. Além destes sistemas, o protocolo de regeneração a partir de folhas de clones comerciais de E. grandis mostrou-se bastante eficiente. Na segunda parte do trabalho desenvolveu-se a metodologia de transformação genética de E. grandis e E. grandis x E. urophylla, via Agrobacterium tumefaciens. O efeito de diferentes tempos de pré-cultivo dos explantes, tempo de sonicação, influência dos meios de co-cultivo e dos agentes seletivos, foram avaliados. As sementes pré-cultivadas durante 2 e 15 dias apresentaram as maiores porcentagens de expressão da???? -glucuronidase (GUS) (21,7 e 37,4 %, respectivamente), quando sonicadas. As sementes pré-cultivadas por 2 dias apresentaram mais de 90 % das áreas transformadas localizadas nos cotilédones e no colo. Já as sementes pré-tratadas por 15 e 17 dias apresentaram 60 % das áreas de transformação localizadas no primeiro par de folhas. A condição mais adequada para a transformação de cotilédones foi a sonicação por 120 s, de sementes pré-cultivadas por 2 dias, sendo o melhor meio de co-cultivo o MS. Plantas transgênicas de E. grandis e E. grandis x E. urophylla foram obtidas, abrindo assim uma grande perspectiva na área de melhoramento do eucalipto, com a introdução das técnicas de transformação gênica. / The pulp and paper industry is an important sector of Brazilian industry, showing good perspectives of expansion, once Brazil is the main cellulose producer from Eucalyptus. Genetic transformation may contribute to increase productivity by the introduction of desirable traits, such as pest resistance and improvement of wood quality. However, the prerequisite for the success of the transformation strategy is the establishment of an efficient, in vitro, regeneration system. The recovery of transgenic plants is only possible from cells that respond to both processes: the integration of the transgene and also the plant regeneration. Hence, this work was divided in two phases. The first study was carried out to develop an efficient and reproducible regeneration system by indirect organogenesis of E. grandis and the hybrid E. grandis x E. urophylla. The experiments were organized to evaluate the effect of the different seedling explants, genotypes, hormonal concentration and regenerative rate of clonal material. The regeneration efficiency of cotyledons and leaves of seedling explants was around 30 % and 25 %, respectively. In addition, an efficient regeneration protocol of Eucalyptus grandis was developed which uses leaf explants from clonal plants. In the second study the procedure for genetic transformation of E. grandis and E. grandis X E. urophylla using Agrobacterium is described. Several experimental parameters were evaluated such as the length of precultivation, the sonication effect, the cocultivation media and the selective agents. Germinating seeds of 2 and 15 days had the highest percentage of ? -glucuronidase (GUS) expression (21.7% and 37.4%, respectively), when sonicated. Germinating seeds imbibed for 2 days showed over 90% of the blue sectors localized in cotyledons and in the intersection of the hypocotyls and roots, whereas, seedlings that had germinated for 15-17 days had an average of 60% of the transformed sectors localized in the first pair of leaves. The best condition for an efficient genetic transformation was 2-day precultivation, associated with 120-s sonication and MS media for cocultivation. Transgenic plants of E. grandis and E. grandis x E. urophylla were obtained by this method, opening an important perspective for the breeding of Eucalyptus through genetic transformation techniques.

The Bt-cotton carries genes that express the toxin Cry1Ac. This creates new interactions in the agroecossystems encouraging researches to answer the questions about the potential impact on nontarget organisms. In the cotton ecosystem, the two spotted spider mite, Tetranychus urticae Koch, acquires and mantain in its body high concentrations of Cry toxins (Cry1Ab from Bt-corn and Cry1Ac from Bt-cotton). This finding leads to the question about the potential long term impact on the phytophagous minte, a nontarget, and their predators. Thus, this work investigated the biology and behavior of T. urticae and its predatory mite, Phytoseiulus macropilis (Banks) on Bt-cotton. The results obtained during three successive generations showed that the Bt-cotton does not affect negatively its development, viability of immature stages and reproductive output. Furthermore, the preference for feeding and oviposition of T. urticae and its predator, P. macropilis, were similar on both cotton types. In addition, P. macropilis exhibited similarpredatory behavior on T. urticae fed on both cotton types. Despite of similar results, T. urticae acquired about 3.97 times more Cry1Ac than that expressed in the Bt-cotton plants. / Plantas de algodoeiro têm sido geneticamente transformadas com genes da bactéria Bacillus thuringiensis Berliner (Bt) os quais conferem resistência a certas espécies de lepidópteros-praga. O algodoeiro Bt BollgardTM possui genes que expressam a toxina Cry1Ac. Com esta nova variável interagindo nos agroecossistemas, algumas questões foram levantadas sobre a ação de toxinas Cry em artrópodos não-alvo, motivando o desenvolvimento de pesquisas para gerar os esclarecimentos necessários. O ácaro rajado, Tetranychus urticae Koch, adquire da planta e mantêm em seu corpo concentrações elevadas de toxinas Cry (Cry1Ab do milho Bt e Cry1Ac do algodoeiro Bt). Este resultado leva ao questionamento sobre os possíveis efeitos da toxina sobre este ácaro fitófago e aos seus predadores. Assim, este trabalho investigou a biologia e comportamento de T. urticae e do seu predador, Phytoseiulus macropilis (Banks) em algodoeiro Bt e não-Bt. Os resultados, obtidos durante três gerações consecutivas, mostram que o algodoeiro Bt não afetou negativamente o tempo de desenvolvimento, a viabilidade das formas imaturas e a reprodução dos dois ácaros. Também, a preferência para a colonização e postura de T. urticae e de P. macropilis foram similares entre os algodoeiros Bt e não-Bt. Além disso, não houve alteração na preferência de P. macropilis, quando predando de T. urticae criado em algodoeiro Bt e não-Bt. O ácaro rajado adquiriu e concentrou em seu corpo aproximadamente 3,97 vezes a quantidade de Cry1Ac expressada na planta de algodoeiro.
