across the geographic distribution of a species. More suitable habitats, in ecological terms,
allow the survival of greater number of individuals, which increases the genetic variability
in the population, while in less suitable habitats, because of smaller population size,
individuals have lower reproductive rate and higher mortality rate, these populations may
lead to loss of genetic variability and extinction. In this context, the goal of this study was
to use meta-analysis techniques to relate the genetic variables (observed and expected
heterozygosity and inbreeding) with the models of potential geographical distribution of
nine species of Cerrado plants. In addition, genetic variables were correlated with human
occupation at sites where plant species occurred. For this work were used 11 studies, in
which genetic data were obtained from literature and the points of occurrence in databases
and also the literature. Five species distribution models (Maxent, Mahalanobis, Euclidian,
Domain and Bioclim) were used and then superimposed on a combined basis (i.e
Ensemble forecasting). Were performed Pearson correlations between genetic variables
and environmental and human occupation. The coefficients of these correlations were
matched using a meta-analysis to find a general correlation studies. The correlations
between genetic variables with the ecological variable (i.e environmental suitability) and
human occupation tended to zero. Although significant values have been obtained with
studies of isozymes for the human index (r = 0.57 P = 0.01) related, independent, with the
observed heterozygosity. A positive relationship exists between the human and the
expected heterozygosity index (r = 0.27, P = 0.01) was found in the relationship between
all the studies together. We also found significant values for studies with isoenzymes in
ecological indices (r = 0.39, P = 0.04) and human (r = 0.50, P = 0.01) and the distance
from the center of the potential distribution species (r = -0.44, P = 0.05) between the
expected heterozygosity. The results of the mean effect between heterozygosity and
occupation indices suggest that the plants were evaluated in studies with isozymes show a
high heterozygosity in places with high human occupation. Due to the occupation of the

The results of studies with isozymes, correlation between heterozygosity and distance

relationship is also negative (r = -0.35, P <0.05), suggesting that adequacy is also lower in
peripheral regions. In general, the methodology used to conduct preliminary assessments
of the distribution of the population structure of Cerrado plants related to genetic variables
proven very useful and practical. / Fatores ecológicos e evolutivos podem explicar os padrões de variabilidade
genética intrapopulacional ao longo da distribuição geográfica de uma espécie. Habitats
mais adequados, em termos ecológicos, permitem a sobrevivência de uma quantidade
maior de indivíduos, o que aumenta a variabilidade genética na população, enquanto que
em habitats menos adequados os indivíduos, por possuírem um tamanho populacional
menor, apresentam uma menor taxa de reprodução e uma maior taxa de mortalidade,
podendo levar estas populações à perda de variabilidade genética e extinção. Neste
contexto, o objetivo desse trabalho foi utilizar técnicas de meta-análise para relacionar as
variáveis genéticas (heterozigosidade esperada e observada e endogamia) com os
modelos de distribuição geográfica potencial de nove espécies de plantas do Cerrado.
Além disso, esse trabalho teve como objetivo relacionar as variáveis genéticas com a
ocupação humana nos locais de ocorrência das espécies de plantas. Foram utilizados 11
estudos, cujos dados genéticos foram obtidos da literatura e os pontos de ocorrência em
bases de dados e também da literatura. Cinco técnicas de modelagem (Maxent,
Mahalanobis, Euclidiana, Bioclim e Domain) foram utilizadas e em seguida, de forma
combinada foram sobrepostas (i.e Ensamble forecasting). Foram feitas correlações de
Pearson entre variáveis genéticas e os índices ecológicos e de ocupação humana. Os
coeficientes destas correlações foram combinados usando uma meta-análise para
encontrar uma correlação geral dos estudos. A espécie que apresentou a maior
distribuição geográfica potencial foi a Solanum lycocarpum com aproximadamente
4.088.960Km2 e a menor foi a Vellozia epidendroides com aproximadamente 69.104 Km2.
As correlações entre as variáveis genéticas com a variável ecológica (i.e. adequabilidade
ambiental) e a ocupação humana tenderam a zero. Embora valores significativos tenham
sido obtidos, com estudos de marcadores isoenzimáticos, para o índice humano (r = 0,57
P = 0,01) relacionados, independentes, com a heterozigosidade observada. Um efeito
positivo entre o índice humano e a heterozigosidade esperada (r = 0,27; P = 0,01) foi
encontrado na relação entre todos os estudos reunidos. Também foram encontrados
valores significativos para os estudos com isoenzimas nos índices ecológico (r = 0,39; P =
0,04) e humano (r = 0,50; P = 0,01) e na distância do centro da distribuição potencial da
espécie (r = -0,44; P = 0,05) entre a heterozigosidade esperada. Os resultados do efeito
médio entre as heterozigosidades e os índices de ocupação sugerem que as plantas
avaliadas nos estudos com marcadores isoenzimáticos apresentam uma alta
heterozigosidade em lugares com alto índice de ocupação humana. Devido à ocupação
do Cerrado ser intensa, porém recente, e devido às espécies de plantas possuírem ciclos
longos de vida as populações das espécies podem ainda não estar estruturadas
geneticamente nesta nova paisagem do Cerrado. Os resultados dos estudos, com
marcadores isoenzimáticos, de correlação entre as heterozigosidades e a distância do
centro de distribuição potencial da espécie corroboraram com as nossas predições.
Plantas mais afastada tendem perder a variabilidade genética por possuírem uma menor
densidade populacional. A relação entre a adequabilidade de habitat e a distância do
centro indicou uma relação também negativa (r = -0,35; P < 0,05), sugerindo que a
adequabilidade é também menor nas regiões periféricas. De maneira geral, a metodologia
utilizada neste trabalho para a realização de avaliações preliminares da distribuição da
estrutura populacional de plantas do Cerrado relacionada às variáveis genéticas
demonstrou ser bastante útil e prática.

Previous issue date: 2009-01-22 / The goal of this study was to evaluate the genetic structure and the spatial
pattern of intra and interpopulational gene flow of Dipteryx alata Vogel, based on nuclear
and chloroplastidial microsatellite markers. Primers were developed based on sequencing
of random fragments from a shotgun genomic library for detection of microsatellite
regions. 12 microsatellites regions were obtained from 688 sequences, which allowed the
construction of pairs of primers. These regions are composed of motifs with two to six
nucleotides, ranging from 136 to 380 base pairs. This shows that the random sequencing
strategy from shotgun libraries is interesting because it allows the achievement of primers
for repetitive regions with different motifs. Two of these loci (Da_E06 e Da_E12) were
polymorphic with three alleles each. He estimation for these loci showed satisfactory values
(0.2946 and 0.2879, respectively), considering the number of alleles. Also, we used a
transferred primer from the species Phaseolus vulgaris (BM164) for D. alata. Moreover,
other two chloroplastidials microsatellite primers were used for molecular analyses of
georeferenced subpopulations, totalizing 775 plants distributed over the natural occurrence
area of Cerrado. 210 of these plants were collected and georeferenced one by one along the
margins of the Araguaia River in the states of Mato-Grosso (RAMT) and Goiás (RAGO)
for spatial distribution of genetic variability in local scale analysis. The relationship
between estimations of genetic diversity parameters with patterns of potential distribution
of species was evaluated. This was used to test the hypothesis that the genetic variability of
D. alata populations is distributed according to the central-periphery model. D. alata
subpopulations showed considerable high levels of genetic variability that was
significantly structured among subpopulations and well structured in space, both for
nuclear and chloroplastidial data. The estimation of the apparent cross-fertilization rate (ta
= 1.0575) indicates that the species is allogamous. Estimations of migration rates by pollen
and by seeds were lower than one, indicating that seed dispersal contributes more effectively for total gene flow. Estimates of the genetic diversity parameters from the
Araguaia River population showed similar values between both margins. The estimation of
the apparent cross-fertilization rate (0.9434) indicated that the Araguaia River is not a
physical barrier to effective gene flow. The effective size of the neighborhood, i.e., the
mean number of individuals in an area where panmixia occurs was 85.64 and 22.99 for
nuclear and chloroplastidial data, respectively, indicating that seed dispersal is more
restricted. The correlogram generated with chloroplastidial data presented a cline pattern of
variance more evident than with nuclear data, suggesting that the presence of spatial
genetic structure is being more influenced by seed dispersal. We observed that the genetic
parameters do not follow a classical central-periphery model, because peripheral
population (according to geographical distribution of sampling locations) tended to
demonstrate higher values for these estimations, mainly the South and Western
subpopulations. The relationship found between the fixation index (f) with human impact
indicated that the subpopulations evaluated can be affected by fragmentation process and
land use, probably caused by the recent human colonization in Cerrado biome. / O objetivo geral deste estudo foi avaliar a estrutura genética e o padrão
espacial do fluxo gênico intra e interpopulacional de Dipteryx alata Vogel, com base em
marcadores microssatélites nucleares e cloroplastidiais. Foi utilizada uma estratégia de
desenvolvimento de iniciadores que se baseia no seqüenciamento aleatório de fragmentos
provenientes de uma biblioteca genômica shotgun para a detecção de regiões
microssatélites, em barueiro. Das 668 sequências obtidas, foram encontradas 12 regiões
microssatélites que possibilitaram a construção dos pares de iniciadores. Estas regiões são
compostas por motivos com dois a seis nucleotídeos, que variam entre 136 a 380 pb. Isto
mostra que a estratégia de sequenciamento aleatório a partir de bibliotecas shotgun é
interessante por possibilitar a obtenção de iniciadores para regiões repetitivas com
diferentes motivos. Dois destes locos (Da_E06 e Da_E12) foram polimórficos,
apresentando três alelos cada. As estimativas de He para estes locos apresentaram valores
satisfatórios, considerando o número de alelos, e foram iguais a 0,2946 e 0,2879,
respectivamente. Isto indica que os iniciadores desenvolvidos e padronizados com sucesso
neste trabalho podem ser utilizados para estudos genético-populacionais. Juntamente com
estes dois iniciadores desenvolvidos, foi utilizado outro transferido da espécie Phaseolus
vulgaris (BM164) para D. alata, além de outros dois iniciadores microssatélites
cloroplastidiais para análises moleculares de 23 subpopulações georeferenciadas,
totalizando 775 plantas, distribuídas ao longo da área de ocorrência natural do Cerrado.
Destas plantas, 210 foram coletadas e georreferenciadas uma a uma, ao longo das margens
do alto rio Araguaia nos estados de Mato-Grosso (RAMT) e Goiás (RAGO), para análises
de distribuição espacial da variabilidade genética em escala local. Para testar a hipótese de
que a variabilidade genética das subpopulações de D. alata se distribui conforme o modelo
central-periférico, foram avaliadas as relações entre as estimativas de parâmetro genético
de diversidade com os padrões de distribuição potencial da espécie. Foi observado que as
subpopulações de D. alata apresentam consideráveis níveis de variabilidade genética que se encontra significativamente estruturada entre as subpopulações, tanto para os dados
nucleares quanto para os cloroplastidiais, apresentando-se também estruturada no espaço.
A estimativa da taxa de fecundação cruzada aparente (ta = 1,0575) indica que a espécie seja
alógama. As estimativas das razões de migração via pólen e via semente foram menores do
que 1, o que indica que a dispersão de sementes contribui mais efetivamente para o fluxo
gênico total em escala regional. As estimativas dos parâmetros genéticos de diversidade
referentes à população contígua ao Rio Araguaia mostraram valores semelhantes entre as
margens do rio. A estimativa da taxa de fecundação cruzada aparente (0,9434) confirma a
reprodução por alogamia no barueiro. O baixo nível de diferenciação entre as
subpopulações das duas margens do rio indica que o rio Araguaia não confere uma barreira
física efetiva ao fluxo gênico. O tamanho efetivo de vizinhança, ou seja, o número médio
de indivíduos numa área onde ocorre panmixia, para os dados nucleares e cloroplastidiais
foi igual a 85,64 e 22,99, respectivamente, indicando que a dispersão de sementes é mais
restrita em escala local. O correlograma gerado a partir dos dados cloroplastidiais
apresenta um padrão clinal de variação mais evidente do que o proveniente dos dados
nucleares, o que sugere que a presença da estrutura genética espacial deve estar sendo mais
influenciada pelo padrão de dispersão da semente. Foi observado que parâmetros genéticos
estimados para D. alata não se enquadram no clássico modelo central-periférico, pois as
populações tidas como periféricas (de acordo com a distribuição geográfica dos pontos de
coleta) tendem a exibir maiores valores para estas estimativas, com destaque para as as
subpopulações das bordas Sul e Oeste do bioma. A relação encontrada entre os valores
estimados para o índice de fixação intrapopulacional (f) com o impacto humano indica que
as subpopulações estudadas podem estar sofrendo as conseqüências dos processos de
fragmentação e uso da terra na variabilidade genética, causado provavelmente pelo recente
processo de ocupação do Cerrado.

Previous issue date: 2009-01-22 / The purpose of this work is to perform the pharmacognostic studies of the species Hyptidendron canum (Pohl ex Benth.) Harley (Lamiaceae) and Eugenia uniflora L. (Myrtaceae), investigate the population variability of the constituents of the essential oil from H. canum leaves and inflorescences, and assess the bioactivity of the crude ethanol extracts and fractions from leaves of this plants against microorganisms (bacteria and the fungus Candida albicans) and on Oreochromis niloticus Linnaeus. For the variability study of the H. canum essential oil, samples obtained from leaves and inflorescences from Hidrolândia, Silvânia, Bela Vista and Goiânia cities were analyzed by gas chromatography and gas
chromatography with mass spectrometry. The anatomic analysis and the phytochemical screening of the leaves from the two species were performed using conventional techniques. The antimicrobial activity was assessed with Gram-positive and Gram-negative bacteria and with the Candida albicans fungus using the well diffusion test and the agar dilution method to determine the minimal inhibitory concentration (MIC). The biological activities of the crude
ethanol extract and ethyl acetate, hexanic and chloroformic fractions of the leaves from the two species were tested in O. niloticus fish hepatopancreas and gill. The H. canum essential oils analysis indicated high chemovariability in the oils from different locations. The phytochemical screening and the thin layer chromatography (TLC) analysis of the H. canum
leaves evidenced the presence of flavonoids, saponins, terpenes and lignans, while the E. uniflora leaves evidenced the presence of tannins, steroids, triterpenes, anthraquinonic
heterosides, saponinic and flavonoids. As for the antimicrobial activity testing, the crude ethanol extracts from both species presented antimicrobial activity against all the microorganisms
tested and the E. uniflora ethyl acetate fraction presented satisfactory activity against resistant Pseudomonas aeruginosa. The H. canum crude extract and fractions
presented vasoactive activity in the hepatopancreas and gill of the tilapias, causing varying degrees of vasodilation and vascular congestion. An apparent proliferation of blood capillaries was detected in the hepatopancreas of the fish that ingested the hexanic fraction. The E. uniflora crude extract and the ethyl acetate, chloroform and hexanic fractions caused vasodilation, vascular congestion and tissue alterations in the O. niloticus hepatopancreas and gills / O presente trabalho teve por objetivo realizae o estudo farmacognóstico das espécies Hyptidendron canum (Pohl ex Benth.) Harley (Lamiaceae) e Eugenia uniflora L. (Myrtaceae),
investigar a variabilidade populacional dos constituintes do óleo essencial das folhas e inflorescências da H. canum e avaliar a bioatividade dos extratos etanólicos brutos e das
frações das folhas das referidas plantas contra icrorganismos (bactérias e fungo) e em Oreochromis niloticus Linnaeus. Para o estudo da variabilidade do óleo essencial da H. canum, as folhas e inflorescências foram coletadas nos municípos de Hidrolândia, de Silvânia, de Bela Vista e de Goiânia e seus óleos essenciais foram analisados por cromatografia gasosa
e cromatografia gasosa acoplada a espectrometria de massa. A análise anatômica e a prospecção fitoquímica das folhas das duas espécies foram realizadas utilizando as técnicas convencionais. A atividade antimicrobiana foi avaliada com bactérias Gram-positivas e Gramnegativas e com o fungo Candida albicans, usando o teste de difusão em poço e o método de diluição em ágar, para determinar a concentração inibitória mínima (CIM). As atividades biológicas do extrato etanólico bruto e das frações acetato de etila, hexânica e clorofórmica das folhas das duas espécies foram avaliadas no hepatopâncreas e brânquias do peixe O. niloticus L. A análise dos óleos essenciais da H. canum indicou elevada quimiovariabilidade nos óleos dos diferentes locais. A triagem fitoquímica e a cromatografia em camada delgada (CCD) das folhas da H. canum evidenciaram a presença de flavonóides, saponinas, terpenos e lignanas e das folhas da E. uniflora, a presença de taninos, esteróides, triterpenos, heterosídeos antraquinônicos, saponínicos e flavonóides. Na avaliação da atividade antimicrobiana, observou-se que os extratos etanólicos brutos das duas espécies demonstraram atividade antimicrobiana contra todos os microrganismos testados, e a fração acetato de etila da E. uniflora apresentou boa atividade contra Pseudomonas aeruginosa resistentes aos antibióticos convencionais. O extrato bruto e as frações da H. canum apresentaram atividade vasoativa no hepatopâncreas e brânquias das tilápias, promovendo, em maior ou menor grau, vasodilatação e congestão vascular. Detectou-se ainda aparente proliferação dos capilares sanguíneos no hepatopâncreas dos peixes tratados com a fração hexânica. Verificou-se que tanto o extrato bruto, quanto as frações acetato de etila, clorofórmica e hexânica da E. uniflora promoveram, no hepatopâncreas e nas brânquias de O.
niloticus, vasodilatação, congestão vascular e alterações teciduais
