Avaliações histológica e histomorfométrica do reparo de cavidades ósseas preenchidas por osso bovino anorgânico ou vidro bioativo associados ao plasma rico em plaquetas em mandíbula de cão /

Foram utilizados, neste trabalho, 4 cães, machos, e neles foram confeccionadas 4 cavidades bilaterais na mandíbula, e depois divididos em 4 grupos: controle; PRP; PRP/Bio-Oss®; PRP/Bio-Gran®. Após 120 dias, os animais foram sacrificados e a análise histológica demostrou que o processo de reparo foi completo somente para o grupo Controle. Nos demais grupos não foi possível verificar o completo reparo das cavidades. Em nível biológico, os melhores resultados obtidos estão apontados em seqüência; PRP/Bio- Oss®; PRP/Bio-Gran®; e PRP. Esses achados foram confirmados por meio de análise histomorfométrica, onde as diferenças entre os grupos se mostraram estatisticamente signicantes na comparação do grupo Controle. Foi possível concluir neste estudo que o PRP isolado ou associado ao vidro bioativo e ao osso bovino anorgânico não favoreceu o processo de reparo. / Abstract: This study evaluated the effect of PRP associeted to bovine anorganic bone or bioglass on bone healing. From adult male dogs leid form bone cavities prepared in earl side of the mandible, which were divided into four groups, according to the fillig material, as follows: control, PRP, PRP/ Bio-Oss, PRP/Bio-Gran. The animals were sacrified after 120 days, and healing was complete only in the control cavities. Among the treat groups, the better results were found for PRP/Bio-Oss, followed by PRP/Bio-Gran and PRP. Those findings were confirmed by histometric analysis, which showed statistically significant difference in the amount of bone formation favorable to the control cavities. Thus, PRP, associetd or not with bovine anorganic bone or bioglass did not favorable affect bone healing. / Doutor

Mecanismo de ação das plaquetas na insuficiência hepática aguda

A insuficiência hepática aguda é caracterizada por morte celular maciça de hepatócitos. As primeiras células que se ativam após uma lesão hepática são as células de Kupffer. Estas células secretam citocinas e produzem espécies reativas de oxigénio, causando a apoptose de hepatócitos. Em um estudo anterior, mostramos que plaquetas encapsuladas aumentam a sobrevida de animais em um modelo de insuficiência hepática aguda em 10 dias. Aqui investigamos como as plaquetas exercem o seu efeito benéfico precoce nesse modelo. Para isso, plaquetas encapsuladas ou cápsulas vazias foram implantadas em ratos submetidos a hepatectomia parcial de 90%. Os animais foram eutanasiados 1, 3, 6, 12, 24, 48 e 72 horas (n=6/grupo/hora) após hepatectomia. O fígado remanescente foi coletado para avaliar o estresse oxidativo, a atividade da caspase 3 e a expressão de genes relacionados ao estresse oxidativo ou função hepática. Além disso, os níveis de lipopolissacarideos no soro e no tecido foram medidos. O número de células de Kupffer do fígado remanescente foi avaliado. A interação de plaquetas encapsuladas e células de Kupffer foi investigada utilizando um sistema de co-cultura. Foi observado que os níveis de lipopolissacarideos foram semelhantes em ambos os grupos, assim como a expressão do gene de Tlr4 e Myd88, mas o Lbp foi maior no grupo plaquetas. O número de células de Kupffer no grupo plaquetas estava aumentado 1 hora após hepatectomia, voltando a níveis normais em seguida. No grupo controle estava aumentado às 6 até 72 horas. Além disso, as plaquetas modulam a expressão de interleucina-6 e interleucina-10 em células de Kupffer após 24 horas de co-cultura. Além disso, as plaquetas aumentam a atividade de superóxido dismutase e catalase e reduzem a peroxidação lipídica. Além disso, a atividade da caspase 3 também foi reduzida em animais que receberam plaquetas encapsuladas às 48 e 72 horas. A expressão da óxido nítrico sintase endotelial, do fator nuclear kappa B e interleucina-6 estavam elevados no grupo de plaquetas. A expressão do gene de albumina e do fator V também estavam aumentados no grupo plaquetas. Além disso, estes efeitos são mediados por fatores parácrinos imediatamente após a lesão hepática. / Acute liver failure is characterized by massive hepatocyte cell death. Kupffer cells are the first cells to be activated after liver injury. They secrete cytokines and produce reactive oxygen species, leading to apoptosis of hepatocytes. In a previous study, we showed that encapsulated platelets increase survival in a model of acute liver failure. Here we investigate how platelets exert their early beneficial effect in this model. For that, encapsulated platelets or empty capsules were implanted in rats submitted to 90% partial hepatectomy. Animals were euthanized at 1, 3, 6, 12, 24, 48 and 72 hours (n = 6/group/time) after hepatectomy. Liver was collected to assess oxidative stress, caspase activity, and gene expression related to oxidative stress or liver function. Also, lipopolysaccharide (LPS) levels in serum and tissue were assessed. The number of Kupffer cells in the remnant liver was evaluated. Interaction of encapsulated platelets and Kupffer cells was investigated using a co-culture system. It was observed that LPS levels were similar in both groups, as well as gene expression of Tlr4 and Myd88, but Lbp was higher in platelet group. Avaliação da degradação do RNA viral do ZIKA vírus em diferentes compartimentos biológicos

Orientador: Rejane Maria Tommasini Grotto / Resumo: O vírus Zika pertence a família Flaviviridae, gênero Flavivirus e tem seu genoma viral constituído de uma molécula de RNA de fita simples de polaridade positiva, com aproximadamente 10.700 pb. As interações de Flavivirus com compartimentos biológicos como as plaquetas foram relatadas pela literatura, sendo demostrado que o vírus é carreado para outros tecidos e sua viabilidade é mantida por um maior tempo, no entanto, este tipo de estudo ainda não foi reportado para o ZIKV. O objetivo deste trabalho foi comparar a carga viral do vírus Zika entre o plasma rico em plaquetas (PRP) e o plasma pobre em plaquetas (PPP). As amostras utilizadas foram coletadas de doadores saudáveis e posteriormente incubadas com o vírus ZIKA à 37°C na presença de CO2 durante 12 dias consecutivos, sendo retiradas alíquotas no momento do preparo e após 2, 4, 6, 8, 10 e 12 dias de incubação. Os resultados demostraram que a carga viral no PRP foi estatisticamente superior a do PPP depois de 12 dias de incubação com o vírus, sugerindo que as plaquetas podem carrear e preservar o vírus durante maior tempo comparado ao plasma isento de plaquetas. / Abstract: The Zika virus belongs to the Flaviviridae family, Flavivirus genus, and the viral genome consists of the molecule single-stranded, positive-sense RNA of approximately 10,700 bp. The interaction of Flavivirus with biological compartments such as platelets has been reported in the literature, and have shown that the virus is carried to other tissues and its viability is maintained for a longer time. However, this kind of evaluation was not reported for ZIKV yet. The aim of this study was to compare the viral load of Zika virus between platelet rich plasma (PRP) and platelet poor plasma (PPP). The samples were collected from healthy donors and subsequently incubated with ZIKA virus at 37 ° C with CO2 atmosphere for 12 consecutive days. Aliquots were evaluated at the zero time and after 2, 4, 6, 8, 10 and 12 days of incubation Results showed that the viral load in PRP is statistically higher than in PPP after 12 days of incubation with the virus, suggesting that platelets can carry and preserve the virus longer than in platelet-free plasma. / Mestre

Urb ("Brazilian jalapa") (Convolvulaceae) is a common species of the Brazilian Northeast. It is popularly used because of its laxative and purgative properties. The tincture of Operculina macrocarpa tubers (major constituent) and Convolvulus scammonia composes the raw materials of Aguardente Alemã®, herbal medicine, referred to as antithrombotic in folk medicine. The objective of this study was to investigate the antiplatelet and anticoagulant potential for Operculina macrocarpa in human plasma, including the bioprospection study and determination of possible mechanism of action. The evaluation of antiplatelet activity of drugs (tincture and fractions of O. macrocarpa, tincture of C. scammonia and AAL) in human platelet-rich plasma (PRP) was measured by the turbidimetric method where the aggregation was induced by the addition of agonists (adenosine diphosphate (ADP), epinephrine (EPI), arachidonic acid (AA) collagen (COL) or thrombin (TROM). Unlike tincture of C. scammonia and AAL, both O. macrocarpa tincture (TOM/preciptade (P) and supernatant (S)) and one of the organic fractions obtained from O. macrocarpa (TOM/1-F4) showed antiplatelet activity in human plasma where TOM/1-F4 (100 µg/mL) presented comparable effect to AAS. Physicochemical characterization (HPLC and spectrophotometer) of TOM/PS (content of resins 1,38g%) and TOM/1-F4 allowed the identification of phenolic acids (chlorogenic, galic and caffeic) as well as the determination of total phenols of TOM (0,14g%). The antiplatelet effect of TOM/PS and TOM/1-F4 seems to result from many intracellular actions. However, TOM/PS in presence of AAS, L-ARG, ODQ, pentoxifilin (PTX) or ticlopidine (TIC) had its antiplatelet effect modified mainly because of TIC, suggesting an important role of purinergic receptors in TOM/PS. TOM/PS presented antioxidant activity determined by DPPH test. Furthermore, TOM/PS and TOM/1-F4 did not showed anticoagulant action in human plasma but TOM/PS increased the bleeding time in mice. It was observed a relative toxicity of TOM/PS in human neutrophil. Therefore, this study demonstrated, as never before, the antiplatelet potencial of tincture and the organic fraction of "jalapa" and that at least related to the presence of phenols, particulary phenolic acids, and resins in plant. / Operculina macrocarpa (L.) Urb. (“jalapa brasileira”) (Convolvulaceae) é uma espécie comum do Nordeste brasileiro, sendo popularmente utilizada pelas suas propriedades laxativa, purgativa e “depurativa”. A tintura do tubérculo de jalapa, matéria-prima majoritária, ao lado da tintura de Convolvulus scammonia (TCS) compõem o fitoterápico Aguardente Alemã® (AAL) ou Tintura de Jalapa Composta, indicada como antitrombótico na medicina popular. O objetivo do presente trabalho foi investigar o potencial antiagregante plaquetário e anticoagulante de O. macrocarpa em plasma humano, incluindo estudo de bioprospecção e determinação do possível mecanismo de ação. A avaliação da atividade antiagregante plaquetária das drogas testes (tintura e frações de O. macrocarpa, TCS e AAL) em plasma humano rico em plaquetas (PRP) foi mensurada por método turbidimétrico, sendo a agregação induzida por vários agonistas como difosfato de adenosina (ADP), epinefrina (EPI), ácido araquidônico (AA), colágeno (COL) ou trombina (TROM). Ao contrário da TCS e da AAL, tanto a tintura de O. macrocarpa (TOM/precipitado(P) e sobrenadante(S)) quanto uma das frações orgânicas obtidas da planta (TOM/1-F4) mostraram atividade antiagregante plaquetária, onde a TOM/1-F4 (100 µg/mL) apresentou efeito comparável ao ácido acetilsalicílico (AAS). Caracterização físico-química (CLAE e espectrofotometria) da TOM/PS (teor de resinas 1,38g%) e da TOM/1-F4 permitiu a identificação de ácidos fenólicos (clorogênico, gálico e caféico), bem como a determinação do teor de fenóis totais da TOM (0,14g%). O efeito antiagregante plaquetário da TOM/PS e da TOM/1F-4 parece resultar de várias ações intracelulares. Contudo, a TOM/PS na presença do AAS, L-ARG, ODQ, pentoxifilina (PTX) ou ticlopidina (TIC) teve seu efeito antiagregante plaquetário modificado principalmente pela TIC, sugerindo um papel importante dos receptores purinérgicos na bioatividade da TOM/PS. A TOM/PS apresentou atividade antioxidante determinada através do teste do DPPH. Além disso, a TOM/PS e a TOM/1F-4 não mostraram ação anticoagulante em plasma humano, mas a TOM/PS aumentou o tempo de sangramento em camundongos. Foi observada uma toxicidade relativa da TOM/PS em neutrófilo humano. Dessa forma, o presente estudo comprovou, de maneira inédita, o potencial antiagregante plaquetário da tintura e da fração orgânica de jalapa, que estão pelo menos em parte relacionados à presença de fenóis, particularmente ácidos fenólicos, e resinas na planta.

O papel das plaquetas na regeneração hepática em modelo de hepatectomia 90%

A hepatectomia de 90% em murinos é um modelo de insuficiência hepática aguda. Recentemente, tem sido demonstrado que as plaquetas possuem um efeito importante na regeneração hepática. As plaquetas promovem a regeneração de tecidos através da liberação de fatores de crescimento tais como o Fator de Crescimento Derivado de Plaquetas (PDGF) e a Serotonina. O objetivo deste estudo foi avaliar o papel do plasma rico em plaquetas (PRP) em modelo de hepatectomia de 90% em ratos. Para isso, PRP ou medula óssea total (WBM) foram imobilizados em microcápsulas de alginato de sódio implantadas no peritônio de ratos imediatamente após a hepatectomia. O grupo controle recebeu cápsulas vazias (EC). Os animais foram sacrificados às 6, 12, 24, 48, 72 horas pós-hepatectomia (n=5-8/grupo/tempo) para avaliar a taxa de regeneração hepática, o índice mitótico, o índice de marcação com 5-bromo-2’-deoxiuridina (BrdU), HGF sérico, e a expressão no fígado remanescente de 5Ht2b e Pdgf. A taxa de sobrevida em 10 dias foi avaliada em outro grupo de animais (n=20/grupo). Foi observado que o grupo PRP apresentou a maior taxa de sobrevida aos 10 dias apesar da menor taxa de regeneração hepática em todos os tempos. Já o grupo EC mostrou uma taxa de regeneração maior nas primeiras 72 horas. O índice mitótico foi maior para o grupo PRP às 48 horas, porém o índice de BrdU não foi diferente entre os grupos em nenhum tempo. A expressão de 5Ht2b e Pdgf foi elevada no grupo PRP às 72 horas, entretanto o HGF sérico não apresentou diferenças entre os grupos. Estes resultados sugerem que o efeito benéfico do PRP pode ser mediado pelo incremento gradual e uniforme da taxa de regeneração e por uma resposta a fatores mitogênicos como a serotonina e o PDGF. / Ninety percent hepatectomy in rodents is a model for acute liver failure. Recently, it has been reported that platelet has a strong effect enhancing liver regeneration, due to its several growth factors such as Platelet Derived Growth Factor (PDGF) and serotonin. The aim of this study was to investigate the role of Platelet-Rich Plasma on 90% hepatectomy in rats. Platelet rich-plasma (PRP) or Whole Bone Marrow (WBM) were microencapsulated in sodium alginate and implanted in the peritoneum of rats immediately after 90% hepatectomy. Control group received empty capsules (EC). Animals were sacrificed at 6, 12, 24, 48, and 72 hours post hepatectomy (n=5-8/group/time) to evaluate liver regeneration rate, mitotic index, 5-bromo-2’-deoxyuridine (BrdU) labeling index, serum HGF, and 5Ht2b and Pdgf expression in liver. Survival rate in 10 days was also evaluated in a different set of animals (n=20/group). PRP group showed the highest survival rate at 10 days despite the lowest liver regeneration rate at any time point. EC group showed the highest regeneration rate in the first 72 hours. Mitotic index was highest in PRP group at 48 hours although BrdU index did not show any difference among groups. Gene expression of 5Ht2b and Pdgf was both up-regulated at 72 hours in PRP group, whereas serum HGF did not show any differences among groups. These results suggest that the beneficial effect of PRP may be mediated by a gradual increase in the regeneration rate and a response to mitogenic factors such as serotonin and PDGF.

As plaquetas foram estudadas do ponto de vista funcional, através da agregação plaquetária espontânea, e morfológico, através da ultra-estrutura e da morfometria plaquetária. Também foram analisadas plaquetas de indivíduos normais, de faixa etária inferior a 50 anos (GC1), para verificar eventual influência do fator idade na função plaquetária. Para a análise morfológica e morfométrica, sedimentos plaquetários foram processados para microscopia eletrônica de transmissão (MET) e varredura (MEV), segundo técnicas convencionais. Para a prova funcional, o plasma rico em plaquetas foi agitado em agregômetro de plaquetas, sem a adição de agonistas. Todos os indivíduos foram avaliados clínica e laboratorialmente. Com relação à função plaquetária, não houve diferença significativa na agregação plaquetária espontânea, entre os grupos analisados (p>0,05). A morfometria das plaquetas também não acusou diferença na forma das plaquetas, entre os grupos estudados (p>0,05). À MEV, as plaquetas dos indivíduos do grupo GC1 apresentaram freqüência de alterações na forma significativamente maior (42,70±4,54), quando comparadas às dos outros grupos, GC2 (32,31±6,22) e GAOP (37,76±8,91), p>0,05. A dosagem de fibrinogênio do grupo GAOP (262,90±47,86) revelou-se significativamente maior que do grupo GC1 (209,80±47,02), p>0,05; a dosagem de HDL colesterol do grupo GAOP (30,00±5,33) foi significativamente menor que do grupo GC2 (42,70±12,58), p>0,05. Nossos resultados mostram que as plaquetas circulantes dos pacientes portadores de AOP não... / This study aims to analyze the platelets of patients with peripheral obliterative atherosclerosis (POAG), at the intermittent claudication stage and comparing this platelets with the control group of healthy age and sex-matched subjects (CG2). The platelets were studied in relation to: their function, by the spontaneous platelet aggregation, and their morphology, by the platelet ultrastructure and morphometry. We also analyzed the platelets of younger healthy subjects aged less than 50 years (CG1) to find out a probable influence of age factor on platelet function. Morphological and morphometric analyses were performed using platelet pellets processed by transmission electron microscopy (TEM) and scanning electron microscopy (SEM), according to conventional procedures. The functional test was performed using the platelet-rich plasma, mixed in platelet aggregometer without the addition of stimulating agents. All the healthy subjects and patients submitted to clinical and laboratory evaluations. Platelet function did not show significant difference in the studied groups (p>0.05). Platelet morphometry did not show difference in the analysed groups (p.>0.05). The CG1 platelets showed major alteractions in shape (42.70±4.54) than the other groups: CG2 (32.31±6.22) and POAG (37.76±8.91), p>0.05. The POAG fibrinogen determination (262.90±47.86) was significantly higher than CG1 group (209.80±47.02), p>0.05. The POAG HDL determination (30.00±5.33) was significantly higher than CG2 (42.70±12.58), p>0.05. Our results reveal: the POAG circulating platelets do not show functional or morphological alteractions when compared to the age-matched healthy subjects; and the freqüency of circulating platelet with alteractions in shape using SEM preparations were greater in CG1. This suggests an influence of the age factor on function platelet.

RNA extraído foi utilizado para detectar a presença do vírus por RT-PCR e, avaliar as diferenças quantitativas na carga viral de acordo com o perfil HPA por qRT-PCR. Todas as plaquetas foram positivas para o RNA do VHC após a incubação com o vírus, o que demonstra que o vírus interage com as plaquetas in vitro. Foi observado um desvio no equilíbrio de Hardy-Weinberg no sistema HPA-1.
