Příprava vrstev metodou plazmové polymerace a jejich charakterizace / Preparation of layers by plasma polymerization and their characterization

The diploma work deals with the preparation of biocompatible hydrophilic layers from a propane-butane mixture by plasma polymerization. Subsequent characterization of the layers was performed using both biological (antibacterial tests and biocompatibility test) and physico-chemical diagnostics (SEM, OES and surface energy determination). The theoretical part presents the basic principle of dielectric barrier discharge, its use and various types, including surface dielectric barrier discharge (SDBD), which was used during deposition. Next, cell cultures, cell morphology and clarification of the term biocompatibility were described. At the end of the theoretical part, all used during the experimental part of the diagnostics were also described. In the experimental part, the procedure of deposition of hydrophilic layers and the overall apparatus, which was used during deposition, were presented. Prior to the characterization of the samples, the discharge was diagnosed using optical emission spectroscopy. Furthermore, the level of hydrophilicity of the layers and their elemental composition on the surface was determined. Biological tests for biocompatibility and antibacterial properties provided information on the viability of the cells on the layers and their antibacterial effects.

The thesis is focused on evaluation and comparison of welds made by welding technologies using laser and plasma. For the purpose of comparing those technologies, were chosen austenitic stainless steel x5CrNi18-18 and ferritic stainless steel X6Cr17. These materials were chosen for their diversity in properties gained by high temperatures that go along with most welding processes. First part of the thesis is focused on description of welded materials and technologies of laser and plasma welding. In following experimental section, the thesis is focused on evaluation of welded samples, made for this purpose. Samples were examined for metallography, specifically macrostructure and microstructure. Followed by tensile test of mechanical properties and microhardness test.

Ovládací a měřicí jednotka pro generátory plazmatu / Control and measurement unit for plasma generators

The diploma thesis deals with the issue of plasma generators. Specificantlly, the work focuses on the design of HMI devices for plasma generators, including the design of the control part of the generator. The main advantage of controlling the plasma generator using an HMI device is the ability to quickly change important parameters. The HMI device allows convenient switching of the generator output frequency without the need to interfere with the wiring, which leads to higher efficiency when working with the plasma generator. Another advantage of the HMI device is the display of all important information on the LCD display. The work deals with the basic theory of plasma and its use, the division of microcontrollers and the description of their peripherals, the selection of components used and their wiring. The last part of the work is devoted to the software of the plasma generator.

Srovnání kvantitativního měření ocelových standardů dvoupulzní a jednopulzní LIBS / Comparison of quantitative LIBS analysis of steel standards utilizing single-pulse and double-pulse LIBS

In this diploma thesis the differences between single pulse and double pulse laser induced breakdown spectroscopy (LIBS) methods are discussed. The time evolution of laser induced plasma (LIP) parameters, such electron density, plasma temperature, density of ions and atoms is shown. Selected methods for evaluation of plasma parameters (Boltzmann equation, Saha-Boltzmann equation etc.) are also stated. The theoretical bases of local thermodynamic equilibrium (LTE), which is necessary condition for using of previously stated methods for computation of plasma parameters, are discussed. Diploma thesis also contains the description of each component of our LIBS experimental set up and the way of their optimization. Presented theoretical advantages of DP LIBS method are demonstrated on results of experimental measurements of steel standards.

Rekombinace iontů s elektrony v nízkoteplotním plazmatu / Electron Ion Recombination in Low Temperature Plasma

The presented work focuses on study of electron - positive ion recombination in low temperature plasma. The principal method used were Flowing afterglow with Langmuir probe and Stationary afterglow with Cavity Ring-Down Spectrometer. For the first time electron and neutral assisted collisional radiative recombination of Ar+ ions was studied in temperature range of 50 - 200 K. Resulting ternary recombination rate coefficients are in good agreement with theoretical predictions. Recombination of H3 + and D3 + ions with electrons was studied in the temperature range of 50 - 300 K and binary and ternary recombination rate coefficients were obtained. The effect of nuclear spin on recombination of H3 + ions with electrons was studied in the temperature range of 80 - 200 K and state selective recombination rate coefficients were obtained for ortho- and para-H3 + . Results show that at 80 K para-H3 + recombines with electrons substantially faster than ortho-H3 + .

Šio darbo literatūros apžvalgoje aptariami plazmos tipai, plazmos charakteristikos bei sąveika su medžiaga. Aptariama plazminės implantacijos technologija. Trumpai apibūdinama vandens garų plazma ir jos panaudojimas. Apžvelgiama titano oksido struktūra bei jo panaudojimas katalizatorių gamybai, kurie gali būti skirti skaldyti vandens molekules ir gaminti vandenilį. Darbe paaiškinamas magnetroninis nusodinimas bei jo privalumai. Darbo metu buvo oksiduojamas titanas vandens garų plazmoje. Titano oksidacija priklauso nuo daugybės plazmoje vykstančių procesų (adsorbcija, sulaikymas, vakansijų susidarymas ir pan.). Titano oksido panaudojimas yra labai platus dabartiniu metu. Aptariama šio eksperimento technologija bei atliekama oksiduotų titano dangų analizė. SEM, XRD, AES, GDOES analizės metodais buvo tiriama titano dangos oksidacija ir aiškinamas oksidacijos mechanizmas. / Types of plasma, characteristics and plasma interaction with solids are discussed in the literature review of this paper. Also, the plasma immersion ion implantation are described. Water vapour plasma are briefly discussed. Titanium oxide structure and it‘s usage for catalyst which could split water molecules are reviewed. Magnetron deposition are explained in this paper. The titanium film was oxidized by water vapour plasma on experiment. The oxidation of titanium depends on many processes in plasma (adsorption, trapping, formation of oxygen vacancies and etc.). Appliance of titanium oxide is very large in recent times. Experimental technology are discussed and plasma treated films are analysed. Titanium oxidation was analysed by SEM, XRD, AES, GDOES. Oxidation mechanism was explained in this paper.

Studium dohasínajícího plazmatu ve směsích N2-H2 / Study of post-discharge in N2-H2 mixtures

The presented Thesis deals on the nitrogen-hydrogen DC post-discharge observations by optical emission spectroscopy. The plasma was generated in Pyrex tube in flowing regime at pressure of 1 kPa at discharge current of 100 mA. The optical emission spectra were recovered at discharge times up to 50 ms in nitrogen containing 0 – 92 % of hydrogen; the gas mixture volume, i.e. the gas speed in the system was conserved for all mixtures. The experiments were carried out at two reactor wall temperatures at the spectra collecting point – at ambient temperature and at the wall temperature of liquid nitrogen (temperature in plasma was about 150 K). The nitrogen first and second positive and first negative spectral systems were identified in the spectra, the hydrogen atomic Balmer series lines were recorded, too. No molecular hydrogen emission was determined during the post-discharge. Some non-identified bands (but with high probability bands of nitrogen Herman infrared system) at 690 and 780 nm were detected, too, mainly at low temperature. The intensities of all determined radiating species decreased exponentially or more than exponentially with the decay time. The experimental data showed strong quenching of all nitrogen radiative states even at very a few percent hydrogen additions. The shape of the selected intensity dependencies on both hydrogen concentration, and the decay time are nearly the same for N2(C) and N2+(B) states, dependencies for N2(B) levels differs of them. The hydrogen line emission was nearly independent on the hydrogen content in the gas mixture up to about 50%, at highest hydrogen concentrations it slightly increased. The results obtained at the decreased wall temperature were very similar, only intensities of all nitrogen spectra increased by the factor about 3, the intensities of levels populated by the recombination of nitrogen atoms increased by factor about five. The atomic hydrogen alpha line (at 656 nm) was the most sensitive on temperature decrease; its intensity increased over one order in whole observed time interval. The obtained results will be confronted with numeric model of kinetic processes in the near future. After that, the specific conditions applicable for the technological applications of nitrogen-hydrogen gas mixtures under post-discharge conditions will be proposed.

Aplikace metod počítačové fyziky při studiu interakce plazmatu s pevnými látkami / Application of methods of computational physics for the study of plasma-solid interaction

Low-temperature plasma and its interaction with immersed solids is studied in this work. The research of the physical processes on this interface is performed by two-dimensional particle computer model. The model uses molecular dynamic method. Mutual forces between particles are computed by Particle- in-Cell method. The main application of the model is in the area of the probe diagnostic of plasma. Simple problems are compared with theory and two dimensional effects are discused. Contribution of particle modeling to plasma research is showed on the problem of interaction of sheaths around cylidrical probes. We deal with question whether we are able to get some information about unevennesses at the surface of solid immersed in plasma by measuring probe characteristics in its surroundings. PharmDr. Martin Beránek, Ph.D. Title of diploma thesis: Free circulating nucleic acids in blood plasma Outline: The aim of this thesis was to compare various commercially available DNA extraction kits and choose the universally optimal method for routine isolation of free circulating DNA from blood plasma samples according to their optical characteristics, the qPCR results and yield of short fragments. Methods: Four commercially available methods of extraction have been chosen to compare: QIAmp DNA Blood Mini Kit, QIAmp DSP Virus Spin Kit (both Qiagen), NucleoSpin Plasma XS (Macherey-Nagel) and Agencourt Genefind v2 (Beckman Coulter). DNA was isolated from aliquots of a pooled blood plasma of healthy individuals. Plasmatic DNA was quantified after extraction by spectrophotometry, fluorimetry and quantitative polymerase chain reaction. Fragmentation analysis on selected samples by capillary electrophoresis was also realized. Results: The best yield and purity provided the method from Qiagen QIAmp DSP Virus Spin Kit.
