Biological significance of colour pattern and variation in the green poison frog, Dendrobates auratus

The assumption that the colours of the green poison frog, Dendrobates auratus, serve as a warning of unpalatability is not supported because the requirements of the theory of aposematism are not met. The theory predicts that the aposematic coloration be conspicuous to potential predators, yet this cannot be demonstrated. The only predator found for D. auratus was the theraphosid spider, Sericopelma rubronitens, which hunts using vibrational cues. Only after sampling all frogs offered did spiders reject the toxic D. auratus over nontoxic Physalaemus pustulosus. The spider's decision to reject D. auratus as prey was made without visual input, and therefore the colours and toxicity of the frogs are decoupled with respect to this predator. Discovering that the crab Armases angustum is a predator of D. auratus tadpoles suggests that D. auratus is most vulnerable to predation during the nontoxic tadpole stage. Indirect methods of estimating predation were unsuccessful. The use of plasticine model frogs to estimate avian predation yielded no useful information and although traumatic injury is usually attributed to attempted predation, this is not true for tropical anurans. The levels of traumatic injury for D. auratus and a cryptic nontoxic frog Physalaemus pustulosus were similar when the life span of the frogs was taken into account. The traumatic injuries in these frogs are a function of interactions with small invertebrates that are neither predator nor prey. The theory of aposematism also predicts that there should be low pattern variation within an aposematic species so that a single search image can be formed and avoided. The patterns of D. auratus are individually distinct and there is significant interpopulational variation in appearance due to differences in colour, pattern and animal size. O município de Juiz de Fora – MG é considerado área endêmica para serpentes, aranhas e escorpiões e carente de estudos neste campo. Este estudo descritivo retrospectivo teve por objetivo caracterizar os acidentes com peçonhas notificados no referido município. Foram avaliados os 1.500 envenenamentos ocorridos na região de abrangência sanitária do município sede, entre 2002 e 2007. Os dados foram coletados nas fichas de investigação do Sistema de Informação de Agravos de Notificação (SINAN), disponível no Departamento de Vigilância Epidemiológica local e registros de pacientes obtidos no Hospital de Pronto Socorro (HPS) Doutor Mozart Geraldo Teixeira. Tais dados foram avaliados por estatística descritiva, com auxílio do Software Statistical Package for the Social Sciences 14.0 e o georreferenciamento realizado com a utilização do Software ArcGIS 9.2. As análises evidenciaram maior frequência de acidentes por aranhas (28,9%, n = 434), escorpiões (25,3%, n = 379) e serpentes (19,8%, n = 297). A maioria dos atendimentos foi efetuada no Pronto Atendimento Central (79,3%, n = 1189) e HPS (19,9%, n = 299). Os acidentes ofídicos foram mais frequentes na zona rural (63,0%, n = 187), com aumento de casos no período quente e úmido do ano (70,7%, n = 210). O gênero Bothrops foi o agressor mais envolvido (67,0%, n = 199) e os membros inferiores os mais acometidos (64,6%, n = 192). As vítimas tinham, em média, 34 ± 18,3 anos. A taxa de letalidade foi de 0,34%. Os acidentes escorpiônicos ocorreram, em sua maioria, na zona urbana (81,8%, n = 310), com maior frequência no período quente e úmido (57,3%, n = 217). Tityus serrulatus foi o mais envolvido (63,2%, n = 213) e os membros superiores os mais acometidos (49,8%, n = 189). As vítimas tinham, em média, 37 ± 20 anos. A taxa de letalidade foi de 0,26%. Os acidentes por aranhas ocorreram na zona urbana em 50,0% (n = 217) dos casos e a frequência de ocorrência permaneceu estável ao longo do ano. O gênero Phoneutria foi o mais envolvido (45,4%, n = 197) e os membros superiores os mais acometidos (44,6%, n = 193). As vítimas tinham, em média, 37 ± 19 anos. Nos três tipos de acidente, os envenenamentos foram considerados leves (68,6%, n = 761), as vítimas foram atendidas em menos de 3 horas (64,1%, n = 712) e evoluíram para cura (80,5%, n = 894). A soroterapia foi realizada em 63,3% (n = 197) dos casos de ofidismo; 2,1% (n = 8), de escorpionismo e 3,2% (n = 14), de araneísmo. A sintomatologia local mais frequente foi a dor (81,4%, n = 904). A maioria dos acometidos era do sexo masculino (67,7%, n = 752) e a atividade laboral foi a circunstância mais relacionada ao acidente (36,5%, n = 405). O número de acidentes no período analisado confirma a área como endêmica para estes animais e justifica estudos que permitam o conhecimento do perfil epidemiológico da região e desenvolvimento de políticas públicas locais. / Accidents involving venomous animals are usually severe, therefore resulting in high morbidity and mortality. The city of Juiz de Fora - MG is considered endemic area for snakes, spiders and scorpions, but there is a lack of studies on these subjects. This retrospective descriptive study aimed to characterize the venom accidents reported in that city. We evaluated the 1,500 poisonings occurred in the region, between 2002 and 2007. Data were collected in the investigation forms of the Information System for Notifiable Diseases, available at the Department of Epidemiological Surveillance and the patient registers obtained from the Hospital de Pronto Socorro (HPS) Doctor Mozart Geraldo Teixeira. These data were analyzed by descriptive statistics, with the aid of Software Statistical Package for Social Sciences 14.0 and the georeferencing was performed using the software ArcGIS 9.2. The results showed higher frequency of accidents was caused by spiders (28.9%, n = 434), scorpions (25.3%, n = 379) and snakes (19.8%, n = 297). Most of the calls was made at the Emergency Care Center (79.3%, n = 1189) and HPS (19.9%, n = 299). The snake bites were more frequent in rural areas (63.0%, n = 187), mostly during the h ot and humid periods of the year (70.7%, n = 210). The genus Bothrops was the aggressor more involved (67.0%, n = 199) and the lower limbs were the most affected (64.6%, n = 192). The victims aged 34 ± 18.3 years. The mortality rate was of 0.34%. Scorpion stings occurred mostly in urban areas (81.8%, n = 310), with higher frequency in hot and humid seasons (57.3%, n = 217). Tityus serrulatus was the more involved (63.2%, n = 213) and the arms were the most affected part of the body (49.8%, n = 189). The victims were 37 ± 20 years. The mortality rate was of 0.26%. The accidents by spiders occurred in urban areas in 50.0% (n = 217) of the cases and the frequency of occurrence have remained stable throughout the year. The genus Phoneutria was the more involved (45.4%, n = 197) and the arms were the most affected (44.6%, n = 193). The victims were 37 ± 19 years. In all three types of accidents, poisonings were considered mild (68.6%, n = 761), because the victims were treated up to 3 hours (64.1%, n = 712) and all of them got cured (80.5%, n = 894). The antivenin was performed in 63.3% (n = 197) of cases in snakebite, 2.1% (n = 8) in the scorpion cases and in 3.2% (n = 14) of the spider ones. The local symptoms more frequently were pain (81.4%, n = 904). Most affected victims were male (67.7%, n = 752) and work activity was the fact more related to the accident (36.5%, n = 405). The number of accidents in the period under review confirms the area as endemic for these animals and justifies studies to the knowledge of the epidemiological profile of the region and development of local public policies.

Efeito da Abarema cochliacarpos (Gomes) na lesão muscular induzida pelo veneno de Bothrops leucurus / Effect of Abarema cochliacarpos (Gomes) in muscle injury induced by Bothrops leucurus

The snakebite is a public health problem worldwide, especially in tropical countries. The objective of this study was to investigate the mechanisms involved in the effect of hydroethanolic extract of the stem bark Abarema cochliacarpos (EAc) in muscle injury induced by Bothrops leucurus. Male swiss mice were used (28-32g ; n= 6 groups), where they received perimuscular injection Bothrops leucurus venom (BlV 1 mg/Kg/paw Volume 50 ìl) the right hind limb, treated orally (po), with vehicle (saline) or EAc (100, 200 or 400 mg / kg). In the mechanical hypernociception animals were evaluated in time 2, 4 and 6 hours using digital analgesymeter (von Frey). Edema activity in the animals were evaluated at 15, 30, 60 and 90 minutes, using a digital caliper. Have motor activity was assessed by the rota -rod test and the animals were evaluated at 1, 3 and 7 days. Histological evaluation extensor digitorum longus muscle (EDL) was isolated, removed, fixed, paraffin emblocado (Optical Microscopy) and resin (Electron Microscopy) and cut. Tissues were stained with hematoxylin and eosin and observed under optical and electron microscopy and subsequently photographed. The experimental protocols were approved by the ethical committee for animal research at UFS (CEPA: 61/12). The results were analyzed using followed by Student.s t-test. The treatment orally with EAc (400 mg / kg ) inhibited mechanical hypernociception (2h 5.1 ± 0.76, 5.70 ± 0.65 4h, 6h 5.93 ± 0.49, (p < 0, 05) compared with the BlV venom group (2h 2.08 ± 0.33; 4h 2.28 ± 0.18; 6h 2.52 ± 0.24). The inhibition of edema was also seen in activity with EAc (400 mg / kg) (15 min 15.35 ± 0.27; 30 min 12.63 ± 0.69; 60 min 9.38 ± 0,29 and 90 min 6.83 ± 0.66, p < 0.05) compared with the BlV venom group (15 min 29.7 ± 0.17; 30 min 25.8 ± 0.26; 60 min 20.15 ± 0.24 and 90 min 14.76 ± 0.21). Regarding motor activity, the EAc (400 mg / kg) preserved motor ability (1day 83.22 ± 0.46; 3 days 98.02 ± 0.20 and 7 days 119.24 ± 0.48, p < 0.05) compared to the BlV venom group (1 day 20.03 ± 0.26, 3 days 35.22 ± 0.36 and 7 days 111.21 ± 0.18). Histological analysis showed an protection of muscle injury after administration of EAc (400 mg / kg), maintaining muscle fibers. Our results demonstrated that EAc inhibited the harmful effects of the venom, suggesting that this compound has biotechnological potential in adjuvant treatment of snakebite. / O ofidismo e um problema de saude publica em todo o mundo, especialmente nos paises tropicais. O objetivo deste trabalho foi de investigar os mecanismos envolvidos no efeito do extrato hidroetanolico da entrecasca da Abarema cochliacarpos (EAc) (popularmente conhecida como gbarbatimao h) na lesao muscular induzida pelo veneno de Bothrops leucurus (BlV). Foram utilizados camundongos Swiss machos (28-32 g; n=6 por grupo), que receberam injecao perimuscular do veneno Bothrops leucurus (BlV . 1 mg/Kg/pata . Volume 50 Êl) no membro posterior direito, tratados por via oral (v.o.), com veiculo (solucao salina) ou EAc (100, 200 ou 400 mg/kg). Na hipernocicepcao mecanica os animais foram avaliados nos tempos 2, 4 e 6 horas, utilizando o analgesimetro digital (Von Frey). Na atividade edematogenica os animais foram avaliados nos tempos 15, 30, 60 e 90 minutos, utilizando o paquimetro digital. Ja atividade motora foi avaliada pelo teste de rota-rod e os animais foram avaliados em 1, 3 e 7 dias. Na avaliacao histologica o musculo Extensor digitorum longus (EDL) foi isolado, retirado, fixado, emblocado com parafina (Microscopia optica) e resina (Microscopia eletronica) e cortados. Os tecidos foram corados com hematoxilina- eosina e observados ao microscopio optico e eletronico e posteriormente fotografados. Os protocolos experimentais foram aprovados pelo comite de etica em pesquisa com animais da UFS (CEPA: 61/12). Os resultados foram analisados utilizando o teste Student.S t-test. O tratamento, por v.o, com EAc (400 mg/Kg) inibiu a hipernocicepcao mecanica, (2h 5,1 }0,76; 4h 5,70 }0,65; 6h 5,93 }0,49; p<0,05) quando comparados aos animais do grupo BlV (veneno) (2h 2,08 }0,33; 4h 2,28 }0,18; 6h 2,52 }0,24). A inibicao tambem foi verificada na atividade edematogenica, com EAc (400 mg/kg), (15 min. 15,35 }0,27; 30 min. 12,63 }0,69; 60 min. 9,38 }0,29 e 90 min. 6,83 }0,66; p<0,05), quando comparados aos animais do grupo BlV (veneno) (15 min. 29,7 }0,17; 30 min. 25,8 }0,26; 60 min. 20,15 }0,24 e 90 min. 14,76 }0,21). Quanto a atividade motora, o EAc (400 mg/Kg) preservou a capacidade motora (1 dia 83,22 }0,46; 3 dias 98,02 }0,20 e 7 dias 119,24 }0,48; p<0,05), comparados ao grupo BlV (veneno) (1 dia 20,03 }0,26; 3 dias 35,22 }0,36 e 7 dias 111,21 }0,18). Na analise histologica verificou-se uma protecao da lesao muscular apos administracao do EAc (400 mg/Kg), preservando as fibras musculares. Nossos resultados demonstraram que o EAc inibiu os efeitos nocivos do veneno, sugerindo que este composto apresenta potencial biotecnologico no tratamento coadjuvante do ofidismo.
