This thesis focuses on the traditional cultivation of an edible root species by Kwaxsistalla, Clan Chief Adam Dick, of the Qawadiliqalla Clan, of the Dzawada ēnuxw, a subgroup of Kwakwaka’wakw, occupying the Kingcome Inlet area on the Central Coast of British Columbia. Kwaxsistalla is a traditionally trained Clan Chief and potlatch speaker with recognized authority to share his detailed knowledge and experiences of his clan’s food production system. This research is centered on his Clan’s tekkillakw (estuarine salt marsh root garden) root gardens of the Kingcome River estuary, and the long-standing practices associated with the large-scale production of tleksem Pacific silverweed [Argentina egedii (Wormsk.) Rydb.; syn. Potentilla pacifica (L.) Howell.], is one of the four cultivated root species. Kwaxsistalla has shared his hands-on knowledge of how root garden cultivation fits into his family’s seasonal patterns of food production as well as detailed accounts of how to construct and use tools for cultivating, weeding, harvesting, and cooking estuarine roots. He has also provided information that has been instrumental in developing a model of aboriginal management of estuarine root gardens (Deur 2005). This thesis builds on Deur’s model by attempting to experimentally replicate tekkillakw management in order to better understand the management effect on the abundance, size, and flavour of Argentina egedii roots. Over the course of the 2008 growing season I randomly subjected 60 ¼ square meter patches of Kwaxsistalla’s fallow tekkillakw to either a “till” or “till + weed” treatment and allocated 30 similar patches as a control. I applied a roto-tilling treatment just prior to the growing season, a weeding treatment mid-summer, and harvested the roots near the end of the growing season. While the short duration of my study and use of a roto-tiller limit the inferential power of my results, I found that tilling and weeding significantly increased the abundance or A. egedii but significantly decreased the root size. Throughout the same 2008 field season I also collected root specimens for analysis of their bitter and sweet constituents and found (bitter) tannins concentrations to be highest in the late summer and lowest in the spring and fall. / Graduate

Lietuvoje augančių Potentilla genties augalų fitocheminė analizė bei vartojimo galimybių tyrimai in vitro / Phytochemical analysis of in Lithuania growing Potentilla genus plants and using possibilities in vitro

Fitoterapija (gr. phyton – augalas, therapeia – gydymas) – tai ligų profilaktika ir gydymas, vartojant augalų dalis ar jų preparatus. Nuo seno lietuvių liaudies medicinoje gerai žinomi sidabražolinių (Potentilla) genties augalai. Lietuvoje auga 17 Potentilla genties augalų rūšių, kai kurios iš jų kultūrinės. Šio darbo tikslas yra atlikti biologiškai veiklių medžiagų – flavonoidų ir raugų – išskyrimą iš Potentilla genties augalų (Potentilla anserina L., Potentilla erecta L., Potentilla fruticosa L.) nustatyti veikliųjų medžiagų kiekio kitimus skirtingu augalų vegetacijos metu (birželio – rugsėjo mėnesiais) ir atlikti šių augalinių žaliavų antioksidacinio bei antimikrobinio poveikio tyrimus. Tyrimo medžiaga ir metodai. Tyrime naudota Potentilla genties augalinė žaliava surinkta Šiaulių rajone, Gruzdžių miške (P.anserina žolė, P.erecta šakniastiebiai ir žolė) bei Šiaulių universiteto botanikos sode (P.fruticosa žolė). Permanganatometrinio titravimo metodu buvo nustatytas rauginių medžiagų kiekis tirtuose Potentilla genties augalinėse žaliavose. Naudojant parūgštintą vandenilio chlorido rūgštimi 70% etanolį ir 70% etanolį spektrofotometriniu ir efektyviosios skysčių chromatografijos (ESC) metodu buvo nustatyti bendri flavonoidų kiekiai tirtose augalinėse žaliavose. Antioksidaciniam aktyvumui įvertinti buvo taikomas fotometrinis 2,2-difenil-1-pikrilhidrazilo (DPPH•) laisvojo radikalo sujungimo metodas. Since ancient times Cinquefoil (Potentilla) family plants have been well-known in Lithuanian non-traditional medicine, comprising over 17 species, some of them are cultivated. Aims and Objectives. To extract biologically active substances – flavonoids and tannins – from Potentilla plants (Potentilla anserina L., Potentilla erecta L., Potentilla fruticosa L.) in order to determine the quantity changes at different times of plant vegetation (June – September), and to research antioxidant and antibacterial effect of those plants. Materials and Methods. Potentilla raw material was collected in the region of Šiauliai, Gruzdžiai forest (Herba Potentillae anserinae, Rhizoma et herba Potentillae erectae) and Šiauliai University Botanical Garden (Folium cum flore Potentillae fruticosae). Permanganatometric titration method was used to find out how much tannin the selected Potentilla contained. Also, spectrophotometric and HPLC methods allowed to measure flavonoid while using 70% of ethanol with hydrochloric acid and 70% of ethanol in the raw vegetation. To evaluate the anti-oxidizing activity photometric 2,2-difenil-1-pikrilhydrosil (DPPH•) free radical method was applied. Systematics and polyploid evolution in Potentilleae (Rosaceae)

This thesis comprises studies of the phylogenetic relationships in the flowering plant clade Potentilleae in Rosaceae. The relationships were elucidated by using DNA sequence data from the nuclear genome as well as from the plastid genome. In particular, the focus of the studies was the investigation of allopolyploidy, i.e. speciation as a result of hybridization and subsequent chromosome doubling. A phylogenetic method was used for identifying allopolyploidy through comparison of trees resulting from the analyses of different DNA sequences. Five sub-clades were investigated. First, both the sister clades that together contain all of Potentilleae: Fragariinae and Potentilla. Secondly, three subclades of Fragariinae, namely Alchemilla in wide sense, Sibbaldia and relatives, and Fragaria. The aim was to unravel the phylogenetic relationships, including instances of allopolyploidy. Classification issues were discussed in relation to the phylogenetic results. The split between Potentilla (=Potentillinae) and Fragariinae received better support than in previous studies. The phylogeny of Fragariinae was found to be consistent with classifying ten genera: Alchemilla in wide sense (incl. Aphanes and Lachemilla), Comarum, Sibbaldia, Sibbaldianthe, Sibbaldiopsis, Chamaerhodos, Drymocallis, Dasiphora, Potaninia, Fragaria, and also including a few orphan Potentilla species. The segregated genera Ivesia, Horkelia, Horkeliella and Duchesnea were found to be nested within Potentilla, corroborating earlier studies, while the segregated genus Argentina (P. anserina and close relatives) showed an ambiguous position. Plastid and nuclear (ribosomal) phylogenies were compared and incongruences were detected as potential instances of allopolyploid speciation. Five strongly supported incongruences were detected in Fragariinae and four of them were considered to be potentially caused by allopolyploidy. In addition, five supported incongruences were found in Potentilla. Alchemilla in the wide sense was found to contain four major clades, African Alchemilla, Eurasian Alchemilla, Lachemilla and Aphanes. Both Lachemilla and Aphanes were nested within Alchemilla and it was suggested that the name Alchemilla should be used in the wide sense, i.e. including both the genera Lachemilla and Aphanes. The genus Sibbaldia as commonly classified was shown to be polyphyletic in five different places in Potentilleae. Three Sibbaldia clades ended up in Fragariinae and two in Potentilla. A phylogeny of Fragaria, based on a nuclear low/single copy DNA region was estimated. The gene copy phylogeny was used to construct a reticulate tree hypothesizing allopolyploid speciation events.
