The thesis has a theoretical and empirical nature. It focuses on issues of teacher's personality and view of 2nd Primary school for her. The first part describes personality traits, teacher and diverse teaching styles. It defines the professional competence of teachers, such as competence and communicative competence reflect on their activities. Attention is paid to define the characteristics of "good" teachers, who are the authority, assertiveness, emotional stability, sense of humor, fair evaluation, creativity and flexibility. In conclusion, the theoretical part of the established requirements and objectives for the current school teachers and the teaching profession The influence of personality. The practical part presents the results of quantitative research, which was written the method of questioning. The data obtained explain the view of 2nd Primary school teacher and a personality show, which features students assessed as significant and which forms and methods of work adopted positively.

Výtvarné studium skutečnosti ve VV na 1. stupni ZŠ / Study of Reality in Art Education at the Primary School

The diploma thesis deals with the art studies of reality at primary school. The thesis is divided into a theoretical and an empirical part. The theoretical part has three chapters in total. The first one is focused on the definition of the reality study and its development from prehistoric times to contemporary art. In the second, the focus is on the historical and contemporary concepts of art education. The last section is devoted to the ontogenetic pupils' development of younger and middle school age. Although reality study is found in the curriculum, there is considerable reserve in its application in art education. In the empirical part, we find out what teachers respond to the study of reality as a concept, and whether they are aware of when this issue is introduced into art lessons, or if they reveal some progress in pupil's speech. Through the research method, an interview was selected, which is a qualitative research.

The diploma thesis deals the phenomenon of education of Roma children in the system of special elementary education. The aim of the thesis is to capture the specifics of this education focusing on the analysis of the civic education on primary practical schools in context of relationships to Roma pupils. The theoretical part is focused on the current situation of the Roma minority in the Czech republic. The work briefly describes the history of the Roma minority and the current situation of Roma people in the modern society. Most attention is paid to the Czech primary educational system, especially teaching Roma pupils on practical primary schools. The work briefly deals with the current changes in the educational system, so called inclusive education too. The practical part is focused on the methodology of the multicultural education themes, on the second degree of an elementary practical school in Vimperk.

This master thesis investigates attitudes of pupils aged 13 to 15 to visual arts. The work is divided into two sections. Within a theoretical section, the focus is on pupils from the point of view of developmental and social psychology. The following section is dedicated to attitudes from the perspective of general and social psychology. Afterwards, the issue of educational institutions and their curricular documents concerning visual arts is discussed - whether the output of the subject fulfils particular goals of visual literacy. The theoretical section also mentions methods, forms and actions which can be used to mediate visual arts. An empirical section researches an answer to the question: "What are the attitudes of pupils aged 13 to 15 to visual arts?" In order to find out the answer, a quantitative research combining questionnaires and a method of semantic differential is conducted.

Didaktické zpracování tématu: Občanská a politická participace / Didactic analysis of the topic of civic and political participation

The diploma thesis deals with the concept of civic and political participation in elementary and secondary school. Introductory part of the thesis defines terms of civic and political participation, there are also findings about civic attitudes of young Czechs based on international comparative researches. The focus of the thesis is to analyze the representation of the theme of participation at different levels of the Czech education system and to compare the concept of the theme in curriculum documents of selected states. This is followed by an analysis of the approach to the topic of participation by authors of approved Czech textbooks. The analyzes show that the theme of participation is insufficiently embedded in the Czech national curriculum and these deficiencies are reflected in the approach to the topic in school educational programs and textbooks. In the light of these findings, a didactic analysis of the topic of participation is carried out. The diploma thesis sets down the theoretical minimum for a given topic. The last part presents proposals of didactic procedures in teaching focused on topics of civic and political participation. Additionally, the thesis recommends projects of non-profit institutions and other initiatives that focus on selected aspects of the topic of participation.

My diploma thesis deals with Cross-curricular themes for teaching in primary school education and especially with the Cross-curricular theme Education to Thinking in European and Global Context. In the theoretical section I have explained the changes in the curriculum, the White paper and the Framework educational program and briefly I have characterized all Cross-curricular themes. Theoretically and within educational areas I have focused on the Cross-curricular theme Education to Thinking in European and Global Context. In the practical section I have done a survey of primary school teachers to examine attitudes to Cross-curricular themes and the integration of the Cross-curricular theme Education to Thinking in European and Global Context into lessons. I have compared relation to each Cross-curricular theme. I have also suggested an intensive instruction leading to the formation of attitudes and opinions by content of the Education to Thinking in European and Global Context.

Structural Studies on DNA Damage Inducible Protein 1 (Ddi1) of Leishmania and the Rotavirus Nonstructural Protein NSP4

Structuraj investigations on the Ddi1 (DNA-damage inducible protein 1) of Leishmania major and on the rotavirus nonstructural protein NSP4 were carried out. Ddi1 belongs to the ubiquitin receptor family of proteins. One of its domains is similar to the retroviral aspartic proteinases. It has been shown that this domain is the target of HIV-protease inhibitors that were being used in the treatment of AIDS and it was observed that these drugs effectively controlled opportunistic diseases caused by many parasitic protozoa such as Leishmania and Plasmodium species. The retroviral protease-like domains present in Ddi1 proteins of these organisms were identified as the targets of these drugs. Structural studies on Ddi1 from L. major have been carried out, in an attempt to provide a platform for the design of anti-protozoal compounds. Rotavirus NSP4, the first viral enterotoxin to be identified, is a multifunctional glycoprotein that plays critical roles in viral pathogenesis and morphogenesis. As part of an ongoing project on the structural characterization of NSP4, we determined the structure of the diarrhea-inducing region of this protein from the rotavirus strain MF66. Chapter 1 presents an overview of Ddi1 and NSP4 of the rotavirus with an emphasis on their structural features. The methods employed during the course of the present work are described in Chapter 2. Structural studies on the retroviral protease-like domain of Ddi1 (Ddi1-RVP) of L. major is presented in Chapter 3. Apart from this domain, Ddi1 of L. major also has a ubiquitin-associated and ubiquitin-like domains whereas P. falciparum has only the ubiquitin-associated domain. Activity of the full length Ddi1 of L. major and the retroviral protease domain of P. falciparum using an HIV protease substrate was shown to be inhibited by an HIV protease inhibitor, saquinavir. Binding of saquinavir to the proteins was also confirmed by Biolayer Interferometry studies. The crystal structure of the retroviral protease domain of L. major Ddi1 has been determined. It forms a homodimeric structure similar to that of HIV protease and the reported structure of the same domain from Saccharomyces cerevisiae. The loops in Ddi1-RVP are similar to the 'flap' regions of the HIV protease which close-in upon substrate/inhibitor binding; they are visible in the electron density maps, unlike the case of the S. cerevisiae protein. Though the native form of the domain shows an open dimeric structure, normal mode analysis reveals that it can take up a closed conformation resulting from relative movements of the subunits. The present structure of Ddi1-RVP of L. major with the defined 'flap'-like loops will be helpful in the design of effective drugs against protozoal diseases, starting with HIV protease inhibitors as the lead compounds. Chapter 4 describes the structural investigations carried out on the diarrhea-inducing region of the nonstructural protein NSP4 of the rotavirus strain MF66 which forms an α-helical coiled-coil structure. Crystal structures of a synthetic peptide and of two recombinant proteins spanning this region showed parallel tetrameric organization of this domain with a bound Ca2+ ion at the core. Subsequently, we determined the structure of NSP4 from a different strain as a pentamer without the bound Ca2+ ion. This new structure provides more insights into understanding some of the functions of NSP4 such as the release of ions into the cytoplasm and binding to the double-layered particle (DLP). We also established conditions responsible for these structural transitions. The crystal structure of the coiled-coil domain of NSP4 presented in this chapter shows an entirely different structure which is an antiparallel tetramer. This explains our failure to determine the structure by the molecular replacement method using known oligomers. The structure was solved by the Sulphur-SAD method using diffraction data collected with Cr Ka radiation. The study reveals that the structural diversity of NSP4 is not limited. We could relate sequence variations and pH conditions to the differences in oligomeric assemblies. Surface properties of the domain suggest that the new form is likely to interact with different sets of proteins compared to those that interact with the parallel tetramers or pentamers. Further investigations are needed to establish this property.

This diploma thesis is focused on pupils' motivation to learning in ICT at vocational secondary schools and colleges. The main educational goal of ICT as a subject is to develop pupils' ICT literacy. The theoretical part of this thesis deals with motivation as a pedagogical concept and appropriate teaching approaches to it. An analysis of ICT subjects in the curriculum of the Framework Education Programmes for vocational secondary schools and colleges contributes to form an integral part of the theoretical base. The empirical part is divided into three mini-surveys carried out at a vocational secondary school and training institution in Liberec. The first was a questionnaire for pupils to discover what motivates them to study ICT. The second was a questionnaire for teachers which aimed to find out if and how teachers apply and exploit pupils' ICT literacy in their teaching of vocational ICT as a specialist subject. The third element was interviews with ICT teachers with the aim of investigating how ICT teachers understand the role of ICT as a subject and how they develop their pupils' ICT literacy. The discourse deals with the potential of implementation of joint teaching of history to pupils of multi-grade class in a primary school with composite classes. The education will proceed through the use of methods and techniques of the theatre education. Therefore the theoretical section pursues the features of the theatre education and its benefit to pupils' education. The discourse also explores whether the curricular documents provide for such teaching. The practical section of the discourse inquires into class education planning, its fruition and reflection, it also summarizes the used methods and techniques of the theatre education. A part of the section is an aggregate chart of pupils evaluating feasibility of the used activities. In conclusion a hypothesis is established that the education by means of theatre education is attractive to pupils.
