Avaliação da transfusão sanguínea homóloga em ovinos / Evaluation of homologous blood transfusion in sheep

Eighteen adult Santa Inez crossbred sheep, males and females, were used, weighing on average 52kg. The animals were submitted to a single phlebotomy to remove 40% of blood volume and equally divided into three experimental groups: G0 - animals receiving fresh blood; G15 receiving blood stored for 15 days; and G35 - receiving blood stored for 35 days. The animals were submitted to clinical examination and blood collection 24 h after induction of anemia (T0), 30 minutes after transfusion (T30), six, twelve, twenty-four, forty-eight, seventy-two and ninety-six hours post-transfusion (T6, T12, T24, T48, T72 and T96, respectively), and eight and sixteen days post-transfusion (T8d and T16d, respectively). The blood bags stored for 35 days showed an increase in K, pCO2, pO2, lactate, plasma hemoglobin and decreased plasma pH, sodium and leukocytes. The sheep transfused from all groups had increased GV, red cell count and total hemoglobin in the T30. The animals of the G35 had higher plasma hemoglobin in T12, a significant decrease in blood pH indicating a mild metabolic acidosis on T96. With respect to oxidative stress, was observed a decreased on catalase values of the G35 at T30, T6, T12 and T24, suggesting the occurrence of hemolysis witch was supported by the concomitant increase in the total bilirubin values at the same periods. Animals that received blood stored for 35 days showed higher alteration on hematological, blood gas, biochemical and oxidative stress parameters / O presente trabalho objetivou avaliar as respostas clínicas, hematológicas, bioquímicas, hemogasométricas e o estresse oxidativo de ovinos induzidos à anemia superaguda e transfundidos com sangue total fresco ou armazenado por dois diferentes períodos (15 e 35 dias). Foram utilizados 18 ovinos, machos e fêmeas, com idade entre 3 e 4 anos, pesando em media 52kg. Os animais foram submetidos a uma única flebotomia para retirada de 40% do volume sanguíneo e divididos em três grupos experimentais, sendo o G0 composto por animais que receberam sangue fresco, G15 e G35 animais que receberam sangue armazenado em bolsas CPDA-1 por 15 e 35 dias, respectivamente. Foi realizado exame clínico e coleta de amostras de sangue 24 horas pós-indução da anemia (T0), 30 minutos pós-transfusão (T30), seis, 12, 24, 48, 72 e 96 horas após à transfusão (T6, T12, T24, T48, T72, T96, respectivamente) e oito e dezesseis dias após à transfusão (T8d e T16d, respectivamente). O sangue armazenado por 35 dias apresentou aumento do potássio, pCO2, pO2, lactato, hemoglobina plasmática e diminuição do pH, sódio e leucócitos. Ovinos transfundidos no T30 apresentaram aumento significativo do VG, hemácias e hemoglobina total. Os animais do G35 apresentaram maiores valores de hemoglobina plasmática no T12 e diminuição do pH sanguíneo, caracterizando leve acidemia metabólica no T96. Com relação ao estresse oxidativo o G35 apresentou redução da catalase no T30, T6, T12 e T24, indicando a ocorrência de hemólise, o que foi corroborado pelo aumento concomitante da bilirrubina. Flow of healthy and sickle red blood cells in microcirculatory conditions : clustering process and self-margination phenomenon / Écoulement de globules rouges sains et drépanocytaires en conditions micro-circulatoires : processus d'agrégation (clustering) et phénomène d'automargination

J'ai caractérisé expérimentalement la formation de clusters au cours du passage de globules rouges (GRs) sains et drépanocytaires dans microcapillaires droites. L'effet de l'agrégation a été également étudié. J'ai montré que la formation des clusters dans des conditions physiologiques est due à la combinaison des interactions hydrodynamiques et des celles causées par les macromolécules du plasma. En effet, les interactions macromoléculaires ne sont pas complètement atténuées sous contraintes de cisaillement physiologiques et au contraire ils contribuent à la stabilité des clusters. En outre, j'ai découvert la présence d’une distribution bimodale en ce qui concerne les distances entre les cellules constituant les clusters hydrodynamiques.En plus, j'ai étudié expérimentalement le comportement collectif des globules rouges drépanocytaires oxygénés et leur distribution radiale le long de microcapillaires cylindriques avec un diamètre comparable à ces des veinules et des artérioles humaines. J'ai trouvé que les GRs montrent une distribution hétérogène en fonction de leur densité: les cellules plus légères ont tendance à rester prés du centre du canal, alors que la plupart des cellules denses (et aussi plus rigides) auto-marginent sous des conditions définies. L'agrégation semble d'inhiber l'auto-margination en fonction des patients et en particuliers des facteurs d’agrégation: le dextrane, par exemple, favorise l'auto-margination dans certains patients et il la diminue dans des autres. Le plasma montre de contraster l'auto-margination des GRs dans tous les sujets, en soulignant l'importance des protéines et des molécules adhésives du plasma dans les phénomènes d'agrégation. Finalement, j'ai observé que l'auto-margination se manifeste naturellement au cours de l’écoulement de globules rouges drépanocytaires. / I experimentally characterized the clustering formation of healthy and sickle red blood cells (RBCs) flowing through straight micro-capillaries. The effect of aggregation was also investigated. I found that cluster formation under physiological conditions is most likely caused by a combination of hydrodynamic and macromolecule-induced interactions. Macromolecule-induced interactions are not fully overcome by shear stresses within the physiological range, and they contribute to cluster stability. Moreover, I found that a pronounced bimodal distribution of the cell-to-cell distances in the hydrodynamic clusters is produced.Additionally, I investigated experimentally the collective behavior of oxygenated sickle RBCs and their distribution along cylindrical micro-capillaries with diameters comparable to a human venule or arteriole. I have shown that there is a heterogeneous distribution of RBCs according to their density: low-density cells tend to stay closer to the center of the channel, while most dense cells (also more rigid) self-marginated under defined conditions. Aggregation seems to inhibit self-margination depending on the aggregative factor and patient: dextran allows self-margination in some patients and inhibits it in others. Plasma inhibits self-margination of cells in all cases, highlighting the importance of the plasma proteins and adhesive molecules in the aggregation phenomena.

Derangements of tonicity and implications for veterinary patients

Master of Science / Department of Clinical Sciences / Thomas Schermerhorn / Tonicity is property of a solution that is defined as the total effective (impermeable) osmole concentration that drives fluid movement across a semipermeable membrane via osmosis. Tonicity is related to but distinct from solution osmolality, which is a summation of all solute concentrations, regardless of the solute membrane permeability. In the mammalian body, tonicity is tightly regulated at both a cellular and systemic level; tonic derangements cause rapid change in cell and tissue volume leading to significant dysfunction. Input from the central nervous, circulatory, endocrine, gastrointestinal, and urinary systems are integral to osmoregulation, so many diseases in veterinary medicine are associated with tonicity disorders. However, because the homeostatic mechanisms that control tonicity overlap with those regulating electrolyte and acid-base balance as well as hydration and vascular volume, tonic consequences of disease can be difficult to isolate. Understanding of disease-associated changes in tonicity is further complicated by the fact that the tonic contributions of many solutes that accumulate in disease are unknown. Additionally, direct assessment of tonicity is difficult because tonicity is not just a physiochemical property, but it implies a physiologic effect. Thus, simple summation of osmole concentrations is an inadequate measurement of tonicity. The following report includes three studies investigating various aspects of tonicity as it applies to veterinary patients. Chapter 2 reports a study that examines the tonic effects of ketoacids and lactate using two different in vitro red blood cell assays. Results demonstrated that the ketoacids, beta-hydroxybutyrate and acetoacetate, behave as ineffective osmoles while the tonic behavior of lactate is variable, implying a more complex cellular handling of this anion. Two additional studies examine whether the mean corpuscular volume difference (dMCV) is a novel clinical marker for hypertonicity in dogs. Results of separate retrospective (Chapter 3) and prospective (Chapter 4) studies provide evidence that dMCV is a useful clinical marker for hypertonicity in dogs.

Dans ce travail, nous développons un cadre de calcul dédié à la simulation d'écoulements à plusieurs fluides. Nous présentons des validations et vérifications de ces méthodes sur des problèmes de capture d'interfaces et de simulations de bulles visqueuses.Nous montrons ensuite que ce cadre de calcul est adapté à la simulation d'objet rigides en écoulement.Puis, nous étendons ces méthodes à la simulation d'objets déformables simulant le comportement des globules rouges : les vésicules. Nous validons aussi ces simulations.Enfin nous appliquons les précédents modèles à des problèmes ouverts de microfluidique tels que la séparation d'une suspension dans une bifurcation microfluidique et la rhéologie en milieu confiné. / In this work, we develop a framework dedicated to the simulation of multi-fluid systems. We present validations and verifications of these methods on interface capture problems and viscous bubbles simulations.We then show that this framework is well fitted for the simulation of the rigid bodies flow.Next, we extend these methods to the simulation of deformable objects reproducing the behavior of red blood cells: the vesicles. We also validate these simulations.Finally, we apply the previous models to open micro-fluidic problems such as the splitting of a suspension at a bifurcation and the rheology in a confined environment.

Orientador: Elenice Deffune / Resumo: As vesículas extracelulares (VEs) são formações membranárias heterogêneas de tamanhos submicrométricos, que possuem funções específicas dependentes de sua biogênese. Os Concentrados de hemácias (CH) sofrem inúmeras alterações bioquímicas e morfológicas durante a estocagem comprometendo o meio intracelular e extracelular. O controle de qualidade das unidade de CH avalia essa lesão de estoque das hemácias principalmente a partir da determinação do grau de hemólise através da dosagem de hemoglobina livre. Não se sabe ao certo a quantidade de vesículas extracelulares que um CH pode gerar durante seu armazenamento mas muitos estudos relacionam as VEs com complicações pós-transfusionais. O objetivo deste estudo foi analisar o controle de qualidade de concentrados de hemácias sob o aspecto da identificação de vesículas extracelulares, correlacionando o grau de hemólise e a identificação de VEs em CH de hemácias em diferentes dias de conservação. Métodos: CH foram obtidos de 20 doadores saudáveis, refrigerados armazenados por 35 dias, e avaliados para o perfil de doação de sangue, grau de hemólise e dosagem de VEs por citometria de fluxo nos dias 5, 15, 25 e 35 de armazenamento. Resultados: Não houve diferença estatística significante entre o perfil da doação e as dosagens de grau de hemólise e VEs. O número total de VEs aumentou significativamente com o armazenamento assim como o grau de hemólise, observando maior correlação entre os métodos em D25 (p=0,0002) e D35 (p<0,0001). O tam... (Resumo completo, clicar acesso eletrônico abaixo) / Abstract: The extracellular vesicles (EVs) are heterogeneous membrane formations of submicrometric sizes, which have specific functions depending on their biogenesis. Red blood cell (RBC) undergo numerous biochemical and morphological changes during storage age compromising the intracellular and extracellular environment. The quality control of these units evaluates this lesion RBCs inventory mainly from the haemolysis determination through the measurement of free haemoglobin. It is unclear how much EVs the RBCs can generate during its storage but many studies related the EVs with post-transfusion complications. The objective of this study was to analyze the RBCs quality control under the aspect of the identification of EVs, correlating the haemolysis and the identification of VEs on different days of conservation. Methods: RBC were obtained from 20 healthy donors, refrigerated and stored for 35 days, and evaluated for blood donation profile, haemolysis and EVs counting by flow cytometry on storage days 5, 15, 25 and 35. Results: There was no statistically significant difference between the donation profile and haemolysis percentual and EVs dosages. The total number of EVs increased significantly with storage as well as the haemolysis, observing a higher correlation between the methods in D25 (p = 0.0002) and D35 (p <0.0001). O uso dos lasers na indústria, para processamento de materiais e em aplicações médicas, como a realização de cirurgias, têm atraído grande interesse nas últimas décadas. Neste trabalho, laser infravermelho foi explorado no corte de bolsas de sangue e no desenvolvimento de sistema de avaliação de elasticidade de hemácias baseado em técnica de aprisionamento óptico. No corte de bolsas de sangue foram determinados os parâmetros da ablação como tempo de perfuração, taxa de ablação e diâmetro dos furos para diferentes fluências e taxas de repetição do laser. Neste trabalho foram utilizados laser pulsados no regime de femtossegundos, com comprimento de onda de 800 nm, com taxa de repetição de 10 Hz e 1 KHz. Os resultados mostraram uma dependência do processo de ablação com o aumento da fluência e da taxa de repetição do laser. Além disso, foram avaliados os resíduos gasosos emitidos durante o processo de ablação das bolsas. A avaliação dos parâmetros de ablação à laser de bolsas de sangue consiste em um estudo pioneiro e introduz um novo direcionamento no processo de corte das mesmas. No desenvolvimento de um sistema para avaliação automática de elasticidade de hemácias aprisionadas opticamente foram utilizados lasers contínuos nos comprimentos de onda de 785 nm e 1064 nm. O sistema permite obter o valor da elasticidade de uma célula em 20 segundos, o que imprime uma redução significativa do tempo do processo de avaliação (60 ×) comparada ao método convencional. O sistema automático pode ajudar a expandir as aplicações de pinças ópticas em Hematologia e Hemoterapia. Além disto, o sistema construído foi utilizado para avaliar danos em hemácias aprisionadas opticamente. Foi verificada uma dependência da elasticidade das células com o comprimento de onda, potência do laser incidente e com tempo de aprisionamento da hemácia. Observou-se que as hemácias, após 2 minutos de exposição ao laser de 785 nm tornaram-se até ~ 104% menos deformáveis que as hemácias controle. A exposição ao laser de 1064 nm de 2 minutos a 10 mW induziu um aumento de até ~ 20% na rigidez celular. Atribuiu-se a dependência do comprimento de onda dos danos ópticos à absorção do laser pela hemoglobina. As modificações observadas nas propriedades elásticas das células estudadas estabelecem novos limites para aplicações utilizando lasers em hemácias. / This work explores the use of lasers in the infrared region of the electromagnetic spectrum in biomedical applications. The use of lasers in industry for materials processing and in medical applications, such as in surgeries, have attracted great interest in recent decades. In this work, infrared lasers were explored in blood bags cutting and in the development of a red blood cells elasticity evaluation system based on optical trapping technique. For the blood bags cutting process, ablation parameters such as drilling time, ablation rate and ablation diameter for different laser fluence and repetition rates were determined. In this work we used pulsed laser in femtosecond regime, with a wavelength of 800 nm, with repetition rate of 10 Hz and 1 KHz. The results showed a dependence of the ablation process with laser fluence and repetition rate. In addition, the waste gases emitted during the blood bags ablation process were evaluated. The evaluation of the blood bag laser ablation parameters consists of a pioneering study and introduces a new direction in the blood bag cutting process. In the development of a system for automatic evaluation elasticity of optically trapped red blood cells (RBCs), continuous laser at a wavelength of 785 nm and 1064 nm were used. The system allows to obtain the value of cell elasticity in 20 seconds, establishing a significant reduction in the assessment process time (× 60) compared to the conventional method. The automated system can help expand the applications of optical tweezers in Hematology and Hemotherapy. In addition, the constructed system was used to evaluate damage to red blood cells optically trapped. A dependence of the cells elasticity with the wavelength and power laser and with cell time trapping was observed. The results shows that the red blood cells was up to ~104% less deformable after 2 minutes of 785 nm laser exposition. The 2 minutes exposure to 10 mW of 1064 nm laser induced an increase up to ~ 20% on cell rigidity. We ascribed the wavelength dependence of the optical damages to the laser absorption by the hemoglobin. Moreover, the increase of RBCs rigidity could be associated to initial changes optically caused in the hemoglobin after irradiation. The results establish new limits for laser applications in RBCs, by identifying considerable modifications on their elastic properties.

Identificação dos fatores interferentes no transporte de concentrado de hemácias / Identification of the interfering factors in packed red blood cells transportation

Introdução. O transporte é um procedimento crítico na manutenção da cadeia do frio do sangue. Falhas neste procedimento podem comprometer a sua qualidade e a segurança transfusional. Objetivo. Avaliar as não conformidades (NC) do Hemocentro de Ribeirão Preto relacionadas ao transporte de concentrado de hemácias (CH). Analisar caixas térmicas e gelos recicláveis submetidos à temperatura de 20°C e 30°C e propor melhorias neste processo. Métodologia. Estudo retrospectivo de NC relacionadas ao transporte de CH no software do sistema da qualidade do Hemocentro de Ribeirão Preto e pesquisa experimental realizada com três caixas diferentes em combinação com três gelos recicláveis distintos, simulando uma situação de transporte, nas temperaturas externas de 20°C e 30°C. Realizou-se uma estatística descritiva com os dados. Utilizou-se a metodologia de análise de variância (ANOVA), com o auxílio do software SAS® 9, utilizando a PROC GLM. Para as comparações foram utilizados contrastes ortogonais baseados na distribuição t. Resultados. Os resultados mostraram diferenças significativas no desempenho entre as caixas e entre os gelos recicláveis. No entanto, as maiores diferenças ocorreram entre as temperaturas externas de 20°C e 30°C, indicando a interferência da temperatura externa na temperatura do conteúdo interno. Outra diferença encontrada foi entre pontos de aferição das temperaturas, dentro da mesma caixa, indicando que a distribuição do ar, no interior da caixa não é homogênea. Conclusão. São vários os interferentes existentes durante o transporte de CH, que podem comprometer a qualidade do sangue. O transporte deve ser validado, padronizando o maior número de variáveis possíveis, para minimizar as adversidades que ocorrem durante o procedimento / Introduction. The transport is a critical procedure in the maintenance of the blood cold chain. Failures in this procedure can compromise the blood quality and transfusion safety. Purpose: Evaluate the non-conformances (NC) of Hemocentro de Ribeirão Preto related to the Red Blood Cell (RBC) transport. Verify coolers, recycle ice at a temperature of 20°C and 30°C and propose improvements in this process. Methodology: Retrospective study of NC related to RBC transport in Hemocentro de Ribeirão Preto quality system software. Experimental research performed with three different insulated boxes combined with three different ice packs, simulating a situation of transport in the external temperatures of 20°C and 30°C. It was made a descriptive statistic of the data. The methodology of analysis of variance was used, through the software SAS® 9, using a PROC GLM. To do the comparisons orthogonal contrasts based on t distribution were used. Results: The results shown significant differences between insulated box and ice packs. However, the larger differences occurred between the temperatures of 20°C and 30°C, indicating that the external temperature interfere in the temperature inside the box. Another difference found was between points of temperature, inside the same insulated box, indicating that the air distribution inside the insulated box is not homogeneous. Conclusion. During RBC transport, many interfering exist that can compromise the blood quality. The transport must be validated, standardizing the largest number of details that is possible, to minimize adversity that occur during the procedure

A transfusão de sangue continua a ser uma importante ferramenta terapêutica, porém como qualquer outra, apresenta riscos. O sangue após coletado é processado e origina os hemocomponentes, dentre eles o Concentrado de Hemácias (CH). O CH é o hemocomponente mais utilizado na prática clínica. É indicado para o tratamento de anemias e em casos de hemorragia aguda ocorridas em cirurgia ou em decorrência de doenças ou traumas. Dentre os principais riscos da transfusão de CH estão a aloimunização e as reações transfusionais, imediatas ou tardias. Deve-se avaliar a real necessidade de se transfundir um paciente, principalmente, no período intra operatório. A transfusão de CH no intra operatório está associada a um pior prognóstico, maior tempo de internação, sobrevida menor e aumento do risco de infecção. Este estudo tem por objetivo analisar as solicitações de reserva de CH, e a partir desta análise, elaborar um protocolo de reservas cirúrgicas de CH com a finalidade de determinar o quantitativo de bolsas a serem reservadas ou não para a realização procedimentos cirúrgicos realizados no Hospital Regional de Ceilândia, no Distrito Federal. Para a elaboração do protocolo, foi realizado o levantamento de dados referentes às solicitações de reservas realizadas neste hospital entre março e agosto de 2017. Baseado nestes dados, calculou-se o Índice de Pacientes Transfundidos (IPT) que definiu o percentual das reservas que foram utilizadas. Verificou-se que a maioria das reservas solicitadas não foi utilizada. Do total de 216 pacientes que tiveram reservas preparadas, para as quais foram compatibilizadas 438 bolsas, apenas 48 destas bolsas foram transfundidas, ou seja, somente 10,95%. A partir do resultado do IPT, foi definida uma conduta hemoterápica cirúrgica para cada tipo de cirurgia. E, a partir destas, foi elaborado, um protocolo de reservas de CH para cirurgias, com o intuito de racionalizar a solicitação de reservas para ser implementado no hospital. Podendo assim ser um instrumento, para a equipe médica e para a agência transfusional, contribuindo para a economia de recursos financeiros e de sangue. / Blood transfusion continues to be an important therapeutic tool but it presents risks. The blood after collection is processed originating the blood components, among them the Red blood cells (RBC). RBC is the most widely used blood component in clinical practice. It is indicated for the treatment of anemias and in cases of acute hemorrhage occurring in surgery or due to pathologies or traumas. Among the main risks of RBC transfusion are alloimmunization and transfusion reactions. The actual need to transfuse a patient, especially during the intraoperative period, should be evaluated. Intraoperative RBC transfusion is associated with a worse prognosis, longer length of stay, shorter survival and increased risk of infection. The aim of this study was to analyze the RBC reserve requests and then to elaborate a protocol of RBC surgical reserves in order to determine the quantity of blood bags to be reserved or not for surgical procedures performed at the Regional Hospital de Ceilândia (HRC), in the Federal District. For the elaboration of this protocol, the data of the reservations requests performed in HRC March 2017 to August 2017 was carried out. The transfusion Index was calculated from the data, which defined the percentage of reservations that were used. It was found that most of the requested reserves were not used. Of the total of 216 patients who had prepared reservations, for which 438 bags were compatibilized, only 48 of these bags were transfused, that is, only 10.95%. From the result of the transfusion Index, so for each elective surgical procedures a guide surgical hemotherapy was defined. In addition, a reserve protocol of RBC for surgeries was elaborated, with the purpose of rationalizing the reservation request to be implemented in the hospital. This can be an instrument for the medical team and for the transfusion agency, contributing to the rational use of financial and blood resources.

Inhibition of the prothrombinase complex on phospholipid vesicles, activated platelets, and red blood cells by a covalently-linked antithrombin-heparin complex

<p>Prothrombinase is composed of a proteinase, factor Xa (Xa), its cofactor Va (Va), Ca<sup>2+</sup> and a zymogen, prothrombin (II), assembled on a phospholipid surface. During coagulation, prothrombinase accelerates II to thrombin conversion; but during anticoagulation, it protects the proteinase from inhibition by antithrombin (AT) ± unfractionated heparin (UFH). Although the degree of Xa protection by prothrombinase varies according to the reports in literature, moderate to significant protective effects have been consistently reported by most investigators. To overcome the limitations of UFH, our laboratory has developed a covalent complex of AT and UFH (ATH) with superior anticoagulant responses. To further understand the mechanisms of enhanced anticoagulant activity of ATH, we proceeded to study inhibition of the prothrombinase complex<em> </em>on synthetic vesicles, activated platelets and red blood cells (RBCs). Using discontinuous inhibition assays, we determined the rate of inhibition of prothrombinase-complexed Xa compared to control Xa. With synthetic vesicles, Xa was protected from inhibition by AT+UFH when in prothrombinase, while only a mild protective effect was observed with ATH. Omission of various components of the prothrombinase led to a reduction in Xa protection for AT+UFH. However, an increased Xa protection against ATH was observed when II was omitted from the prothrombinase. In comparison to the synthetic vesicle system, activated platelets showed a similar trend for protection of Xa in reactions involving prothrombinase ± components, while no protection of Xa was observed for ATH reactions. Alternatively, RBCs showed differences relative to vesicles in that increased protection of Xa occurred with omission of II and Va for AT+UFH, whereas omission of Va increased protection against ATH inhibition. In addition, ATH had improved inhibition of thrombin generation, fibrin formation and plasma coagulation compared to AT+UFH. Studies of fluorescently labelled Xa and inhibitors detailed binding interactions with prothrombinase subunits. Overall, the results suggest that a covalent linkage between AT and heparin improves inactivation of prothrombinase complexed-Xa leading to down-regulation of prothrombinase function.</p> / Doctor of Philosophy (Medical Science)

Σκοπός της διδακτορικής διατριβής είναι η ανάπτυξη κατάλληλων μεθόδων ψηφιακής επεξεργασίας εικόνας και αναγνώρισης προτύπων με τις οποίες θα προσδιορίζονται βιομετρικές και διαγνωστικές παράμετροι μέσω της αλληλεπίδρασης φωτονίων στο ορατό και υπέρυθρο φάσμα. Πιο συγκεκριμένα επιλύεται ένα αντίστροφο πρόβλημα σκέδασης ΗΜ ακτινοβολίας από ένα ανθρώπινο, υγιές και απαραμόρφωτο ερυθρό αιμοσφαίριο. Παρουσιάζονται μέθοδοι εκτίμησης και αναγνώρισης των γεωμετρικών χαρακτηριστικών απαραμόρφωτων υγιών ερυθρών αιμοσφαιρίων με χρήση εικόνων που προσομοιώνουν φαινόμενα σκέδασης ηλεκτρομαγνητικής ακτινοβολίας που διέρχεται από προσανατολισμένα ερυθρά αιμοσφαίρια. Η διαδικασία της ανάκτησης της πληροφορίας περιλαμβάνει, εξαγωγή χαρακτηριστικών με χρήση δισδιάστατων μετασχηματισμών, κανονικοποίηση των χαρακτηριστικών και την χρήση νευρωνικών δικτύων για την εκτίμηση των γεωμετρικών ιδιοτήτων του ερυθροκυττάρου. Παράλληλα σχεδιάστηκε και αξιολογήθηκε σύστημα αναγνώρισης των γεωμετρικών χαρακτηριστικών των ερυθρών αιμοσφαιρίων. Οι εικόνες σκέδασης δημιουργήθηκαν προσομοιώνοντας το πρόβλημα εμπρόσθιας σκέδασης ενός επίπεδου ηλεκτρομαγνητικού (ΗΜ) κύματος, χρησιμοποιώντας την μέθοδο των συνοριακών στοιχείων, λαμβάνοντας υπόψη τόσο την αξονοσυμμετρική γεωμετρία του ερυθροκυττάρου όσο και τις μη αξονοσυμμετρικές οριακές συνθήκες του προβλήματος. Η επίλυση του εν λόγω προβλήματος πραγματοποιήθηκε στα 632.8 nm και εν συνεχεία επεκτάθηκε σε 12 διακριτά ίσου βήματος μήκη κύματος από 432.8 nm έως 1032.8 nm. Επίσης, προτάθηκε μία νέα πειραματική διάταξη για την απόκτηση πολλαπλών εικόνων σκέδασης και την εκτίμηση των γεωμετρικών χαρακτηριστικών των ερυθρών αιμοσφαιρίων, αποτελούμενη από μία πολυχρωματική πηγή φωτός (Led) και πολλαπλά χρωματικά φίλτρα. Επίσης κατασκευάστηκε μέθοδος επίλυσης του σημαντικού προβλήματος εύρεσης της περιεκτικότητας του διαλύματος σε ερυθρά αιμοσφαίρια διαφορετικών μεγεθών στην περίπτωση απόκτησης πολλαπλών εικόνων σκέδασης από διαφορετικές φωτοδιόδους και πολλαπλά χρωματικά φίλτρα. Στα πειράματα αξιολόγησης της μεθόδου που προτείνεται με εικόνες προσομοίωσης δείχνεται ότι είναι ικανή η εύρεση της αναλογίας των ερυθρών αιμοσφαιρίων με πολύ μεγάλη ακρίβεια ακόμα και στη περίπτωση όπου στις εικόνες έχει προστεθεί λευκός κανονικός θόρυβος. Η βασική μεθοδολογία που παρουσιάζεται στην παρούσα δια-τριβή μπορεί να χρησιμοποιηθεί για την αναγνώριση παθολογικών αιμοσφαιρίων ή να χρησιμοποιηθεί στην αναγνώριση μικροσωματιδίων σε υγρά ή αέρια. / The aim of this PhD thesis is the development of digital image processing and pattern recognition methods to estimate biometric and diagnostic parameters using scattering phenomena in the visible and infrared spectrum. More concretely, several reverse scattering problems of EM radiation from a human, healthy and undistorted Red Blood Cell (RBC) is solved. Methods of estimation and recognition of geometrical characteristics of healthy and undistorted RBCs using simulating images are presented. The information retrieval process includes, features extraction using two-dimensional integral transforms, features normalization, and Neural Networks for estimation of three major RBC geometrical proper-ties. Using the same features set, a recognition system of the geometric characteristics of RBCs was developed and evaluated. The scattering images were created simulating the forward scattering problem of a plane electromagnetic wave using the Boundary Element Method, taking into account both axisymmetric geometry of the scatterer and the non-axisymmetric boundary conditions of the problem. Initially, the problem is solved at 632.8 nm and consequently the same problem was solved at 12 different wavelengths, from 432.8 to 1032.8 nm equally spaced. Also, a new device for acquisition of scattering images from RBCs-flow, consisting of a multi-color light source (Led) was proposed, for RBC size estimation and recognition. Finally, a system for the estimation of different RBCs concentration was developed when scattering images acquired using multiple scattering images acquired from multiple Leds and color filters. The system was evaluated using additive white regular noise.
