Orientador: Paola Castro Moraes / Resumo: A catarata situa-se entre as afecções oculares mais frequentes em cães. A facoemulsificação é o método que melhor se adéqua à sua terapia e o preferido entre os oftalmologistas. Dentre as complicações pós-operatórias destacam-se lesões endoteliais que podem produzir descompensação permanente da córnea, alterando a sua transparência. Buscou-se avaliar os efeitos do inibidor de ROCK (Y-27632), na proteção endotelial, em cães submetidos à facoemulsificação. Utilizaram-se 20 olhos de 10 pacientes, machos ou fêmeas, com sete a 12 anos, portando catarata senil bilateral. Dois grupos foram concebidos: Olhos controles (Oc) (n=10), receberam solução salina balanceada e Olhos Tratados (Ot) (n=10), receberam intracameralmente, 0,3 mL de Y-27632, na concentração de 100 µmol/L, imediatamente após a sutura corneal. O endotélio corneal foi avaliado à microscopia especular de não contato, antes e em diferentes momentos após a facoemulsificação. Avaliaram-se a densidade (células/mm2) e a área celular (µm2), a espessura corneal (mm), a hexagonalidade (%) e o coeficiente de variação do tamanho celular (%). Valores de P iguais ou inferiores a 0,05 foram considerados significativos. Com relação à densidade de células endoteliais, ambos os grupos apresentaram perdas, sem diferença estatística. Relativamente à espessura corneal e hexagonalidade, não houve diferença. No Oc, a área celular aumentou significativamente entre os momentos avaliados, o que não foi observado em Ot. Phacoemulsification is the method that best fits its therapy. Among the complications related to this surgery, there are endothelial injuries that can induce permanent corneal decompensation, altering its transparency. The goal of this study was to evaluate the effects of ROCK inhibitor Y-27632 on endothelial protection in dogs undergoing phacoemulsification. Twenty eyes of 10 patients, male or female, aged 7 to 12 years, with bilateral senile cataract were used. Two groups were conceived: Control eyes (Oc) (n = 10), that received balanced saline solution (BSS) and Treated Eyes (Ot) (n = 10), which received intracamerally 0.3 mL of Y-27632 at a concentration of 100 µmol. / L, immediately after corneal suture. The corneal endothelium was assessed via non-contact specular microscopy at multiple time points before and after phacoemulsification. Cell density (cells/mm2 ) and area (µm2 ), corneal thickness (mm), hexagonality (%), and the coefficient of variation of cell size (%) were all assessed. Il est établi depuis maintenant 40 ans que l'interaction nucléaire forte peut être décrite par la Chromodynamique Quantique (QCD). Le confinement des quarks dans cette théorie masque cependant ses degrés de liberté élémentaires dans les processus de basse énergie, qui doivent être calculés non-perturbativement. La QCD sur réseau est à ce jour la seule méthode permettant de mener à bien de tels calculs, et arrive dorénavant à maturité. Cependant, et alors que le spectre des hadrons stables est calculée avec précision, l'étude des désintégrations reste délicate. Nous présentons ici l'application du formalisme de Lüscher au calcul des propriétés de résonances se désintégrant sous l'interaction forte, ainsi qu'un calcul avec précision de la largeur de désintégration dans le cas particulier du meson rho. / It has been established for now 40 years that the strong nuclear interaction can be described by Quantum Chromodynamics (QCD). However, the confinement of quarks in this theory hides its elementary degrees of freedom in low-energy processes, which have to be computed non-pertubatively. Lattice QCD is, to date, the only method allowing to go through such a computation, and is now reaching maturity. However, although the spectrum of stable hadrons is computed with precision, the study of decays is still delicate. We hereby present the application of Lüscher's formalism to the study of strong-decaying resonance properties, along with a precise computation of the decay width in the particular case of the rho meson.

O Corpo Ciliar (CC) é uma fonte de células tronco da retina de animais adultos, mas sua ativação permanece desconhecida. GTPases Rho são proteínas que reorganizam do citoesqueleto de actina, regulam vias de sinalização e transcrição gênica, sobrevivência celular e proliferação. Neste trabalho, investigamos a expressão das GTPases Rho nas células do CC e seu efeito na regulação do ciclo celular. As GTPases RhoA, RhoB e Rac1 foram expressas nas células do CC e sua ativação pelo ácido lisofosfatidico (LPA) aumentou a expressão dos genes progenitores retinianos Pax6 e Chx10. A inibição das proteínas por Toxina A de Clostridium difficile aumentou a proliferação no CC e potencializou o efeito proliferativo dos fatores de crescimento. A inibição especifica destas proteínas diminuiu a expressão dos transcritos de p21cip, p27kip, p16INK4a e p19INK4d e aumentou de Ki67, CiclinaA e D1. O estudo da via de Wnt indicou que Rac1 regulou os genes de componentes da degradação de -catenina e Lef1. Concluímos que a inativação das GTPases Rho induziu a proliferação das células progenitoras retinianas localizadas no CC e regulou a via de Wnt. Sua ativação induziu o perfil de célula progenitora, sugerindo uma nova ferramenta para o mecanismo de reparo retiniano. / Ciliary Body (CB) is a potential source of stem cells in the adult retina, but its activation is still unknown. Rho GTPases play a role in actin-based cytoskeleton reorganization, regulate signaling pathways and gene transcription, cell survival and cell proliferation. In this study we investigated the expression of Rho GTPases in CB cells and their role on cell cycle regulation. The GTPases RhoA, RhoB and Rac1 were present in CB cells and the activation by lysophosphatidic acid (LPA) increased the expression of the progenitor genes Pax6 and Chx10. The inhibition by Clostridium difficile Toxin A increased the proliferation of CB cells and potentiated the proliferative effect of growth factors. The specific inhibition decreased the expression of p21cip, p27kip, p16INK4a and p19INK4d as well as increased Ki67, cyclinA and D1 transcripts. The Wnt pathway study indicated that Rac1 regulated -catenin degradation genes components and Lef1. Taken together, the inactivation of Rho GTPases stimulated the proliferation of progenitor cells located in CB as well as regulate the Wnt signaling pathway. The proteins activation was correlated to progenitor profile induction. These different mechanisms may provide a potential new approach on retinal repair.
